Seed Morphological Characteristics and Dormancy type of Eranthis stellata Maxim., Korea Rare Plant. (희귀식물 너도바람꽃(Eranthis stellata Maxim.) 종자의 형태특성 및 휴면유형 분석)

  • Eranthis stellata Maxim. is a perennial plant that grows around the valley. E. stellata is concerned about the decline in natural habitats due to climate change in KOREA, continues to be observed and protected as an endangered species (Least Concrned, LC). Nevertheless, studies on the characteristics of the seeds of E. stellata are insufficient. So, this study analyzed the morphological characteristics and dormancy types of seeds. Seeds of E. stellata was collected in April at Gyeongsangbuk-do Arboretum and kept at 5 ℃ until using. To investigate the morphology of seeds, an optical microscope and a scanning electron microscope (SEM) were used. GA3 treated or untreated seeds (4 replicates of 25 seeds each) were observed germination and embryo growth for 1 month at 5 ℃ and 25/15 ℃ (12h day/12h night). The seed surface of E. stellata, light brown, was observed as a common characteristic of Eranthis genus, reticulate. The short axis of seeds was 1.11~1.77mm (average 1.44mm), and the long axis was 1.27~1.91mm (average 1.63mm), which was investigated in a slightly round shape (subglose). While no germination was observed at all conditions, Embryo growth was observed at 5 ℃ both in the control group and with GA3treated groups. Thus, seeds of E. stellata are classified as morphological physiological dormancy (MDP), which requires embryonic development and dormant break at the same time. These results can be useful information for determining morphological physiological seed dormancy and germination, and will be an important basic data for seed propagation of E. stellata as a resource.

A Literature Review of Biophilic Design Applied to Healthcare and Welfare Facilities: Comparison of Facilities for the Elderly and Children/Adolescents (건강·복지시설에 적용된 바이오필릭 디자인 문헌연구: 노인과 아동청소년 시설 비교)

  • This study is a literature review analyzing the characteristics of biophilic design in health and welfare facilities for the elderly, children, and adolescents. Studies on biophilic design in such facilities were selected from two Korean databases. Eight studies were analyzed based on biophilic design elements, and the characteristics of biophilic design were compared between facilities for the elderly and those for children/adolescents. Elderly facilities exhibited more patterns of 'direct experience of nature' than their counterparts, while children/adolescents' facilities displayed more patterns of 'space and place experience' compared to facilities for the elderly. The pattern of 'indirect experience of nature' was similar for both the elderly and children/adolescents. We suggest utilizing more 'direct experience of nature' patterns, such as natural light and water, to reduce stress in vulnerable children and younger individuals. Additionally, incorporating more 'experience of space and place' patterns may help reduce stress and enhance the cognition of older individuals.

Chlorophyll contents and expression profiles of photosynthesis-related genes in water-stressed banana plantlets

  • Water scarcity decreases the rate of photosynthesis and, consequently, the yield of banana plants (Musa spp). In this study, transcriptome analysis was performed to identify photosynthesis-related genes in banana plants and determine their expression profiles under water stress conditions. Banana plantlets were in vitro cultured on Murashige and Skoog agar medium with and without 10% polyethylene glycol and marked as BP10 and BK. Chlorophyll contents in the plant shoots were determined spectrophotometrically. Two cDNA libraries generated from BK and BP10 plantlets, respectively, were used as the reference for transcriptome data. Gene ontology (GO) enrichment analysis was performed using the Database for Annotation, Visualization, and Integrated Discovery (DAVID) and visualized using the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway prediction. Morphological observations indicated that water deficiency caused chlorosis and reduced the shoot chlorophyll content of banana plantlets. GO enrichment identified 52 photosynthesis-related genes that were affected by water stress. KEGG visualization revealed the pathways related to the 52 photosynthesisr-elated genes and their allocations in four GO terms. Four, 12, 15, and 21 genes were related to chlorophyll biosynthesis, the Calvin cycle, the photosynthetic electron transfer chain, and the light-harvesting complex, respectively. Differentially expressed gene (DEG) analysis using DESeq revealed that 45 genes were down-regulated, whereas seven genes were up-regulated. Four of the down-regulated genes were responsible for chlorophyll biosynthesis and appeared to cause the decrease in the banana leaf chlorophyll content. Among the annotated DEGs, MaPNDO, MaPSAL, and MaFEDA were selected and validated using quantitative real-time PCR.

