Effects of Calcium and dbcAMP on Sperm Motility in Mouse (생쥐 정자의 운동성에 미치는 Calcium 과 dbcAMP 의 영향)

  • In order to study the effects of $Ca^{++}$ and dbcAMP on sperm motility, cauda epididyal spermatozoa of mouse were incubated in the media containing the various concentrations of $Ca^{++}$ and dbcAMP, and Its motility were observed and analyzed. The rate(%) of the sperm with forward motility which may be fertile, reached at the equilibrium state in 30-60 min. after the beginning of incubation. $Ca^{++}$ requirement for the maintenance of sperm motility obviously appeared in 3hrs. after the beginning of incubation. In the medium containing 0.01mM $Ca^{++}$, the rate of sperm forward motility was highest(30.6-40.6%), whereas in 10mM $Ca^{++}$ medium, the rate was rather reduced because of a severe agglutination phenomenon. On the other hand, 1mM dbcAMP in the medium was significantly effective for the maintenance of sperm forward motility.

Activation of Porcine Oocytes Following Intracytoplasmic Injection of Various Sperm Components and foreign species spermatozoa (여러 가지 정자구성성분 및 이종정자 주입에 의한 돼지난자의 활성)

  • We determined the incidence of activation, male pronuclear formation and apposition of pronuclei in porcine oocytes following intracy-toplasmic injection of various porcine sperm components and foreign species spermatozoa, such as mouse, human or cattle. The porcine oocytes were activated by injection of a spermatozoon or an isolated sperm head. Neither isolated sperm tail nor perinuclear material removed sperm head activated oocytes. Because injection of mouse, bovine or human spermatozoon activated porcine oocytes, the sperm born activation factors is not strict species specific. Male pronuclear formation and pronuclear apposition were observed in the porcine oocytes following injection of porcine, bovine, mouse or human spermatozoa. The electrical stimulation following sperm cell injection did not enhance the incidence of male pronuclear formation nor pronuclear apposition comparent with sperm cell injection alone (p>0.1). Mitosis and two cell division in some oocytes were observed at 20 to 24 h after injection of porcine spermatozoon. However, none of oocytes following injection of mouse, bovine or human spermatozoa developed to the mitotic metaphase or normally divided to the two cell stage. These results suggested that the oocyte activating factor(s) presented in the perinuclear material and it is not species specific for the porcine oocyte.

  • The development of 2-cell mouse embryos to the blastocyst stage in vitro has been used as a quality control for the media empolyed for human in vitro fertilization. There was a comparison between the quality control data of the culture medium as ascertained by 2-cell mouse embryos development and sperm motility and the data from fertilization and cleavage of human oocytes. However, there was no obvious association between fertilization and cleavage of human oocytes and the quality of the medium ascertained by mouse embryo development and sperm motility.

Effects of Gossypol Injection into the Stroma of Testes on Spermatogenesis in Mouse (생쥐 정소 실질내 Gossypol 투여가 조정기능에 미치는 영향)

  • This experiment was conducted to determine the effects of gossypol injection spermatogenesis of mice. Gossypol was injected into the stroma of testes(TS) and the doses of gossypol injected were 5, 10 and 15mg per kg of body weights, respectively. The number of sperm and the weight of testes were gradually reduced(P<0.01) from 2 to 4 weeks after gossypol treatment in all groups of mice treated with gossypol, compared with the control group. The rates of malformation(loss of proacrosome, damage of midpiece and breaking of tail) of sperm were significantly(P<0.01) increased at 2 and 3 weeks after the injection of 10 or 15mg of gossypol. However, the weight of testes and the number of normal sperm were gradually increased and the malformation rate of sperm was decreased between 4 and 6 weeks after injection of 5mg of gossypol. The results of this experiment indicated that probably ireeversible suppression of spermatogenesis could be brought about easily and immediately by the single injection of gossypol into TS.

