흰쥐에서 Curcuminoid 및 이를 함유한 천연식물 혼합물이 심혈관계에 미치는 영향 (Effect of Curcuminoids and Natural Plants Extract Mixture on the Cardiovascular System in Rats)

  • Antiplatelet aggregation, anticoagulant and lipid-lowering drugs are clinically widely used for secondary preventive purpose in the cardiovascular patients, but there is no primary preventive agents to prevent these diseases. With the aim of developing effective primary agents for cardiovascular diseases, we tried to formulate an optimized mixture of natural plants extract containing Theae sinensis, Camelliae sinensis, Vitis vinifera, Gingko folium and curcuminoids from Curcuma longa and to evaluate its anti-thrombotic and anti-hypercholesterolemic effects in vivo. The inhibitory effect of curcuminoids on vascular smooth muscle cell proliferation and migration were also investigated in vitro. in the animal experiments treated with hyperlipidemic diet, oral treatment of curcuminoids and natural plants extracts mixture (100 mg/kg) into male Sprague Dawley rats for 7 week simultaneously inhibited platelet aggregation as well as improved lipid profile in the blood. Compared to control group, both of curcuminoids-treated and mixture-treated groups revealed significantly decrease of total cholesterol (24.4%, 28.6%), free cholesterol (25.1%, 24.0%), cholesterol ester (14.6%, 29.0%), LDL-cholesterol (27.0%, 32.0%) and triglyceride (15.0%, 31.0%), respectively. However, both groups showed increase of HDL-cholesterol (46.6% and 51.5%) . In particular, atherogenic index of curcuminoids and mixture treatment group was significantly decreased to 47.0% and 56.0%, respectively. Furthermore, oral treatment of curcuminoids and mixture significantly inhibited collagen-induced platelet aggregation (21.1% and 29.1%, respectively), compared to control group. The anti-thrombotic values of mixture was almost similar to that of aspirin treatment (100 mg/kg) group. These results suggest that the oral treatment of curcuminoids-based natural plant extract mixture improved cardiovascular conditions in hyperlipidemic rats.

  • HAE-06 extract is a mixture of four medicinal plants, namely Lonicerae Folium et Caulis (Lonicera japonica), Scutellariae Radix (Scutellaria baicalensis), Adenophorae Radix (Adenophora triphylla var. japonica), and Polygonati Oddorati Rhizoma (Polygonatum odoratum var. pluriflorum). The HAE-06 extract demonstrated a concentration-dependent relaxing effect and enhanced cAMP production in bronchial smooth muscle that had been stimulated to contract with acetylcholine. Using a blocker, it was confirmed that the effect was through the β2-adrenergic receptor/cAMP/PKA pathway. In addition, it is thought that the HAE-06 extract has a bronchial smooth muscle relaxation effect by reducing the inflow of Ca2+ through the K+ and Ca2+ channels present in the sarcoplasmic membrane. If research continues in the future, it is believed that it will be possible to use it as a material for pharmaceuticals and functional foods.

양파 수확 후 잔재물과 쇠비름 추출물이 유기농 양파의 수확량 및 품질 특성에 미치는 영향 (Effect of a Mixture of Extracts from Residues of Onion Left after Onion Harvesting and Purslane (Portulaca oleracea) on Productivity and Quality Characteristics of Organic Onions)

  • 본 연구는 수확 후 버려지는 양파 잔재물과 쇠비름으로부터 추출한 수액을 유기농 양파 재배 시 시비하여 유기농 양파의 수확량, 품질 등에 미치는 영향을 평가하였다. 도복기 때의 양파 생육은 초장, 엽수, 엽초경에서 식물추출액 처리구들이 무처리구보다 좋았다. 양파 수확량도 식물 추출액 처리구들이 무처리구보다 유의성 있게 증가되었으며(p<0.05), 구 중 분포에서는 상품성이 있는 300 g 이상의 대구 양파가 양파와 쇠비름 추출액 혼용 처리구에서 다른 처리구들보다 유의성 있게 증가 되었다(p<0.05). 양파의 무기성분 함량은 양파와 쇠비름 추출액 혼용 처리구에서 생산된 양파가 다른 처리구들에서 생산된 양파보다 석회와 황을 제외한 다량원소들이 높게 함유되었다. 양파의 경도는 양파와 쇠비름 추출액 혼용 처리구에서 생산된 양파가 양파 잔재물 추출액 처리구와 무처리구에서 생산된 양파보다 각각 8%와 20% 높게 나와 유의성을 나타내었지만(p<0.05), 당도는 모든 처리구에서 생산된 양파가 비슷하게 나와 유의성을 나타내지 않았다. 양파 수확 후 토양의 무기성분 함량은 양파 잔재물 추출액 처리구 토양에서 인산, 칼슘, 황산, 망간에서 높게 나타났다. 이상의 결과로 양파 수확 후 버려지는 잔재물과 길가에 방치되는 쇠비름의 추출물이 유기 양파 재배 시 천연 친환경 자재로 활용 가치가 있다고 보겠다.