  • This study explores the enhancement of mechanical properties in Polymethyl Methacrylate (PMMA) composites through the incorporation of Methyl Methacrylate (MMA) and Ethylene Glycol Dimethacrylate (EGDMA). Utilizing Digital Light Processing (DLP) technology, we conducted a series of experiments to analyze the impact of varying concentrations of MMA and EGDMA on PMMA. The results indicate that while MMA demonstrates non-linear and variable mechanical strength across different PMMA concentrations, EGDMA consistently improves mechanical strength as PMMA concentration increases. This consistent enhancement by EGDMA suggests a stable and predictable reinforcement effect, which is critical for applications requiring high mechanical strength. Our comparative analysis highlights that EGDMA is a more effective additive than MMA for optimizing the mechanical performance of PMMA composites. Specifically, EGDMA's ability to provide uniform reinforcement across various PMMA concentrations makes it ideal for high-strength applications. These findings are significant for material scientists and engineers focused on the design and development of advanced PMMA-based materials. In conclusion, this research underscores the importance of selecting appropriate additives to enhance the mechanical properties of PMMA composites. The superior performance of EGDMA in reinforcing PMMA suggests its potential for broader applications in fields such as automotive, construction, medical devices, and 3D printing. This study provides valuable insights that can guide future research and development in high-performance composite materials, paving the way for innovative applications and improved material efficiency.

Characterization of the Non-Volatiles and Volatiles in Correlation with Flavor Development of Cooked Goat Meat as Affected by Different Cooking Methods

  • Thai-Native×Anglo-Nubian goat meat cooked by grilling (GR), sous vide (SV), and microwave (MW), was compared to fresh meat (Raw) in terms of flavor development. Non-volatile [i.e., free amino acids, nucleotide-related compounds, taste active values (TAVs) and umami equivalency, sugars, lipid oxidation, Maillard reaction products] and volatile compounds, were investigated. Notably, inosine monophosphate and Glu/Gln were the major compounds contributing to umami taste, as indicated by the highest TAVs in all samples. Raw had higher TAVs than cooked ones, indicating that heat-cooking removes these desirable flavor and taste compounds. This could be proportionally associated with the increase in aldehyde, ketone, and nitrogen-containing volatiles in all cooked samples. GR showed the highest thiobarbituric acid reactive substances (1.46 mg malonaldehyde/kg sample) and browning intensity (0.73), indicating the greatest lipid oxidation and Maillard reaction due to the higher temperature among all cooked samples (p<0.05). In contrast, SV and Raw exhibited similar profiles, indicating that low cooking temperatures preserved natural goat meat flavor, particularly the goaty odor. The principal component analysis biplot linked volatiles and non-volatiles dominant for each cooked sample to their unique flavor and taste. Therefore, these findings shed light on cooking method selection based on desirable flavor and preferences.

