Antioxidant and tyrosinase inhibitory activity of Hypsizygus marmoreus (brown cultivar) methanol extracts (갈색 느티만가닥버섯 메탄올 추출물의 항산화 및 tyrosinase 저해 효과)

  • The objective of this study was to evaluate antioxidant effect and tyrosinase inhibitory activity of methanol extracts from Hypsizygus marmoreus. The Hypsizygus marmoreus was divided into two parts (pileus and stipe) and extracted with methanol. Total polyphenolics and flavonoids in the methanol extracts were measured by spectrophotometric methods and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities have been determined for antioxidant activities. The total polyphenolics and flavonoids contents of methanol extract of the pileus were higher than methanol extract of the stipes. The total polyphenolics contents in methanol extracts of the pileus and stipes were 8.7 ug/mg and 5.6 ug/mg, respectively. The total flavonoids contents in methanol extracts of the pileus and stipes were 2.8 ug/mg and 1.4 ug/mg, respectively. The tyrosinase inhibitory activity was proportional to concentration of methanol extract. The tyrosinase inhibitory activity of the methanol extract (200 mg/ml) of pileus (66.9%) and stipe (57.97%) was lower than those of positive control 2% arbutin. The DPPH radical scavenging activity of the methanol extract (20 mg/ml) of pileus and stipes was 52.55% and 30.35%, respectively. Moreover, the effects of methanol extarcts on cell proliferation of B16BL6 mouse melanoma cells were investigated using WST-1 assay (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate) and B16BL6 mouse melanoma cells treated with methanol extract of 200-2,000 ug/ml were higher proliferation rate than those of 0.04% adenosine.

  • During the screening for inhibitors of melanin biosynthesis from plant extract, Angelica polymorpha MAXIM which showed a high level of inhibition was selected. The inhibiting substances were purified form methanol extract of Angelica polymorpha MAXIM followed by silica gel column chromatography and HPLC. The inhibitors were identified as heraclenin, isosaxalin and heraclenol 3'-Me ether, by spectrescopic methods of ESI-MS, H-NMR, C-NMR, DEPT, HMQC and HMBC. These compounds did not have mushroom tyrosinase inhibitory activity, but showed a highly potent melanin biosynthesis inhibition zone in the plate culture of Streptomyces bikiniensis, a bacterium used as an indicator organism in this work. These compounds did not show any growth inhibition against S. bikiniensis at the same concentration of melanin biosynthesis test.

  • A kojic acid derivative, kojic acid 7-O-$\beta$-D-tetraacetylglucopyranoside(KTG) was synthesized. Regio-and stereo-selective glycosylation at 7-postion in kojic acid with $\beta$-D-pentaacetylglucose was achieved with high yield(80%) by the use of Lewis acid and organic base in nonpolar solvent. KTG was hydrolyzed in methanol by the aid of sodium methoxide to give kojic acid 7-O-$\beta$-D-glucopyranoside(KGP). KGP is freely soluble in water and soluble in methanol and ethanol. Its structure was comfirmed by $^1$H-NMR and $^{13}$ C-NMR. Tyrosinase activity inhibition of KGP was measured with mushroom tyrosinase compared with ascorbic acid, kojic acid and arbutin. KGP showed higher tyrosinase inhibition activity($IC_{50}$/=33.3 $\mu\textrm{g}$/ml) than ascorbic acid(63.2 $\mu\textrm{g}$/ml) and arbutin(91.8 $\mu\textrm{g}$/ml) but lower inhibition activity than kojic acid(8.3 $\mu\textrm{g}$/ml). To test free-radical scavenging activity, we used 1, 1-diphenyl-2-picrylhydrazyl(DPPH) as a free-radical source. Free-radical scavenging activity of KGP was very low($SC_{50}$/>1000 $\mu\textrm{g}$/ml) compared with ascorbic acid($SC_{50}$/=2.68 $\mu\textrm{g}$/ml) and arbutin($SC_{50}$/=180$\mu\textrm{g}$/ml). Melanin formation inhibition of KGP was measured in B16 melanoma, compared with kojic acid, arbutin and Vitamin C. Inhibition activity of KGP for melanin formation was not found within test concentrations.

