서울 시내 산업체 급식소의 plastic용기 및 기구류 위생에 관한 연구 (The Microbiological Assessment of Plastic Container and Kitchen Utensils Used in Employee Feeding Foodservice Operation in Seoul)

  • 서울 시내 산업체 단체 급식소 3곳에서 이용되는 플라스틱 용기, 행주, 도마, 식칼의 미생물 검사결과는 다음과 같았다. 플라스틱 용기의 표준 평판균수는 3곳 모두 1$\times$$10^{*}$ 5 (CFU/100 $cm^2$)으로 높게 나타났고, 대장균군 수는 두 급식소는 양호하나 한 급식소에서 2$\times$$10^1$(CFU/100$cm^2$)으로 높게 나타났다 전자현미경 관찰 결과 구균, 간균 형태의 박테리아가 많이 관찰되었다 행주의 경우는 표준 평판균수는 1$\times$$10^3$-1 $\times$$10^{*}$ 5 (CFU/100 $cm^2$)으로 3곳 모두 높게 나타났고, 대장균군 수는 한곳을 제외한 두곳에서 4$\times$$10^1$-6$\times$$10^1$(CFU/100 $cm^2$)으로 높게 나타났다. 식칼의 표준 평판균수는 1$\times$$10^3$-l$\times$$10^{*}$ 5 (CFU/300$cm^2$)으로 3곳 모두 높게 나타났고, 대장균군 수는 한곳을 제외한 두곳에서 2$\times$$10^1$-3$\times$$10^1$(CFU/100 $cm^2$)으로 높게 나타났다. 도마는 표준 평판균수가 1$\times$$10^{*}$ 5 (CFU/100 $cm^2$)으로 3곳 모두 높게 나타났고 대장균군 수는 한곳을 제외한 두곳에서 4$\times$$10^1$-9)$\times$$10^1$(CFU/100 $m^2$)으로 높게 나타나 기구류의 보다 철저한 위생관리가 요구되어졌다.

Clinical and microbiological effects of adjunctive local delivery of minocycline (Periocline®) in patients receiving supportive periodontal therapy: a pilot study

  • Purpose: This study aimed to evaluate the clinical and microbiological efficacy of adjunctive local delivery of minocycline (Periocline®) in patients receiving supportive periodontal therapy (SPT) after initial treatment. Methods: The participants were 16 men and 8 women (age, 20-65 years) who had at least 15 natural teeth, underwent SPT for more than 1 year due to chronic periodontitis, had 4 or more periodontal pocket sites deeper than 5 mm, and showed >25% gingival bleeding on probing (BoP). They were randomly assigned to the test and control groups. In the test group, mechanical debridement and local antibiotic delivery were performed for all periodontal sulci/pockets; in the control group, mechanical debridement and saline irrigation were performed. In patients who underwent SPT for more than 1 year, clinical and microbiological examinations were performed at baseline and 1 and 3 months after SPT. The clinical examination included an assessment of the periodontal pocket depth, clinical attachment level, plaque index, and BoP. Microbial tests were performed using real-time polymerase chain reaction; the relative ratios of Porphyromonas gingivalis and Fusobacterium nucleatum were determined. Results: Both groups showed significant improvements in clinical parameters at 1 and 3 months from baseline; there were no significant changes between months 1 and 3. Intergroup differences were insignificant. The microbiological analysis revealed no significant differences in P. gingivalis and F. nucleatum ratios across time points. While intergroup differences were insignificant, there was a tendency for the P. gingivalis and F. nucleatum ratios to decrease in the test group. Conclusions: Mechanical debridement in patients receiving maintenance therapy resulted in clinically significant improvement; the effectiveness of additional local delivery of antibiotics was not significant. The ratios of P. gingivalis and F. nucleatum showed a tendency to decrease in the test group, although it was not significant.

