The purpose of this study is to assess the criteria on a floor hinge and door closer for the optimum design of the access door of a smoke control room. The door opening force due to differential pressure is 60.75 N, 40.5 N, 32.91 N and 12.66 N when the differential pressure is 60 Pa, 40 Pa, 32.5 Pa and 12.5 Pa, respectively. The door opening force of the floor hinge and door closer to which the criteria of KS F 2806 are applied is 27.5 N, 40 N, 75 N, 100 N and 125 N for the Nos. 1, 2, 3, 4 and 5 class floor hinges and door closers, respectively. This study compared the differential pressure and opening force limits of floor hinges and door closers with the values specified in NFSC 501A and found that they exceeded the criteria specified in NFSC 501A. Therefore, it is necessary to reflect the differential pressure and smoke control wind speeds as well as the opening forces specified in NFSC 501A on the design of floor hinges and door closers. The installation conditions of floor hinges and door closers of access doors differ depending on the type and name of a smoke control damper. This study found that Nos. 1, 2 and 3 floor hinges and door closers could be installed for access doors with low differential pressure and that Nos. 1 and 2 floor hinges and door closers could be installed for access doors with normal differential pressure.
To investigate if there are tandem repeats in iNOS gene in Korean genome we applied PCR amplification followed by DNA sequencing. Tandem repeats we were looking at were (AAAT)n in the promoter region. Totally, 65 people were subjected for this experiment. Twenty of them were patients with metabolic disease. Only $(AAAT)_4$ was found in all of these Korean samples. This result was somewhat different trom the data for Caucasians and other Asian people. So, we assume this is specific VNTR (variable number of tandem repeat) in Korean which can be used for the purpose of diagnosis and for the differentiation of ethnic groups.
Anti-inflammatory activity of the extracts of Hibiscus manihot was investigated through the evaluation of its inhibitory effect on the production of inflammatory biomarkers (i-NOS, COX-2) in RAW264.7 murine macrophage cells. Among the sequential solvent fractions (hexane, dichloromethane, ethyl acetate, n-butanol and water), the fractions of dichloromethane (1 ${\mu}g/ml$) and ethyl acetate (5 ${\mu}g/ml$) showed potential inhibitory activities on i-NOS and COX-2 activity in RAW264.7 cells. These results suggest that Hibiscus manihot might have an anti-inflammatory activity through the suppression of inflammatory markers.

Two phenolic flavonoids were isolated from the traditional medicine of Eastern Asia, Caesalpinia sappan L (Leguminosae). Brazilin (1) showed a significant inhibitory activity against inducible Nitric Oxide Synthase (iNOS) in lipopolysaccharides (LPS)-induced macrophage RAW 264.7 cells with an IC$\sub$50/ value of 1.68 mg/ml, which is more potent than the positive control, L-N$\^$6/-(1-iminoethyl)lysine (IC$\sub$50/ 3.49 mM). On the other hand, caesalpine J (2) was found to be inactive in the present iNOS assay system despite of their structural similarities. (omitted)
Stimulated alveolar macrophages and neutrophils produce nitric oxide, a free radical by an inducible nitric oxide synthase(iNOS), which reacts with superoxide anion to form peroxynitrite, a more highly reactive toxic species. The objectives of the present study were to evaluate acute inflammatory lung injury and to determine iNOS mRNA induction and nitric oxide production by rat broncho-alveolar lavage cells following intratracheal treatment of silica. After 4 h exposure to silica, differential counts of broncho-alveolar lavage cells and lactate dehydrogenase(LDH) activity as well as total protein in the broncho-alveolar lavage fluid were determined. Broncho-alveolar lavage cells were also assayed for iNOS mRNA and the productions of nitrite and nitrate measured in the cells cultured. Differential analysis of broncho-alveolar lavage cells showed that the number of alveolar macrophages slightly decreased following silica treatment; however, red blood cells, lymphocytes, and neutrophils significantly were increased by 9-, 14-, and 119-fold following silica treatment, respectively, compared with the saline control. It was also found significant increases in the LDH activity and total protein in the lavage fluid obtained from silica-treated rats, indicating silica-induced acute lung injury. Northern blot analysis demonstrated that the steady state levels of iNOS mRNA in broncho-alveolar lavage cells were increased following silica treatment. The productions of nitrite and nitrate in the cultured cells were significantly increased by 2-fold following silica treatment, respectively, which were attenuated by the NOS inhibitor $N{\omega}-nitro-L-arginine-methyl$ ester(L-NAME) and partially reversed by L-arginine. These findings suggest that nitric oxide production in alveolar macrophages and recruited neutrophils is increased in response to silica. Nitric oxide may contribute in part to acute inflammatory lung injury.
Diabetes mellitus is associated with vascular complications, including an impairment of vascular function and alterations in the reactivity of blood vessels to vasoactive substances in various vasculature. In the present study, the authors have observed endothelin-B ($ET_B$) receptor agonist-induced relaxation in precontracted mesenteric arterial segments from streptozotocin (STZ)-induced diabetic rats, which was not shown from control rats or in other arterial segments from diabetic rats. Accordingly, the goal of this study was to investigate in what way STZ-induced diabetes altered reactivity of the mesenteric arterial bed and to examine the causal relaxation, if any, between this $ET_B$ receptor-mediated relaxation and endothelial paracrine function, especially nitric oxide (NO) production. The relaxation induced by $ET_B$ agonists was not observed in mesenteric arteries without endothelium. The relaxation to $ET_B$ agonists was completely abolished by pretreatment with BQ788, but not by BQ610. $N_{\omega}-nitro-L-arginine$ methyl ester and soluble guanylate cyclase inhibitors, methylene blue or LY83583 significantly attenuated the relaxant responses to $ET_B$ agonists, respectively. When the expression of eNOS and iNOS was evaluated on agarose gel stained with ethidium bromide, the expression of eNOS mRNA in diabetic rats was significantly decreased, but the expression of iNOS was increased compared with control rats. Furthermore, the iNOS-like immunostaining was densely detected in the endothelium and slightly in the arterial smooth muscle of diabetic rats, but not in control rats. These observations suggest that $ET_B$ receptor may not play a role in maintaining mesenteric vascular tone in normal situation. However, the alterations in $ET_B$ receptor sensitivity were found in diabetic rats and lead to the $ET_B$ agonist-induced vasorelaxation, which is closely related to NO production. In the state of increased vascular resistance of diabetic mesenteric vascular bed, enhanced NO production by activation of iNOS could lead to compensatory vasorelaxation to modulate adequate perfusion pressure to splanchnic area.

