Isolation and Characterization of New Family Genes of DNA Damage in Fission Yeast

  • The SNF2 family includes proteins from a variety of species with roles I cellular processes such as transcriptional regulation, recombination and various types of DNA repair. Several proteins with unknown function are also included in this family. Here, we report the cloning and characterization of hrp 2+ gene (helicase related gene from S. pombe) which was isolated by PCR amplication using the conserved domain of SNF2 motifs within the ERCC6 gene which encodes a protein involved in DNA excision repair. The hrp2+ gene was isolated by screening with yeast S. pombe genomic library. The isolated cloned contained 6.5 kb insert DNA. Southern blot analysis confirmed that S. pombe chromosome contains the same DNA as hrp2+ gene and this gene exists as a single copy in S. pombe genome. The 4.7 kb transcript of mRNA was identified by Northern blot. To examined the transcriptional regulation of hrp2+ gene, DNA damaging agents were treated. These results indicated that the hrp2+ gene may not be directly involved in DNA replication, but may be involved in damage response pathway.

Emerging functions for ANKHD1 in cancer-related signaling pathways and cellular processes

  • ANKHD1 (ankyrin repeat and KH domain containing 1) is a large protein characterized by the presence of multiple ankyrin repeats and a K-homology domain. Ankyrin repeat domains consist of widely existing protein motifs in nature, they mediate protein-protein interactions and regulate fundamental biological processes, while the KH domain binds to RNA or ssDNA and is associated with transcriptional and translational regulation. In recent years, studies containing relevant information on ANKHD1 in cancer biology and its clinical relevance, as well as the increasing complexity of signaling networks in which this protein acts, have been reported. Among the signaling pathways of interest in oncology regulated by ANKHD1 are Hippo signaling, JAK/STAT, and STMN1. The scope of the present review is to survey the current knowledge and highlight future perspectives for ANKHD1 in the malignant phenotype of cancer cells, exploring biological, functional, and clinical reports of this protein in cancer.

Specific bovine antibody response against a new recombinant Cryptosporidium parvum antigen containing 4 zinc-finger motifs

  • A Cryptosporidium parvum sporozoite and oocyst λgt11 cDNA library was screened with a hyperimmune rabbit serum that was developed against insoluble fragments of ultrasonicated oocysts. A clone named Cp22.4.1 encoding a protein of 231 amino acids with 4 zinc-finger domains characterized by a Cys-X2-Cys-X4-His-X4-Cys motif was isolated and characterized. There was a complete match between the sequencing data of the coding region of Cp22.4.1 and the corresponding gene at chromosomal level. Cloning in a pBAD-TOPO-TA expression vector permitted to evaluate the antigenicty of the recombinant His-tagged antigen. This antigen was recognized by 2 out of 5 sera from Cruptosporidium immune calves and not by sera from parasite naive animals.

  • For a good design, it is important to understand consumers and trend of textile and fashion design. Previous researches on textile pattern design with CAD have been done. They have studied on Korean traditional floral motives/hand painting or some geometric patterns. Few studies have been done on conversational design. Therefore the purpose of study was to observe types of textile pattern design for the recent fiver years and determine fabric pattern trends, especially on the conversational motifs. The types of patterns were analysed with professional panels from the 2005~2009 collection sources, using SPSS program. This study was also develop textile patterns creating using CAD. We conducted design process on motif creating, repeat, and mapping jobs, using textile special design program Prima vision and Photoshop CS3.

Cell cycle regulatory element in the promoter of the human thymidine kinase gene and its binding to factors

  • When quiescent cells ate stimulated to enter the cell cycle, the thymidine kinase(TK) gene is transcriptionally activated at the border of Gl and 5. In this report we show that the human TK promoter contains multiple protein-binding sites. By site-directed mutagenesis, we identified a protein-binding site on the human TK promoter requited for conferring Gl-S-regulated transcription to a heterologous promoter and dissociated it functionally from an adjacent protein-binding domain containing an inverted CCAAT motif requited for high basal level expression. Substitution-mutation of this site results in constitutive expression of the neo reporter gene in serum-stimulated fibroblasts, as well as in cells arrested in mid-Gl by a temperature-sensitive mutation. The regulatory domains for the human TK promoter exhibit interesting symmetrical features, including a set of CCAAT motifs and sites similar to the novel Yi protein-binding site recently discovered in the mouse TK promoter. Thus, components of the hTK complex is important for hTK gene regulation.

Bridging a Gap between DNA sequences and expression patterns of genes

  • The completion of sequencing human genome would motivate us to map millions of human cDNAs onto the unique ruler "genome sequence", in order to identify the exact address of each cDNA together with its exons, its promoter region, and its alternative splicing patterns. The expression patterns of some cDNAs could therefore be associated with these precise gene addresses, which further accelerate studies on mining correlations between motifs of promoters and expressions of genes in tissues. Towards the realization of this goal, we have developed a time-and-space efficient software named SQUALL that is able to map one cDNA sequence of length a few thousand onto a long genome sequence of length thirty million in a couple of minutes on average. Using SQUALL, we have mapped twenty thousand of our Bodymap (http://bodymap.ims.u-tokyo.ac.jp) cDNAs onto the genome sequences of Chr.21st and 22nd. In this talk, I will report the status of this ongoing project.

한국복식(韓國服飾)에 표현(表現)된 흔들림과 떨림의 미(美)

  • This study is focused on the swinging and trembling effect expressed in Korean costume from ancient days to the present. The swinging is a term standing for gentle movements that are created and implied by the long ties, strings, tassels, pendants and hanging ornamentations. These attatched elements of the costume move as the wearer moves. Trembling is a term expressing small vibrating movement which is created by the tiny decorative motifs attatched to Korean costume. This little decorative elements were attatched to the royal crown, earings, necklaces even the sole of the metalic shoes for a change and to draw viewers attention. The Swinging and Trembling have been used mainly to express the decorative desires. The Swinging on shaman costume is a expression of the aesthetics of evil's eye, while the Swinging shown on the costume of a schalor is expressing the beauty of personality and nature. In General during the Choson Period, the Swinging of the tassels, ribbons, string belts is a expression of the beauty of nature. Consequently the Swinging and the Trembling are the typical examples of the "Meot"(멋), Korean traditional aesthetics.

Molecular Cloning and Characterization of the doxA Cytochrome P-450 Hydroxylase Gene in Streptomyces peucetius subsp. caesius ATCC 27952

  • DNA sequence analysis of doxA from Streptomyces peucetius subsp. caesius ATCC 27952 revealed a $95\%$ amino acid identity with that of Streptomyces strain C5. DoxA from S. peucetius subsp. caesius ATCC 27952 encodes a peptide with both conserved heme-binding and dioxygen-binding motifs. Expression of this gene in S. lividans 1326 resulted in bioconversion of daunorubicin to doxorubicin.

Discovering cis-regulatory motifs by combining multiple predictors

  • The computational discovery of transcription factor binding site is one of the important tools in the genetic and genomic analysis. Rough prediction of gene regulation network and finding possible co-regulated genes are typical applications of the technique. Countless motif-discovery algorithms have been proposed for the past years. However, there is no dominant algorithm yet. Each algorithm does not give enough accuracy without extensive information. In this paper, we explore the possibility of combining multiple algorithms for the one integrated result in order to improve the performance and the convenience of researchers. Moreover, we apply new high order information that is reorganized from the set of basis predictions to the final prediction.