메타분석에 의한 식중독 원인 미생물들의 최소감염량 분석 (The Analysis for Minimum Infective Dose of Foodborne Disease Pathogens by Meta-analysis)

  • 본 연구는 정량적 미생물 위해평가(Quantitative microbial risk assessment: QMRA)에 절대적으로 필요한 9종의 식중독 세균, 2종의 바이러스, 1종의 원생동물에 대한 최소 감염량(minimum infective dose)을 선정한 연구이다. 주요 식중독 미생물들의 최소 감염량을 선정하기 위하여, 1980년부터 2012년까지 PubMed, ScienceDirect database 등에서 주요 식중독 미생물들의 최소 감염량 및 위해평가 자료 82종을 수집하였다. 수집된 자료는 메타분석(mata-analysis)에서 사용되고 있는 relative frequency(fi, 상대빈도 값)를 계산하여 가장 적정한 최소 감염량을 추정 및 선정하였다. 주요 식중독 미생물들의 최소 감염량은, B. cereus $10^5cells/g$ (fi = 0.32), C. jejuni 500 cells/g (fi = 0.57), Cl. perfringens $10^7cells/g$ (fi = 0.56), Pathogenic E. coli 중 EHEC 10 cells/g (fi = 0.47), ETEC $10^8cells/g$ (fi = 0.71), EPEC $10^6cells/g$ (fi = 0.70), EIEC $10^6cells/g$ (fi = 0.60), L. monocytogenes $10^2{\sim}10^3cells/g$ (fi = 0.23), Salmonella spp. 10 cells/g (fi = 0.30), Shigella spp. 100 cells/g (fi = 0.32), S. aureus $10^5cells/g$ (fi = 0.45), V. parahaemolyticus $10^6cells/g$ (fi = 0.64), Hepatitis A virus $10{\sim}10^2particles/g$ (fi = 0.33), Noro virus 10 particles/g (fi = 0.71), C. pavum $10{\sim}10^2oocyst/g$ (fi = 0.33)으로 나타났다. 본 연구결과는 향후 국내 QMRA를 통한 위해수준 추정결과의 정확성을 향상시키는데 기여할 수 있을 것으로 기대된다.

주요 식중독 원인 미생물들에 대한 용량-반응 모델 연구 (A Study on Dose-Response Models for Foodborne Disease Pathogens)

  • 본 연구는 정량적 미생물 위해평가(Quantitative microbial risk assessment: QMRA)에 절대적으로 필요하지만 국내의 경우 관련 정보 및 자료가 부족한 주요 식중독 원인 미생물에 대한 용량-반응모델(dose-response models) 관련 자료를 수집 정리하여 가장 적합한 용량-반응 모델을 분석 및 선정하였다. 1980년부터 2012년까지 식중독 발생과 관련이 있는 26종의 세균, 9종의 바이러스, 8종의 원생동물관련 용량-반응 모델 및 위해평가 자료들을 중심으로 국내 NDSL (National Digital Science Library), 국외 PubMed, ScienceDirect database에서 총 193개의 논문을 추출하여 정리하였다. 조사된 자료로부터 세균별, 바이러스별, 원생동물별 용량-반응 모델의 미생물 위해평가 활용여부를 확인하고, 위해평가에 활용된 모델들을 메타분석(meta-analysis)에서 사용되고 있는 Relative frequency (fi, 상대빈도 값)를 계산하여 가장 적정한 용량-반응 모델을 제시하였다. 주요 식중독 원인 미생물들인 Campylobacter jejuni, pathogenic E. coli O157:H7 (EHEC / EPEC / ETEC), Listeria monocytogenes, Salmonella spp., Shigella spp., Staphylococcus aureus, Vibrio parahaemolyticus, Vibrio cholera, Rota virus, Cryptosporidium pavum의 적정 용량-반응 모델은 beta-poisson (${\alpha}=0.15$, ${\beta}=7.59$, fi = 0.72), beta-poisson (${\alpha}=0.49$, ${\beta}=1.81{\times}10^5$, fi = 0.67) / beta-poisson (${\alpha}=0.22$, ${\beta}=8.70{\times}10^3$, fi = 0.40) / beta-poisson (${\alpha}=0.18$, ${\beta}=8.60{\times}10^7$, fi = 0.60), exponential ($r=1.18{\times}10^{-10}$, fi = 0.14), beta-poisson (${\alpha}=0.11$, ${\beta}=6,097$, fi = 0.09), beta-poisson (${\alpha}=0.21$, ${\beta}=1,120$, fi = 0.15), exponential ($r=7.64{\times}10^{-8}$, fi = 1.00), beta-poisson (${\alpha}=0.17$, ${\beta}=1.18{\times}10^5$, fi = 1.00), beta-poisson (${\alpha}=0.25$, ${\beta}=16.2$, fi = 0.57), exponential ($r=1.73{\times}10^{-2}$, fi = 1.00), and exponential ($r=1.73{\times}10^{-2}$, fi = 0.17)로 각각 선정하였다. 본 연구에서 제시된 용량-반응 모델들은 향후 국내 QMRA 관련 연구 및 진행에 많은 도움이 될 것으로 기대된다.