Melanin Inhibitory Effect of Tuber himalayense Isolated in Incheon, Korea

  • There has been a growing interest in skin beauty and antimelanogenic products. Melanogenesis is the process of melanin synthesis whereby melanocytes are activated by UV light or hormone stimulation to produce melanin. Melanogenesis is mediated by several enzymes, such as tyrosinase (TYR), microphthalmia-associated transcription factor (MITF), tyrosinase-related protein-1 (TRP-1), and TRP-2. In this study, we investigated the effect of Tuber himalayense extract on melanin synthesis in α-melanocyte-stimulating hormone (α-MSH)-treated B16F10 melanoma cells. We confirmed that T. himalayense extract was not toxic to α-MSH-treated B16F10 melanoma cells and exhibited a significant inhibitory effect on melanin synthesis at concentrations of 25, 50, and 100 ㎍/ml. Additionally, the T. himalayense extract inhibited melanin, TRP-1, TRP-2, tyrosinase, and MITF, which are enzymes involved in melanin synthesis, in a concentration-dependent manner. Furthermore, T. himalayense extract inhibited the mitogen-activated protein kinase (MAPK) pathways, such as extracellular signal-regulated kinase-1/2 (ERK), c-Jun N-terminal kinase (JNK), and p38. Therefore, we hypothesized that various components of T. himalayense extract affect multiple factors involved in melanogenesis in B16F10 cells. Our results indicate that T. himalayense extract could potentially be used as a new material for preparing whitening cosmetics.

Field Survey on the Maintenance Status of Greenhouses in Korea (온실의 유지관리 실태조사 분석)

  • The purpose of this study was to investigate greenhouse maintenance by farms by looking into greenhouses across the nation for greenhouse specification, disaster-resistance greenhouse construction, types and degree of damage due to natural disasters, pre-inspection in case of typhoon or heavy snow forecast, and fire-fighting facilities to prevent a fire. The findings were summarized as follows: as for greenhouse specification, the highest proportion of them were 90 m or longer both in single- and multi-span greenhouses in terms of length; 8 m or wider and 7.0~7.9 m in single- and multi-span greenhouses, respectively, in terms of width; 1.5~1.9 m and 2.0~2.9 m in single-and multi-span greenhouses, respectively, in terms of height; and 3.0~3.9 m and 6 m in single- and multi-span greenhouses, respectively, in terms of diameter. As for disaster-resistance greenhouses, farmers were reluctant to install such greenhouses. The low distribution of disaster-resistance greenhouses was attributed to the greenhouses built dependent on the old practice, the greenhouses already completed, and relatively high construction costs. As for damage by natural disasters, greenhouses were subject to more damage by typhoons than heavy snow. They mainly inspected the ceiling and side windows, entrances, and fixation bands for covering materials in case of typhoon forecast and the heating devices in case of heavy snow forecast. As for repair methods for greenhouse pipe corrosion, they preferred partial replacement to painting and did not use stiffeners for structures to prevent a natural disaster in most cases. As for the maintenance of greenhouse covering materials, most farmers inspected their sealing property but did not clean the coverings for light transmission. The destruction of structural materials can be prevented by eliminating greenhouse covering materials during a typhoon, but they were not able to do so because of the covering material replacement costs and the crops they were growing. The study also examined whether greenhouse farms had fire-fighting facilities to prevent a fire and found that they lacked the perception of greenhouse fire prevention to a great degree.

  • Elder-berries are economical plants and grown commercially in the United States. Most of the crop is sold to processors and made into natural colour, jellies, jam, port wine or elder berry wine, elder berry juice or used to manufacture of pie mixes. Processors are showing increasing interest in cultivation elder berries. In the present study was wish to find the effect of various factors and practical methods on the processing for the true fruit colour made from fresh elder berries juice as the clearing by the treated pectinase, added sugar, vaccum concentration, extraction of anthocyanosid powder and its property with used prospects. In this study obtained results can be summarized as follows. 1. Pectinase is useful and effected to make clear juice from fresh elder berry juice treated on the $34{\sim}45^{\circ}C$. 2. P.V.C. drum is most useful stored without unfading for clearing elderberry juice and its exposed redish dark purple colour. 3. Clearing elderberry juice stored in the stainless steel drum, changed colour from redish dark purple to dark violets during the stored at the room temperature. 4. Added sugar (Brix $48{\sim}53^{\circ}C$) to the fresh elderberry juice is useful and suitable prospects for to make home juice and wine. 5. The elderberry juice of vaccum concentration is change colour from dark purple to black brown colour during the stored at the room temperature. 6. Its possible to make anthocyanosid powder extracted made from fresh elderberry juice 7. Property of anthocyanosid is an follows. 1) The spectrum of elderberries juice is $523{\sim}530m{\mu}$ 2) The colouring stability is less than $pH_4$ 3) The colouring stability by the heating is less than $pH_4$, and then the more than $pH_4$ is changed colour from redish dark purple to dark brown. 4) By the light colouring stability is less than $pH_4$ 5) The colouring most stability in the pH is $pH_3$ 8. In the future, natural colour made from elderberry juice on the useful prospects is suitable used to make natural fruit juice, ice cream, candy, commercial and home wine, gum, and manufacture of jelly jam, pie maxes, ect.

