다공질 지르코니아 세라믹스의 기공율 제어 (Porosity Control of Porous Zirconia Ceramics)

  • A simple pressing process using zirconia and microbead for fabricating porous zirconia ceramics is demonstrated. Effects of microbead content and sintering temperature on microstructure, porosity, compressive and flexural strengths were investigated in the processing of porous zirconia ceramics using microbead as a pore former. By controlling the microbead content and the sintering temperature, it was possible to produce porous zirconia ceramics with porosities ranging from 43% to 70%. Typical compressive and flexural strength values at ${\sim}50%$ porosity were ${\sim}150\;MPa$ and ${\sim}35\;MPa$, respectively.

  • We present a microfluidic chip designed for the detection of antibody by using quantum dots fluorescence and a microbead-based assay. A custom designed PDMS microfluidic chip with multi-layer channel is utilized for capturing microbeads; antibody injection into each micro-well; QD injection; and fluorescence detection. The experiment using the fabricated microfluidic chip has been performed on solutions with various concentrations of antibody and has shown correlated fluorescent intensities.

스크럽 화장품의 사용 전과 후의 피부상태 비교와 이에 첨가된 마이크로비드의 현미경적 특성 (Comparison of Skin Condition Before and After Use of Scrub Cosmetics and Microscopic Characteristics of Microbeads)

  • 본 연구는 스크럽 화장품을 사용하여 피부상태의 변화를 피부분석장비로 분석하고 마이크로비드의 형태적 특징을 더마스코프와 주사전자현미경으로 관찰하였다. 더마스코프 관찰에서 클렌징을 하는 과정 중의 얼굴피부에 붙어있는 마이크로비드는 서로 밀착되어 존재하고 있거나 분산되어 있었다. 클렌징 한 피부는 깨끗하고 매끄럽게 관찰되었으며 표피 각질세포들 사이의 미세한 주름이 줄어든 것을 확인하였다. 주사전자현미경 관찰에서 구슬모양의 마이크로비드 표면은 심한 굴곡이나 거친 표면을 가지고 있지 않았다. 스크럽을 하기 전과 후의 유분, 수분, pH의 변화를 비교분석한 결과 스크럽을 하기전의 피부가 스크럽 후의 피부보다 수분과 유분의 함량이 높게 나타났고 pH의 변화는 큰 차이가 없었다.

Gray-scale photolithography를 이용한 바이오칩 제작 (Fabrication of Biochip Using Gray-scale Photolithography)

  • Biochip, which implements bioanalytical process on a tiny surface, is one of candidates for medical diagnosis, drug screening, and molecular sensing. In general, a type of biochip based on microfluidics is composed of microcomponents including microchannel, pump, and valve, which require complicated processes. In this study, gray-scale photolithography(GSPL) was applied to fabricate a biochip with multiple layers. A mould for casting PDMS(polydimethylsiloxane) channel, was fabricated using GSPL. A gray-photomask was prepared by printing gray patterns on a high-quality glossy paper followed by photoreducing by 10:1 onto the photo-film. The formation of multiple layers was studied according to the change of gray level of pattern and the developing time. A biochip composed of a weir(multiple layer structure) and a reaction chamber in a single microchannel was fabricated in a glass plate. Finally, we investigated the application of biochip to antigen-antibody reaction by packing the microbead coated with antibody.

Effect of pH on Swelling Property of Hyaluronic Acid Hydrogels for Smart Drug Delivery Systems

  • Hyaluronic acid(HA) hydrogels were synthesized by immersing HA microbeads in phosphate buffered saline solutions having different pH levels to assess the effect of pH on the swelling ratio of HA hydrogels for smart drug delivery systems. No beads were formed when the HA solution(below pH 9) was crosslinked with divinyl sulfone(DVS) because DVS is a basic solution. The variation regarding the size of the microbead was not significant, suggesting that the bead size is not a function of pH(10 ~ 14). However, the pore size of the microbeads decreased with increasing pH from 10 to 14, leading to the surface smoothness and dense network as a result of higher crosslinking. The swelling ratio of hydrogels increased when the pH rose from 2(acidic) to 6(neutral). Afterwards, it decreased with further increasing pH(basic). The lower swelling ratio may be due to the lack of ionization of the carboxyl groups. On the other hand, a higher swelling ratio is likely due to the increased electrostatic repulsions between negatively charged carboxyl groups on different chains. Experimental results suggested that pH-responsive HA hydrogels can be applicable to the controlled drug delivery systems.

양자점이 고밀도화된 마이크로 비드의 제조 및 특성 (The Synthesis and Characterization of Mesoporous Microbead Incorporated with CdSe/ZnS QDs)

  • The spherical mesoporous silica is synthesized and incorporated with CdSe/ZnS quantum dots(QDs) for preparing micro beads to detect toxic and bio-materials with high sensitivity. The spherical silica beads with the brunauer-emmett-telle(BET) average pore size of 15 nm were prepared with a ratio 1, 3, 5-trimethylbenzen, as a swelling agent, to the block-copolymer template surfactant of over 1 and under vigorous mixing condition. The surface of spherical mesoporous silica is modified using octadecylsilane for incorporating QDs. Based on photoluminescence(PL) spectra, the relative brightness of mesoporous silica beads incorporated with 10 nM of QDs is 79,000 times brighter than that of Rodamine 6 G.

