Different statistical distribution functions were examined to find an adequate distribution function to describe the microbial contamination behavior of a Korean side dish product, seasoned soybean sprouts for different seasons and market groups. The triang distribution was the best for any market groups in winter, while the logistic distribution could describe the microbial contamination in log CFU/g for all the market groups in spring and summer. From parametric bootstrapping based on the fitted distributions, it was found that a normal distribution could describe the distribution of mean microbial count in log CFU/g for all the seasons and market groups. Statistical parameters for each season/market group are presented to estimate the confidence interval.
To determine structure and activities of microbial communities in a food wastewater treatment system, biofilm of RABC (rotating activated Bacillus contactor) and samples of aeration tanks were analyzed. Heterotrophic bacterial concentrations were similar between biofilm and stage 1 aeration tank and decreased 2-log at stage 3 aeration tank as dissolved oxygen decreased, however portions of Bacillus groups were increased at stage 3 aeration tank. It was revealed by quantitative and qualitative analysis of metabolic fingerprinting patterns of Biolog GN2 plate that RABC represented much higher activities and a different microbial community structure compared to aeration tanks. Metabolic fingerprinting showed the carbon sources that isolated Bacillus groups could or could not use, were used similarly meaning that not only Bacillus groups but also other microbial groups would contribute to the treatment of wastewater.

Permafrost is frozen soil below $0^{\circ}C$ for two or more years. Surface of permafrost is called as active layer that seasonally thaws during the summer. Although the thawing of permafrost may deepen the active layer and consequently increase the microbial activity, the microbial community structure in this habitat has not yet been well described. In this study, we presented bacterial and archaeal diversity in the active layer soil from Resolute, Canada using pyrosequencing analysis. The soil sample was collected from the surface of the marsh covered with moss and Carex. A total of 7,796 bacterial reads for 40 phyla and 245 archaeal reads for 4 phyla were collected, reflecting the high diversity of bacteria. Predominant bacterial groups were Proteobacteria (37.7%) and Bacteroidetes (30.0%) in this study. Major groups in Archaea were Euryarchaeota (51.4%) and Thaumarchaeota (46.1%). Both methane producing archaea and consuming bacteria were detected in this study. Although it might be difficult to characterize microbial community with only one sample, it could be used for the basis of assessing the relative importance of the specific groups with a high resolution on the bacterial and archaeal community in this habitat.
The present study investigated the effect of ammonia load on microbial communities in mesophilic anaerobic digestion of propionic acid. A laboratory-scale continuous anaerobic digester treating propionic acid as a sole organic substrate was operated under non-inhibitory condition and inhibitory conditions with ammonia (1.5 g and 3.5 g ammonia-N/L, respectively), and bacterial and archaeal communities in the steady states of each ammonia condition were analyzed using high-throughput sequencing. Thirteen bacterial families were detected as abundant bacterial groups in mesophilic anaerobic digestion of propionic acid. Increase in ammonia concentration resulted in significant shifts in microbial community structures. Syntorophobacter, Pelotomaculum, and Thermovigra were determined as the dominant groups of (potential) propionate oxidizing bacteria in the non-inhibitory condition, whereas Cryptanaerobacter and Aminobacterium were the dominant groups of (potential) propionate oxidizing bacteria in the ammonia-inhibitory condition. Methanoculleus and Methanosaeta were the dominant methanogens. Acetate-oxidation coupled with hydrogenotrophic methanogenesis might be enhanced with increases in the relative abundances of Methanoculleus and Tepidanaerobacter acetatoxydans under the ammonia-inhibitory condition. The results of the present study could be a valuable reference for microbial management of anaerobic digestion systems that are exposed to ammonia inhibition and propionic acid accumulation.
Objective: The objective of this study was to investigate how cellulase or/and lactic acid bacteria (LAB) affected the fermentation characteristic and microbial community in wet brewer's grains (WBG) and corn stover (CS) mixed silage. Methods: The WBG was mixed thoroughly with the CS at 7:3 (w/w). Four treatment groups were studied: i) CON, no additives; ii) CEL, added cellulase (120 U/g fresh matter [FM]), iii) LAB, added LAB (2×106 cfu/g FM), and iv) CLA, added cellulase (120 U/g FM) and LAB (2×106 cfu/g FM). Results: All additive-treated groups showed higher fermentation quality over the 30 d ensiling period. As these groups exhibited higher (p<0.05) LAB counts and lactic acid (LA) content, along with lower pH value and ammonia-nitrogen (NH3-N) content than the control. Specifically, cellulase-treated groups (CEL and CLA) showed lower (p<0.05) neutral detergent fiber and acid detergent fiber contents than other groups. All additives increased the abundance of beneficial bacteria (Firmicutes, Lactiplantibacillus, and Limosilactobacillus) while they decreased abundance of Proteobacteria and microbial diversity as well. Conclusion: The combined application of cellulase and LAB could effectively improve the fermentation quality and microbial community of the WBG and CS mixed silage.
