Surface Modification of PDMS for Hydrophilic and Antifouling Surface Using PEO-PPO-PEO Block Copolymer (PEO-PPO-PEO 블록 공중합체를 이용한 PDMS의 친수성 표면 개질 방법)

  • In this study, we optimized a method of PEO-PPO-PEO block copolymer embedding, for solving non-specific protein and biomolecular adsorption and high hydrophobicic surface property, which is widely known as problems of poly (dimethylsiloxane) (PDMS) that has frequently been used in basic biological and its applied research. We assessed its surface modification by controlling the concentration of embedded block copolymer, water-soaking time, and recovery time as variables by contact angle measurements. In order to evaluate its antifouling ability, adsorption of FITC-BSA molecules was quantified. Furthermore, we generated oil-in-water (O/W) emulsion as a proof-of-concept experiment to confirm that the optimized surface modification works properly.

  • In this paper, a novel fluid controlled phase tunable line using inkjet printed microfluidic composite right/left-handed(CRLH) transmission line(TL) is proposed. A CRLH-TL prototype has been inkjet-printed on a paper substrate using silver nano particle ink. In addition, a laser-etched microfluidic channel in poly methyl methacrylate(PMMA) has been integrated with the CRLH TL using inkjet-printed SU-8 as a bonding material. The proposed TL provides excellent phase-tuning capability that is dependent on the different fluidic materials used. As the fluid is changed, the proposed TL can have negative-phase, zero-phase, and positive-phase characteristics at 900 MHz and reflection coefficient is maintained to below -10 dB. The performance of the proposed TL is successfully validated using simulation and measurement results.

  • The flow in a microchannel is usually characterized as a low Reynolds number (Re) so that good mixing is quite difficult to be achieved. In this regard, we developed a novel chaotic micromixer, named Serpentine Laminating Micromixer (SLM) in the present study, Part 1. In the SLM, the higher level of chaotic mixing can be achieved by combining two general chaotic mixing mechanisms: splitting/recombination and chaotic advection. The splitting and recombination (in other term, lamination) mechanism is obtained by the successive arrangement of 'F'-shape mixing units in two layers. The chaotic advection is induced by the overall three-dimensional serpentine path of the microchannel. Chaotic mixing performance of the SLM was fully characterized numerically. To compare the mixing performance, a T-type micromixer which has the same width, height and length of the SLM was also designed. The three-dimensional numerical mixing simulations show the superiority of the SLM over the T-type micromixer. From the cross-sectional simulation results of mixing patterns, the chaotic advection effect from the serpentine channel path design acts favorably to realize the ideal lamination of fluid flow as Re increases. Chaotic mixing mechanism, proposed in this study, could be easily integrated in Micro-Total-Analysis-System, Lab-on-a-Chip and so on.

  • In this paper, Part II, we realized the Serpentine Laminating Micromirer (SLM) which was proposed in the accompanying paper, Part I, by means of the injection molding process in mass production. In the SLM, the higher level of chaotic mixing can be achieved by combining two general chaotic mixing mechanisms of splitting/recombination and chaotic advection by the successive arrangement of 'F'-shape mixing units in two layers. Mold inserts for the injection molding process of the SLM were fabricated by SU-8 photolithography and nickel electroplating. The SLM was realized by injection molding of COC (cyclic olefin copolymer) with the fabricated mold inserts and thermal bonding of two injection molded COC substrates. To compare the mixing performance, a T-type micromixer was also fabricated. Mixing performances of micromixers were experimentally characterized in terms of an average mixing color intensity of a pH indicator, phenolphthalein. Experimental results show that the SLM has much better mixing performance than the I-type micromixer and chaotic mixing was successfully achieved from the SLM over the wide range of Reynolds number (Re). The chaotic micromixer, SLM proposed in this study, could be easily integrated in Micro-Total-Analysis- System , Lab-on-a-Chip and so on.

  • Formation and characterization of self-assembled monolayers (SAMs) on various surfaces are the essential basis for many other applications, including molecular switches, biosensors, microfluidics, and fundamental studies in surfaces and interfaces. To improve the performance at these applications, it is a key to control the quantity of each molecule in various mixed SAMs on the surface. In this study, using mixed SAM of carbamate-based hydroquinone (HQ)-PhBr and11-mercaptoundecanol, the quantitative mass spectrometric method of mixed SAM was developed based on comparison study with XPS and FT-IR methods. In addition, our method was applied to another mixed SAM of biotinylated PEG alkane thiol and 11-mercaptoundecanol for verification purpose. Time-of-flight secondary mass spectrometry (ToF-SIMS) analysis was performed to identify and quantify each molecule of mixed SAM along with principal component analysis (PCA). Since there is no matrix effect in the X-ray photoelectron spectroscopy (XPS) and Fourier transform-infrared (FT-IR) techniques, we compared ToF-SIMS results with XPS and FT-IR results. Because PCA results from ToF-SIMS analysis are well matched with XPS and FT-IR results from both mixed SAMs, we are expecting that our method will be useful to identify and quantify each molecule in various mixed SAMs.