형광분광측정법에 의한 항산균의 생명력 평가 (Assessment of Mycobacterial Viability by Fluorospectrophotometry)

  • 나균(M. leprae)을 비롯한 항산균(mycobacterium)의 시험관내 생리력 또는 생명력평가법(in vitro assessment of physiological potential or viability)으로 형광분광측정법(fluorospectrophotometry)를 고안하였다. 균액을 비형광성의 fluorescein diacetate(FDA)로 처리, 균체의 생대사능에 의해 FDA로부터 유리된 체내 fluorescein 량을 Aminco-Bowman 형광분광기로 측정함으로 균체의 생리능을 fluorounit로 표기해 보았다. Fluorounit로 표기된 균체의 생명력을 균배양의 광학밀도(optical density, colony forming unit, 체내 ATP 량 intracellular ATP content)등으로 표기되는 항산균의 생명력고 비교 검토함으로 형광분광법에 의한 시험관내 항산균의 생명력 평가법의 적합성을 살펴보았다. 형광분광측정에 의한 생명도의 평가는 객관성을 띤 기기정량법으로 조작이 간단하고 신속하게 결과를 얻을 수 있어 시험관내 배양의 속도가 완만한 균이나(slow growing mycobacteria, I.e.M. lerpae, Listeriae sp)등의 생명력의 상대적 평가에 활용될 수 있다고 사료된다.

Safety Assessment of Potential Lactic Acid Bacteria Bifidobacterium longum SPM1205 Isolated from Healthy Koreans

  • The safety assessment of Bifidobacterium longum SPM1205 isolated from healthy Koreans and this strain's inhibitory effects on fecal harmful enzymes of intestinal microflora were investigated. The overall safety of this strain was investigated during a feeding trial. Groups of SD rats were orally administered a test strain or commercial reference strain B. longum $1\times10^9\;CFU/kg$ body weight/day for four weeks. Throughout this time, their feed intake, water intake and live body weight were monitored. Fecal samples were periodically collected to test harmful enzyme activities of intestinal microflora. At the end of the four-week observation period, samples of blood, liver, spleen, kidney, and gut tissues were collected to determine for hematological parameters and histological differences. The results obtained in this experiment demonstrated that four weeks of consumption of this Bifidobacterium strain had no adverse effects on rat's general health status, blood biochemical parameters or histology. Therefore, it is likely to be safe for human use. Fecal harmful enzymes such as $\beta-glucosidase,\;\beta-glucuronidase$, tryptophanase and urease, were effectively inhibited during the administration of the B. longum SPM1205. These results suggested that this B. longum SPM 1205 could be used for humans as a probiotic strain.

Microplate Reader를 이용하여 측정한 혈액의 엽산 농도와 실측량 기록법에 의한 엽산 섭취량 (Blood Folate Level Determined by a Microplate Reader and Folate Intake Measured by a Weighted Food Record)

  • Microbiological method using a 96-well microplate reader for folate assay was established, and folate intake and blood folate concentrations of 23 female college students were assessed. To evaluate folate intake, dietary data were collected by a 3-day weight food record, and serum and RBC folate concentrations were measured by the new method. The coefficient of variation for the new method was less than 10%. Mean daily folate intake of the subjects was 126.7${\mu}g$ which is only 50.7% of the RDA. The mean concentrations of serum and RBC folate were 7.46ng/ml and 294.4ng/ml, respectively, which were within the normal range. These results indicate that folate intake seems to be underestimated due to incomplete food composition database. Therefore, folate database should be appropriately in order to asses folate intake accurately.

