Acanthophysium sp. KMF001, a New Strain with High Cellulase Activity

  • Cellulase is an eco-friendly biocatalyst, and its demand is growing in many industrial applications such as food, textile, paper, and bioenergy. Strains with a high cellulase activities are the starting point for the economic production of cellulase. In a previous study, Acanthophysium sp. KMF001 with high cellulase production ability was selected among 54 wood-rotting fungi. In this study, we evaluated the cellulase productivity of Acanthophysium sp. KMF001 quantitatively and analyzed its taxonomic location using a genetic method. Acanthophysium sp. KMF001 showed high cellulase productivity similar to that of Acanthophysium bisporum and was much better than A. bisporum in specific enzyme activity. The 28S rRNA sequence of Acanthophysium sp. KMF001 was similar to that of Acanthophysium lividocaeruleum MB1825, with 98.40% homology. Phylogenetic analysis suggested that Acanthophysium sp. KMF001 is a new strain. In this study, we propose a new strain with high cellulase productivity.

A Prediction Model for Low Cycle Fatigue Life of Pre-strained Fe-18Mn TWIP Steel (Fe-18Mn TWIP강의 Pre-strain에 따른 저주기 피로 수명 예측 모델 연구)

  • The influence of pre-strain in low-cycle fatigue behavior of Fe-18Mn-0.05Al-0.6C TWIP steel was studied by conducting axial strain-controlled tests. As-received plates were deformed by rolling with reduction ratios of 10 and 30%, respectively. A triangular waveform with a constant frequency of 1 Hz was employed for low cycle fatigue test at the strain amplitudes in the range of ${\pm}0.4{\sim}{\pm}0.6$ pct. The results showed that low-cycle fatigue life was strongly dependent on the amount of pre-strain as well as the strain amplitude. Increasing the amount of prestrain, the number of reversals to failure was significantly decreased at high strain amplitudes, but the effect was negilgible at low strain amplitudes. A new model for predicting fatigue life of pre-strained body has been devised adding a correction term of ${\Delta}E_{pre-strain}$ to the energy-based fatigue damage parameter.

  • To prevent appearance of resistant mosquitoes against $\delta$-endotoxin of bacillus thuringiensis subsp. israelensis (Bti) in field, a mosquitocidal Bacillus thuringiensis strain 21-2(Bt21-2) producing a new type of $\delta$-endotoxin was isolated. The strain Bt 21-2 belongs to H serotype 43, B. thuringiensis subsp. guiyangiensis (Btg). The $\delta$-endotoxins from the strain Bt 21-2 and the strain Bti were a cuboid shape morphologically, but the $\delta$-endotoxin of the strain Bt 21-2 was composed of 150, 90 and 70kDa proteins on SDS-PAGE, and the antigenicity of $\delta$-endotoxin of the strain Bt 21-2 was different from that of the strain Bti on immunoblot. The $\delta$-endotoxin gene of the strain Bt 21-2 was not amplified with specific primers of $\delta$-endotoxin gene (cry4A and cry4B) of the strain Bti on PCR.

Comparative Analysis of Meat Quality Traits of New Strains of Native Chickens for Samgyetang (삼계용 신품종 토종닭의 육질특성 비교분석)

  • The carcass and meat quality of new native chicken strains (A, C, and D) being developed in the Golden Seed Project for Samgyetang were compared with a commercial native chicken (Hanhyup No. 3, H) and Baeksemi (white semi broiler, W) strains. In total, 250 male chicks were raised under the same feeding and raising conditions for 5 weeks. After slaughter, the carcass weight and percentage yield of parts from each strain were measured and the physicochemical quality traits (general composition, color values, fatty acid composition, and sensory evaluation) of breast muscles were comparatively analyzed. The carcass weight (875.10 g) of the W strain was significantly heavier than that of the other native chicken strains, while that of A strain (537.54 g) was the lowest. The percentage yield of breast meat was also higher in the W strain than in the other strains. The fat content of W strain was higher than that of the other native chicken strains. The new native chicken strains (A, C, D) had higher contents of linoleic acid (C18:2) than the H strain. The A and D strains especially showed relatively high content of arachidonic acid (C20:4), a characteristic fatty acid in native chicken meats. On the other hand, the fatty acid composition of C strain was similar to that of the W strain. This study provides important information on specific quality characteristics than can be used to select new native chicken lines for breeding program.

