Review of Experimental Studies on Estradiol Valerate-induced Polycystic Ovarian Syndrome in Herbal Medicine (Estradiol Valerate로 유발된 다낭성 난소 증후군의 한약치료에 대한 국내 실험연구 고찰)

  • Objectives: To observe the effects of Herb medication on Polycystic Ovary (PCO) in rats. Methods: Riss (Research Information Sharing Service) was searched for Oriental medicine about PCO on 2000~2014. So, Experimental studies using PCO model induced by Estradiol Valerate (EV) was selected to analyze how effects they were. Consequently, 10 studies published on 2000~2014 were selected to analyze about weight of model and both ovaries, the number aries, the number of follicle and corpus of follicle and corpus luteum, the level of serum Androstenedione (ADD) and total estrogen, the level of Nerve Growth Factor (NGF) and Corticotropin Releasing Factor (CRF), the crossover rate and the number of implantation. Results: Among them, there was significantly difference between treatment group and control group in weight of both ovaries, the number of follicle and corpora lutea, the crossover rate and the number of implantation. Conclusions: The effects of Herb medication was observed on making weight of ovaries with PCO to promote. The study about effect of Herb medication on NGF and CRF with PCOS had to be conducted. The further research have to focus on treatment of subfertility and infertility, that is field showing the advantage of Oriental medicine.

Involvement of Cytochrome c Oxidase Subunit I Gene during Neuronal Differentiation of PC12 Cells

  • It is becoming increasingly evident that significant changes in gene expression occur during the course of neuronal differentiation. Thus, it should be possible to gain information about the biochemical events by identifying differentially expressed genes in neuronal differentiation The PC12 cell line is a useful model system to investigate the molecular mechanism underlying neuronal differentiation and has been used extensively for the study of the molecular events that underlie the biological actions of nerve growth factor (NGF). In this study, we report an application of the recently described mRNA differential display method to analyze differential gene expression during neuronal differentiation. Using this technique, we have identified several cDNA tags expressed differentially during neuronal differentiation. Interestingly, one of these clones was cytochrome c oxidase subunit I (COX I) gene. The differential expression of COX I gene was confirmed by Northern blot analysis as well as RT-PCR. Southern blot analysis of the genomic DNA of PC12 cells revealed that COX I is a single gene. Induction of the oxidative enzyme might reflect the energy requirement in neuronal differentiation.

  • Brain-derived neurotrophic factor (BDNF) is a small secretory protein and a member of the nerve growth factor (NGF) gene family. We cloned the flounder BDNF gene from a flounder brain cDNA library. The nucleotide sequence of the cloned gene showed an open reading frame (ORF) consisting of 810 bp, corresponding to 269 amino acid residues. The tissue distribution of flounder BDNF was determined by reverse transcription-polymerase chain reaction (RT-PCR) in brain, embryo, and muscle tissues. To express fBDNF using a eukaryotic expression system, we constructed the vector mpCTV-BDNF containing the fBDNF gene and transformed this vector into Chlorella ellipsoidea. Stable integration of introduced DNA was confirmed by PCR analysis of genomic DNA, and mRNA expression in C. ellipsoidae was confirmed by RT-PCR analysis.

Effect of Reserpine on the Behavioral Defects, Aβ-42 Deposition and NGF Metabolism in Tg2576 Transgenic Mouse Model for Alzheimer's Disease (알츠하이머질환 모델동물인 Tg2576마우스의 행동, Aβ-42 침적, 신경성장인자 대사에 미치는 reserpine의 영향)

