Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85 (황색종 연초 돌연변이 계통 KF8832-85의 특성)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars.   • Drosophila melanogaster is a useful model organism that offers essential insights into developmental and cellular processes shared with humans, which has been adapted for large scale analysis of medically important microbes and to test the toxicity of heavy metals, industrial solvents and other poisonous substances. We here give a brief review of the use of the Drosophila model in medical mycology, discuss the volatile organic compounds (VOCs) produced by the opportunistic human pathogen, Aspergillus fumigatus, and give a brief summary of what is known about the toxicity of some common fungal VOCs. Further, we discuss the use of VOC detection as an indirect indicator of fungal growth, including for early diagnosis of aspergillosis. Finally, we hypothesize that D. melanogaster has promise for investigating the role of VOCs synthesized by A. fumigatus as possible virulence factors.

2-Undecanone derived from Pseudomonas aeruginosa modulates the neutrophil activity

  • Pseudomonas aeruginosa (P. aeruginosa) is a well-known Gramnegative opportunistic pathogen. Neutrophils play key roles in mediating host defense against P. aeruginosa infection. In this study, we identified a metabolite derived from P. aeruginosa that regulates neutrophil activities. Using gas chromatography-mass spectrometry, a markedly increased level of 2-undecanone was identified in the peritoneal fluid of P. aeruginosa-infected mice. 2-Undecanone elicited the activation of neutrophils in a Gαi-phospholipase C pathway. However, 2-undecanone strongly inhibited responses to lipopolysaccharide and bactericidal activity of neutrophils against P. aeruginosa by inducing apoptosis. Our results demonstrate that 2-undecanone from P. aeruginosa limits the innate defense activity of neutrophils, suggesting that the production of inhibitory metabolites is a strategy of P. aeruginosa for escaping the host immune system.

Development of RT-PCR Kit for Diagnosis of Pathogenic Agent of Ginseng Root Rot in the Ginseng Field (인삼포장에서 뿌리섞음병원균의 진단을 위한 RT-PCR KIT의 개발)

  • Cylindrocarpon destructans is the major pathogen inducing the root rot disease in ginseng. Up to now, there is no reliable and convenient method to analyze the spore density or population of this pathogen in ginseng-growing soil or any contaminated farmlands. Therefore, it will be very valuable to develop a new and reliable method in detecting the spore of this pathogen. In this study, a molecular biological technique using two step nested PCR method, was developed. Two universal ITS primers, ITS5F and ITS4R were used in the first round of PCR to amplify a fragment of ITS region from the genomic DNA of C. destructans. The specific prmers Nest 1 and Nest 2 were designed and used in the second round of PCR to amplify a inner fragment from the first round PCR product of C. destructans. C. destructans spore, only soil samples from the diseased ginseng farm produced the positive bands, suggesting its usefulness in detecting the C. destructans spores in soil samples. Thus it is recommended to first extract the whole genomic DNA from soil samples and use it for the PCR reaction, thereby eliminating the inhibitory activity of soil components.

The fucose containing polymer (FCP) rich fraction of Ascophyllum nodosum (L.) Le Jol. protects Caenorhabditis elegans against Pseudomonas aeruginosa by triggering innate immune signaling pathways and suppression of pathogen virulence factors

  • Brown algal extracts have long been used as feed supplements to promote health of farm animals. Here, we show new molecular insights in to the mechanism of action of a fucose containing polymer (FCP) rich fraction from the brown seaweed Ascophyllum nodosum using the Caenorhabditis elegans-Pseudomonas aeruginosa PA14 infection model. FCP enhanced survival of C. elegans against pathogen stress, correlated with up-regulation of key immune response genes such as: lipases, lysozyme (lys-1), saponin-like protein (spp-1), thaumatin-like protein (tlp-1), matridin SK domain protein (msk-1), antibacterial protein (abf-1), and lectin family protein (lfp). Further, FCP caused down regulation of P. aeruginosa quorum sensing genes: (lasI, lasR, rhlI, and rhlR), secreted virulence factors (lipase, proteases, and elastases) and toxic metabolites (pyocyanin, hydrogen cyanide, and siderophore). Biofilm formation and motility of pathogenic bacteria were also greatly attenuated when the culture media were treated with FCP. Interestingly, FCP failed to mitigate the pathogen stress in skn-1, daf-2, and pmk-1 mutants of C. elegans. This indicated that, FCP treatment acted on the regulation of fundamental innate immune pathways, which are conserved across the majority of organisms including humans. This study suggests the possible use of FCP, a seaweed component, as a functional food source for healthy living.

