Measurement of Ion Concentrations in Denitrifying Biofilm by Microelectro-sensor (미소전극 센서를 이용한 탈질 생물막내의 이온 농도측정)

  • Microelectrode probe was made and applied to the biofilm in the biological treatment process as the state-of-art technology in order to actually measure the biofilm thickness, ionic concentration gradient, and material transport, etc. instead of classical theoretical approach. The working microelectrode, one of the main components of microelectrode probe, was easily contaminated and broken when determining the differences in the ionic concentrations through the measurement of biofilm's EMF (electromotive force). As a demonstration, two microelectrode probes were constructed in our lab for the measurement of the pH and $NO_3{^-}$ concentration in denitrifying biofilm. The microelectrode probe through the inner biofilm ($350{\mu}m$ from the surface of biofilm) showed that the pH was increased from pH 8 in the bulk solution to pH 8.3, on the other hand, the $NO_3{^-}$ concentration was decreased from 30 fig N/L in the bulk solution to 4 fig N/L.

Thalamotomy without Microelectrode Recording

  • Objective: Tremor, either essential tremor or Parkinsonian tremor, has been effectively and safely treated by lesioning the ventral intermediate(Vim) nucleus of the thalamus with or without mircroelectrode recording. The authors evaluate the treatment outcome of sixteen tremor patients who had been treated with thalamotomy without microelectrode. Methods: Between September, 2001, and December, 2003, sixteen tremor patients were treated with thalamotomy without microelectrode recording. Twelve patients suffered from Parkinsonian tremor and four patients were essential tremor patients. The male to female ratio was 1.6 to 1 with median age of 59.6 years (range; 39-74 years). Under local anesthesia, a 3mm hole was made using a hand-held twist drill, and the dura mater was penetrated with a 1.2mm sharp drill beat. Radiofrequency(RF) electrode was placed in the Vim nucleus of thalamus. With intraoperative macrostimulation, RF lesion was made. Postoperative CT scan and/or MR imaging was performed to confirm the localization of the target lesioned. Preoperative and postoperative tremor was evaluated with simple tremor severity scale and the development of complications related with the procedure was closely reviewed at the immediate postoperative period and the last follow-up. Results: It produces immediate relief in up to 98.4% of the patients. There were no development of complications related with procedure, all patients discharged one or two days after surgery. Conclusion: Vim thalamotomy without microelectrode recording is a safe and effective procedure to control the tremor with minimal morbidity. Intraoperative macroelectrode stimulation safely localizes the Vim nucleus target of the thalamus for the treatment of patients with tremor.

  • The electrocatalytic reduction of dioxygen is investigated by cyclic voltammetry and chronoamperometry at glassy carbon electrode and carbon microelectrode coated with a variety of cobalt phenylporphyrins. The n value obtained at carbon microelectrode is slightly different from that determined at glassy carbon electrode. Dioxygen reduction catalyzed by the monormeric porphyrin Co(II)-TPP mainly occurs through the $2e^-$ reduction pathway resulting in the formation of hydrogen peroxide, electrocatalytic process carries out $4e^-$ reduction pathway of dioxygen to $H_2O$ at the electrodes coated with bis-cobalt phenylporphyrins. The electrocatalytic reduction of dioxygen is irreversible and diffusion controlled.

Estimation of Oxygen Consumption Rates by Using an Oxygen Microelectrode in Ganghwa Intertidal Flat (강화도 남부 갯벌퇴적물에서 산소 미세전극을 이용한 산소소모율 추정)

  • We measured oxygen microprofiles using an oxygen microelectrode in Ganghwa intertidal flat in April and September. Oxygen consumption rate was calculated by using three different methods based on the oxygen microprofiles. The method using the PROFILE software was thought to be the most reliable among the three methods. The oxygen consumption rates calculated at station D1 by using the PROFILE software were 10.5 and 6.27mmol $m^{-2}d^{-1}$ in April and September, respectively. At station D2, they were 10.9mmol $m^{-2}d^{-1}$ in April and 5.39 mmol $m^{-2}d^{-1}$ in September. There was little spatial variation, but large seasonal variation, with almost two times larger values in April than in September. The higher rate in April is ascribed to higher oxygen concentration in the seawater and higher organic carbon content in the surface sediments, which probably accelerate oxygen consumption for organic matter decomposition in the sediments. Aerobic remineralization rates estimated from the oxygen consumption rates ranged from 4.14 to 8.07 mmol C $m^{-2}d^{-1}$ in Ganghwa intertidal flat, which were much lower than the anaerobic remineralization rate.