  • Hyaluronic acid(HA) microbeads were synthesized by dropping the sodium hyaluronate(Streptococcus) solutions in NaOH into a solution mixture of divinyl sulfone(DVS) in 2-methyl-1-propanol, followed by stirring, cleaning and drying process at room temperature. The initial experimental conditions are crosslinking time(CLTi) of 5 h, crosslinking temperature(CLTe) of room temperature, injection air pressure(IAPr) of 5 psi, and DVS concentration( DVSc) of 0.2 vol%, respectively. Then, parametric studies were performed by varying the parameters to investigate the morphology, the porosity, the swelling ratio and the size of the beads. The microbead size pattern was not regular to function of the degree of crosslink. It was observed that the swelling ratio, the degree of crosslink, and the pore size can be controlled by adjusting the CLTi, CLTe and DVSc. Among the parameters investigated, the smallest bead size can be achieved by varying the CLTi parameter. The lowest swelling ratio, as an indication of the highest degree of crosslink, can be obtained by varying CLTe.

프릿을 이용한 다공질 SiC 세라믹스의 저온 제조 공정 및 물성 (Low Temperature Processing and Properties of Porous Frit-Bonded SiC Ceramics)

  • Porous frit-bonded SiC ceramics were successfully prepared at a temperature as low as $800^{\circ}C$ from SiC, frit, and microbeads (glass or polymer). The effects of SiC starting particle size and microbead addition on microstructure, porosity, and flexural strength were investigated. The addition of hollow glass microbead improved the strength of frit-bonded SiC ceramics without the loss of porosity by acting additional binder phase between SiC grains. The 65 ${\mu}m$-sized SiC resulted in lower porosity and higher strength than 50 ${\mu}m$-sized SiC because of higher packing density. Typical flexural strengths of frit-bonded SiC were 23 MPa at 46% porosity and 19 MPa at 49% porosity.

전기 검출 시스템을 이용한 Microfluidic Immuno-Sensor Chip (Microfluidic Immuno-Sensor Chip using Electrical Detection System)

  • Bio-MEMS를 기반으로 microfilter와 백금 전극이 내재되어 있는 microbiochip을 제작하였다. 우리는 이 chip으로 microbead에 indirect ELISA 방법으로 항원-항체를 반응시키고 전기 신호 검출 방법을 이용하여 반응 여부를 판단하였다. 이 때 신호 증폭을 위해 silver enhancer를 사용하였다. Chip 상에서 항원-항체 반응 조건을 최적화하기 위해 pH, temperature, flow time, flow rate, silver enhancer time을 결정하였다. 이렇게 최적화된 조건을 바탕으로 짧은 시간 안에 소량의 시료로 immunoassay를 성공적으로 수행할 수 있었다. 전기 신호 검출 방식을 사용함으로써 biosensor 장비의 소형화와 다중 시료 측정과 자동화를 biosensor에서 적용할 수 있는 가능성을 제시하였다.

Chitosan Microbeads에 의한 효소고정화 (Immobilization of an Enzyme with Chitosan Microbeads)

  • Chitosan의 산업적 응용분야 중 효소의 고정화담체로서의 이용 가능성을 확인하기 위한 방법으로 알코올 발효공업에서 다량으로 사용되어지고 있는 담화효소인 amyloglucosidase를 공유결합에 의한 고정화를 시도, 여러가지 반응조건에서의 활성변화를 free enzyme과 비교해 보았으며, 생산공정에서 사용되어지고 있는 액화전분용액에 대한 반응을 실시하여 효소고정화를 위한 담체로서의 chitosan 적용가능성을 검토한 결과, 제조되어진 chitosan bead의 직경은 약 1.2mm 정도였으며 고정화시에 AMG의 농도 6mg/ml가 최적의 반응액농도로서 더 이상의 농도 증가에도 불구하고 거의 일정한 수준의 고정화율을 나타내었으며 pH 4.2인 0.01 M sodium acetate 완충액, 반응온도 $55^{\circ}C$, 15min의 조건에서 free enzyme에 대한 상대효소활성도는 $97.8\%$에 달하였고, 고정화효소의 효소활성의 최적온도조건은 $55^{\circ}C$로서 free enzyme의 그것과 동일한 것으로 나타났다. 고정화효소의 최적 활성조건은 pH 4.2로서 free enzyme과 동일하였으며 중성 이후의 영역에서는 free enzyme에 비하여 상대적으로 높은 효소활성을 나타내었고, 열에 대한 내성에 있어서, 가열온도 $30^{\circ}C$를 기점으로 free enzyme에 비하여 상대적으로 소폭 증대되어진 활성을 가지고 있는 것으로 나타났다. 알코올발효용 액화전분용액에 대한 free enzyme 및 고정화효소의 반응시, 두 시료 모두 $55^{\circ}C$에서 최대의 활성을 나타내었으나 고정화효소의 free enzyme에 대한 상대활성도는 $62.6\%$ 였다.