숙주 동물과 동물의 장내 미생물의 건강 또는 생산성에 대한 연구결과를 미루어 볼 때, 가축 동물의 장내 미생물에 대한 연구는 매우 중요하다. 본 연구는 국내에서 사육되는 젖소 중 홀스타인 종과 저지종 젖소의 장내 미생물을 분석하고 차세대 염기서열 분석을 통해 젖소 종에 따른 장내 미생물 군집 구조의 차이를 규명하고자 하였다. 젖소의 원유 생산과 관련있는 것으로 알려진 종 풍부도와 종 다양성 지수 분석 결과 대부분의 풍부도 및 다양성 지수가 홀스타인 종 보다 저지 종에서 유의한 수준으로 높은 것으로 나타났으나, 종 간의 계통학적 거리를 합산하여 산출되는 phylogenetic diversity 지수는 낮은 것으로 나타났다. 미생물 분포 분석 결과 홀스타인과 저지 종의 두 집단 장내 미생물 군집 구조가 다른 것으로 나타났다. 두 종의 젖소에서 과(family) 수준의 다양한 장내 미생물간의 분포에 상관관계가 있는 것으로 나타났으며, 특히 저지 종의 장내 미생물은 다양한 미생물 분포 사이에 매우 유의한 수준의 상관관계가 있는 것으로 나타났다. 두 종의 젖소 장내 미생물 구조에 차이가 있는지 확인하기 위해 beta-diversity 분석을 수행하였으며, PCoA 분석과 UPGMA clustering 분석 결과 두 그룹의 cluster가 명확히 분리되는 것을 시각적으로 확인하였으며, PERMANOVA 분석 결과 두 종의 장내 미생물 군집 구조가 통계적으로 매우 유의한 수준의 차이가 있는 것으로 나타났다. 두 젖소 종의 장내 미생물 군집 구조 차이에 기여하는 미생물을 확인하기 위해 LEfSe 분석을 수행하였으며, 그 결과 Firmicutes, Bacilli, Moraxellaceae, Pseudomonadales 등의 상대적인 미생물 분포 차이가 두 그룹간 장내 미생물 군집 구조 차이에 가장 큰 영향을 미치는 것으로 나타났다.
본 연구는 우리나라 전통 장류인 된장과 청국장의 미생물 분포와 시료간의 미생물학적 차이에 대해 차세대 염기서열 분석법(NGS)을 이용하여 분석하였다. α-diversity 분석 결과 된장에서 종 추정치와 풍부도가 통계학적으로 유의한 수준으로 높은 것으로 나타났다. 세균 분포를 분석한 결과 문 수준에서 Firmicutes가 된장에서 97.02%, 청국장에서 99.67%를 차지하여 공통적으로 가장 우점하는 것으로 나타났으며, 속 수준에서는 된장과 청국장에서 Bacillus가 각각 71.70%, 59.87%를 차지하여 가장 우점하는 것으로 확인되었다. 된장과 청국장의 미생물 분포에 차이가 있는지 분석하기 위해 PERMANOVA 분석을 수행한 결과 된장과 청국장의 미생물 분포에 통계학적으로 유의한 수준으로 차이가 나는 것으로 나타났다. 각 된장과 청국장 미생물 군집을 대표하는 biomarker를 분석하기 위해 LEfSe분석을 수행한 결과 된장에서 Bacillus subtilis, Tetragenococcus halophilus, Clostridium arbusti가 상대적으로 많이 분포하였으며, 청국장에서는 Bacillus thermoamylovorans, Enterococcus faecium, Lactobacillus sakei가 상대적으로 많이 분포하는 것으로 나타났다. 본 연구를 통해 콩을 주원료로 하는 우리나라 대표 전통장류인 된장과 청국장의 시료별 유사성과 차이점에 대한 미생물 분포를 정의하고 전통장류의 생화학적, 생리학적 특성과 미생물 분포의 상관관계를 규명하기 위한 기초 연구자료로 활용할 수 있을 것으로 기대된다.
