In Vivo Kinetics and Biodistribution of a HIV-1 DNA Vaccine after Administration in Mice

  • In this study we have investigated the pharmacokinetics and tissue distribution of GX-12, a multiple plasmid DNA vaccine for the treatment of HIV-1 infection. Plasmid DNA was rapidly degraded in blood with a half-life of 1.34 min and was no longer detectable at 90 min after intravenous injection in mice. After intramuscular injection, plasmid DNA concentration in the injection site rapidly declined to less than 1 % of the initial concentration by 90 min post-injection. However, sub-picogram levels (per mg tissue) were occasionally detected for several days after injection. The relative proportions of the individual plasm ids of GX-12 remained relatively constant at the injection site until 90 min post-injection. The concentration of plasmid DNA in tissues other than the injection site peaked at 90 min post-injection and decreased to undetectable levels at 8 h post-injection. The rapid in vivo degradation of GX-12 and absence of persistence in non-target tissues suggest that the risk of potential gene-related toxicities by GX-12 administration, such as expression in non-target tissues, insertional mutagenesis and germline transmission, is minimal.

  • Polyethylene has been prepared via metallocene catalysts [(TMDS)$Cp_2$]$ZrCl_2$, 1, and [$(n-Bu)_2Cp_2$]$ZrCl_2$, 2, in the presence of hydrogen as a chain transfer reagent. Increase of hydrogen flow to the polymerization reactor resulted in the drop of catalytic activity, reduction of molecular weight of polyethylene, getting narrow of molecular weight distribution of polyethylene, and melting point of the polyethylene wax. It should be noticed that it was possible to control molecular weight down to 1500 and melting temperature to 60 $^\circ$C of polyethylene wax using the catalyst 1 that has been developed by authors as well as the catalyst 2 from Exxon.

Gintonin regulates inflammation in human IL-1β-stimulated fibroblast-like synoviocytes and carrageenan/kaolin-induced arthritis in rats through LPAR2

  • Background: In ginseng, there exists a glycolipoprotein complex with a special form of lipid LPAs called Gintonin. The purpose of this study is to show that Gintonin has a therapeutic effect on rheumatoid arthritis through LPA2 receptors. Methods: Fibroblast-like synoviocytes (FLS) were treated with Gintonin and stimulated with interleukin (IL)-1β. The antioxidant effect of Gintonin was measured using MitoSOX and H2DCFDA experiments. The anti-arthritic efficacy of Gintonin was examined by analyzing the expression levels of inflammatory mediators, phosphorylation of mitogen-activated protein kinase (MAPK) pathways, and translocation of nuclear factor kappa B (NF-κB)/p65 into the nucleus through western blot. Next, after treatment with LPAR2 antagonist, western blot analysis was performed to measure inflammatory mediator expression levels, and NF-κB signaling pathway. Carrageenan/kaolin-induced arthritis rat model was used. Rats were orally administered with Gintonin (25, 50, and 100 mg/kg) every day for 6 days. The knee joint thickness, squeaking score, and weight distribution ratio (WDR) were measured as the behavioral parameters. After sacrifice, H&E staining was performed for histological analysis. Results: Gintonin significantly inhibited the expression of iNOS, TNF-α, IL-6 and COX-2. Gintonin prevented NF-κB/p65 from moving into the nucleus through the JNK and ERK MAPK phosphorylation in FLS cells. However, pretreatment with an LPA2 antagonist significantly reversed these effects of Gintonin. In the arthritis rat model, Gintonin suppressed all parameters that were measured. Conclusion: This study suggests that LPA2 receptor plays a key role in mediating the anti-arthritic effects of Gintonin by modulating inflammatory mediators, the MAPK and NF-κB signaling pathways.

A Study on YHB RDF via Density Dependent Pressure Calculation of Ethene Gas (Ethene 기체의 밀도변화에 따른 압력 계산에 의한 YHB 동경분포함수에 대한 연구)

  • The radial distribution functions of a linear spheroidal type gas molecule $C_2H_4$ were calculated by using the Stockmayer molecular molel, which assumed the $C_2H_4$ molecule as simple quadrupolar molecule. To examine the validity of the calculated radial distribution of $C_2H_4$ gas, the density dependent pressures of $C_2H_4$ gas at several temperatures were calculated and compared with literarily known experimental pressure data of $C_2H_4$ gas. The temperatures examined was 50, 100 and $150^{\circ}C$ and the densities were up to $0.02/{\AA}^3$ (maximum pressure = 1500 atm). The radial distribution function expression used was derived by Yoon, Hacura, and Baglin (YHB) with Baker and Henderson's perturbation theory. The calculated results by a computer showed that the $C_2H_4$ pressures calculated agreed well with the experimental values within ${\pm}5%$ of error range. This indicated that the YHB radial distribution function expression is good enough to obtain various physical quantities sensitive to density such as pressure with resonable accuracy.

  • This research article explores the application of Polymer of Intrinsic Microporosity (PIM-1) as a cutting-edge material for CO2 gas separation membranes in response to the escalating global concern over climate change and the imperative to reduce greenhouse gas emissions. The study delves into the synthesis, molecular weight control, and fabrication of PIM-1 membranes, providing comprehensive insights through various characterization techniques. The intrinsic microporosity of PIM-1, arising from its unique crosslinked and rigid structure, is harnessed for selective gas permeation, particularly of carbon dioxide. The article emphasizes the tunable chemical properties of PIM-1, allowing for customization and optimization of gas separation membranes. By controlling the molecular weight, higher molecular weight (H-PIM-1) membranes are demonstrated to exhibit superior CO2 permeability and selectivity compared to lower molecular weight counterparts (L-PIM-1). The study's findings highlight the critical role of molecular weight in tailoring PIM-1 membrane properties, contributing to the advancement of next-generation membrane technologies for efficient and selective CO2 capture-an essential step in addressing the pressing global challenge of climate change.

