Effects of Feral Haw (Crataegus pinnatifida BUNGE) Seed Extracts on the Antioxidant Activities (야생산사(Crataegus pinnatifida BUNGE)씨 생리활성물질의 산화억제 효과)

  • Haw (Crataegus pinnatifida $B_{UNGE}$) is a medicinal plant commonly used in Korea due to its beneficial health effects. This study was performed to investigate the antioxidative activity of bioactive components from haw seed. Freeze-dried seed was processed with various solvents such as 70% methanol, 70% ethanol, a mixture of chloroform and methanol (2:1, v / v), n-butanol and ethyl acetate (EA). DPPH, reducing power, ferric reducing antioxidant power (FRAP), and ABTS radical scavenging activity decreased in this order: 70% methanol, 70% ethanol, EA, CM, n-butanol extracts. NO radical scavenging activity was high in the n-butanol extract (36.73% in 0.8 mg/ml concentration) and low in the 70% ethanol extract (24.96% in 0.8 mg/ml concentration). $OH^-$ radical scavenging activity of n-butanol extract (40.42% in 0.8 mg/ml concentration) was higher than the other extracts, but EA extract was the lowest (13.39%) at the same concentration. The results show that each extract contained various antioxidative substances and it seemed that different influence at each antioxidative activity.

Antioxidative Effects of Campanula takesimana Nakai Extract (섬초롱(Campanula takesimana Nakai) 용매 추출물의 항산화 효과)

  • The study analyzed the total polyphenolic contents, DPPH radical scavenging activities, ABTS radical scavenging activities, reducing power levels, SOD-like activities, and tyrosinase inhibition activities of fractions from Campanula takesimana Nakai extract. Total polyphenolic content of the ethyl acetate fraction from Campanula takesimana Nakai extract was the highest. The ethyl acetate fraction of Campanula takesimana Nakai extract showed the lowest $IC_{50}$ value (0.13 mg/mL) as well as greater than 98% ABTS radical scavenging activity, which was similar to ascorbic acid. Further, reducing power was significantly higher in the ethyl acetate fraction. The SOD-like activity of edible plant was 70.56% (n-butanol fraction) and 62.27% (ethyl acetate fraction) at 1 mg/mL. Tyrosinase inhibition activity of Campanula takesimana Nakai extract was reduced to 34.77% in the n-butanol fraction and 30.85% in the ethyl acetate fraction at 5 mg/mL. These results suggest that the ethyl acetate and n-butanol fractions of Campanula takesimana Nakai extract could be applicable as food additives or cosmeceutical ingredients.

Oenanthe javanica extract accelerates ethanol metabolism in ethanol-treated animals

  • The effect of water dropwort (Oenanthe javanica DC) extract in eliminating ethanol was evaluated in New Zealand white rabbit and ICR mice. When a hot-water extract of water dropwort extract and ethanol was injected into New Zealand white rabbit, the plasma ethanol level was rapidly reduced, similar to metadoxine treatment. Specifically, the n-butanol fraction of hot-water extract was the strongest in eliminating plasma alcohol in ICR mice. When ethanol was orally ingested, administration of the hot-water extract eliminated up to 44% of the plasma ethanol in mice while the n-butanol fraction eliminated around 70%. Alcohol removal behaved in a dose-dependent manner in response to 50-200 mg/kg of n-butanol fraction. These data show O. javanica extract is effective in overcoming alcohol intoxication by the accelerating ethanol metabolism.

Effect of Garlic (Allium sativum L.) Extracts on Formation of N-Nitrosodimethylamine (마늘 추출물이 N-Nitrosodimethylamine의 생성에 미치는 영향)

  • This study was conducted to investigate the effect of garlic extracts on electron donating ability nitrite-scavenging activity and N-nitrosodimethylamine (NDMA) formation. Content of total phenolic compounds was the highest in ethyl acetate extract. Electron donating ability was significantly increased with increasing sample concentration, as $88.0{\pm}1.53%$ from ethyl acetate extract and $77.5{\pm}0.8%$ from butanol extract in concentration of 10 mg/mL. Butanol extract from garlic with highest nitrite scavenging activity and inhibition effect of NDMA formation exhibited $96.9{\pm}0.75%$ and $68.6{\pm}1.15%$ in concentration of 10 mg/mL, respectively. The butanol extract was fractionated into 5 fractions using prep-LC. Nitrite-scavenging activity and inhibition effect of NDMA formation were excellent in fraction III, which further fractioned into 3 subfractions (III-a, III-b and III-c) using prep-LC. Of the 3 subfractions, its activity was the highest in the subfraction III-c. Therefore butanol extract of garlic was suggested to be effective inhibitor of NDMA formation.

