Development of Axenic Culture and Astaxanthin Production in Microalgae (미세조류를 이용한 무균분리법 개발 및 astaxanthin 생산)

  • Microalgae are a renewable natural resource that requires only sunlight, carbon dioxide, phosphorus, and nitrogen for rapid growth. They produce a broad variety of basic chemical substances―such as vitamins, fatty acids and carotenoids―that have high added value potential for the pharmaceutical and food industries. The aim of this study was to develop axenic culture and to establish a cell growth assay for microalgae. A further experiment was carried out to determine the yield of astaxanthin derived from microalgae. The axenic culture was developed using a mixture of antibiotics [ampicillin (100 ${\mu}g/ml$), streptomycin (10 ${\mu}g/ml$), chloramphenicol (10 ${\mu}g/ml$), penicillin (10 ${\mu}g/ml$), neomycin (50 ${\mu}g/ml$), gentamycin (50 ${\mu}g/ml$), kanamycin (10 ${\mu}g/ml$), and nystatin (1.5 ${\mu}g/ml$)] and then used to extract a variety of useful components from the microalgae. The optimal concentration for the antibiotic mixture was 1-3 percent. A spectrophotometric cell growth assay was also established. Astaxanthin was extracted from Haematococus lacustris with a yield of $1.9{\times}10^{-3}{\mu}g/l$ per 1 ml of culture medium. In conclusion, the axenic culture method developed here allows extraction of high-quality astaxanthin and other useful components from microalgae.

Influences of Extraction pH on the Functionality of Soybean Protein Isolate (추출 pH가 분리대두단백질의 기능성에 미치는 영향)

  • This study was conducted to investigate the functionality of soybean protein isolates extracted in acidic range (pH 2.0 and 3.0), neutral range (pH 7.0) and alkaline range (pH 10.0 and 12.0). The protein content of soybean protein isolates extracted at pH 3.0 was maximum (93.31%), but that of pH 7.0 was minimum (73.93%). The extraction yield of soybean protein isolates extracted at pH 3.0 was minimum (0.36%), but that of pH 12.0 was maximum (47.54%). The functionality (solubility, water absorption, oil absorption, foam capacity, foam stability, emulsion capacity and gelation) of soybean protein isolates was significantly influenced by pH of extraction medium. The soybean protein isolates extracted at pH 2.0 and 3.0 were more soluble at acidic ranges and those of pH 3.0 and 7.0 were more soluble at neutral ranges, but those of pH 2.0, 3.0, 7.0, 10.0 and 12.0 were more soluble at alkaline ranges than other ranges. The soybean protein isolates extracted at pH 2.0 and pH 12.0 gave greater water absorption, oil absorption and foam capacity than those extracted at pH 3.0, pH 7.0 and pH 10.0. And the emulsion capacity of soybean protein isolates was increased by the increase of extraction pH.

  • The various conditions of photographs, especially kinds of films, combinated filters and seasons are important factors for forestry purpose aerial photography. In this paper the variations of tones were compared between color and color infrared, panchromatic black and white and infrared black and white, and among false color photographic images created by using 3 kinds of filters when prints are made. Color infrared film was good for identifying tree species, for its spectral signatures had a greater range of tones and hues than color signatures. In that case taken in May were more effective than taken April. False color photographs were not so good as color photographs because they were mostly dark and indistinct. Infrared black and white film with medium red filter showed potential for separating broad-leaved forests from conifers. MSS composed photographs, when composed with proper bands and densities, were proved useful for distinguishing land use types but not applicable to more detailed practices such as forest type separation and tree species identification.

Discovery of a Faint Quasar at z ~ 6 and Implications for Cosmic Reionization

  • Recent studies suggest that faint active galactic nuclei may be responsible for the reionization of the universe. Confirmation of this scenario requires spectroscopic identification of faint quasars ($M_{1450}$ > -24 mag) at z > 6, but only a very small number of such quasars have been spectroscopically identified so far. Here, we report the discovery of a faint quasar IMS J220417.92+011144.8 at z ~ 6 in a $12.5deg^2$ region of the SA22 field of the Infrared Medium-deep Survey (IMS). The spectrum of the quasar shows a sharp break at ${\sim}8443{\AA}$, with emission lines redshifted to $z=5.944{\pm}0.002$ and rest-frame ultraviolet continuum magnitude $M_{1450}=-23.59{\pm}0.10$ AB mag. The discovery of IMS J220417.92+011144.8 is consistent with the expected number of quasars at z ~6 estimated from quasar luminosity functions based on previous observations of spectroscopically identified low-luminosity quasars. This suggest that the number of $M_{1450}$ ~ -23 mag quasars at z ~ 6 may not be high enough to fully account for the reionization of the universe. In addition, our study demonstrates that faint quasars in the early universe can be identified effectively with a moderately wide and deep near-infrared survey such as the IMS.

