Tumor necrosis factor-inducible gene 6 interacts with CD44, which is involved in fate-change of hepatic stellate cells

  • Tumor necrosis factor-inducible gene 6 protein (TSG-6) is a cytokine secreted by mesenchymal stem cells (MSCs) and regulates MSC stemness. We previously reported that TSG-6 changes primary human hepatic stellate cells (pHSCs) into stem-like cells by activating yes-associated protein-1 (YAP-1). However, the molecular mechanism behind the reprogramming action of TSG-6 in pHSCs remains unknown. Cluster of differentiation 44 (CD44) is a transmembrane protein that has multiple functions depending on the ligand it is binding, and it is involved in various signaling pathways, including the Wnt/β-catenin pathway. Given that β-catenin influences stemness and acts downstream of CD44, we hypothesized that TSG-6 interacts with the CD44 receptor and stimulates β-catenin to activate YAP-1 during TSG-6-mediated transdifferentiation of HSCs. Immunoprecipitation assays showed the interaction of TSG-6 with CD44, and immunofluorescence staining analyses revealed the colocalization of TSG-6 and CD44 at the plasma membrane of TSG-6-treated pHSCs. In addition, TSG-6 treatment upregulated the inactive form of phosphorylated glycogen synthase kinase (GSK)-3β, which is a negative regulator of β-catenin, and promoted nuclear accumulation of active/nonphosphorylated β-catenin, eventually leading to the activation of YAP-1. However, CD44 suppression in pHSCs following CD44 siRNA treatment blocked the activation of β-catenin and YAP-1, which inhibited the transition of TSG-6-treated HSCs into stem-like cells. Therefore, these findings demonstrate that TSG-6 interacts with CD44 and activates β-catenin and YAP-1 during the conversion of TSG-6-treated pHSCs into stem-like cells, suggesting that this novel pathway is an effective therapeutic target for controlling liver disease.

  • Background: Multiple complex pathways are operable in the evolution of cutaneous T cell lymphomas (CTCLs). These pathways involve interaction between neoplastic T cells and cells of the immune system (especially dendritic cells and the non-malignant T cells). Granulysin is a proinflammatory antimicrobial peptide which has an immune alarmin function, activating dendritic cells, as well as an active role in tumor immunology and prognosis. FOXP3+ regulatory T cells Tregs are an important player in the immune system. Much controversy is found in the literature about the role of Tregs in CTCL. Aim: The present study aimed to investigate the expression of granulysin and FOXP3 in mycosis fungoides (MF), its precursor lesion large plaque parapsoriasis and its leukemic form ;$s\acute{e}ezary$ syndrome (SS). Materials and Methods: Immunohistochemical expression of granulysin and FOXP3 were assessed in lesional skin biopsies taken from 58 patients (4 large plaque parapsoriasis, 48 MF and 6 SS). Results: Granulysin positivity was cytoplasmic and higher in MF than in parapsoriasis en plaque and higher in the more advanced stages of MF (p<0.001). All groups showed significant differences between each other except between MF tumor stage and SS. FOXP3 positivity was nuclear and higher in early stage MF (plaque and patch stages) than in tumor stages and SS (p<0.001). However the FOXP3 count was lower in parapsoriasis en plaque than in other stages of MF. All the groups showed significant differences between each other except between parapsoriasis and SS and between patch and plaque stages of MF. Conclusions: The present study supports a role for granulysin in MF progression and proposes a novel hypothesis about the effect of FOXP3 +veTregs in the suppression of the activity of the neoplastic cells in MF.

  • Versatile and innovative interdisciplinary professionals refer to those who can engage in an efficient cooperation with experts in other fields or to those who can themselves put knowledge of different fields together. This article aims to look into Galileo Galilei as an example of historic figure that made remarkable achievements by merging knowledge in multiple fields of study. It also shows that Galileo, who had active exchange with painters during the Renaissance, presented the findings from his telescope observations in the form of drawings and that he used them to build core logics that criticizes the traditional Aristotelian cosmology. Galileo drew the critical logics, hardly achievable from a simple observation report or mathematical demonstration, from his hand drawing. The Galileo case well proposes the goals and direction of how the modern society should nurture its interdisciplinary professionals today.

Complexity Estimation Based Work Load Balancing for a Parallel Lidar Waveform Decomposition Algorithm