Preservation Conditions of Aqueous Samples Containing silver Nanomaterials (은나노물질을 포함한 수질시료의 보관조건)

  • A prerequisite for precise quantification of nanomaterials contained in environmental samples is to prepare suitable preservation conditions of samples. This study was initiated to suggest preservation conditions of aqueous samples for analyses of metal nanomaterials. Variation in the size of silver nanomaterial (cit-AgNP) was observed according to change in various conditions, such as pH, electrolyte concentration, temperature, nanomaterial concentration, and time. Aggregation of AgNP was characterized for each environmental condition, and finally proper preservation conditions of samples were proposed based on experimental results on AgNP aggregation. In addition, the preservation period of sample was computed by the doublet time of AgNP. The results indicate that the aggregation rate of cit-AgNP was close to 0 at the conditions of pH of ${\geq}7$, electrolyte ($Ca(NO_3)_2$) concentration of ${\leq}3mM$, temperature of $4^{\circ}C$, and cit-AgNP concentration of ${\leq}2mg/L$. Furthermore, the experimental results on doublet time of cit-AgNP suggest that maximum preservation period was evaluated to be 15.79~17.53 days when the concentration of 100 nm cit-AgNP is assumed to be $1{\mu}g/L$ which is considered as an environmentally-relevant concentration of engineered nanomaterials. Our results suggest that samples should be preserved at $4^{\circ}C$ and analyzed within 2 weeks.

Multiple Monoclonal Antibodies Produced in a Single Transgenic Plant (형질전환 식물체에서의 복합 단일 항체 단백질 생산)

  • Production of highly valuable immunotherapeutic proteins such as monoclonal antibodies and vaccines using plant biotechnology and genetic engineering has been studied as a popular research field. Plant expression system for mass production of such useful recombinant therapeutic proteins has several advantages over other existing expression systems with economical and safety issues. Immunotherapy of multiple monoclonal antibodies, which can recognize multiple targeting including specific proteins and their glycans highly expressed on the surface of cancer cells, can be an efficient treatment compared to a single targeting immunotherapy using a single antibody. In this study, we have established plant production system to express two different targeting monoclonal antibodies in a single transgenic plant through crossing fertilization between two different transgenic plants expressing anti-colorectal cancer mAbCO17-1A and anti-breast cancer mAbBR55, respectively. The F1 seedlings were obtained cross fertilization between the two transgenic parental plants. The presence, transcription, and protein expression of heavy chain (HC) and light chain (LC) genes of both mAbs in the seedlings were investigated by PCR, RT-PCR, and immunoblot analyses, respectively. Among all the seedlings, some seedlings did not carry or transcribe the HC and LC genes of both mAbs. Thus, the seedlings with presence and transcription of HC and LC genes of both mAbs were selected, and the selected seedlings were confirmed to have relatively stronger density of HC and LC protein bands compared to the transgenic plant expressing only each mAb. These results indicate that the F1 seedling plant with carrying both mAb genes was established. Taken together, plant crossing fertilization can be applied to generate an efficient production system expressing multiple monoclonal antibodies for immunotherapy in a single plant.