Impacts of Various Solvent Extracts from Wild Haw(Crataegus pinnatifida Bunge) Pulpy on the Antioxidative Activities (야생산사(Crataegus pinnatifida Bunge)과육 용매별 추출물의 항산화 효과)

  • Haw(Crataegus pinnatifida Bunge) belongs to the rose family and has beneficial health effects such as anti-obesity and gastroprotective activities. This study compared the antioxidative activities of various solvent extracts from haw. After removing seeds, pulpy was extracted with 70% methanol, 70% ethanol, chloroform :methanol(CM, 2:1, v/v), n-butanol, and ethyl acetate(EA). Total phenol contents were 71.46, 70.41, 47.63, 47.49 and 45.95 mg/g in the 70% methanol, 70% ethanol, CM, n-butanol and EA extracts, respectively. Total flavonoid contents of CM and n-butanol extracts were higher compared to other extracts at 1.63 mg/g and 1.59 mg/g, respectively, whereas EA extract had the lowest flavonoid content at 1.12 mg/g. Corresponding to total phenol contents, among $NO_2$ radical scavenging activity, antioxidant activity by ${\beta}$-carotene bleaching assay, and superoxide dismutase(SOD)-like ability, 70% methanol and 70% ethanol extracts showed higher antioxidative activities than the other extracts. However, the lowest activities were n-butanol extract in $NO_2$ radical scavenging activity as well as EA extract in antioxidant activity by ${\beta}$-carotene bleaching assay and SOD-like ability. These results suggest that haw extracts are available as a new natural antioxidant, and its activities are attributed to antioxidant substances such as phenolic compounds and flavonoids.

Effects of Godulbaegi Leaf Extracts on $CCl_4$-Induced Hepatotoxicity in Rats (고들빼기 잎추출물이 흰쥐의 사염화탄소에 의한 간손상에 미치는 영향)

  • This study was carried out to investigate the effect of godulbaegi leaf extracts on detoxication of liver injury in $CCl_4$-treated rats. We divided normal group(5) and $CCl_4$-treated group and then $CCl_4$- treated group was divided into 6 groups; only $CCl_4$-treated group(CS), aqueous extract pretreated group (CSA), n-butanol extract pretreated group(CSB), Tween 80 pretreated group(CT), n-hexane extract pretreated group(CTH) and ethyl acetate extract pretreated group(CTE). The ratio of liver weight per body weight and the activity of GPT in hexane extract group(CTH) were lowest, similar to the results of S. Godulbaegi n-hexane extract(IS) groups intoxicated by $CCl_4$ had lower values of MDA than CS and CT which are control groups. Histological finding of liver tissue revealed less of necrosis in IS extracts groups than in control groups(CS, CT). From these results, IS extracts could predominently prevent hepat-otoxicity of rats. Especially, hexane extract was effective on the detoxication of liver injury among the other extracts.

  • This study was carried out to develop antitumor effect of the n-butanol soluble fraction of Perilla frutescens on (KB cells) human oral epitheloid carcinoma cells. The cytotoxictty of methanollc extract of Perilla frutescens on KB cells was evaluated by 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide(MTT) assay. The antitumor activity of various fractions obtained from n-butanol soluble fraction of Perilla frutescens was evaluated in human oral epithelold carcinoma cells. The antitumor acavity of the n-butanol soluble fraction on human oral epitheloid carcinoma cells was evaluated by MTT assay of colorimetric method. The light microscopic study was carried out to observe morphological changes of cultured human oral epitheloid carcinoma cells. These results were obtained as follows; 1. The fractions 1,2 and 3 of the n-butanol soluble fraction of Perilla frutescens were shown significant antitumor activities. 2. The number of human oral epitheloid carcinoma cells were decreased and tend to form cell cluster by treatment with fractions 1,2,3 and 4 of the n-butanol soluble fraction of Perilla frutescens. 3. The fraction 1 of the n-butanol soluble fraction of Perllla frutescens showed the highest antitumor activity on Perilla frutescens. It has been selected as a lead fraction for further examinations.