  • Reserpine, an anti-hypertensive drug, is able to positively modulate several phenotypes associated with $A{\beta}$ toxicity in a Caenorhabditis elegans model of Alzheimer's disease (AD). We investigated into the therapeutic effects of reserpine on mammalian neurodegenerative disorders, and found that significant alteration of the key factors influencing AD was detected in Tg2576 mice after reserpine treatment for 30 days. The aggressive behavior of Tg2576 mice was significantly improved upon reserpine treatment, whereas their social contact was consistently maintained. Furthermore, the levels of $A{\beta}$-42 peptide in the hippocampus of the brain and blood serum were lower in the reserpine-treated group than in the vehicle-treated group. Among g-secretase components, the expression levels of PS-2, Pen-2, and APH-1 were slightly lower in reserpine-treated Tg2576 mice, although a significant change in nicastrin (NCT) expression was not detected. Furthermore, the serum level of nerve growth factor (NGF) increased in reserpine-treated Tg2576 mice compared with vehicle-treated mice. Among down-stream effectors of the NGF receptor TrkA signaling pathway, reserpine treatment induced elevation of TrkA phosphorylation and reduction of ERK phosphorylation. In addition, in the NGF receptor $p75^{NTR}$ signaling pathway, the expression levels of $p75^{NTR}$ and Bcl-2 were enhanced in reserpine-treated Tg2576 mice compared with vehicle-treated mice, whereas the expression level of RhoA declined. Overall, these results suggest that reserpine can help relieve AD pathogenesis in Tg2576 mice through downregulation of $A{\beta}$-42 deposition, alteration of ${\gamma}$-secretase components, and regulation of NGF metabolism.

  • Erinacine which is produced from Hericium erinaceus mycelia is known as stimulating activity of nerve growth factor(NGF) synthesis. Thus. this work was concentrated on the maximum production of Hericium erinaceus mycelia. Media compositions were studied as the chemical conditions that affected cell growth. The modified media was ditermined from the results, including glucose 20g/L and yeast extract 10g/L.

  • Erinacine which is produced from Hericium erinaceus mycelia is known to have a stimulating activity of nerve growth factor(NGF) synthesis. Thus, this work was concentrated on the maximum production of Hericium erinaceus mycelia. In order to maximize cell growth, the cultivation was performed with varying the agitation rate and applying the additional medium. As the results, the mycelium concentration was 24.2g/L.

Neurotrophic Factors Mediate Memory Enhancing Property of Ethanolic Extract of Liriope platyphylla in Mice

  • The roots of Liriope platyphylla (Liliaceae) are widely used in traditional Chinese medicine. In the present study, we investigated the effects of ethanol (70%) extract of the roots of Liriope platyphylla (ELP70) on learning and memory using behavioral and immunohistochemical methods in mice. Control animals were treated with vehicle (10% Tween 80). With sub-chronic treatments of ELP70 (p.o.) for 14 days, the latency time was significantly increased compared with that of the vehicle-treated control group (50, 100 and 200 mg/kg; P<0.05). Moreover, immunopositive cells for brain derived neurotrophic factor (BDNF) were significantly increased in the hippocapmpal dentate gyrus and CA1 regions after ELP70 treatments for 14 days (50, 100 and 200 mg/kg; P < 0.05). In addition, those cells for nerve growth factor (NGF) were also increased in the hippocapmpal dentate gyrus region (50, 100 and 200 mg/kg; P<0.05). These results suggest that the sub-chronic administration of ELP70 improves learning and memory, and that their beneficial effects are mediated, in part, by the enhancement of BDNF or NGF expression.

Effects of Jeongkyeong-Tang on the Estradiol Valerate-induced Polycystic Ovary in Rats (정경탕(定經湯)이 Estradiol Valerate로 유도된 흰쥐의 다낭성 난소에 미치는 영향)