Comparative Activities of Novel $\beta$-Lactamase Inhibitors, 6-Exomethylene Penamsulfones (CH1240, CH2140) in Experimental Mouse Infection Model

  • The antibacterial activity of novel ${\beta}$-lactamase inhibitors, 6-exomethylene penamsulfones (CH1240, CH2140), has been compared in vivo with that of sulbactam and clavulanic acid against b-lactamase producing strains. In vivo microbiological assessment was used as experimental mouse infection model by gram negative strains. Against Pseudomonas aeruginosa F0013, cefoperazone/CH 1240 was slightly less active than sulbactam. ampicillin/CH 2140 was less effective than sulbactam against escheriachia coli 3457. Especially against Citrobacter diversus 2046E, amoxicillin/CH 2140 was the most potent and amoxicillin/CH 1240 was slightly more active than clavulanic acid. consequently the difference in efficacy between the drug combinations appers to be related to the degree of protection afforded the animals by the b-lactamasse inhibitors. CH1240 and CH2140 are promising new agents and should undergo further investigations.

조미김의 HACCP 시스템 구축을 위한 생물학적 위해도 평가 연구 (A Study on Microbiological Risk Assessment for the HACCP System Construction of Seasoned Laver)

  • Objectives: The purpose of this study was to apply the Hazard Analysis Critical Control Point (HACCP) system to the production of seasoned laver products. The hazard analysis examined microbial evaluations and developed a HACCP management plan through the heating process. Methods: In this study we chose three companies and performed the analysis thrice. During this study, general bacteria along with other food poisoning bacteria such as Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus, E.coil, O157:H7, Vibrio parahaemolyticus, were studied at varying temperatures from 100 to $300^{\circ}C$. Results: The presence of general bacteria was detected in raw laver in the samples analyzed from all the three companies, and the number ranged from $10^5-10^7$. Bacillus cereus was detected in samples from only two of the three companies analyzed. However, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, E.coil, O157:H7, and Vibrio parahaemolyticus were all negative. General bacteria was reduced to $10^5$ after being subjected to temperatures of $100-250^{\circ}C$, but heating to over $270^{\circ}C$ reduced the number to below $10^3$, and the other microbes such as Bacillus cereus were not detected. Conclusions: In conclusion, the heating process ($270-280^{\circ}C$) along with RPM of 100-1200 were identified as CCP to reduce biological hazards.

한국형 건강편의식 개발을 위한 두부조림의 Cook/Chill 생산 및 포장방법에 따른 품질 평가 (Quality Assessment of Cook/chilled Soy Sauce Glazed Soybean Curd Packaged with Different Methods for the Development of Health-oriented Convenience Foods)

  • The purpose of this study was to assess the qualities of cook/chilled soy sauce-glazed soybean curd with various packaging conditions as a trial to develop health-oriented convenience foods. The effects of three packaging methods, linear low density-polyethylene (LLD-PE), top sealing, and modified atmosphere packaging(MAP) on the shelf-life of HACCP-based cook/chilled soy sauce-glazed soybean curd were evaluated during 20 days of chilled storage in terms of time-temperature, microbiological (total aerobic plate count, psychrotrophic plate count, coliform, and fecal coliform count), chemical(pH and peroxide value(POV)), and sensory evaluation. The results of microbiological and chemical analyses were within the limits of the microbial and chemical standards for all phases after cooking. No significant differences were detected in microbial counts of the samples for all three packaging methods. However, sensory evaluation indicated that the top sealing and MAP methods showed a longer shelf-life than LLD-PE packaging. Recommended shelf-life of the product was 12 days for LLD-PE packaging, and 16 days for both top sealing and MAP. In conclusion, MAP was considered as the most effective packaging method for assuring microbial and sensory quality of this cook/chilled product.