  • LIDAR (LIght Detection And Ranging) is an active remote sensing technology which provides 3D coordinates of the Earth's surface by performing range measurements from the sensor. Early small footprint LIDAR systems recorded multiple discrete returns from the back-scattered energy. Recent advances in LIDAR hardware now make it possible to record full digital waveforms of the returned energy. LIDAR waveform decomposition involves separating the return waveform into a mixture of components which are then used to characterize the original data. The most common statistical mixture model used for this process is the Gaussian mixture. Waveform decomposition plays an important role in LIDAR waveform processing, since the resulting components are expected to represent reflection surfaces within waveform footprints. Hence the decomposition results ultimately affect the interpretation of LIDAR waveform data. Computational requirements in the waveform decomposition process result from two factors; (1) estimation of the number of components in a mixture and the resulting parameter estimates, which are inter-related and cannot be solved separately, and (2) parameter optimization does not have a closed form solution, and thus needs to be solved iteratively. The current state-of-the-art airborne LIDAR system acquires more than 50,000 waveforms per second, so decomposing the enormous number of waveforms is challenging using traditional single processor architecture. To tackle this issue, four parallel LIDAR waveform decomposition algorithms with different work load balancing schemes - (1) no weighting, (2) a decomposition results-based linear weighting, (3) a decomposition results-based squared weighting, and (4) a decomposition time-based linear weighting - were developed and tested with varying number of processors (8-256). The results were compared in terms of efficiency. Overall, the decomposition time-based linear weighting work load balancing approach yielded the best performance among four approaches.

  • In 2012, the International Council on Archives(ICA) distributed AtoM public release, an archival description software based on web, for free to support small archival institutions. The purpose of AtoM project was to let a single or multiple repositories describe archival materials based on ICA descriptive standards and access via a web browser. The value of AtoM project is to promote actual business experience by sharing technical expertise and form active community between users and developers. Recently, there is a growing interest in open source software in the field of domestic record management. This interest is to not only reduce cost, but to escape technology dependencies and to address requirements on the field. The critical mind of this study lies in the fact that there is no practical methodology to establish AtoM system. Even when using AtoM itself, basic tasks such as the migration of the existing data or the improvement of user interface are required. That is why this study suggests the process and methodologies to establish and customize archival information system based on AtoM software.

  • This study was to conducted on the general public living in the Busan area to determine their knowledge and awareness of radiation. A questionnaire written in Google Form was distributed, and 193 questionnaires were analyzed. Independent samples t-test and one-way ANOVA were conducted to verify differences in variables according to the characteristics of the study subjects. Correlation analysis was conducted to determine the relationship between variables. Multiple linear regression analysis was conducted to confirm the correlation between variables. As a result of the study, firstly, The knowledge and awareness of radiation and confidence level of radiological information were found to be high among men and unmarried group. Second, as a result of analysis according to occupation, it was found that students had a positive perception of radiation. Third, the Internet and TV are the most commonly used channels for obtaining information about radiation. Lastly, the thing that has the greatest impact on the awareness of radiation is the reliability of the obtained radiation information. In order to relieve excessive anxiety about radiation and to gain a proper understanding of radiation, the reliability of information about radiation is important. Therefore, it is believed that active efforts are needed to provide accurate information about radiation by utilizing media that are widely used by citizens.

Liposome Formation and Active Ingredient Capsulation on the Supercritical Condition (초임계 상태에서 리포좀의 생성 및 약물봉입)

  • This study is to produce multiple layers of liposomes in a supercritical state and encapsulates active ingredients in order to stably encapsulate thermodynamically unstable active ingredients. In order to form a liposome in a supercritical state, a mixed surfactant development including vegetable-derived hydrogenated phosphatidyl choline and their delivative, hydrogenated sucrose distearate was synthesized as high purity. It describes a manufacturing method of injecting liquid carbon dioxide into a reactor to create a supercritical state and stirring to produce a giant liposome, and adding and loading genistein and quercetin. The HLB of the mixed lipid complex (SC-Lipid Complex) was 12.50, and multiple layers of liposome vesicles were formed even at very low concentrations. This surfactant had a specific odor with a pale yellow flake, the specific gravity was 0.972, and the acid value was 0.12, indicating that it was synthesized with high purity. As a result of the emulsifying capacity experiment using 20 wt% capric/capric triglyceride and triethylhexanoin using SC-Lipid Complex, it was found to have 96.2% emulsifying power. SC LIPOSOME GENISTEIN was confirmed that a multi-layer liposome vesicle was formed through a transmission electron microscope (Cryo-TEM) for the supercritical liposome encapsulated with genistein. The primary liposome particle size in which genistein was encapsulated was 253.9 nm, and the secondary capsule size was 18.2 ㎛. Using genistein as the standard substance, the encapsulation efficiency of supercritical liposomes was 99.5%, and general liposomes were found to have an efficiency of 93.6%. In addition, the antioxidant activity experiment in which quercetin was sealed was confirmed by the DPPH method, and it was found that the supercritical liposome significantly maintained excellent antioxidant activity. In this study, thermodynamically unstable raw materials were sealed into liposomes without organic solvents in a supercritical state. Based on these results, it is expected that it can be applied to various forms such as highly functional skincare cosmetics, makeup cosmetics, and scalp protection cosmetics.