  • The methanol extract of Euphorbia milii exhibited strong inhibitory effect on platelet aggregation in the cource of our search for anti-platelet component from succulent plants. Two components, components 1 and 2 were isolated from this plant. 1 was the mixture of 72% of 1-octacosanol (1a) and 28% of 1-triacontanol (1b), and 2 was identified as ${\beta}-sitosterol$. 2 ($IC_{50}$: $195\;{\mu}M$, and $170\;{\mu}M$ respectively) was about two fold stronger than ASA ($IC_{50}$: $420\;{\mu}M$ and $340\;{\mu}M$ respectively) on both collagen and U46619 induced aggregation, while the effect of 1 to platelets was negligible.

호박잎에서 Paraquat 활성 억제 물질의 분리, 동정 및 특성 구명 (Isolation, Identification and Characterization of Paraquat Activity-Inhibiting Substances in Squash (Cucurbita moschata Duchesne ex Poiret) Leaves)

  • 선행연구를 통해 18종의 호박 품종에 대한 엽위별 paraquat에 대한 내성 반응을 조사한 결과, 1엽<2엽<3엽<4엽 순이었다. 이들 품종 중에서 엽령간 뚜렷한 내성차이가 있는 중앙애호박의 경우 4엽은 1엽에 비해 paraquat에 대해 3배 이상의 높은 내성을 보였다. 또한 이들 4엽의 물 추출물과 paraquat를 혼용하여 옥수수 경엽에 처리시 paraquat 약해를 경감시켰다. 따라서 본 연구는 paraquat 약해를 경감시키는 물질이 어린 호박잎에 존재하는지를 알아보기 위하여 수행하였다. MeOH 추출물보다 물 추출물에서 paraquat 활성경감이 더 높아서 물 추출물을 실리카겔과 Sephadex LH-20 칼럼크로마토그래피와 TLC, HPLC를 이용하여 paraquat 활성 경감 효과를 보이는 순수물질을 얻어 NMR을 통해 분석한 결과 malic acid로 추정되었다. 따라서 malic acid가 paraquat 활성경감 물질여부를 확인하기 위하여 malic acid와 paraquat를 옥수수에 동시 처리하였다. 처리 후 1일째 $100{\mu}M$ paraquat를 단독으로 처리한 경우 옥수수 잎 피해가 62% 발생하였으나 $100{\mu}M$ paraquat에 malic acid를 각각 0.1, 0.3, 0.5 및 1% 혼용처리한 경우 잎 피해가 전혀 발생하지 않았다.

천연식물자원 지모와 황백피 혼합 수추출물의 안전성평가 (Evaluation of Genotoxicity and 14-day Repeat Dose Toxicity of Water Extract of the Mixture of Natural Plants, Anemarrhena and Phellodendron)

  • The safety of a new natural plant composition (ADP) was assessed on the genotoxicity study and 14-day repeat dose toxicity study. ADP contains a mixed water extract obtained from the mixture of Phellodendron cortex (Phellodendron amurense) and Anemarrhena rhizoma (Anemarrhena asphodeloides), and poses the contractile properties mediated by alpha-adrenoceptor of the prostate and urethra as well as antioxidant and anti-inflammatory properties. In order to evaluate genetic safety, in vivo micronucleus test was performed in ICR mice orally administered with three dose levels of 1250, 2500, 5000 mg/kg body weight, and vehicle and positive control. In the 14 days study, Sprague-Dawley rats were treated with ADP at the dose levels of 500, 1000, 2000 mg/kg once a day, and clinical signs, body weights, hematology, serum biochemistry, necropsy findings and organ weights were monitored and examined. In experimental results, ADP treatment, compared with vehicle control, did not induce the micronucleated erythrocytes from mouse bone marrow. In the 14 days study, any significant and toxicological differences in all measurements of parameters were not observed in ADP treatment groups of animals, compared with vehicle treatment. The No-Observed-Adverse-Effect-Level (NOAEL) of ADP in the 14 days study was determined to be greater than 2000 mg/kg/day in both sexes.