A Study on Whitening and Anti-inflammatory Effects of Eriobotrya Japonica Leaf Extracts with Different Extraction Methods (추출 방법에 따른 비파엽 추출물의 미백 및 항염활성에 관한 연구)

  • In this study, we prepared Eriobotrya japonica leaf extracts by several extraction processes and then evaluated their biological activities for their potential application as a new raw material of functional cosmetic. Their whitening effects were measured by tyrosinase inhibitory activities, and anti-inflammatory effects were determined by inhibitory activities of nitric oxide (NO) production. Among the several extracts obtained from E. japonica leaf, supercritical fluid extract showed tyrosinase inhibitory activities at the concentration of 10%. Inhibitory activity on NO production effect related to anti-inflammatory efficacy was in the order: supercritical fluid extract > ethanol extract > hot water extract. According to the results of MTT assay, cell cytotoxicity was not observed at all concentrations except for a 5% concentration of the 70% ethanol extract. For whitening effects, 30% ethanol and 70% ethanol extract showed mushroom tyrosinase inhibitory activity at the concentration of 5%. These results indicated that E. japonica leaf extracts could have the functional effects when they are added as ingredients in cosmetics.

Characteristics of the Fermentation products of Paecilomyces tenuipes Fermented Using Different Microorganisms (발효 미생물에 따른 누에동충하초 발효산물의 특성)

  • In order to develop fermented silkworm "Dongchunghacho" (Paecilomyces tenuipes) with improved absorption and increased effectiveness, we fermented Dongchunghacho using four kinds of microorganisms, viz., lactic acid bacteria, Bacillus subtilis, Natto bacillus, and yeast. A total of 15 samples were fermented using a combination of microbial inoculation culture and conditions to produce fermentation products. The contents of basic components such as sugar, reducing sugar, protein, total polyphenol, and total flavonoid were examined as well as the antioxidant, tyrosinase inhibitory, and thrombolytic activities of the fermented products were analyzed. We observed that reducing sugar and protein contents decreased in most of the fermented products, but the products fermented using yeast exhibited higher sugar content and, thus, higher sweetness. Total polyphenol content and antioxidant activity did not increase in fermented products compared to non-fermented Dongchunghachos, but total flavonoid content and tyrosinase inhibitory and thrombolytic activities increased by fermentation. In particular, total flavonoid content and tyrosinase inhibitory and thrombolytic activities primarily increased in the products fermented using yeast and lactic acid bacteria. However, it was not possible to confirm the increase in these activities in samples fermented by single fermentation using only yeast. Therefore, we propose that it will be possible to develop fermented food from silkworm Dongchunghacho (P. tenuipes) with excellent health benefits through additional study of multiple fermentation conditions using lactic acid bacteria and yeast.

Studies on antioxidant, anti-inflammatory and tyrosinase inhibitory activity of Ganoderma lucidum fermented Artemisia capillaris extract (인진쑥 영지버섯 균사 발효 추출물의 항산화, 항염 및 티로시나제억제 활성 연구)

  • This study investigated whether Ganoderma lucidum (Y2)-mediated fermentation of Artemisia capillaris extract (ACE) could synergistically enhance its antioxidant, anti-inflammatory, and tyrosinase-inhibiting activities. Both G. lucidum extract and fermented ACE exhibited 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability, but with poorer efficacy than ACE (even at a low ACE concentration). Viability of RAW264.7 macrophages was significantly reduced in the presence of ACE (150 mg/mL and above). However, this effect was greatly mitigated upon G. lucidum-mediated ACE fermentation. Additionally, relative to the same concentration ($25{\mu}g/mL$) of G. lucidum mycelial extract, ACE exhibited an improved ability to significantly inhibit RAW264.7 macrophage nitric oxide (NO) production. Finally, relative to the same concentration ($200{\mu}g/mL$) of a positive control (arbutin), fermented ACE exhibited an approximately 3.66 times higher capacity for tyrosinase inhibition. These results suggest that G. lucidum-fermented ACE possesses enhanced tyrosinase-inhibiting activity and may be of utility as a skin-lightening agent.

