소에서 Coxiella burnetii의 검출 (Detection of Coxiella burnetii in Cattle)

  • Coxiella burnetii는 세포 내 기생하는 리케치아의 한 종류로 전세계에서 발병하는 인수공통 전염병인 큐열의 원인체이다. 국내에서 C. burnetii의 감염이 사람과 동물에서 보고되고 있지만 제한적인 지역에서만 실시되었다. 본 연구의 목적은 real-time PCR을 이용하여 소의 혈액과 유즙 시료에서 C. burnetii의 전국적인 검출률을 조사하는 것이다. 총 1,325 개 시료(한우 혈액 622 개, 젖소 혈액 185 개, 젖소 개체 유즙 170 개 및 젖소 집합유 348 개)를 전국적으로 채취하였다. 채취한 시료에서 Genomic DNA를 추출하였으며 C. burnetii 검출을 위한 real-time PCR을 실시하였다. 한우 혈액 622 개 중 17(2.7%) 개에서 C. burnetii가 검출되었고 젖소 혈액 185 개 중 2 (1.1%) 개에서 C. burnetii가 검출되었다. 젖소의 개체별 유즙 170 개 중 27(15.9%) 개에서 C. burnetii가 검출되었으며 집합유 348 개 중 84(24.1%) 개에서 C. burnetii가 검출되었다. 본 연구는 국내의 소가 C. burnetii에 높게 감염되어 있는 것을 확인하였으며, 인수공통질병인 큐열에 의한 가축과 사람의 피해를 예방하기 위하여, 가축과 사람의 큐열에 대한 추가적인 역학적 연구가 필요하다고 생각된다.

Identification of Genetic Markers for Korean Native Cattle (Hanwoo) by RAPD Analysis

  • In order to develop the specific genetic marker for Korean native cattle (Hanwoo), randomly amplified polymorphic DNA (RAPD) analysis of 6 different cattle breeds was attempted by using 38 decamer primers. In comparison of RAPD patterns, two distinctive DNA bands specific for Hanwoo were detected. One was 296 bp of DNA fragment found to be specific only for female Hanwoo when primer GTCCACACGG was employed. In individual analysis of this RAPD marker was observed only in female individuals with the possibility of $85.3\%$. The other was 521 bp of RAPD marker amplified using TCGGCGATAG and AGCCAGCGAA primers, which showed $83.0\%$ of genetic frequency in 85 male and 68 female individuals tested. Nucleotide sequencing of these genetic markers revealed that 296 bp marker has a short micro satellite-like sequence, ACCACCACAC, and a tandem repeat sequence of microsatellite GAAAAATG in the determined sequence. Two distinctive tandem repeats of microsatellite sequences, MC and GAAGA, were also appeared in 521 bp DNA marker. In BLAST search, any gene having high homology with these markers was not found.

한우사 내부 위치 및 TMR 배합 작업에 따른 분진 모니터링 (Dust Concentration Monitoring in Korean Native Cattle Farm according to Sampling Location and TMR Process)

  • Many parts of problems in livestock industry today are associated with organic dust. Endotoxin and toxic gasses on the surface of dust and dust itself can cause aesthetic displeasure and respiratory disease. It also reduces livestock productivity by suppressing immunity of animals and carrying microbes causing animal disease. However, dust level of cattle farm was rarely reported in Korea, and regulation for cattle farm worker does not exist. In this paper, dust concentration and environmental condition were regularly monitored in a commercial Korean native cattle farm. The measurement was conducted according to location and working activities. From the measurement, distribution of dust concentration was affected by wind environment, as the result of natural ventilation. TMR mixer was a major source of dust in target cattle house. The maximum inhalable dust concentration was 637.8 times higher than exposure limit as feed dropped into the TMR mixer. It was expected that dust generation could be affected by particle size and drop height of feed. This study suggests potential risk of dust in cattle farm, and necessity for latter study. Effect of aerodynamic condition and TMR processing should be investigated for dust reduction study.

