Nucleotide Binding Component of the Respiratory Burst Oxidase of Human Neutrophils

  • The respiratory burst oxidase of neutrophils is a multicomponent enzyme, domant in resting cells, that catalyzes the reduction of oxygen to $O_{2}^{-}$ at the expense of NADPH. In the resting neutrophil, some of the components of the oxidase, including proteins p47 and p67, are in the cytosol, while the rest are in the plasma membrane. Recent evidence has suggested that at least some of the cytosolic oxidase components exist as a complex. The cytosolic complex with a molecular weight of ~240 kDa was found to bind to blue-agarose and 2',5'-ADP-agarose, which recognize nucleotide requiring enzymes. In order to identify the nucleotide binding component of the cytosolic complex we purified recombinant p47 and p67 fusion proteins using the pGEX system. Pure recombinant p47 was retained completely on 2',5'-ADP-agarose, whereas pure recombinant p67 did not bind to these affinity beads. On the basis of these results, we infer that p47 may contain the nucleotide binding site.

Antioxidant and Anti-inflammatory Activities of Allium victorialis subsp. platyphyllum Extracts

  • This study was conducted to investigate antioxidant activity and anti-immunological inflammatory effect of Allium victorialis subsp. platyphyllum extracts (AVPEs). Antioxidant activities of AVPEs were determined by free radical scavenging assay and reducing power test. Leaf-part extract had comparatively better antioxidant activity than other-part extracts. Antioxidant activity of extracts had protective effect for human umbilical vein endothelial cells (HUVECs) against superoxide anions secreted from activated neutrophils. Also, we observed AVPEs had inhibitory effects on the adherence of monocytic THP-1 to HUVEC monolayer to the basal level. Inhibitory effect on cell adhesion was caused by suppression of tumor necrosis factor-${\alpha}\;(TNF-{\alpha})-upregulated$ expression of vascular cellular adhesion molecule-1 (VCAM-1) and E-selectin in HUVECs. From these results, we expect to support the evidence of anti-immunological inflammatory effects of Allium victorialis subsp. platyphyllum (AVP) as a Korean traditional pharmaceutical.

The Effects of Sojeokbaekchoolsan on the Immune Cells, the Chemotactic and Adherent Ability of Macrophages (소적백출산(消積白朮散)이 면역세포(免疫細胞)의 동태(動態), 대식세포(大食細胞)의 走化性(주화성) 및 부착성(附着性)에 미치는 영향(影響))

  • In order to investigate the effects of Sojeokbaekchoolsan on the various immune responses, the chemotactic and adherent ability of macrophages were studied in vivo and in vitro. The results obtained were follows: 1. The administration of Sojeokbaekchoolsan enhanced significantly the Chemotactic activity of macrophages and neutrophils. 2. The administration of Sojeokbaekchoolsan enhanced significantly the adherent activity of macrophages and neutrophils. From above findings, it is suggested that Sojeokbaekchoolsan seem to produce the increase of immune response such as the chemotactic activity and adherent activity of macrophage. According to the above results, it could be suggested that Jukyeopseokgo-tanggagambang extract has indirect autitumor effects by strengthening the effects of MMC on tumor cells.

  • Chronic obstructive pulmonary disease (COPD) is a chronic and progressive inflammatory disease of the airways and lungs that results in limitations of continuous airflow and is caused by exposure to noxious gasses and particles. A major cause of morbidity and mortality in adults, COPD is a complex disease pathologically mediated by many inflammatory pathways. Macrophages, neutrophils, dendritic cells, and CD8+ T-lymphocytes are the key inflammatory cells involved in COPD. Recently, the non-coding small RNA, micro-RNA, have also been intensively investigated and evidence suggest that it plays a role in the pathogenesis of COPD. Here, we discuss the accumulated evidence that has since revealed the role of each inflammatory cell and their involvement in the immunopathogenesis of COPD. Mechanisms of steroid resistance in COPD will also be briefly discussed.

Inhibitory Effect of S100A8 on Neutrophil Apoptosis by Cytokine Release of Normal and Allergic Monocytes

  • S100A8 functions as an essential factor in inflammatory response. Cytokine release of monocytes and regulation of neutrophil apoptosis are important steps in pathogenesis of allergy. This study aims to examine the relation between cytokine release of monocytes due to S100A8 and neutrophil apoptosis. S100A8 enhanced the release of IL-6 and IL-8 in monocytes of normal and allergic subjects. Treatment of supernatants of normal and allergic monocytes with S100A8 blocked neutrophil apoptosis by inhibition of caspase 9 and caspase 3 activation. The secretion signal induced by S100A8 is involved in TLR4, Src family protein, PKCδ, ERK1/2, p38 MAPK, JNK, and NF-κB. These findings may contribute to understanding the complex pathogenesis of allergic diseases by determining inflammatory responses associated with S100A8, monocytes, and neutrophils.