Separation of Human Breast Cancer and Epithelial Cells by Adhesion Difference in a Microfluidic Channel

  • A simple, label-free microfluidic cell purification method is presented for separation of cancer cells by exploiting difference in cell adhesion. To maximize the adhesion difference, three types of polymeric nanostructures (50nm pillars, 50nm perpendicular and 50nm parallel lines with respect to the direction of flow) were fabricated using UV-assisted capillary moulding and included inside a polydimethylsiloxane (PDMS) microfluidic channel bonded onto glass substrate. The adhesion force of human breast epithelial cells (MCF10A) and human breast carcinoma (MCF7) was measured independently by injecting each cell line into the microfluidic device followed by culture for a period of time (e.g., one, two, and three hours). Then, the cells bound to the floor of a microfluidic channel were detached by increasing the flow rate of medium in a stepwise fashion. It was found that the adhesion force of MCF10A was always higher than that of MCF cells regardless of culture time and surface nanotopography at all flow rates, resulting in a label-free detection and separation of cancer cells. For the cell types used in our study, the optimum separation was found for 2 hours culture on 50nm parallel line pattern followed by flow-induced detachment at a flow rate of $300{\mu}l/min$.

Flexible Microfluidic Metamaterial Absorber for Remote Chemical Sensor Application (원격 화학 센서로 활용 가능한 플렉서블 미세유체 메타물질 흡수체)

  • In this paper, a novel flexible microfluidic metamaterial absorber is proposed for remote chemical sensor applications. The proposed metamaterial absorber consists of a periodic of split-ring-cross resonators(SRCRs) and a microfluidic channel. The SRCR patterns are inkjet-printed using silver nanoparticle inks on paper. The microfluidic channels are laser-etched on polydimethylsiloxane(PDMS) material. The proposed absorber can detect change of the effective permittivity at different liquids. Therefore, the absorber can be used for a remote chemical sensor by detecting change of the resonant frequencies. The performance of the proposed absorber is demonstrated with full-wave simulation and measurement results. The experimental results shows that the resonant frequency is 10.49 GHz at the empty channel. When ethanol and DI-water are injected into the channel, the resonant frequencies are 10.04 GHz and 8.9 GHz, respectively.

Fabrication of Polymeric Microcapsules in a Microchannel using Formation of Double Emulsion (마이크로채널 내 이중유화 액적 형성을 통한 마이크로캡슐 제조)

  • In this study, we present simple microfluidic approach for the synthesis of monodisperse microcapsules by using droplet-based system. We generate double emulsion through single step in the microfluidic device having single junction while conventional approaches are limited in surface treatment for the generation of double emulsion. First, we have injected disperse fluid containing FC-77 oil and photocurable ethoxylated trimethylolpropane triacrylate (ETPTA) and water containing 3 wt% poly(vinyl alcohol) (PVA) as continuous phase into microfluidic device. Under the condition, we easily generate double emulsion with high monodispersity by using flow focusing. The double emulsion droplets are transformed into microcapsules under the UV irradiation via photopolymerization. In addition, we control thickness of double emulsion's shell by controlling flow rate of ETPTA. We also show that the size of double emulsions can be controlled by manipulation of flow rate of continuous phase. Furthermore, we synthesize microcapsules encapsulating various materials for the application of drug delivery systems.

  • Colloids are a heterogeneous system in which particles of a few nanometers to hundreds micrometers in size are finely dispersed in liquid medium, but show homogeneous properties in macroscopic scale. They have attracted much attention not only as model systems of natural atomic and molecular self-assembled structures but also as novel structural materials of practical applications in a wide range of areas. In particular, recent advances in colloidal science have focused on nano-bio materials and devices which are essential for drug discovery and delivery, diagnostics and biomedical applications. In this review, first we introduce nano-bio colloidal systems and surface modification of colloidal particles which creates various functional groups. Then, various methods of fabrication of colloidal particles using holographic lithography, microfluidics and virus templates are discussed in detail. Finally, various applications of colloids in metal inks, three-dimensional photonic crystals and two-dimensional nanopatterns are also reviewed as representative potential applications.

Increase in Voltage Efficiency of Picoinjection using Microfluidic Picoinjector Combined Faraday Moat with Silver Nanoparticles Electrode (은 나노입자 전극과 패러데이 모트를 이용한 미세유체 피코리터 주입기의 전압효율 상승)

  • This study presents modified microfluidic picoinjector combined Faraday moat with silver nanoparticle electrode to increase electrical efficiency and fabrication yield. We perform simple dropping procedure of silver nanoparticles near the picoinjection channel, which solve complicate fabrication process of electrode deposition onto the microfluidic picoinjector. Based on this approach, the microfluidic picoinjector can be reliably operated at 180 V while conventional Faraday moat usually have performed above 260 V. Thus, we can reduce the operation voltage and increase safety. Furthermore, the microfluidic picoinjector is able to precisely control injection volume from 7.5 pL to 27.5 pL. We believe that the microfluidic picoinjector will be useful platform for microchemical reaction, biological assay, drug screening, cell culture device, and toxicology.