천연약재로부터 문화재보존용 방충방균제 개발연구 (The utilization of fungicide and insecticide from medicinal plants for conservation of cultural properties)

  • The germicidal and insecticidal properties of volatile components extracted from star anise(Illicium verum Hooker filius) and clove (Eugenia caryophyllata THUNBERG)were evaluated against five microorganisms and three insects for the purpose of developing biocidal active substances from medicinal plants. The volatile components of star anise and clove showed strong antimicrobial effect against Aspergillus niger, Penicillium funiculosum, Mucor hiemalis, Trichoderma viride, and Aureobasidium pullulans. The extracts of each medicine also showed insecticidal effects against Sitophilusoryzae L., Lyctus linearis GOZE, and Reticulitermes spertus kyushuensis Morimoto. Fumigant toxicities to adult insects were determined. In the case of fumiganttoxicity, the extract of star anise showed 100% mortality against R. spertus, S.oryzae, and L. linearis at rates of $2.5\mu\ell$, $50\mu\ell$, $250\mu\ell$/filter paper, respectively but showed no killing effects by clove. The volatile components of star anise and clove were investigated by means of GC/MS. The main constitute, anethole among 20components from star anise and eugenol among 9 components from clove were identified. The mixture of star anise and clove as the volume ratio of 2 : 1 showed higher properties for antimicrobial and insecticidal effect than each volatile component. A. niger was inhibited by the mixture(125ml/$m^3$) for up to 10 days of exposure. Also, from the result of observing state change of organic materials by volatile extracts of star anise and clove, volatile extracts effects have no effect on natural organic materials of organic cultural properties and can be used as biological control agent. As research contents as above, the insecticidal and germicidal agents from star anise and clove and the mixture of them were more efficient and high level to prevent biological damage for conservation of organic cultural properties. So they may be used in new development of biologicalinsecticidal and germicidal agents for conservation of cultural properties.

Extraction and Bioassay of Allelochemicals in Jerusalem Artichoke

  • Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.

Inhibitory Effects of Collagen Coated Coffee Bean Intake on Skin Aging

  • To evaluate the protective effect of collagen peptide-coated coffee extract on skin aging, cell viability was measured with a MTT assay using cultured CCD-986sk fibroblasts, and its effect on wrinkles in the skin of hairless mice induced by UVB-irradiation was examined. In addition, its effect on procollagen synthesis and anti-oxidative, and its inhibitory activity against collagenase, elastase, tyrosinase and MMP-1 were analysed. After the 30-minute topical treatment, the animals were exposed to UVB irradiation (60-100 mJ/cm2) for 4 weeks and its intensity increased during the period. Under the experimental conditions set in this study, the skin thickness of hairless mice significantly decreased (11.8-21.3%) compared to the control group. Based on these results, the prolonged oral intake of a collagen peptide mixture with coffee is expected to significantly increase the synthesis of procollagen in dermal fibroblasts, thereby contributing to the alleviation of wrinkling and lowered elasticity due to structural damage to the dermal layer caused by UV. The oral intake of collagen-coated coffee contributes to increasing collagen biosynthesis in a dose-dependent manner and alleviates the symptoms of thickened keratin caused by UV irradiation. However, it did not inhibit the enzymes involved in skin aging, whitening, wrinkle improvement, and antioxidation. Based on the these results, it can be concluded that the intake of collagen peptide-coated coffee extract can be utilized as an alternative material for the prevention or treatment of diseases associated with photoaging.

Phytonutrient Effects of Date Pit Extract against Azoxymethane-Induced Oxidative Stress in the Rat Colon

  • Plants and their by-products offer a diverse mixture of chemical constituents like natural antioxidants. Date-pits are rich in phenolic compounds that have antioxidant potential. The main objective of this study was to investigate the protective effect of a date-pit extract (DPE) against AOM-induced colonic carcinogenicity and oxidative stress. Thirty-two weanling male Sprauge-Dawley rats were randomly divided into four groups (eight rats in each group). All rats were fed basic diet and water ad libitum, and randomly distributed per treatment groups as follows: negative controls injected with normal saline once a week for two weeks, a cancer group injected intra-peritoneally with azoxymethane (15mg/kg body weight) for two consecutive weeks, and DPE treated groups receiving the extract via the oral route (1.5ml/day) for the entire experiment in the presence or absence of AOM injection. Results showed that DPE contained phytonutrients that were capable of inhibiting chemically-induced oxidative stress in the rat colonic cells. In those animals that consumed DPE, a protective effect was observed against AOM-induced oxidative stress in rat colonic cells as evident by a significant decrease in MDA and oxidized DCF formation in AOM injected and DPE fed groups. It is concluded that DPE has potential antioxidant and anticarcinogenic properties.