Antimicrobial Activities of Organic Extracts from Fruit of Thuja orientalis L. (측백나무 열매 추출물의 항균활성)

  • This study was carried out to investigate the antimicrobial activities of organic extracts obtained from the fruit of Thuja orientalis L. The native fruits in Korea were collected and extracted by 80% ethanol, and the extract was sequentially fractionated with n-hexane, chloroform, ethylacetate, and butanol. The fraction yields of n-hexane, chloroform, ethylacetate, butanol and water of ethanol extract were 10.15%, 10.05%, 1.45%, 45.35% and 27.55%, respectively. n-Hexane-soluble fraction showed the highest antibacterial activity against gram positive bacteria, while the chloroform, ethylacetate, butanol and aqueous fractions did not show any antibacterial activity. Minimum inhibitory concentration (MICs) on Staphyloycoccus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction were $100\;{\mu}g$, $500\;{\mu}g$ and $50\;{\mu}g$/disc, respectively. The antibacterial activity was not destroyed by heating at 80, 100, $120^{\circ}C$ for 30 min and was not affected by pH. In the inhibitory test against the Staphylococcus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction showed potent growth inhibition at the concentration of 0.1 and $0.5\;{\mu}g/mL$ for 12~24 hours and n-hexane-soluble fraction did not show any mutagenic activity.

Antioxidative and Physiological Activities of Fractions from Pleurospermum kamtschaticumin Extracts (누룩치(Pleurospermum kamtschaticumin) 추출물의 항산화 효과 및 생리활성)

  • This study investigated the antioxidant activities, whitening effects, and antimicrobial activities of fractions from Pleurospermum kamtschaticumin extract. Total polyphenolic contents of fractions from Pleurospermum kamtschaticumin extract were 116.44~382.73 mg/g GAE (gallic acid equivalent), with the highest value in the ethyl acetate fraction. Fractions of Pleurospermum kamtschaticumin showed the highest DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity ($IC_{50}$=0.04 mg/mL) and ABTS radical scavenging activity (98.86% at 0.5 mg/mL), which was similar to ascorbic acid. Further, reducing power and FRAP activity were significantly high in the ethyl acetate and n-butanol fractions. The ethyl acetate and n-butanol fractions showed significantly high SOD-like activities (1 mg/mL, 86.93%, and 78.23%, respectively) compared to other fractions. Tyrosinase inhibition activities of the n-butanol fraction and 80% ethanol extract were 43.52% and 41.64%, respectively. Antimicrobial activity against Escherichia coli was only observed in the chloroform fraction (16.7 mm in 0.5 mg/disc). These results suggest that fractions from Pleurospermum kamtschaticumin extract show comparatively high antioxidant activity and thus could be used as a food additive or cosmeceutical ingredient.

Antioxidant Activities of Bamboo (Sasa Borealis) Leaf Extract according to Extraction Solvent (추출용매에 따른 조릿대 잎 추출물의 항산화활성)

  • This study was carried out to investigate the antioxidant activity of bamboo (Sasa borealis) leaf extract by measuring electron donating ability, superoxide dismutase (SOD)-like activity, reducing power, and lipid peroxidation inhibitory activity. Two crude extracts by water or 70% EtOH and five fractions of n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous from the crude extract of 70% EtOH were prepared for this study. The crude extracts of water and 70% EtOH yielded 8.5% and 11.4%, respectively and the yields of n-hexane, chloroform, ethyl acetate, n-butanol, and aqueous fractions were 5.1% to 0.6%. Total polyphenol contents of the water and the 70% EtOH crude extracts were not significantly different; however, their total flavonoid contents were significantly greater in the 70% EtOH than in the water crude extract. Total polyphenol contents were the highest in chloroform fraction followed by ethyl acetate and n-butanol fractions and total flavonoid contents were the highest in ethyl acetate fraction followed by chloroform and n-hexane fractions. The two crude extracts as well as the five fractions showed election donating ability, SOD-like ability, reducing power, and lipid peroxidation inhibitory activity. Most of the antioxidant activities of each crude extract or fractions increased proportionally with the concentration. These results indicate that bamboo (Sasa borealis) leaf extracts show antioxidant activities due to its substantial content of polyphenol including flavonoid. Thus, it could be concluded that crude extracts by water or 70% EtOH and the fractions from the 70% EtOH extract, especially chloroform, ethyl acetate and n-butanol, would be useful as natural antioxidant substances.