Whitening and Antioxidative Effect of Rice Bran Fermented by Bacillus subtilis (발효된 미강의 항산화 효과 및 미백 효과)

  • In this study, we investigated rice bran fermented by Bacillus subtilis (RFB) at $40^{\circ}C$ for 36 h to develop a new natural antioxidant and whitening agent for new natural cosmetics. RFB showed reactive oxygen species (ROS) scavenging activities in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide radical scavenging activities and hydroxyl radical scavenging activities at a concentration of $100 \;{\mu}L$/mL. DPPH, superoxide radical scavenging activities and hydroxyl radical scavenging activities were higher in the RFB than in the RB. Inhibitory activity on auto-oxidation of linoleic acid is also highest at a concentration of $100 \;{\mu}L$/mL. RFB showed the higher inhibitory activity than RB in auto-oxidation. RFB reduced intracellular tyrosinase activity about 80 % at a concentration of $100 \;{\mu}L$/mL. Therefore, we suggest that RFB could be used as a useful antioxidant and whitening agent.

Preparation of Natural Compounds-tripeptides Derivatives and Their Melanogensis Inhibitory Activity (천연유래물질 펩타이드 유도체의 제조와 멜라닌 생성 저해 효과 탐색)

  • Derivatives of a novel natural compunds, melanostatin (PLG-$NH_2$) were prepared by solid phase synthesis[1,2] and assayed to evaluate their melanogensis inhibitory activity. Also, a small library (natural compound-XLG-$NH_2$, natural compound-X LG-OH) was prepared with same method for increasing synthetic yield and cost-reduction. PLG-$NH_2$ (Proline-Leucine-Glycine-$NH_2$) was well-known tripeptide as its $\alpha$-MSH release-inhibiting activity and tyrosinase inhibitory activity[3-5]. In order to choose best candidate for peptide derivatization, various natural compounds were screened by their tyrosinase inhibitory activity. As a result, caffeic acid and coumaric acid were selected. Most of these derivatives showed better activities than the parent natural compound, melanostatin.

Physiological Characteristics and Tyrosinase Inhibitory Activity of Lactobacillus plantarum M23 Isolated from Raw Milk (원유에서 분리한 Lactobacillus plantarum M23의 Tyrosinase 활성 저해 및 생리적 특성조사)

  • In order to develop a new starter for fermented milk, Lactobacillus plantarum M23 was isolated from raw milk and investigated for physiological characteristics. It showed good tyrosinase inhibitory activity compared with commercial lactic acid bacteria. The optimum growth temperature of Lactobacillus plantarum M23 was $40^{\circ}C$ and cultures took 17 hr to reach pH 4.3. Lactobacillus plantarum M23 showed more sensitivity to Penicillin-G, Oxacillin, Novobiocin, Chloramphenicol in a comparison of 12 different antibiotics, and showed most resistance to Vancomycin. It showed higher leucine arylamidase and ${\beta}-galactosidase$ activities compared to 16 other enzymes. It was comparatively tolerant to bile juice and able to survive at pH 2 for 3 hours. It showed high resistance to Escherichia coli, Salmonella typhimurium and Staphylococcus aureus with rates of 77.8%, 86.5% and 83.8%, respectively. Based on these and previous results, Lactobacillus plantarum M23 could be an excellent starter culture for fermented milk with high resistance to melanin.

Analysis of nutritional compositions and biological activity of Agrocybe aegerita (버들송이(Agrocybe aegerita) 영양성분 및 생리활성 분석)

  • The mushroom Agrocybe aegerita was analyzed to evaluate the nutritional value of this potential food and to enhance the cultivation and consumption. Among the nutritional compositions of Agrocybe aegerita, contents of crude proteins, crude fats and ashes were 38.3%, 3.0% and 8.2% respectively. The contents of potassium and phosphorus were higher than that of other minerals. Total phenolics contents were $65.2{\mu}g/m{\ell}$ and $46.0{\mu}g/m{\ell}$ in water extract and ethanol extract, respectively. Total flavonoids were estimated as $12.5{\mu}g/m{\ell}$ contents of water extract and ethanol extract were $7.1{\mu}g/m{\ell}$. Free radical scavenging activity against DPPH of ethanol extract, 79.2% was higher than that of water extract(58.2%). The contents of total phenolics, flavonoids and antioxidant activity by the DPPH scavenging activity of Agrocybe aegerita were higher than those of Pleurotus eryngii and Pleurotus ostreatus. The tyrosinase inhibitory activity showed 63.2% and 65.0% in water extract and ethanol extract, respectively. Angiotensin converting enzyme inhibitory activity of water extract was 78.7% and 75.0% in ethanol extract. In this study, Agrocybe aegerita has abundant essential nutrients and thus is good source of functional health food.