국내 한우의 소바이러스성 설사 바이러스 지속감염우에 대한 실태 조사 (Prevalence for persistently infected cattle with bovine viral diarrhea virus in Korea)

  • Bovine viral diarrhea virus (BVDV) is very important disease in domestic and wild ruminants and has a world wide distribution. Cattle persistently infected with BVDV (BVDV-PI) are the primary reservoir for BVDV infection in Korean native cattle herds. The prevalence of cattle persistently infected with BVDV (BVD-PI) was determined using 4,260 heads from 29 Korean native cattle farms at 8 districts from 2011 to 2012. The sera and ear nothches were collected for each sample. We surveyed BVD-PI cattle using antibody ELISA and antigen capture ELISA for detection of antibody and antigen respectively. Three thousand seventy-six cattle (72.2%) were positive for BVDV antibody and a total of 27 BVD-PI cattle were found in 12 farms. 11 cattle (40.7%) out of the total 27 BVDV-PI cattle were six months old or under. The positive rate of BVDV antibody (83.2%) from 12 farms with BVD-PI cattle was higher than the positive rate of BVDV antibody (63.6%) from 17 farms without BVD-PI cattle.

소의 친자감정을 위한 Microsatellite markers의 유전적 다양성 분석 (Analysis of genetic diversity for cattle parentage testing using microsatellite markers)

  • The objective of present study was to ascertain genetic diversity for cattle parentage testing. A total of 59 random cattle samples(29 Korean native cattle and 30 dairy cows) were genotyped by using 11 microsatellite loci(BM1824, BM2113, ETH10, ETH225, EH3, INRA23, SPS115, TGLA122, TGLA227, TGLA53, and TGLA126). This method consisted of multiplexing PCR procedure and showed reasonable amplification of all PCR products. Genotyping was performed with an ABI 310 genetic analyzer. The number of alleles per locus varied from 5 to 11 with a mean value of 6.73 in the Korean native cattle(KNC), 4 to 9 with a mean of 5.91 in dairy cows(DC). Expected heterozygosity was ranged 0.534~0.855(mean 0.732), 0.370~0.866(mean 0.692) in the KNC and DC, respectively. PIC value was ranged 0.485~0.821(mean 0.684), 0.336~0.834(mean 0.640) in the KNC and DC, respectively. Of the 11 markers, 7 markers(ETH10, EH3, INRA23, SPS115, TGLA122, TGLA227, TGLA53) and 3 markers(INRA23, TGLA227, TGLA53) have relatively high PIC value (>0.7) in the KNC and DC, respectively. The total exclusion probability of 11 microsatellite loci was 0.9997 and 0.9991 in the KNC and DC, respectively. These results present basic information for developing a system for parentage verification and individual identification in the KNC and DC.

전북지역 한우의 소 바이러스성 설사 바이러스 감염 현황 조사 (Infection status of bovine viral diarrhea virus in Korean native cattle in Jeonbuk State, Korea)

  • Bovine viral diarrhea virus (BVDV) is distributed in cattle worldwide and causes significant economic losses to the livestock industry. Identification and remove of BVDV persistently infected (PI) cattle is very important to control BVDV infection in cattle herd. The objective of this study is to investigate the infection status of BVDV infection in Korean native cattle (Bos taurus coreanae) farms located in Jeonbuk State. From 2021 to 2022, a total of 1,497 samples were collected from 17 cattle farms and tested for BVDV antigen using a commercial ELISA kit. By the first ELISA testing, 24 cattle from six farms were positive for BVDV antigen, showing the farm-level or cattle-level prevalence of 35.3% or 1.6%, respectively. By the second ELISA testing which carried out with the first ELISA-positive samples after three-weeks, 12 cattle (0.8%) from five farms (29.4%) were positive for BVDV antigen, indicating these cattle were PI cattle. Genotypes of BVDV were determined with 12 BVDV-positive samples using a previously described RT-PCR assay and the results showed that 3 (25.0%) and 9 (75.0%) were confirmed to be type 1 and type 2, respectively. These results will be helpful to establish the effective control strategy for BVDV in cattle farms in Jeonbuk State.