  • NELL2, a neural tissue-enriched protein, is produced in the embryo, and postembryonically in the mammalian brain, with a broad distribution. Although its synthesis is required for neuronal differentiation in chicks, not much is known about its function in the adult mammalian brain. We investigated the distribution of NELL2 in various regions of the adult rat brain to study its potential functions in brain physiology. Consistent with previous reports, NELL2-immunoreactivity (ir) was found in the cytoplasm of neurons, but not in glial fibrillary acidic protein (GFAP)-positive glial cells. The highest levels of NELL2 were detected in the hippocampus and the cerebellum. Interestingly, in the cerebellar cortex NELL2 was observed only in the GABAergic Purkinje cells not in the excitatory granular cells. In contrast, it was found mainly in the hippocampal dentate gyrus and pyramidal cell layer that contains mainly glutamatergic neurons. In the dentate gyrus, NELL2 was not detected in the GFAP-positive neural precursor cells, but was generally present in mature neurons of the subgranular zone, suggesting a role in this region restricted to mature neurons.

Research of stone pagoda weathering degree from contaminated distribution analysis of 10-storied pagoda in the temple of Kyoungchunsa (경천사십층석탑의 오염물분포별 현황조사를 통한 석탑 풍화도에 관한 연구)

  • The conservation treatments of 10-storied pagoda in the temple of Kyoungchunsa is progressed favorably to come to the finish since 1995. Most of all, the important conservation treatments are stone crack, exfoliation and partial form modification. This pagoda has been made up a complicated structure constructed using 142 stones. Therefore, the stone crack and exfoliation have been occurred as very varied forms. Especially, the stylobate of the pagoda has 19 substitution stones of 48 stones, which is40% of modern stone substitution. It is supposed that there is serious weathering on the stones. The other stones, which are no substitution stones, have some kinds of weathering and so we carried out inside replenishment and partial outside reconstruction of crack stones using high molecular resin and substitution stone. The stone of each storey has also partial difference, but the condition of serious weathering on the whole. We can detect general degree through treatment of high molecule resin or distribution research of contaminated material on 10-storied pagoda in the temple of Kyoungchunsa. Especially, we are able to analyze general weathering degree and know details weathering of each storey or direction for the difference of replenishment quantity and different pattern of stone crack degree. Also, the distribution research will accomplish very important parts of examining environment effects and use as research data of cultural heritage conservation field.

  • The distribution and genotypes of the sea nettle Chrysaora pacifica have been reported in the South Sea of Korea; however, little research work has been attempted in the East Sea. Here, we collected similar jellyfishes from the East Sea coasts (Goseong, Yangyang and Sokcho), and identified them to the sea nettle morphologically. In addition, the genotypes of these sea nettle were compared with those from the South Sea (Tongyeong and Geoje). Phylogenetic analysis by using the mitochondrial COI sequences showed that the genus Chrysaora was clearly separated from other taxa to be formed a monophyletic group, with each species distinctly separated. C. pacifica in the East and South Seas was separated geographically by the COI phylogeography, representing potentially different populations. The COI gene of the Korean C. pacifica had approximately 7 times more genetic variation than the nuclear ITS rDNA, and thus it might be considered as a useful marker for genetic analysis of the jellyfish population.

Effect of High-Molecular Weight Organic Compounds on Improvement of Pore Structure of Cement Materials

  • Carbon dioxide emissions involved in global warming are one of the most important issues in the world, and carbon dioxide emissions from the cement industry are about 7% of total carbon dioxide emissions. Thus, reduction in the amount of utilized cement can contribute to a reduction of carbon dioxide emissions. The average life of concrete is 20 ~ 30 years, and if concrete life can be improved by ten years, cement use will be much lower. In this study, we examined the use and effect of fructan from microbes as a method for the densification of the pore structure of cement. The effect of fructan on the hydration reaction and pore distribution, as well as the water absorption of hardened cement mortar were studied. Pores distribution increased in mesopore OPC, and absorption rate was found to decrease with the use of fructan, which has a glue-like and swelling character.

Dynamics of OH Production in the Reaction of O(1D2) with Cyclopropane

  • The OH($X^2{\Pi}$, ${\upsilon}^{\prime\prime}=0,1$) internal state distribution following the reaction of electronically excited oxygen atom ($O(^1D_2)$) with cyclo-$C_3H_6$ has been measured using laser-induced fluorescence, and compared with that following the reaction of $O(^1D_2)$ with $C_3H_8$. The overall characteristics of the OH internal energy distributions for both reactions were qualitatively similar. The population propensity of the ${\Pi}(A^{\prime})$ ${\Lambda}$-doublet sub-level suggested that both reactions proceeded via an insertion/elimination mechanism. Bimodal rotational population distributions supported the existence of two parallel mechanisms for OH production, i.e., statistical insertion and nonstatistical insertion. However, detailed analysis revealed that, despite the higher exoergicity of the reaction, the rotational distribution of the OH following the reaction of $O(^1D_2)$ with $C_3H_8$ was significantly cooler than that with cyclo-$C_3H_6$, especially in the vibrational ground state. This observation was interpreted as the effect of the flexibility of the insertion complex and faster intramolecular vibrational relaxation (IVR).
