Isolation and Anti-thrombotic Activity of Citric acid 1,5-dimethyl ester from Gastrodia elata

  • In the course of continous work for the discovery of anti-thrombotic constituents from tubers of Gastrodia elata, citric acid 1,5-dimethyl ester was isolated from ethylacetate soluble fraction prepared from the methanol extract. The structure of the compound was determined by the spectroscopic data. The anti-thrombotic effect of this compound was observed with prolonging the bleeding time in thrombin-induced thrombosis model of mice.

  • The natural flavonoid macakurzin C (1) exhibited adiponectin biosynthesis-inducing activity during adipogenesis in human bone marrow mesenchymal stem cells and its molecular mechanism was directly associated with a pan-peroxisome proliferator-activated receptor (PPAR) modulator affecting all three PPAR subtypes α, γ, and δ. In this study, increases in adiponectin biosynthesis-inducing activity by macakurzin C derivatives (2-7) were studied. The most potent adiponectin biosynthesis-inducing compound 6, macakurzin C 3,5-dimethylether, was elucidated as a dual PPARα/γ modulator. Compound 6 may exhibit the most potent activity because of the antagonistic relationship between PPARδ and PPARγ. Docking studies revealed that the O-methylation of macakurzin C to generate compound 6 significantly disrupted PPARδ binding. Compound 6 has therapeutic potential in hypoadiponectinemia-related metabolic diseases.

  • Ammonia is an inorganic compound that may cause severe odor problem. In this study the effectiveness of applying natural neutralizer to destroy and remove the odor-causing compound from gas streams was studied. Experimental result evaluated with a bench-scale apparatus via the neutralization of gas phase. This indicates that the natural neutralization depends on the gas concentration, gas residence time, temperature and pH. Removal efficiency of ammonia from gas stream was achieved by 95% using theconvection in the packed bed. This study proved the chemical neutralization technology was effective for controlling inorganic odor-causing compound.

  • A component, $C_{20}H_{32}O_3$, m.p. $207.5^{\circ}$, was isolated from Siegesbeckia pubescens $M_{AKINO}$. This compound was identified as $17-hydroxy-16{\beta}-(-)-kauran-19-oic\;acid$, using spectroscopies and synthesizing various derivatives of the compound.

  • The chemistry of secondary products from Acanthopanax species and their pharmacological activities were reviewed. A nitrogenous compound, a furan compound, a quinoid, benzoids, coumarins, phenylpropanoids, lignans, flavonoids, terpenoids, phytosterols, polyacetylenes, a pyrimidine, cyclitols, monosaccharides and an aliphatic alcohol have been isolated from Acanthopanax species and have been shown to have various levels of activities such as anti-bacterial, anti-cancer, anti-gout, anti-hepatitis, anti-hyperglycemic, anti-inflammatory, anti-leishmanicidic, anti-oxidant, anti-pyretic, anti-xanthine oxidase, choleretic, hemostatic, hypocholesterolemic, immunostimulatory and radioprotectant effects, etc.

  • Two ergosterins and two triterpenoids were isolated from the dried fruit bodies of Ascomyce Bulgaria inquinans. By means of chemical (hydrolysis) and spectroscopic methods (NMR, EIMS), their structures were established as betuinic acid (1), cerevisterol (2), (24R)ergosta-7, $22E­diene-3\beta,\;5\alpha,\;6\beta-triol-3-O-palmitate$ (3) and ursolic acid (4). Compound 3 is a new compound.

  • This study is performed to evaluate freezing and thawing properties of polypropylene fiber reinforced eco-concrete using soil, natural coarse aggregate, soil compound and polypropylene fiber. The mass loss ratio is decreased with increasing the content of natural coarse aggregate and soil compound, but it is increased with increasing the content of polypropylene fiber. The ultrasonic pulse velocity, dynamic modulus of elasticity and durability factor are increased with increasing the content of natural coarse aggregate and soil compound, but it is decreased with increasing the content of polypropylene fiber. The mass loss ratio, ultrasonic pulse velocity, dynamic modulus of elasticity and durability factor are $1.49{\sim}3.32%,\;1,870{\sim}2,465\;m/s,\;77X10^2{\sim}225X10^2\;MPa\;and\;84.6{\sim}92.8$ after freezing and thawing 300 cycles, respectively. These eco-concrete can be used for environment-friendly side walk and farm road.

  • This study is performed to examine the physical and mechanical properties of eco-concrete with polypropylene fiber. Test results show that the strengths are increased with increase of the content of natural gravel, excellent soil compound and polypropylene fiber. The coefficient of permeability is increased with increase of the content of the polypropylene fiber but it is decreased with increase of the content of natural gravel and excellent soil compound. The lowest coefficient of permeability is achieved by eco-concrete which it is 100 times lower than that of the control. The durability factor is increased with increase of the content of natural gravel and excellent soil compound, and decreased with increase of the content of polypropylene fiber.

  • An antihyperglycemic compound in a model of alloxan induced diabetic rats was isolated from the root bark of Aralia elata. The compound identified to be oleanolic acid $28-O-{\beta}-_D-glucopyranoside$ was active at a dose of 100 mg/kg p.o. in the rats. It also has increased the contents of liver glycogen which were lowered by injection of alloxan in the rats.

  • A cytotoxic compound was purified from the flowers (46 g) of Paulownia tomentosa by normal column chromatography. As a result of the structure analysis by spectroscopic methods, the compound was identified as isoatriplicolide tiglate, which shows in vitro cytotoxicity.
