몰로키아(Corchorus olitorius) 잎에서 추출한 mucilage의 이화학적 및 기능적 특성

  • 본 연구에서는 국내산 몰로키아 잎에서 추출한 mucilage의 이화학적 특성, 동물실험을 통한 콜레스테롤 대사에 미치는 영향가 임상실험에서의 비만 및 체지방대사에 미치는 영향을 검토하였다. 몰로키아 잎은 식이섬유의 함량이 37.4%, 단백질의 함량은 24.4%를 차지하고 있으며, 칼슘의 함량이 2,000mg% 이상으로 대단히 높았다. 몰로키아 앞에서 추출된 mucilage의 경우 단백질의 총당에 대한 비율이 $50^{\circ}C$에서 57.6%를 차지하였으며, 구성다당류의 분자량은 약 500,000 dalton의 크기를 나타내었고 40,000 dalton 이하의 것도 일부 존재하는 것으로 나타났다. 동물실험에서는 혈장의 총 콜레스테롤 함량은 대조구인 셀룰로오스 첨가군과 유의적인 차이가 나타나지 않았지만, 간에서의 지방 농도는 몰로키아 분말과 mucilage 첨가군이 유의적으로 감소되었고 변으로의 콜레스테롤 배설량도 몰로키아 분말 첨가군과 mucilage 첨가군 순으로 높았다. 몰로키아의 섭취가 다이어트와 변비개선에 미치는 효과를 검토한 결과, 체중의 감소는 아주 낮은 유의성을 보였고, 혈액 내 생화학적 수치에서 초기치와 8주 후의 값에는 유의한 차이를 보이지 않았지만, 체지방율의 경우에는 현저히 줄어들었다. 또한 몰로키아의 섭취로 배변이 2배 이상 증가하였으며, 배변시의 통증도 상당히 완화시키는 것으로 나타났다.

  • Functional properties of yam mucilage were investigated by physicochemical analysis. Yam mucilage was extracted from the root of yam and then separated into two fractions by its selective aggregation with isopropanol concentration. Each mucilge fraction showed the excellent binding properties with heavy metal ions Co, Cr and Cu. Cr showed the higher affinity with mucilage than Co and Cu at pH 6.3. In ACE inhibition, IC50 values of mucilage fraction 1 and 2 showed 8.99$\mu\textrm{g}$/${mu}ell$ and 7.1$\mu\textrm{g}$/${mu}ell$, respectively.

Effect of mucilage from yam on activation of lymphocytic immune cells

  • The immunostimulating activities of mucilage fraction from yam were investigated. The proliferation of BSA-primed lymph node cells was enhanced between 4.1- to 10.9-fold compare to control, when cultured with 1 to $25{\mu}g/mL$ of yam-mucilage fraction. It showed strong immunostimulating activity than ginseng extract and as remarkable as Bifidobacterium adolescentis M101-4 known as a positive immunostimulator. Mitogenicity to lymph node cells was fully induced by concanavalin A and lipopolysaccharide. The proliferation of splenocytes and Peyer's patch cells was enhanced between 5.0- to 14.1-fold and 2.4- to 6.4-fold, respectively, when cultured with 1 to $25{\mu}g/mL$ of yam-mucilage fraction. It enhanced the production of cytokines such as tumor necrosis $factor-{\alpha}$ and IL-6 in the culture of RAW 264.7 macrophage cells. In the culture of lipopolysaccharide-stimulated RAW 264.7 cells, production of cytokines was as similar as compared to controls. In unstimulated RAW 264.7 cells, both tumor necrosis $factor-{\alpha}$ and IL-6 production were enhanced between 15.6- to 60.1-fold and 2.3- to 9.1-fold, respectively. Mucilage fraction from yam is expected to be a safe immunopotentiator to maintain the host immunity and develop a physiologically functional food.

Chongkukjang Mucilage Stimulates Immunohistochemical Activities of Gastrointestinal Tract in Rats

  • We investigated the effect of a viscous substance from chongkukjang (chongkukjang mucilage) on immunohistochemical reactions in rat gastrointestinal (GI) tracts, Rats fed a steady diet of chongkukjang mucilage showed an increase in the immunoreactive densities of gastrin and serotonin in the pyloric region of their stomachs and duodenal villi, The number of gastrin and serotonin immunoreactive cells was significantly higher in the experimental group than in the control group. Feeding on dietary chongkukjang mucilage increased the immunohistochemical densities of $CD4^+$ and $CD8^+$ lymphocytes in the mucosa and submucosa of the rats' gastroduodenal region. The universal nitric oxide synthase (uNOS)-immunoreactive neurons and nerve fibers were strongly stained in the vascular walls of the submucosa and myenteric plexus in rats fed the test diet. The results indicate that the intake of chongkukjang mucilage could increase mucosal immune activity, gastrointestinal motility, and blood circulation in the GI tract.

  • The application of the dispersant is indispensable to the manufacture of Korean traditional paper (Hanji). However the mucilage of which extracted from Abelmoschus manihot root has viscosity drop problem in summertime and synthetic dispersant such as polyacrylamide (PAM) and polyethyleneoxide (PEO) have some problems that under the influence of the quality of. water, cohesion, and bad solubility. This study was carried out not only to develop new natural dispersant that can solve such problems but also to investigate the viscosity and papermaking characteristics of Hydrangea paniculata mucilage. There were no viscosity changes between control and treated mucilage that adjusted to pH 9, heated 6 hours at 40 $^{\circ}C$, and stored heating treatment one for a week at 5 $^{\circ}C$. The treatment of mucilage that adjusted to pH 9 and aged for 120 hours at 4$0^{\circ}C$ resulted in viscosity drop. In the hydrolysis of mucilage, galacturonic acid and glucuronic acid contents were decreased by heating and pH adjusting treatments. Wet web stripping quality and physical properties of Korean traditional paper, which used Hydrangea paniculatamucilage were same or superior to the Abelmoschus manihot root.

