Phellinus linteus was artificially cultivated in kangwon province in Korea. The air-dried phellinus linteus was frozen in liquid nitrogen tank and powdered in jar. 10g of the powder was extracted with each 200g of ethanol, methanol, distilled water and 1,3-butylene glycol/distilled water 4 hours under refluxing and then the liquidextract was concentrated under reduced pressure. As a result of analysis by high performance liquid chromatography and thin layer chromarography, many kinds of sugar and flavonoids were detected. Also we knew that phellinus linteus' extract had a strong UV-ray absorption. In the efficacy test for applying to cosmetics, free radical scavenging effect was confirmed. As a result, 2% of sample was the most potent inhibitory effect and the free radical savenging activity, was 0.31%. This is more effective than any other meterial. In the test of antioxidative activity against lipid autoxidation, phellinus linteus' extract had a good effect by 46% while vitamine E was 42.3%. The immunological activity of phellinus linteus was showed through the activation of macrophage cell. Actually, phellinus linteus activated macrophage function of 1.1-1.8 times including nitrite production compared to control. The whitening effect of phellinus linteus was showed through the inhibition of tyrosinase activity, melanin biosynthesis of S. bikiniensis and B-16 melanoma cells. Phellinus linteus' extract was showed strong mushroom tyrosinase inhibitory activity with IC50 value of 0.5% and inhibited melanin biosynthesis with 28mm inhibition zone at 0.005%/paper disc in S. bikinniensis, a bacterium used as an indicator organism in this work. Also it inhibited melanin biosynthesis in B-16 melanoma cells with a minimum inhibitory concentration of 0.134%.

Plants extracts are good resources to find functional compounds for human health. The following eight plants were collected and total phenolic contents were determined. Acer psedo-siebolianum showed the highest phenolic contents, 16.4 mg/g, whereas Cercidiphyllum japonica showed the lowest contents, 1.9 mg/g. The DPPH free radical scavenging capacities of the plant extracts showed high activity in following order : Acer ginnala ($21.3\;{\mu}g/mL$) > Cornus walteri ($23.9\;{\mu}g/mL$) > Distylum racemosum ($29.2\;{\mu}g/mL$) > Castanopsis cuspidata var. Thunbergii ($31.7\;{\mu}g/mL$) > Acer psedo-siebolianum ($34.6\;{\mu}g/mL$) > Thuijopsis dolabrata cv. Aurea ($53.1\;{\mu}g/mL$) > Cercidiphyllum Japonica ($115.2\;{\mu}g/mL$). Also the mushroom tyrosinase inhibitory activities of total extracts were determined at different concentration. D. racemosum extract showed highest (49.1% at 1,000 mg) in inhibitory activity than other seven extracts. The ethanol fraction $IC_{50}$ value: $118.1\;{\mu}g/mL$) from D. racemosum showed more inhibitory activity than ethyl acetate fraction ($IC_{50}$ value: $203\;{\mu}g/mL$). The ethanol fraction on showed no significant cytotoxicity in B16/F1 cells line up to $60\;{\mu}g/mL$. Over $80\;{\mu}g/mL$ of ethanol fraction showed cytotoxicity in B16/F1 cells. The melanin contents of cells were significantly attenuated by ethanol fraction in a dose-dependent manner. The $IC_{50}$ value of ethanol fraction was $75.4\;{\mu}g/mL$.
Bioactive substances, antioxidant activities and tyrosinase inhibitory activities of seed extracts were evaluated to discover new functional materials, using 13 edible or medicinal plants. Total polyphenol and flavonoid contents were highest in Taxus cuspidata, with $57.51mg{\cdot}g^{-1}\;and\;7.98mg{\cdot}g^{-1}$, respectively. Seed extract of Vitis coignetiae${\times}$Vitis labruscana($RC_{30}=0.030mg{\cdot}ml^{-1}$) was found to be the most effective in DPPH radical scavenging test, and the highest ABTS radical scavenging activity was shown in Diospyros lotus($RC_{30}=0.044mg{\cdot}ml^{-1}$). Inhibitory effects on peroxidation of linoleic acid determined by ferric thiocyanate (FTC) method were similar to BHT in Diospyros kaki, Diospyros lotus, Magnolia officinalis, Styrax obassia, Vitis coignetiae, Vitis coignetiae${\times}$Vistis labruscana, Zizyphus jujuba, Zizyphus jujuba var. inermis. For the inhibition against mushroom tyrosinase, Diospyros kaki, Diospyros lotus, Poncirus tritoliata, Prunus serrulata var. spomtanea, Zizyphus jujuba and Zizyphus jujuba var. inermis exhibited inhibitory activity, and especially Diospyros lotus showed the strongest inhibition.

