Objectives The aim of this study is to evaluate the fracture healing effect of Jinmu-tang (JM) on femur fractured rats. Methods Rats were randomly divided into 5 groups (normal, control, positive control, JM extract with low concentration and JM extract with high concentration). All group except normal group went through both femur fracture. Normal and control group received no treatment at all. Positive control group were medicated with tramadol (20 mg/kg) once a day for 14 days. Experimental group was orally medicated with JM extract (10 mg/kg for low concentration, 50 mg/kg for high concentration) once a day for 14 days. In order to investigate fracture healing process, plasma and serum were obtained. Also, micro-computed tomography was conducted to see the frature site visually. Immunohistochemistry for transforming growth factor-β1, Ki67, alkaline phosphatase, runt-related transcription factor 2, receptor activator of nuclear factor kappa-β, tartrate resistant acid phosphatase was conducted to observe bone healing progress after 14 days since fracture occured. Aspartate aminotransferase, alanine aminotransferase, blood urea nitrogen and creatinine levels were measured in plasma, for hepatotoxicity and nephrotoxicity of JM extract. Osteocalcin was measured to observe activity of osteoblast. Results Through Micro-CT, more fracture healing was observed on both experimental group than control and positive control group. Through Hematoxylin & Eosin and safranin O staining showed bone cell proliferation and bone formation in the experimental group. RANK was significantly increased in the experimental groups. JM with high concentration showed statistically significant of TGF-β and Osteocalcin. NO, TRAP and ALP were not significantly changed. Liver toxicity was not significantly observed. Creatinine significantly increased in both experimental groups after 28 days. Conclusions As described above, JM extract showed anti-inflammatory effect, promoted fracture healing by stimulating the bone regeneration factor, and showed little hepatotoxicity and nephrotoxicity. In conclusion, JM extract can promote fracture healing and it can be used clinically to patients with fracture.

The general characteristics, anti-mutagenicity, and in vitro anti-cancer effects of Chungkukjang, Shuidouchi, and Natto were studied. Representative brands of the three types of soybean-based short-term-fermented foods were chosen and evaluated. Chungkukjang showed the highest amino and ammonia type nitrogen contents, although Natto exhibited lower levels, and Shuidouchi showed the lowest pH and high acidity. Anti-mutagenicity was assessed by the Ames test and in vitro anti-cancer effects were assessed in HT-29 human colon cancer cells. Chungkukjang exhibited the highest anti-mutagenicity and anti-cancer effects. Based on these results, Chungkukjang showed the best functional effects, and Shuidouchi showed better functional effects than Natto. Even if such efficacies depend on raw materials and processing methods, our comparison of three types of foods from representative brands shows that Chungkukjang was the best soybean-derived product in terms of overall quality, followed by Shuidouchi and Natto. These results might be due to differences in duration of fermentation, processing method, and quality of raw materials, but further research on the basis of ethnic culture and administration method of each nation should follow.
The production of extracellular mannitol by a new mannitol-producing bacterium, Leuconostoc sp. KY-002 was studied in shake flask cultures. The new isolate has a capability of utilizing fructose and sucrose for mannitol formation. Maximum mannitol production was obtained with fructose as the sole carbon source. Under the optimal culture conditions, within 70 hours of incubation, a final concentration of 26 g/L of mannitol from 50 g/L fructose was obtained with an indicated yield of 52% based on fructose consumed. However, higher concentrations of fructose ranging from 100 to 250 g/L could not effectively be transformed to mannitol due to a lack of osmotolerance. The strain produced no other polyols such as glycerol and sorbitol as by-products. Yeast extract was the best nitrogen source and high levels of inorganic phosphate up to 10 g/L promoted mannitol formation. Any mineral ions and salts did not play important role in both cell growth and mannitol production. Nicotinic acid enhanced mannitol production by 16%. The optimum culture temperature and initial pH were $35^{\circ}C$ and 6, respectively.