  • Purpose : This study was designed to investigate the effects of Jeongkyeong-Tang(JKT) on the polycystic ovary(PCO) induced by estradiol valerate(EV) in rats. Methods : PCO was induced by single intramuscular injection with EV(4mg) in female rats. Normal group(n=8) were injected with sesame oil and orally administrated distilled water for sixty days. PCO control group(n=8) were injected with EV and orally administrated distilled water for sixty days. JKT treated group(n=8) were injected with EV and orally administrated JKT for sixty days. Then we measured weights of body, ovaries and adrenal glands, and measured content of serum estrogen. The histomorphometrical changes of ovaries were also evaluated. The expressions of nerve growth factor(NGF) were analyzed in the central nervous system, adrenal glands and ovaries by immunohistochemistry. Results : - The weights(mg) of ovaries in JKT treated group (69.7${\pm}$6.7) were significantly increased( p<0.001) compared with PCO control group(46.7${\pm}$12.2). - The numbers of secondary follicles in JKT treated group(4.00${\pm}$l.31) were significantly increased(p <0.05) compared with PCO control group(2.25${\pm}$1.39). - The numbers of mature follicles in JKT treated group(5.50${\pm}$1.51) were significantly increased(p<0.01) compared with PCO control group(2.88${\pm}$1.13). - The numbers of atretic follicles in JKT treated group(2.75${\pm}$l.16) were significantly decreased(p<0.001) compared with PCO control group(6.88${\pm}$2.03). - The numbers of corpora lutea in JKT treated group(4.13${\pm}$1.46) were significantly increased(p<0.01) compared with PCO control group(2.13${\pm}$1.46). - The contents(pg/ml) of serum estrogen in JKT treated group(115.18${\pm}$18.29) were significantly decreased(P<0.01) compared with PCO control group(153.06${\pm}$29.47). - The expressions of NGF-immunoreactive cells in the ovarian granulosa cells in JKT treated group were lesser observed than PCO control group. Conclusion : From the above results, we concluded that Jeongkyeong-Tang has inhibitory effect on the development of EV-induced polycystic ovary. And it's effect may be related with decreased NGF activities in the ovary.

Effects of (+)-Eudesmin from the Stem Bark of Magnolia kobus DC. var. borealis Sarg. on Neurite Outgrowth in PC12 Cells

  • (+)-Eudesmin [4,8-bis(3,4-dimethoxyphenyl)-3,7 -dioxabicyclo[3.3.0]octane] was isolated from the stem bark of Magnolia kobus DC. var. borealis Sarg. and found to have neuritogenic activity. $50\;{\mu}M$ (+)-eudesmin induced neurite outgrowth and enhanced nerve growth factor (NGF)-mediated neurite outgrowth from PC12 cells. At this concentration, (+)-eudesmin also enhanced NGF-induced neurite-bearing activity and this activity was partially blocked by various protein kinase inhibitors. These included PD98059, a mitogen-activated protein kinase (MAPK) kinase inhibitor. GF109203X, a protein kinase C (PKC) inhibitor and H89, a protein kinase A (PKA) inhibitor. These results suggest that (+)-eudesmin can induce neurite outgrowth from PC12 cells by stimulating up-stream MAPK, PKC and PKA pathways.

Anti-inflammatory and Neurotrophic 2H-1-Benzopyran Derivatives of Chaenomeles sinensis

  • Two 2H-1-benzopyran derivatives, methyl 8-hydroxy-2,2-dimethyl-2H-1-benzopyran-5-carboxylate (1) and methyl 8-hydroxy-2,2-dimethyl-2H-1-benzopyran-6-carboxylate (2), including a new compound (1) were isolated from the twigs of Chaenomeles sinensis. Their chemical structures were characterized based on analysis of NMR data including 1H and 13C, COSY, HSQC, and HMBC and HRMS data. The isolated compounds (1 and 2) were assessed for their anti-neuroinflammatory activity by measuring inhibition levels of nitric oxide (NO) production in lipopolysaccharide (LPS)-activated BV-2 cells and for their neurotrophic activity by the secretion of nerve growth factor (NGF) in C6 cells. Compounds 1 and 2 exhibited powerful anti-neuroinflammatory effects with IC50 values of 17.14 and 19.30 μM, respectively, without cell toxicity, and also showed moderate effects on the stimulation of NGF secretion levels with 113.15 ± 3.54 and 130.20 ± 8.03%, respectively. The biosynthetic pathway of 1 and 2 was proposed that they would be derived from a protocatechuic acid and an isoprenyl unit.