The water extracts of Samul-tang(SMT) has been used for treatment of ischemic brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extracts of SMT rescues brain cells from ischemic damages. To elucidate the protective mechanism on ischemic induced cytotoxicity, I investigate the regulation of LPS and PMA induced iNOS expression in C6 glial cells. LPS and PMA treatment for 72 h in C6 glial cells markedly induce nitric oxide(NO), but treatment of the cells with the water extracts of SMT decrease. dose dependently nitrite formation. In addition, LPS and PMA treatment for 72 h induce severe cell death and LDH release in C6 glial cells. However treatment of the cells with the water extracts of SMT dose not induce significant changes compare to control cells. Furthermore, the protective effects of the water extracts of SMT is mimicked by treatment of $N^{G}MMA$, a specific inhibitor of NOS. LPS and PMA induced iNOS activation in C6 glial cells cause chromosomal condensation and fragmentation of nuclei by caspase activation. The treatment of the cells with the water extracts of SMT may suppress apoptosis via caspase inhibition by regulation of iNOS expression. Taken together, I suggest that the protective effects of the water extracts of SMT against ischemic brain damages may be mediated by regulation of iNOS during ischemic condition.
Agrimonia pilosa Ledeb. has long been used for a useful natural agent ameliorating inflammation related symptoms in the folk medicine recipe. This study was performed to investigate effects of Agrimonia pilosa Ledeb.(AP) on the expression of inflammation related genes such as the inducible nitric oxide synthase (iNOS) in macrophage cell line, RAW 264.7 cells. The AP (whole plants) was extracted with 80% ethanol and sequentially partitioned with solvents in order to increase polarity. Among the various solvent extracts of AP, the n-butanol (BuOH) fraction showed the most powerful inhibitory ability against nitric oxide (NO) production in lipopolysaccharide (LPS)-induced RAW 264.7 cells without affecting cell viability. Reverse transcriptase-polymerase chain reaction and Western blot analysis revealed that the BuOH fraction provided a primary inhibitor of the iNOS protein and mRNA expression in LPS-induced RAW 264.7 cells. The DPPH and OH radical scavenging activities of the several fractions of 80% ethanol extracts of AP significantly increased by EtOAC and BuOH fractions. Thus, the present study suggests that the response of a component of the BuOH fraction to NO generation via iNOS expression provide an important clue to elucidate anti-inflammatory mechanism of AP.

The fallowing are the result of the experimental studies of KH-304 on the penile nitric oxide synthase (NOS) activity, SOD and the level of blood testosterone in old rat. KH-304 was tested for the expression and activity of eNOS, nNOS, SOD/Mn, caveol in-1, 3 in penis of old-rat. The penile expression level of the five enzyme were increased significantly after oral administration of the KH-304 100mg/kg for 100ays and the concentration of testosterone in the blood were increased. In vitro, the effect of SOD and free radical scavenging were increased significantly. Conclusively, KH-304 is capable of improving of sexual ability in old-rat.

Through molecular cloning, five muscarinic receptors have been identified. The muscarinic receptors can be generally grouped according to their coupling to either stimulation of phospholipase C (m1, m3, and m5) or the inhibition of adenylate cyclase (m2 and m4). Each m1, m3, and m5 receptors has the additional potential to couple to the activation of phospholipase A$_2$, C, and D, tyrosine kinase, and the mobilization of Ca$\^$2+/. However, the differences in coupling efficiencies to different second messenger systems between these receptors have not been studied well. Ectopic expression of each of these receptors in mammalian cells has provided the opportunity to evaluate the signal transduction of each in some detail. In this work we compared the coupling efficiencies of the m1, m3 and m5 muscarinic receptors expressed in chinese hamster ovary (CHO) cells to the Ca$\^$2+/ mobilization and the stimulation of neuronal nitric oxide synthase (nNOS). Because G protein/PLC/PI turnover/[(Ca$\^$2+/])i/NOS pathway was supposed as a main pathway for the production of nitric oxide via muscarinic receptors, we studied on ml, m3 and m5 receptors. Stimulation of guanylate cyclase activity in detector neuroblastoma cells was used as an index of generation nitric oxide (NO) in CHO cells. The agonist carbachol increased the cGMP formation and the intracellular [Ca$\^$2+/] in concentration dependent manner in three types of receptors and the increased cGMP formation was significantly attenuated by scavenger of NO or inhibitor of NOS. m5 receptors was most efficiently coupled to stimulation of nNOS, And, the coupling efficiencies to the stimulation of neuronal nitric oxide synthase in three types of receptors were parallel with them to the Ca$\^$2+/ mobilization.