Twenty apple germplasm accessions from the Korean Genebank were successfully cryopreserved using two-step freezing to back up genetic resources maintained by field collections. This study examined the morphological and genetic stability of cryopreserved dormant apple buds that were stored in liquid nitrogen, and then rewarmed and regrown. Whole plants were regenerated directly from dormant buds through budding without an intermediary callus phase. The cryopreserved buds produced high levels of shoot formation (76.2-100%), similar to those of noncryopreserved buds (91.3-100%), with no observed differences between cryopreserved and noncryopreserved materials. Three of the twenty cryopreserved apple germplasm accessions were used to assess morphological and genetic stability. No differences in morphological characteristics including shoot length, leaf shape, leaf width/length ratio, and root length were observed between controls (fresh control and noncryopreserved) and cryopreserved plantlets. The genetic stability of regenerants (before and after cryopreservation) was investigated using inter simple sequence repeat (ISSR) markers. The ISSR markers produced 253 bands using four primers, ISSR 810, SSR 835, ISSR 864, and ISSR 899. These markers showed monomorphic banding patterns and revealed no polymorphism between the mother plant and regenerants before and after cryopreservation, suggesting that cryopreservation using two-step freezing does not affect the genetic stability of apple germplasm. These results show that two-step freezing cryopreservation is a practical method for long-term storage of apple germplasms.
Objectives : This study was performed to inquire into the protective effects of acupuncture on oxidative stress caused by cadmium accumulation in the kidney. Methods : Sprague-Dawley male($150{\pm}30g$) rats were stabilized for 1 week and divided into 5 groups: normal, control, $LR_3$ acupuncture, $BL_{23}$ acupuncture and sham acupuncture. For three days experimental groups received oral doses of cadmium 2 mg/kg twice a day. Acupuncture was applied bilaterally at each point 10 times for two weeks. The depth of stimulation was 1 mm at right angles and torsion of acupuncture was produced 2 times per second for 1 minute. The kidneys were extracted and weighed after two weeks, and renal function was confirmed through blood urea nitrogen(BUN). We measured reactive oxygen species of the serum and kidney, and compared expression levels of superoxide dismutase(SOD), catalase, glutathione peroxidase(Gpx), nuclear factor erythroid derived 2-related factor 2(Nrf-2), heme oxygenase-1(HO-1), nuclear factor-${\kappa}B$(NF-${\kappa}B)$, cyclooxygenase-2(COX-2), inducible nitric oxide synthase (iNOS), Bax and Cytochrome c. Results : The $LR_3$ acupuncture group and $BL_{23}$ acupuncture group experienced significantly increased kidney weight, and decreased BUN compared to control group. In terms of oxidative stress, the $LR_3$ acupuncture group and $BL_{23}$ acupuncture group experienced significantly reduced reactive oxygen species compared to the control group. Conclusions : The $LR_3$ acupuncture group and $BL_{23}$ acupuncture group experienced showed the effects of antioxidant, anti-inflammatory and apoptosis protection. The $BL_{23}$ acupuncture group was more effective than $LR_3$ acupuncture group.
Soil respiration has been recognized as a key factor of the change of organic matter and fertility due to the carbon and nitrogen mineralization. In this study, we evaluated the effect of soil respiration on the light fraction-C and inorganic N content depending on temperature in soil applied with organic matter. Soil respiration was calculated by using total $CO_2$ flux released from soil applied with $2Mg\;ha^{-1}$ of rice straw compost and rye for 8 weeks incubation at 15, 25, $35^{\circ}C$ under incubation test. After incubation test, light fraction and inorganic N content were investigated. Rye application dramatically increased soil respiration with increasing temperature. $Q_{10}$ value of rye application was 1.69, which was higher 27% than that of rice straw compost application. Light-C and $NO_3-N$ contents were negatively correlated to soil respiration. Light-C in rye application more decreased than that in rice straw compost with temperature levels. These results indicate that temperature sensitivity of soil respiration could affect soil organic mater content and N availability in soil due to carbon availability. Also, light fraction would be useful indicator to evaluate decomposition rate of organic matter in soil under a short-term test.

To find out the optimum fertilizer levcl for Job's Tears, Seungju local cultivar, experiment with six fertilizer levels was conducted on the experimental field of forage crop in Sunchon National University from Apr. 1989 to Aug. 1990. The results obtained were summarized as follows: I .Raising nitrogen(N) application rate up to 13.5 kg/lOa increased dry matter yield linearly. However, at high N application rate, 18 kg/lOa, plants were too leafy and parts of leaves and lodging were decreased. so that dry matter yield was decreased.2. The optimum application rate of fertilizers turned out to be 13.5-6-6 kg/lOa of $P_2O_5$, $K_2O$. Content of crude protein, yield of fresh and dry matter were the highest and that of crude fiber such as NDF. ADF and cellulose were the lowest at this rate.