  • The changes of hydrogen ton concentration, viscosity and free sugars in the water-soluble mucilage of Ulmus Coreana, Nakai root upon storage at 10^{\circ}C were studied. Hydrogen ion concentration in the mucilage changed $pH7{\rightarrow}pH4{\rightarrow}pH6$ or 7. The viscosity of mucilage decreased very slowly. The mucilage contained glucose and galacturonic acid. The hydrolyzed products of mucilage consisted of rhamnose, mannose, galactose, arabinose, xylose, glucose and galacturonic acid.

  • The viscosity of mucilage of Abelmoschus Manihot, Medic root decreased by the influence of various mechanical, physical and chemical conditions. It was experimented by viscosity decrease of mucilage connected with hydrogen ion concentration, bacterial multiplication, disinfection with 70% ethanol, some antibiotics, such as streptomycin, penicillin, ganamycin and chloramphenicol and mucilage derived from autoclaved Abelmoschus Manihot, Medic root. The results obtained were as follows: 1. It was clear that the viscosify of mucilage decreased notably under the influence of infected bacteria and bacterial multiplication. 2. By the inoculation of Bacillus subtilis ATCC 6633 and Escherichia coli ML 1410 to the mucilage the viscosify decrease fast but the viscosity of mucilage derived from autoclaved, Abelmoschus Manihot, Medic root. 3. The small quantity of reducing sugar in the mucilage was detected. 4. Hydrogan ion concentration in the mucilage remained $6.5{\sim}8.0$ in spite of the viscosity decrease.

  • The amino acids in the mucilage and its hydrolyzed products from Ulmus coreana, Nakai root were detected by thin layer chromatography. The mucilage contained 10 kinds of amino acids; alanine, aspartic acid, glutamine, glutamic acid, hydroxyproline, isoleucine, leucine, proline, phenylalanine and tyrosine. In the hydrolyzed mucilage 14 kinds of amino acids were detected; alanine, aspargine, arpartic acid, glutamine, glutamic acid, hydroxyproline, isoleucine, leucine, methionine, proline, phenylalanine, serine, tyrosine and valine.

  • In order to estimate accurate originality of the important crude drugs, Gastrodia, Dendrobium, Bletilla and Cremastra spp. were investigated comparing their morphological, anatomical and physicochemical characteristics and antibacterial, antifungal activities. The results of the studies as follows: 1. In morphological and anatomical studies, Korean Gastrodia contained more mucilage duct and symbiotic fungi than imported Gastrodia. Korean Dendrobium contained starch grains but without starch grains in the imported Dendrobium from China enclosed with thicken silicified wall. The corm of Cremasta appendiculata contained raphides of bundles with mucilage while the bulbs of Tulipa edulis contained several various starch grains form. 2. In physiological and TLC analysis, crude drugs in Orchidaceae contained common constituents with fluorescence and much mucilage. 3. The EtOH extracts of Gastrodia, Dendrobium, Bletilla, Cremastra showed antibacterial activities against B. subtilis and E. coli (Dendrobium>Gastrodia>Bletilla>Cremastra). But, no antifungal activities against C. albicans, A. niger were observed.

Production of Carrot Pomace Fortified with Mucilage, Fibrinolytic Enzyme and Probiotics by Solid-state Fermentation Using the Mixed Culture of Bacillus subtilis and Leuconostoc mesenteroides

  • Bioactive compounds were produced from carrot pomace by solid-state fermentation using Bacillus subtilis HA and Leuconostoc mesenteroides. The carrot pomace (CP) fermented by B. subtilis HA with 3% monosodium glutamate (MSG) showed higher production of various bioactive compounds, with 1.64 Pa·sn of consistency, 2.31% of mucilage content, 16.95 unit/g of fibrinolytic enzyme activity, 35.3 unit/g of proteolytic activity and 37.5 mg% of tyrosine content. The mucilage production was greatly dependent upon the concentration of MSG added. Most MSG added in CP was converted into mucilage (2.3%) including 0.83% poly-$gamma$-glutamic acid (PGA) with 1,505 kDa of molecular weight. The CP fermented secondly by Leuc. mesenteroides showed acidic pH and lower consistency. However, the fibrinolytic and proteolytic activities were increased. The secondly fermented CP showed the viable cell counts with $2.5{\time}108$ CFU/g of B. subtilis HA and $3.7{\time}109$ CFU/g of Leuc. mesenteroides, respectively. The freeze-dried fermented CP showed 2.88 Pa·sn of consistency, 24% of mucilage content and 104.9 unit/g of fibrinolytic enzyme activity, respectively. Also, the powder of fermented CP indicated viable cell counts of $8.0{\time}107$ CFU/g of B. subtilis and $4.0{\time}108$ CFU/g of Leuc. mesenteroides. Therefore, the fermented CP that was fortified with dietary fibers, fibrinolytic enzyme and probiotics could be utilized as valuable ingredients of functional foods in food or cosmetic industries.