Characteristics and breeding of a new variety Pleurotus eryngii, Gongi No.3 (신품종 큰느타리버섯 '곤지3호' 육성 및 특성)

  • We bred a new strain of Pleurotus eryngii. It's name is 'Gongi No.3' and it was bred by mating monokaryotic strain isolated from E08-5D2 and dikaryotic strain GMPE25016 from 2006 to 2010 in Mushroom Research Station, Gyonggi province A.R.E.S. The characteristics of a new strain 'Gongi No.3' is as follows ; The optimum temperature for mycelium growth was from 26 to 29 degrees celsius on PDA medium and those for the premodium formation and the growth of fruit body were from 14 to 18 degrees celsius. The period of spawn running was around 30days at 22 degrees celsius and the period taken from scratching old spawn to make premodium were 8 days. The color degree of cap surface was measured by color difference meter and that of a new strain 'Gongi No.3' was 54.4 by L-value. it was seem to be dark, compared with 'Keunneutari No.2'. The hardness of fruit body of a new strain was higher than 'Keunneutari No.2'. The yield was about 180g per bottle(1100cc). it was 10g more than 'Keunneutari No.2'.

Development of Pressuremeter for Evaluation of Deformation Characteristic at Whole Strain Ranges (전체변형률 범위에서 변형특성 평가를 위한 공내재하시험 장치 개발)

  • In order to analyze the deformational behavior accurately, the in-situ testing technique which provides the reliable deformational characteristics at whole strain ranges, needs to be developed. The pressuremeter is a unique method for assessing directly the in-situ shear modulus of soils with strain amplitude. This paper introduces a new alternative, the cavity strain measuring system in pressuremeter designed for whole strain ranges of $10^{-5}$%∼20%. Not only in the synthetic calibration chamber but also in the field, the pressuremeter tests were performed to determine the compliance of the new developed pressuremeter system. The variation in shear modulus with strain amplitude above $5\times 10^{-2}$% was reliably determined by the developed pressuremeter. It is concluded that the major cause of error in small cavity strain measuring is not from the cavity strain measuring system but from the friction between measuring arm and membrane during unloading-reloading loops.

  • A size-dependent novel hyperbolic shear deformation theory of simply supported functionally graded beams is presented in the frame work of the non-local strain gradient theory, in which the stress accounts for only the nonlocal strain gradients stress field. The thickness stretching effect (${\varepsilon}_z{\neq}0$) is also considered here. Elastic coefficients and length scale parameter are assumed to vary in the thickness direction of functionally graded beams according to power-law form. The governing equations are derived using the Hamilton principle. The closed-form solutions for exact critical buckling loads of nonlocal strain gradient functionally graded beams are obtained using Navier's method. The derived results are compared with those of strain gradient theory.

  • In real design of the high & interim pressure turbine casing, it is one of the important things to figure out its thermal strain exactly. In this paper, with the establishment of the new concept for the heat transfer coefficient of steam that is one of the factors in analysis of the thermal stress for turbine casing, an analysis was done for one of the high & interim pressure turbine casings in operating domestically. The sensitivity analysis of the heat transfer coefficient of steam to the thermal strain of the turbine casing was done with a 2-D simple model. The analysis was also done with switching of the material properties of the turbine casing and resulted in that the thermal strain of the turbine casing was not so sensitive to the heat transfer coefficient of steam. On the basis of this, 3-D analysis of the thermal strain for the high and interim pressure turbine casing was done.

Assumed strain finite strip method using the non-periodic B-spline

  • An assumed strain finite strip method(FSM) using the non-periodic B-spline for a shell is presented. In the present method, the shape function based on the non-periodic B-splines satisfies the Kronecker delta properties at the boundaries and allows to introduce interior supports in much the same way as in a conventional finite element formulation. In the formulation for a shell, the geometry of the shell is defined by non-periodic B3-splines without any tangential vectors at the ends and the penalty function method is used to incorporate the drilling degrees of freedom. In this study, new assumed strain fields using the non-periodic B-spline function are proposed to overcome the locking problems. The strip formulated in this way does not posses any spurious zero energy modes. The versatility and accuracy of the new approach are demonstrated through a series of numerical examples.

  • The analysis model is the infinite power law creep material containing the rigid inclusion with crack shape. The present analysis is performed using the complex pseudo-stress function method. The strain rate intensity factor is developed as new fracture mechanics parameter which represents the stress and strain rate distribution near a crack tip in power law creep material. The strain rate intensity factor is developed in terms of Kolosoff stress functions.