Purpose: This study was performed to determine the inhibitory effect of Soyosangagamhwajae(SYG) on melanin synthesis in B16F10 mouse melanoma cell. Methods: The Inhibitory effects of Soyosangagamhwajae(SYG) on melanin synthesis were determined by in-vitro assay. To elucidate inhibitory effects of SYG on melanin synthesis, we determined the melanin release in B16F10 cell. And to investigate the action mechanism, we assessed the gene expression of tyrosinase, TRP-1, TRP-2. PKA, $PKC{\beta}$ in B16F10 cell. Results: 1. SYG significantly inhibited melanin-release in B16F10 cell. 2. SYG significantly inhibited mushroom tyrosinase activity in vitro. 3. SYG significantly suppressed the expression of tyrosinase in B16F10 cell. 4. SYG significantly suppressed the expression of TRP-1, TRP-2 in B16F10 cell. 5. SYG significantly suppressed the expression of PKA, $PKC{\beta}$ in B16F10 cell. Conclusion: From these results, it may be concluded that SYG has the antimelanogenetic effect.

Purpose: This study was performed to determine the inhibitory effect of Sihosogansangagambang (SS) on melanin synthesis in B16F10 melanoma cells (B16F10). Methods: The inhibitory effects of Sihosogansangagambang on melanin synthesis were used by in vitro assay. To elucidate inhibitory effects of SS on melanin synthesis, we determined the melanin release in B16F10. And to investigate the mechanism of inhibitory effect of SS, we assessed the gene expression of tyrosinase, TRP-1, TRP-2 and ERK-1 in B16F10. Results: 1. SS decreased the release of melanin in B16F10 melanoma cells. 2. SS inhibited mushroom tyrosinase activity in vitro. 3. SS decreased the expression of tyrosinase, TRP-2 in B16F10 melanoma cells, but did not decreased the expression of TRP-1 in B16F10 melanoma cells. 4. SS decreased the expression of ERK-1 in B16F10 melanoma cells. Conclusion: From these results, it may be suggested that SS is possesed of the antimelanogenetic effects.

Studies on seaweed-based materials have been progressing steadily day by day. In this experiment, we checked the anti-oxidant, whitening, and moisturizing activities of fermented extract from a mixture of Undariapinnatifida, Saccharina japonica, and Gloiopeltis furcate. Also, Lactobacillus sakei strains of kimchi were used as the lactic acid bacteria. The physiological status of the combined seaweed extracts was also investigated. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging results showed that the inhibitory effects of the combined seaweed extracts were higher than the positive control. Furthermore, 3,4-dihydroxy-L-phenylalanine (L-DOPA) and Mushroom tyrosinase tests were conducted during the whitening efficacy experiment. Hence, it was confirmed that the whitening activity of fermented extracts was greater than the extracts without fermentation. HPLC analysis of fucose (an active ingredient of seaweed) was also performed and a standard method for solvent conditions was newly established. This study suggests that the composite of algae extract has potentials to be used as anti-oxidant and whitening agents in cosmetics.