한우 결핵의 신속 감별진단을 위한 등온증폭법 개발 (Development of loop-mediated isothermal amplification method for the rapid and sensitive detection of bovine tuberculosis in Korea native cattle)

  • Loop-mediated isothermal amplification (LAMP) was developed to detect Mycobacterium tuberculosis complex (MTC) and non-tuberculous mycobacterium (NTM) genomic DNA in blood samples of Korea native cattle. A set of four primers, two outer and two inner, were designed from M. bovis and M. avium genomic DNA targeting the IS6110 and 16S rRNA gene, respectively. Based on 85 Intradermal Tuberculin Test (ITT) positive blood sample and using conventional PCR and LAMP, the agreement quotient (kappa), which measures agreement beyond chance were 0.93 (conventional PCR) and 0.97 (LAMP), respectively. The detection limit of the LAMP method was $2.0{\times}10^2$ copy/ml M. bovis and M. avium cells, compared to $2.0{\times}10^3$ copy/ml M. bovis and M. avium cells for conventional PCR. These results suggest that the LAMP is a powerful tool for rapid, sensitive, and practical detection of MTC and NTM in blood samples of Korea native cattle.

Peri-estrus activity and mounting behavior and its application to estrus detection in Hanwoo (Korea Native Cattle)

  • This study was conducted to investigate the change in activity and mounting behavior in Hanwoo (Korean Native Cattle) during the peri-estrus period and its application to estrus detection. A total of 20 Hanwoo cows were fitted with a neck-collar accelerometer device, which measured the location and acceleration of cow movements and recorded the number of instances of mounting behavior by the altitude data. The data were analyzed in three periods (24-, 6-, and 2-h periods). Blood samples were collected for 5 days after the prostaglandin F2α (PGF2α) injection, and the concentrations of estradiol, progesterone, follicle-stimulating hormone, and luteinizing hormone were determined by enzyme-linked immunosorbent assays. Activity and mounting behavior recorded over 2-h periods significantly increased as estrus approached and were more efficient at detecting estrus than over 24- and 6-h periods (p < 0.05). Endocrine patterns did not differ with the variation of individual cows during the peri-estrus period (p > 0.05). Activity was selected as the best predictor through stepwise discriminant analysis. However, activity alone is not enough to detect estrus. We suggest that a combination of activity and mounting behavior may improve estrus detection efficiency in Hanwoo. Further research is necessary to validate the findings on a larger sample size.

Demographic Trends in Korean Native Cattle Explained Using Bovine SNP50 Beadchip

  • Linkage disequilibrium (LD) is the non-random association between the loci and it could give us a preliminary insight into the genetic history of the population. In the present study LD patterns and effective population size (Ne) of three Korean cattle breeds along with Chinese, Japanese and Mongolian cattle were compared using the bovine Illumina SNP50 panel. The effective population size (Ne) is the number of breeding individuals in a population and is particularly important as it determines the rate at which genetic variation is lost. The genotype data in our study comprised a total of 129 samples, varying from 4 to 39 samples. After quality control there were ~29,000 single nucleotide polymorphisms (SNPs) for which $r^2$ value was calculated. Average distance between SNP pairs was 1.14 Mb across all breeds. Average $r^2$ between adjacent SNP pairs ranged between was 0.1 for Yanbian to 0.3 for Qinchuan. Effective population size of the breeds based on $r^2$ varied from 16 in Hainan to 226 in Yanbian. Amongst the Korean native breeds effective population size of Brindle Hanwoo was the least with Ne = 59 and Brown Hanwoo was the highest with Ne = 83. The effective population size of the Korean cattle breeds has been decreasing alarmingly over the past generations. We suggest appropriate measures to be taken to prevent these local breeds in their native tracts.