  • Microelectrodes for measuring nitrogenous compounds (${NH_4}^+$, ${NO_3}^-$) that were applied into the microfluidics chips was investigated, and the effect of temperature was especially examined. In this specific research, microelectrodes were first calibrated to check the function, and then microsensor that was combined microelectrode with microfluidic chip was re-calibrated. Experimental results showed that there are no change in the function between microelectrode and microfluidic chip. The electro motive force (EMF) for the ${NH_4}^+$ microsensor was similar to the one theoretically calculated from Nernst equation, but the EMF for ${NO_3}^-$ showed minor change.

Fabrication of Depth Probe Type Semiconductor Microelectrode Arrays for Neural Recording Using Both Dry and wet Etching of Silicon (실리콘 건식식각과 습식식각을 이용한 신경 신호 기록용 탐침형 반도체 미세전극 어레이의 제작)

  • 대뇌 피질에 삽입하여 깊이에 따라 신경 신호를 기록하기 위한 탐침형 반도체 미세전극 어레이(depth-type silicon microelectrode array, 일명 SNU probe)를 제작하였다. 붕소를 확산시켜 생성된 고농도 p-type doping된 p+ 영역을 습식식각 정지점으로 사용하는 기존의 방법과 달리 실리콘 웨이퍼의 앞면을 건식식각하여 원하는 탐침 두께만큼의 깊이로 트렌치(trench)를 형성한 후 뒷면을 습식식각하는 방법으로 탐침 형태의 미세 구조를 만들었다. 제작된 반도체 미세전극 어레이의 탐침 두께는 30 $\mu\textrm{m}$이며 실리콘 건식식각을 위한 마스크로 6 $\mu\textrm{m}$ 두께의 LTO(low temperature oxide)를 사용하였다. 탐침의 두께는 개발된 본 공정을 이용해서 5~90 $\mu\textrm{m}$ 범위까지 쉽게 조절할 수 있었다. 탐침의 두께를 보다 쉽게 조절할 수 있게 됨에 따라 여러 신경조직에 필요한 다양한 구조의 반도체 미세전극 어레이를 개발할 수 있게 되었다. 본 공정을 이용해서 개발된 4채널 SUN probe를 사용하여 흰쥐의 제1차 체감각 피질에서 4채널 신경 신호를 동시에 기록하였으며, 전기적 특성검사에서 기존의 탐침형 반도체 미세전극, 텅스텐 전극과 대등하거나 우수한 신호대 잡음비(signal to noise ratio, SNR)특성을 가짐을 확인하였다.

Bismuth Coated Carbon Fiber Microelectrode with Gallic Acid n-Propyl Ester for Trace Copper Analysis (비스무스코팅 탄소섬유전극과 갤릭산 착물을 사용한 구리 이온의 흔적량 분석)

  • A bismuth-coated carbon fiber microelectrode was prepared using cyclic voltammetry (CV). An analytical application was performed for the copper analysis with Square Wave Stripping Voltammetry (SWSV). Gallic acid n-propyl ester (PG) was used for the complex formation with a copper ion, and electrochemical measurements were performed with a pre-amplifier of a low-current module for nano am per detection. The effects of various parameters on the response were optimized. Analytical working ranges of $0.03-25.9\;{\mu}gl^{-1}$ and $0-25\'mgl^{-1}$ Cu(II) were obtained. The relative standard deviation at $13\;mgl^{-1}$ Cu was 0.9% (n = 12) in optimum conditions. The detection limit was found to have been $0.019\;{\mu}gl^{-1}$, with a 30-sec accumulation time. The developed methods were applied to a copper assay in water samples.

Electrochemical Gene Detection Using Microelectrode Array on a DNA Chip

  • In this study, a DNA chip with a microelectrode array was fabricated using microfabrication technology. Several probe DNAs consisting of mercaptohexyl moiety at their 5' end were immobilized on the gold electrodes by a DNA arrayer. Then target DNAs were hybridized and reacted with Hoechst 33258, which is a DNA minor groove binder and electrochemically active dye. Linear sweep voltammetry or cyclic voltammetry showed a difference between target DNA and control DNA in the anodic peak current values. It was derived from Hoechst 33258 and concentrated at the electrode surface through association with the formed hybrid. This suggested that this DNA chip could recognize the sequence specific genes.

  • In this paper we describe the status of a silicon-based microelectrode for neural recording and an advanced neural interface. We have developed a silicon neural probe, using a combination of plasma and wet etching techniques. This process enables the probe thickness to be controlled precisely. To enhance the CMOS compatibility in the fabrication process, we investigated the feasibility of the site material of the doped polycrystalline silicon with small grains of around 50 nm in size. This silicon electrode demonstrated a favorable performance with respect to impedance spectra, surface topography and acute neural recording. These results showed that the silicon neural probe can be used as an advanced microelectrode for neurological applications.