The Beacon Frame-Based Node Grouping Algorithm for Improving the Performance between MCT devices in the Home Wireless Network (가정 무선 네트워크 내 MCT 디바이스 간 성능 향상을 위한 Beacon frame 기반 노드 그룹화 알고리즘)

  • Recently, M2M (Machine to Machine) communication is possible the development of MTC (Machine Type Communication) devices becomes active. MCT devices in the form of home appliances have a low power consumption, low cost, short-range wireless communication in wireless home network. For purpose, MTC devices based on IEEE 802.15.4/Zigbee are composed in the form of cluster-tree topology, which consists of one PAN (Personal Area Network), one or other router and end of nodes. It happens that transmission delay, packet drop, and lacking data resulted from collision originated by a competition for allocating channels among MTC devices that greatly increased. At last performance of entire network can be degradated. This paper proposes that the beacon frame-based grouping algorithm using multiple channels in a MTC devices in the presence of wireless home network interference. The proposed algorithm decreases the transmission delay, dropped packet and throughput is more increase, so the proposal algorithm is more efficient than the IEEE 802.15.4/ Zigbee standard.

Factors Influencing the Intention of Admission into a Charged Nursing Home for the Elderly (유료노인전문요양원 입주의사 결정요인에 관한 연구)

  • Objectives : To gather information about the factors which influence the interest and intention of admission into charging nursing hones for the elderly(CNH), as these homes represent an important method for resolving the problems related to the rapid population aging occurring in Korea. Methods : A face-to-face interview survey was carried out with 320(men 159, women 165) patients over 60 years old who were admitted at 2 university hospitals and 5 general hospitals in the Busan area between December 1998 and March 1999. Data were analyzed through t-test, ANOVA, correlation and multiple regression analysis. Results : The mean age and years of education of the study population was 67.0 and 7.7 years, and the types of chronic degenerative disease included musculoskeletal disorders(20.1%), cerebrovascular disease(17.1%), and diabetes(14.3%). The major forms of household living arrangement prior to admission were elderly alone(22.6%), and elderly couple(33.5%), while about half of them(55.5%) didn't want to live with their children id the future. Almost half were paying medication fees by themselves(46.6%). The level of actual intention of admission$(3.07{\pm}1.39)$ into a CNH was lower than that of interest$(3.22{\pm}1.33)$(p<0.01). Multiple analysis revealed that the intention of admission increased with decreasing number of future supportive persons$(\beta=-0.107)$, lower level of activity in daily life$(\beta=-0.447)$, and longer years of education$(\beta=-0.447)$ with 32.7% of $R^2$. As for the factors which determined the admission into a CNH, the fee and facilities were considered to be most important, and professional nursing and physician's care were the most desired services. In nomenclature, they preferred 'elderly hospital' or 'elderly health center' to CNH. Conclusions : Interest in CNH is increasing recently, but existing studies about patient experiences in CHN are still limited. This study may form a basis for future examinations of the needs and uses for CNH. Active financial support and public information are considered by the authors to be important factors for the induction of welfare services for the elderly, though CNH.

Effects of Perceived Stress, Sleep, and Depression on Resilience of Female Nurses in Rotating Shift and Daytime Fixed Work Schedules (순환교대근무와 주간고정근무 여성 간호사의 회복탄력성에 대한 스트레스, 수면, 우울증의 영향)

  • Objectives: Healthy sleep is important and can have a positive effect on resilience. The aim of the present study was to compare the differences in resilience between two group nurses in rotating shift and daytime fixed work schedules and to investigate stress perception, coping factors, social and psychological health, and sleep factors that may affect resilience. Methods: A total of 400 female nurses having rotating shift and daytime fixed work schedules at two hospitals was surveyed from June 12, 2017 to June 12, 2018. All participants completed perceived stress scale (PSS), stress coping short form (Brief COPE), psycho-social wellbeing Index short form (PWI-SF) or general health questionnaire-18 (GHQ-18), center for epidemiologic studies depression scale (CES-D), STAI-X-1 in state-trait anxiety inventory (STAI), Pittsburgh sleep quality index (PSQI), Epworth sleepiness scale (ESS), insomnia severity index (ISI), Conner Davidson resilience scale (CD-RISC). Independent t-test, paired t-test, Pearson correlation analysis, and multiple regression analysis were applied to the results of the final 373 questionnaires of 400 nurses in two general hospitals. Results: Comparing the variable statistics between the two groups of rotating shift and daytime fixed work nurses, showed statistically significant differences in all variables except perceived stress, sleep quality, and daytime sleepiness. Factors that had a significant correlation with resilience were stress coping strategies, depression, and insomnia severity (p < 0.001). In multiple regression analysis, larger positive reframing1 (β = 0.206, p < 0.001), severe less depression (β = -3.45, p < 0.001), and higher psychosocial health (β = 0.193, p < 0.001). As acceptance coping2 increased (β = 0.129, p < 0.05), as daytime sleepiness decreased (β = -1.17, p < 0.05), and as active coping2 increased (β = 0.118, p < 0.05), as the positive reframing2 increases (β = 0.110, p < 0.05), the resilience increased. Conclusion: This study, it was found that resilience was higher in daytime fixed workers than in shift workers. In addition, specific stress coping strategies, psycho-social health, sleep, and depression factors were associated with resilience.