Effect of Toluene Treatment on the Xanthine Oxidase and Superoxide Dismutase Activities in Leukocyte of Bacterial Infected Rats (흰쥐에 toluene 투여가 백혈구 xanthine oxidase 및 superoxide dismutase 활성에 미치는 영향)

  • This study was designed to observe the effect of tohiene pretreatment on leukocyte variation in whole blood and the oxygen free radical generating, scavenging enzyme activities in neutrophil of bacteria infected rats. Toluene was administered 7 times intraperitoneally at levels of 9.45 mM/kg body weight to the rats and then infected with S. aureus $2\times10^7$ cfu/ml. The toluene treated-rats showed the significantly decreased numbers of lymphocyte and monocytes, but the similiar numbers of neutrophils with the control. Furthermore the increased neutrophils in blood of bacteria infected rats were reduced by the toluene pretreatment. Concomitantly the increased activities of xanthine oxidase and superoxide dismutase in neutrophil of bacteria infected rats were also decreased by the toluene pretreatment. On the other hand, injection of benzaldehyde to rats also led to similiar results in the count of leukocytes, xanthine oxidase and superoxide dismutase activities of neutrophil with those of toluene treated rats. These data suggest that toluene and its intermediate metabolite, benzaldehyde influence on the phagocytosis and defence mechanism of neutrophil.

Effect of Cyclohexane Application to Rat Skin on the Skin Toxicity (흰쥐의 피부조직에 있어서 Cyclohexane의 독성)

  • To evaluate the skin toxicity of topical cyclohexane application (25mg/$\textrm{cm}^2$) was sequentially applied to the rat skin for four days. On the histopathological findings in the light micrographs, neutrophils and engulfed neutrophils are seen, and many cytoplasmic processes were appeared in proliferated layer whereas in the dermis area, increased numbers of fibroblast, accumulation of neutrophil and lipid droplets are demonstrated. On the other hand, applying the cyclohexane to the rat skin led to the remarkable rise of cutaneous xanthine oxidase activity and similar activities of superoxide dismutase and glutathione peroxidase and glutathione content and declined activity of glutathione S-transferase compared with control group. Especially the remarkably decreased activity of aniline hydroxylase (AH) was appeared in skin as little as scarcely determined. Furthermore, the applying the cyclohexane to skin led to the significantly increased activity of hepatic AH and alcohol dehydrogenase. These results indicate that oxygen free radical and intermediate metabolite of cyclohexane may be responsible for structural changes in skin by cyclohexane application to rat skin.

Effects of Jiyutang with Fructus Immaturus Ponciri on the immune response in the Mouse (지유탕가(地楡湯加) 지실(枳實)이 생쥐의 면역반응(免疫反應)에 미치는 영향(影響))

  • This study was done to know the effect of Giyuetang on the inflammatory response, contact hypersensitivity (CH), and rosette forming capacity of spleen cells. The effects of Jiyutang with Fructus Immaturus Ponciri on the inflammatory response were evaluated by measuring the production of such reactive oxygen intermediate(ROI) as $O_2^-$ and $H_{2}O_2$ in the peritoneal neutrophils and macrophages. The effects of Jiyutang with Fructus Immaturus Ponciri on the CH were evaluated by checking the ear swelling response against dinitrofluorobenzene(DNFB). The administration of Jiyutang with Fructus Immaturus Ponciri on the mouse decreased the amount of ROI in both neutrophils and macrophages. Jiyutang with Fructus Immaturus Ponciri depressed CH without affecting the rosette forming capacity of spleen cells. The results of this study showed that Jiyutang with Fructus Immaturus Ponciri might have anti-inflammatory effects by decreasing the amounts of ROI in the phagocytes and suppressing the CH, without affecting the rosette-forming capacity of spleen cells.

Effects of Gossypol Injection into the Stroma of Testis on Blood Constituents in Mice (생쥐 정소실질내 Gossypol 투여가 혈액성상에 미치는 영향)

  • We have previously shown that gossypol injection into the stroma of testis was found to effectively inhibit the process of spermatogenesis. In this study, properties and chemistry of blood were investigated weekly in ICR mouse after the injection of gossypol into the testicular stroma(5, 10 or 15mg per kg body weight). There were no significant differences in red blood cell(RBC), hematocrit, white blood cell(WBC), basophils, eosinophils and monocytes during first 4 weeks after injection of gossypol between treatments, but neutrophils increased and lymphocytes decreased, respectively(P<0.05). Total content of protein, albumin and globulin were not different, compared with control. However, the concentration of glucose after injection of gossypol increased significantly(P<0.05). In conculsion, the results of this study indicated that injection of gossypol into stroma of testis might affect both properties and chemistry of blood in mice.

Subcorneal pustular dermatosis in a dog (개에서 발생한 각질하 농포성 피부병 1례)

  • Canine Subcorneal pustular dermatosis (CSPD) represents a sterile, superficial, pustular skin disease of unknown cause but may be a variant of pemphigus foliaceus. A 7-year-old, intact female, mixed dog presented with 3-month history of pruritic multiple pustules and crusts. Direct smears from intact pustules revealed numerous nondegenerate neutrophils, some acantholytic cells, and bacterial culture was negative. Histologic examination of lesional skin showed subcorneal pustules filled with neutrophils and acantholytic cells. The direct immunofluorescence tests stained with IgG, IgA, IgM, C3 were negative. Oral administration of dapsone (1 mg/kg/q8h) was initiated and it was reduced to 1 mg/kg/q12h with good control of the lesions.