Objectives: This study aims to correlate caries-causing microorganism load, lactic acid estimation, and blood groups to high caries risk in diabetic and non-diabetic individuals and low caries risk in healthy individuals. Materials and Methods: This study includes 30 participants divided into 3 groups: Group A, High-risk caries diabetic individuals; Group B, High-risk caries non-diabetic individuals; and Group C, Low-risk caries individuals. The medical condition, oral hygiene, and caries risk assessment (American Dental Association classification and International Caries Detection and Assessment System scoring) were documented. Each individual's 3 mL of saliva was analyzed for microbial load and lactic acid as follows: Part I: 2 mL for microbial quantity estimation using nutrient agar and blood agar medium, biochemical investigation, and carbohydrate fermentation tests; Part II: 0.5 mL for lactic acid estimation using spectrophotometric analysis. Among the selected individuals, blood group correlation was assessed. The χ2 test, Kruskal-Wallis test, and post hoc analysis were done using Dunn's test (p < 0.05). Results: Group A had the highest microbial load and lactic acid concentration, followed by Groups B and C. The predominant bacteria were Lactobacilli (63.00 ± 15.49) and Streptococcus mutans (76.00 ± 13.90) in saliva. Blood Group B is prevalent in diabetic and non-diabetic high-risk caries patients but statistically insignificant. Conclusions: Diabetic individuals are more susceptible to dental caries due to high microbial loads and increased lactic acid production. These factors also lower the executing tendency of neutrophils, which accelerates microbial accumulation and increases the risk of caries in diabetic individuals.
This study aimed to evaluate the effects of dietary microbial-fermented molasses on egg production and egg quality in laying hens.In total, 90 Hy-line Brown laying hens were divided into two treatment groups (control and 1% microbial-fermented molasses)with three replicates of 15 birds each. During the experimental period, supplementation of hen diets with 1% microbial-fermented molassesdid not influence egg weight, hen-day egg production, egg mass, and feed conversion ratio (p > 0.05), except for feed intake. Regarding egg quality, diets containing 1% microbial-fermented molasses significantly affected eggshell thickness, Haugh unit, and albumen height (p < 0.05). However, there were no remarkable differences between control and 1% microbial-fermented molasses in eggshell color and egg yolk color (p > 0.05). These results indicate that supplementing 1% microbial-fermented molasses to the diet of laying hens improved egg quality parameters such as eggshell thickness, Haugh unit, and albumen height rather than egg production.
우리나라 전통 콩 발효식품은 탄수화물을 주식으로 하는 한국인의 식생활에 중요한 단백질 급원임에도 불구하고 콩 발효식품의 미생물 다양성과 군집 구조에 대해서는 거의 알려진 바가 없다. 본 연구는 16S rDNA 유전자 서열 분석 기반의 차세대 염기서열 분석법을 이용하여 한국 전통 발효식품인 된장과 간장의 미생물 군집 구조를 밝히고자 하였다. Alpha-diversity 분석 결과 미생물 다양성 지표인 Shannon과 Simpson에서 된장과 간장의 미생물 다양성에 통계학적인 차이가 있는 것으로 나타났으나, 종 풍부도 지표인 ACE, CHAO, Jackknife에서는 차이가 없는 것으로 나타났다. 된장과 간장의 미생물 분포 분석 결과 된장과 간장의 공통적인 우점균은 Firmicutes로 나타났으나, 속 수준에서의 미생물 분포를 분석한 결과 된장에서 Bacillus, Kroppenstedtia, Clostridium, Pseudomonas가 간장보다 높은 비율을 차지하고 있는 것으로 나타났으며, 간장에서는 Tetragenococcus, Chromhalobacter, Lentibacillus, Psychrobacter와 같은 호염성 또는 내염성 세균이 된장보다 높은 비율을 차지하는 것으로 나타났다. 된장과 간장의 미생물 군집구조에 통계학적인 차이가 있는지 확인하기 위해 paired-PERMANOVA 분석을 수행하였으며, 그 결과 통계학적으로 매우 유의한 수준의 차이가 있는 것으로 나타났다. 된장과 간장의 미생물 군집구조 차이에 큰 영향을 미치는 biomarker를 분석하기 위해 LEfSe 분석을 수행하였으며, 그 결과 Bacillus와 Tetragenococcus가 된장과 간장의 미생물 군집 구조에 차이를 나타내는 biomarker로 분석되었다.