Sperm Component Inducing 2nd Polar Body Extrusion in Mouse Oocytes (생쥐 난자의 제2극체 방출을 유발하는 정자 성분)

  • This study was carried out to elucidate whether sperm contain a factor inducing second polar body extrusion and to search for an effective collection method of the sperm factor Thus, sperm extract, dialyzed sperm-extract or liquid chromatographic fractions of sperm extract was microinjected into ovulated oocytes. And the microinjected oocytes were incubated for 24 hours to investigate about the extrusion of second polar body. The results obtained were as follows; 1. Sperm extract significantly increased the second polar body extrusion. 2. Sperm extract showed five major fractions at retention volumes (RVs) 1.25, 1.37, 1.84, 2.10 and 2.67ml after separation with Superose 12 column. These sperm extract fractions did not significantly increase the second polar body extrusion. 3. Dialyzed sperm-extract significantly increased the second polar body extrusion 4. Dialyzed sperm-extract showed three maior fractions at RVs 1.88, 2.14 and 2.77ml after separation with Superose 12 column. Of these fractions, the fraction RV2.14 significantly increased the second polar body extrusion. In conclusion, sperm extract contained a factor inducing the second polar body extrusion and the factor was contained largely in fraction RV2.14 after dialysis and liquid chromatographic fractionation of sperm extract.

  • The possible role of Na$^{+}$/H$^{+}$ antiporter in both the capacitation and the acrosome reaction (AR) was examined in mouse epididymal spermatozoa. Spontaneous acrosome reaction was inhibited by dimethyl amiloride (DMA), a specific inhibitor of Na$^{+}$/H$^{+}$ antiporter, with dose dependent manner. Follicular fluid- or A23l 87-induced acrosome reaction was not inhibited by DMA. It suggests that change in pH$_{i}$ by monovalent cation transport through the Na$^{+}$/H$^{+}$ antiporter is possibly engaged in the capacitation and that agonist- as well as A23l87-induced AR in capacitated sperm might be independent from the Na$^{+}$/H$^{+}$ antiporter. Conclusively, changes in pH$_{i}$ through the Na$^{+}$/H$^{+}$ antiporter might be important for sperm capacitation and it virtually occurs upstream of the $Ca^{2+}$ influx which precedes the acrosome reaction in mouse epididymal spermatozoa.pididymal spermatozoa.

ADAM7 Is Associated with Epididymosomes and Integrated into Sperm Plasma Membrane

  • During epididymal transit, mammalian sperm acquire selected proteins secreted by the epididymis. We previously showed that a disintegrin and metalloprotease (ADAM) 7 is expressed specifically in the epididymis and transferred to the sperm surface during epididymal transit. Here, we show that mouse ADAM7 secreted to the epididymal lumen is associated with membranous vesicles known as epididymosomes. Furthermore, we found that ADAM7 can be transferred directly from epididymal vesicles to sperm and that it is an integral plasma membrane protein in sperm. Thus, our study provides new information regarding the unique mode of secretion and interaction of ADAM7 during the epididymis-to-sperm transfer process.

  • Excretory-secretory products (ESP) of T. vaginalis have been shown to inhibit sperm motility, viability, and functional integrity, leading to a decreased fertilization rate in vitro. This study investigated whether T. vaginalis induce apoptosis and ultrastructural changes of sperm using flow cytometry and electron microscopy. Incubation of sperm with T. vaginalis ESP increased phosphatidylserine externalization and DNA fragmentation, and decreased mitochondrial membrane potential. Transmission electron microscopy of sperm incubated with ESP revealed abnormal features such as distorted heads, broken necks, and acrosomes exocytosis. This is the first report that demonstrates a direct impact of T. vaginalis ESP on sperm apoptosis and architecture in vitro.

Delayed Sperm Penetration into Frozen-Thawed Zona-Free Hamster Oocytes (동결.융해시킨 햄스터 난자에서 장자침입의 지연)

  • Frozen storage of the oocytes has been used in a few mammalian species including mouse, hamster, human and cattle. However, frozen4hawed oocvtes show different sperm penetration on the levels of the zona pellucida and the plasma memhrane when compared with fresh oocytes. To elucidate biological changes occurring during freezing and thawing, we examined the kinetics of sperm penetration into frozen-thawed hamster oocytes. Oocytes obtained from superovulated female golden hamsters were frozen-thawed in an autofreezer according to an established method. Fresh and frozen4hawed oocytes were fertilized in vitro with capacitated hamster spermatozoa after removing the zona pellucida. The oocytes were examined at 1, 2, 3 and 6 h postinsemination. Sperm penetration found to be 1 h delayed in frozen-thawed oocytes. Other parameters such as degree of polyspermy and decondensing sperm heads were not affected by freezing and thawing. The results suggest that freezing and thawing may cause changes in the egg membrane surface and subsequently which leads to delay in the sperm-egg fusion.