Kojic acid is well known for its anti-pigmentation effect with tyrosinase inhibition activity. However, kojic acid is a unstable compound. In order to improve stability, kojic acid derivative, kojic acid $6-O-2',3',4',6'-tetraacetyl-{\beta}-D-glucopy-ranoside\;(KTGP)$, was synthesized with $O-pentaacetyl-{\beta}-D-glucose$ through the regio- and stereo-selective glycosylation of 6-OH group of kojic acid. High yield $(80\%)$ was obtained by the use of Lewis acid and organic base in nonpolar solvent. Hydrolysis of KTGP with the aid of sodium methoxide in methanol afforded kojic acid $6-O-{\beta}-D-glucopyranoside$ (KGP), which was confirmed by $^1H-NMR\;and\;^{13}C-NMR$ KGP is freely soluble in water and soluble in methanol and ethanol. Inhibition activity of KGP for tyrosinase was investigated by measuring the activity of mushroom tyrosinase compared with those of ascorbic acid, kojic acid, and arbutin. The free radical-scavenger activity was determined by the 1,1-diphenyl- 2-picrylhydrazyl (DPPH) assay. In toxicity assay, KGP was much less toxic than kojic acid and arbutin, Therefore, glycosylation of kojic acid may be useful for the development of stable kojic acid derivatives.

Hot water extracts from 16 domestic edible mushrooms including Pleurotus ferulae (Lanzi) X.L. Mao, Lentinula edodes (Berk.) Pegler, and Hypsizygus marmoreus (Peck) H.E. Bigelow, which are commercially available, were used for determining the cosmetic potential of these mushrooms. In this study, we carried out in vitro functional experiments to determine the inhibitory effects of these extracts on L-DOPA oxidation of tyrosinase and melanin synthesis. Based on the results of these experiments, H. marmoreus (Peck) H.E. Bigelow No. 10 and No. 15 were selected for further analysis. We analyzed the melanin synthesis inhibitory activity, TRP1 and MITF expression via real-time PCR, and Fontana Masson staining in artificial skin $Neoderm^{(R)}-ME$. Taken together, we observed that the hot water extract from H. marmoreus (Peck) H.E. Bigelow (No. 15) had better whitening effect than the extracts of other mushrooms. Thus, it can be a potential source of skin-whitening agent.
During the screening of inhibitors of melanin biosynthesis from microbial secondary metabolites, a fungal strain MR304 which was capable of producing high level of an inhibitor was selected. Based on taxonomic studies, this fungus could be classified as Trichoderma harzianum. The active compound (MR304-1) was purified from culture broth by Diaion HP-20 column chromatography, ethylacetate extraction, Sephadex LH-20 column chromatographv and HPLC. The inhibitor was identified as 3-(1,5-dihvdroxy-3-isocyanocyclopent-(E)-3-envl)prop-2-enoate by spectroscopic methods of UV, ESIMS, $^{1}$H-NMR, $^{13}$C-NMR, NOE, HMQC and HMBC. MR304-1 showed strong mushroom tyrosinase inhibitory activity with IC$_{50}$ value of 0.25 $\mu $g/ml. It inhibited melanin biosynthesis with 15 mm inhibition zone at 30 $\mu $g/paper disc in Streptomyces bikiniensis, a bacterium used as an indicator organism in this work. It also inhibited melanin biosynthesis in B16 melanoma cells with a niinimum inhibitory concentration of 0.05 $\mu $g/ml.

In this study, Camellia sinensis Leaf cultivated in Boseong was extracted by vacuum distillation and hydrothermal extraction methods, and we performed in vitro experiments such as antioxidant, anti-wrinkle, whitening, and antibacterial activity test and HPLC analysis. Wrinkle improvement (elastase inhibition assay) and whitening effect (mushroom tyrosinase inhibition assay) of Boseong vacuum distilled extract have excellent results and IC50 values of vacuum distillation extract was measured at a lower concentration than that of the hydrothermal extract. The antioxidant activity of the Boseong hydrothermal extract was excellently measured in DPPH radical and ABTS+ radical scavenging activity, SOD-like activity, and total polyphenol content. In the antibacterial experiment, the Boseong hydrothermal extract formed a clear zone for the Staphylococcus aureus and Cutibacterium acnes strain. As a result of HPLC analysis, EGCG and caffeine in the hydrothermal extract were 40.29 mg/g, 9.94 mg/g respectively, and caffeine in the vacuum distilled extract was 3.40 mg/g. Therefore, we examined that Boseong Camellia sinensis vacuum distilled extract has excellent anti-wrinkle and whitening effect, and its hydrothermal extract has good antioxidant and antibacterial efficacy.