Decomposition of green manure is necessary for the nutrient supply in farm soil. Hairy vetch as a green manure is superior to other winter legumes in terms of wintering ability and N accumulation. This experiment was carried out to investigate the decomposition and N release of hairy vetch and its effect on rice production as the following crop in paddy field. Decomposition of hairy vetch placed by soil depth of 0, 10 and 20cm at transplanting time was investigated by mesh bag method, which was enclosed chopped residue in mesh bags. The fate of $^{15}$ N derived from $^{15}$ N-labeled hairy vetch was investigated at harvest in three levels of N fertilization. Grain yield of the transplanted paddy rice cultured with hairy vetch as starter N were compared with that of applying urea as starter N in the field. Hairy vetch residue decomposed very rapidly both in transplanted and dry-seeded paddy field. In transplanted paddy field, hairy vetch residue lost 72-81 % and 86-90% of its weight after one and five month, respectively, as affected by incorporation depth. The C/N ratio of the decomposing vetch residue increased sharply during the early stages and after then, decreased slowly. The amounts of N and P released from the vetch were about 90% and 97% of initial content after one month, respectively. Recoveries of hairy vetch-$^{15}$ N by rice plant were 30.6, 34.6 and 35.7% in 0, 6 and 12 kg urea-N 10 $a^{-l}$ application, respectively, indicating that N fertilization increased the recovery of hairy vetch. $^{15}$ N. Hairy vetch residue incorporated as starter maintained significant N $H_4$$^{+}$-N concentration in soil water of plow layer until effective tillering stage. Grain yield in the plot applied with hairy vetch was not significantly different from that in the plot with urea. We concluded that hairy vetch incorporation could substitute starter N fertilization and showed possibility to reduce N amount of top-dressing.g.g.
The rice straw managements are essential for maintaining soil fertility as well as reducing chemical fertilizer application in paddy field. A field experiment was conducted on moderately well draining alluvial paddy soil to investigate the decomposition pattern of rice straw. The mesh bags containing the rice straw harvested in the previous year were placed at soil surface and buried into around 10cm depth and recovered periodically for determining the straw decomposition. Pot experiments were conducted to investigate the fates of N released from $^{15}$ N-labeled rice straw under different levels of N fertilizer application. The overall decomposition patterns of rice straw were similar for the two incorporation depths in transplanted paddy field. The straw incorporated at transplanting date showed weight loss of about 50%, 70% and 90% after 2 months, 5 months, and 2 years, respectively. The decompositions of straw cell wall components showed somewhat different pattern. The decompositions of cellulose and silica were similar to that of dry weight while the decomposition of lignin was slower than that of cellulose and silica. N was released from rice straw 42% and 65 % of the initial N after one month and after five months, respectively. P release was faster than N release. Recoveries of rice straw-$^{15}$ N by rice plants were 10.2, 13.4 and 14.9% in 0, 120 and 240 mg N pot$^{-1}$ , respectively. Soil recoveries of rice straw $^{15}$ N were 17.3, 20.6 and 18.9% in 0, 120 and 240mg N pot$^{-1}$ , respectively.

We have grown AlN nanorods and AlN films using plasma-assisted molecular beam epitaxy by changing the Al source flux. Plasma-assisted molecular beam epitaxy of AlN was performed on c-plane $Al_2O_3$ substrates with different levels of aluminum (Al) flux but with the same nitrogen flux. Growth behavior of AlN was strongly affected by Al flux, as determined by in-situ reflection high energy electron diffraction. Prior to the growth, nitridation of the $Al_2O_3$ substrate was performed and a two-dimensionally grown AlN layer was formed by the nitridation process, in which the epitaxial relationship was determined to be [11-20]AlN//[10-10]$Al_2O_3$, and [10-10]AlN//[11-20]$Al_2O_3$. In the growth of AlN films after nitridation, vertically aligned nanorod-structured AlN was grown with a growth rate of $1.6{\mu}m/h$, in which the growth direction was <0001>, for low Al flux. However, with high Al flux, Al droplets with diameters of about $8{\mu}m$ were found, which implies an Al-rich growth environment. With moderate Al flux conditions, epitaxial AlN films were grown. Growth was maintained in two-dimensional or three-dimensional growth mode depending on the Al flux during the growth; however, final growth occurred in three-dimensional growth mode. A lowest root mean square roughness of 0.6 nm (for $2{\mu}m{\times}2{\mu}m$ area) was obtained, which indicates a very flat surface.