  • Biosynthesis of curdlan( ${\beta}$-1,3 glucan) was shown by fluorscence on cellufluor medium. The highest production of curdlan was produced when glucose was used as a carbon source and ($NH_4$)$_2$$SO_4$ was used as a nitrogen source. ${\beta}$ -form of curdlan was detected in the fingerprint region (890 $cm^{-1}$) by FT-IR spectrum and shown homogeneous ${\beta}$ -1,3 glucan by $^{13}C$ NMR spectrum ($C_1$-103 ppm, $C_2$-73.2 ppm, $C_3$-86.4 ppm, $C_4$-68.7 ppm, $C^{5}$-76.63 ppm, $C_{6}$-61.2 ppm). Transition of structure from triple helix coil form to random coil form was appeared at 0.1 ∼0.25 M NaOH concentration. It was shown that natural curdlan is a triple helix form in neutral but becomes weak in alkaline condition.

The Dose-Dependent Effects of Nitric Oxide on Human Sperm Cell Function (Nitric Oxide가 인간 정자세포의 기능에 미치는 효과에 대한 연구)

  • This study was performed to determine the effects of nitric oxide on human sperm cell function. Semen samples were obtained from normal healthy volunteers. Motile spermatozoas collected by swim-up method were incubated up to 24 hours in Ham's F-10 medium supplemented with a various concentration of sodium nitroprusside (nitric oxide releasing agent). Sperm motility, hyperactivation, acrosome reaction rate, and acrosin activity were determined. The results are as follows; 1. 1mM of SNP resulted in a significant decrease in sperm motility ($44.8%{\pm}8.9%:78.1%{\pm}6.3%$, and hyperactivation $(10.4%{\pm}6.4%:47.7%:{\pm}9.5%)$ after incubation for 3 hours compared with the control group (Ham's F-10 alone), but had no effect on acrosome reaction. 2. At $100{\mu}M$ SNP, sperm motility was reduced after incubation for 6 hours $(54.8%{\pm}3.2%)$ compared with that of the control group $(82.7%{\pm}8.9%)$, but hyperactivation and acrosome reaction were not affected. 3. However, a lower concentration (less than $10{\mu}M$) of SNP had no effect on sperm motility and hyperactivation for 8 hours of incubation but significantly decreased them when incubation periods were increased up to 24 hours compared with the control group. On the other hand, $1{\mu}M$ and $10{\mu}M$ SNP significantly increased the acrosome reaction rate in both acrosomal status ($17.3%{\pm}5.2%$, $23.5%{\pm}4.7%$, respectively) and acrosin activity ($34.3{\mu}IU{\pm}10.5{\mu}IU,\;45.6{\mu}IU{\pm}5.6{\mu}IU$, respectively) as compared with the control group $(7.0%{\pm}4.0%,\;9.5{\mu}IU{\pm}3.4{\mu}IU)$. These results indicate that SNP, NO releasing agent, has a dose-dependent effects on the sperm cell function. Therefore it may positively affect the fertilization by promoting acrosomal reaction at a lower concentration (less than $10{\mu}M$).

Studies on the Natural Mortality of the Young Short Necked Clam, Tapes Japonica-II . Resistance to Temperature , Salinity and Exposure for the Young Short Necked Clam (바지락치패의 폐사에 관한 연구-II 바지락 치패의 온도, 염분, 노출에 대한 저항성)

  • In a previous paper, the dirunal and monthly changes of the tidal flat temperature and chlorinity were observed. In this paper the resistance of the young short necked clam in various growth stages to the temperature, to the chlorinity and to the exposure were studies. The young clam used were in early (E), medium (M) and late (L) young stages whose shell lengths were 2.0-3.5mm, 9.0-11.0mm. and 14.0-16.0mm., respectively. The results were : 1. At various sea water temperatures , the chlorinity resistance of the young clam was directly proportional to the shell length. 2. When both inadequate sea water temperature and abnormal chlorinity are simultaneously applied, the resistance of these young clams was more markedly reduced than the case of applying either one of these conditions. 3. In clams of M and L, no lethal effect were observed when daily immersion of four to eight hours for a week into the sea water of any concentration of chloriity at 26-34$^{\circ}C$, whereas in E, 37-90% of mortalities were obtained by immersion of eight hours daily into both the fresh water and the sea water of higher chloriniites (more than 23.5$\textperthousand$ Cl) at the same temperature above. 4. The lower critical thermal maximum for lethal to the young clam was 38$^{\circ}C$. With four hours immersion daily at the water temperature of 38$^{\circ}C$, the mortalities of E, M and L to the lower chlorinities (less than 6.7$\textperthousand$Cl) were 100, 70-100 and 27-37% respectively ; to the higher chlorinities (more than 23.5$\textperthousand$Cl) 10-70, 10-37 and 3 % respectively ; to the normal range of chlorinities (13.4-16.8$\textperthousand$Cl) 0-13, 3 and 0 % respectively. 5. No lethal effects were observed in E and M clams immersed continuously for seven days in sea water with chlornities of 7.2 -21.7$\textperthousand$Cl at 18-24$^{\circ}C$, while notable mortalities were observed in E which had been kept at lower (less than 4.8$\textperthousand$ Cl) and higher (more than 24.1$\textperthousand$ Cl) chlorinites. 6. Although the resistance of the young clam to the chlorinity may have to be related closely to the life history of the clam prior subject to the experiment, the adapted chlorinity range was 7.2-19.3$\textperthousand$ Cl and the optimum range was 13.4-16.8$\textperthousand$Cl. 7. Remarkable lethal effects were observed for the E and M clams to the exposure temperature of 38$^{\circ}C$ whereas the L and had no such fatal results.