Occurrence of Leaf Spot on Eriobotrya japonica Caused by Entomosporium mespili in Korea (Entomosporium mespili에 의한 비파나무 점무늬병 발생)

  • Since 2008, a new leaf spot disease has been found in loquat tree (Eriobotrya japonica) at a nursery station in Goheung, Korea. Above 50% of the seedlings developed the spot symptoms on the leaves. The infected plants were generally reduced in the seedling vigour and early defoliation was occurred. The infected leaves developed grayish lesion with dark reddish brown margin. The pathogen developed round to ellipsoidal acervuli on the lesion. Based on morphological characteristics of the pathogen, the fungus was identified as Entomosporium mespili. The cultured pathogen successfully reproduced the same disease symptom on the leaves of loquat tree and found to be E. mespili. A monoconidial culture was deposited in Korean Agricultural Culture Collection (KACC 44727). This is the first report of E. mespili causing leaf spot of loquat tree in Korea.

Screening Resistance to Crown Gall Disease by Pathogen Inoculation in Muscadine and Florida Hybrid Grapes (병원균 접종에 의한 Muscadine 및 플로리다 교잡종 포도의 줄기혹병 저항성 검정)

  • It is necessary to evaluate the resistance to disease among genetic resources for development of disease resistant grapes. This study was conducted to screen the resistance to crown gall in muscadine and Florida hybrid bunch grapes by pathogen inoculation. In order to compare the responses to infection with different pathogen strains, muscadine and Florida hybrid grapes were inoculated with 3 strains of Agrobacterium vitis. Although there were different levels crown gall formation among grape cultivars, there little variation in response to inoculated strains. Among 29 muscadine cultivars tested by inoculation of A. vitis 'C4612', most of them were shown to be susceptible, and 'Gold Isle' and 'Africa Queen' were highly susceptible, and two cultivars, 'Welder' and 'Jumbo' were found to be resistant to crown gall disease. Among Florida hybrid grapes, 'Daytona', 'Stover', and 'Swanee' were susceptible and 'Blanc du Bois' was moderately susceptible to crown gall. Because muscadine grapes have been actively utilized as useful genetic resources for development of new grape varieties by intersub-genus cross, this result from the screening of resistance among muscadine grapes can provide valuable information in breeding programs of grape resistant to crown gall.

Avirulence Gene AVR-Pita1 in the Rice Blast Fungus (벼도열병균의 비병원성 유전자 AVR-Pita1)

  • The rice blast fungus, Magnaporthe oryzae, is one of the most economically important crop diseases. In addition, rice-M. oryzae interaction is a classical gene-for-gene host-pathogen system. Race variation in pathogen groups was proposed as the main mechanism for rapid break-down of resistance in newly introduced rice cultivars. These new pathogen race variations may be caused by changes in an avirulence gene, such as (i) point mutations, (ii) insertion of transposons, and (iii) frame shifts. The avirulence gene AVR-Pita1 is representative avirulence gene in which all of these mutations are reported. In this review, we present a useful information for avirulence gene AVR-Pita1 and its homologous genes AVR-Pita2 and AVR-Pita3. We also review examples that cause mutations in these evolutionarily significant genes.

  • Listeria monocytogenes is a high-risk foodborne pathogen responsible for foodborne listeriosis outbreaks, and is particularly dangerous to immuno-compromised people with mortality rate of about 30%. This review summarizes subtyping of L. monocytogenes using Pulsed Field Gel Electrophoresis, widely used to trace origin of foodborne outbreaks and to determine relationship between isolates.

Studies ell the strawberry leaf flight Caused by Dondrophoma obscurans (Ell. & Ev.) Anderson (우리나라의 딸기 신병해 겹무늬병 (Dondrophoma obscurans)에 관한 연구)

  • The study has been carried to describe a new disease of strawberry in Korea, which was found in 1977 at Kimhae. Symptom of the disease occurred mainly on leaves as showing large annular brown spots or 'V' shaped brown lesions. Many of dark pycnidia were observed from the both side of old lesions. The pathogen was identified as Dendrophoma obscurans (Ell. & Ev) Anderson which has not been described in Korea as a pathogen of strawberry disease. The common name of the disease was given, temporaly, as Annular leaf blight of strawberry. In the laboratory study, light was necessary for the production of pycnidia and potato dextrose agar was the best media for tile pycnidia formation. There were some difference on resistance to the disease among 48 tested strawberry varieties though none of them shelved highly resistant reaction, and the older leaves showed more susceptible reaction than younger ones.