Phorbol Ester-induced Contraction Through p38 Mitogen-activated Protein Kinase is Diminished in Aortas from DOCA-Salt Hypertensive Rats

  • The role of mitogen-activated protein kinase (MAPK) in the decreased contractile response to phorbol ester in aortic smooth muscle strips from deoxycorticosterone acetate (DOCA)-salt hypertensive rats was examined. Norepinephrine (NE) evoked greater contractility in aortic strips from DOCA rats than in those of sham-operated rats. 12-Deoxyphorbol 13-isobutyrate (DPB) induced contraction in $Ca^{2+}-free$ medium, which was diminished in strips from DOCA rats compared to sham-operated rats. Vasoconstrictions induced by these stimulants were inhibited by SB203580 and PD098059, inhibitors of p38 MAPK and extracellular signal-regulated kinase (ERK) 1/2, respectively, in both strips. The phosphorylation of p38 MAPK and ERK1/2 induced by NE was greater in strips from DOCA rats compared to those from sham-operated rats, and this phosphorylation was inhibited by the kinase inhibitors. DPB increased the phosphorylation of p38 MAPK and ERK1/2 in strips from both animals, and the increment of p38 MAPK phosphorylation by the stimulant was diminished in strips from DOCA rats compared to sham-operated rats. These findings suggest that the $Ca^{2+}-independent$ contraction evoked by DPB results from the activation of MAPKs in rat aortic smooth muscle and that the attenuated contractility by DPB in DOCA rat appears to be associated with diminished p38 MAPK activity.

Induction of Muscle Atrophy by Dexamethasone and Hydrogen Peroxide in Differentiated C2C12 Myotubes (C2C12 근관세포에서 dexamethasone 및 hydrogen peroxide에 의한 근위축 유도)

  • Muscle atrophy due to aging, starvation, and various chronic diseases leads to a decrease in muscle fiber area and density due to reduced muscle protein synthesis and increased protein breakdown. This study investigated the effect of dexamethasone and hydrogen peroxide on the induction of muscle atrophy and expression of atrophy-related genes in differentiated C2C12 myotubes. C2C12 myoblasts were differentiated into myotubes in differentiation medium. During myoblast differentiation, muscle-specific transcription factors, such as myogenin, and MyoD expression increased. Differentiated C2C12 myotubes exposed to noncytotoxic levels of dexamethasone and hydrogen peroxide showed a decrease in myotube diameter, which was associated with up-regulation of muscle-specific ubiquitin ligases, such as muscle atrophy F-box (MAFbx)/atrogin-1 and muscle RING finger-1 (MuRF1), and down-regulation of myogenin and MyoD. These results demonstrated that dexamethasone and hydrogen peroxide induced atrophy through regulation of muscle-specific ubiquitin ligases and muscle-specific transcription factors in C2C12 myotubes. In this study, we confirmed the process of differentiation of C2C12 myoblasts into myotubes in in vitro experiments in the presence of atrophy. This muscle atrophy model of C2C12 cells induced by dexamethasone or hydrogen peroxide seems suited to studies of the mechanism of muscle atrophy suppression and to exploit the experiment for excavating new muscle atrophy.

Isolation and Characterization of a Paenibacillus incheonensis YK5 with Antimicrobial Activity aginst MRSA (항MRSA 활성을 보이는 Paenibacillus incheonensis YK5의 분리 및 특성)

  • Various bacteria were isolated from Korean soil samples based on their capability inhibiting the growth of MRSA strains. Among them, strain YK5 with the highest activity was a Gram positive sporulative bacillus with motility. It did not produce indole and no acid was formed from mannitol by the bacterium. The 16S rRNA sequence of the strain showed $95{\sim}98%$ homology with those of Paenibacillus spp.. The bacterial isolate shared the highest homology with that of P. elgii (98%), but was named as Paenibacillus incheonensis YK5 due to differences in physiological properties. Butanol extract of the P. incheonensis YK5 culture grown in SST medium at $37^{\circ}C$ for 96 hr showed a broad antimicrobial activity against Gram-positive (MRSA and Streptococcus pneumoniae) and negative (Pseudomonas aeruginosa, Salmonella spp., Shigella spp., Escherichia coli, Klebsiella pneumoniae) pathogenic bacteria and fungi (Cryptococcus neoformans and Trichophyton). The antimicrobial activity in the crude extract was stable in a broad range of temperature and pH, $20{\sim}100^{\circ}C$ and $3.0{\sim}6.0$, respectively. Therefore, the antimicrobial activity of P. incheonesis YK5 had potential as a novel antibiotics for pathogens including MRSA.
