Photosynthesis and respiration rate of microalgae are important factors during advanced wastewater treatment research using microalgae, There are several equipments and measurement methods for measuring photosynthesis and respiration, with different challenges that occur during pretreatment and stabilization of the analysis process. Therefore, in this study, for accurate Photosynthesis and Respiration (P&R) analyzer measurement, the analysis process was divided into pre-processing, DO stabilization, and analysis stages and each was optimized to enable accurate evaluation. For this purpose, the effect of DO saturation of the sample on P&R analysis, DO stabilization according to the degassing flow rate, and photoinhibition of the OD level on photosynthesis was investigated. Based on our study results, when DO was supersaturated, photosynthetic efficiency decreased due to photorespiration, making it inappropriate as a P&R sample. In addition, 0.5 L-N2/min level was the optimal nitrogen degassing flow rate for DO desaturation. The inhibition of photosynthetic efficiency by self-shading caused by the increase in OD was observed from OD 2.0, and it was found that P& R analysis is preferably performed on samples with OD less than 2.0. In addition, based on the above three optimization results, an optimized P&R Analyzer instruction for accurate P&R analysis was also presented.
본 연구에서는 해수담수화의 효율을 높이기 위한 효율적인 전처리 기법을 찾기 위해 응집제 투입과 같이 마이크로 공기를 주입하여 전처리 효율을 측정하였다. $AlCl_3{\cdot}6H_2O$와 PGA 응집제 주입량에 따른 탁도 제거효율을 알아보았다. 탁도 유발 물질은 해저퇴적토와 해양미세조류를 선정하여 실험에 필요한 탁도를 구현하였다. 퇴적토 함유 인공해수는 $AlCl_3{\cdot}6H_2O$ 0.5g/L 주입 시 제거율 73.7%를 얻은 반면 PGA 경우 0.05g/L 주입만으로 92.4% 제거율을 얻었다. 미세조류 함유 인공해수 또한 PGA 응집제 0.05g/L의 주입 만으로 95% 이상의 높은 제거효율을 보였다. 퇴적토를 함유 인공 해수에 비해 미세조류를 함유한 인공해수에서 효율이 더 좋았으며, 두 경우 모두 0.1g/L의 PGA 응집제 투입 후 1분간의 응집과정을 거친 후 마이크로 공기를 5초간 주입하였을 때 최적의 제거효율을 보여주었다. 마이크로 공기 주입 후 약 10분 후 최고의 탁도 제거율에 도달하였다. 본 연구결과를 통해 높은 탁도를 가진 해수일지라도 응집제와 마이크로 공기를 주입할 시 신속히 제거가 되었으므로, 본 연구결과는 해수 전처리에 효과적으로 사용될 수 있을 것으로 판단된다.
This study aimed to determine the effects of dietary microalgae (Tetradesmus sp. (TO)) on intestinal immunity and microbiota of pre-starter broilers. One hundred and twenty 1-day-old birds (Ross 308) were allocated to two dietary treatment groups with six blocks in a randomized complete block design. The two experimental diets consisted of a corn-soybean meal-based basal diet and a diet with 0.5% TO powder instead of cornstarch in the basal diet. After feeding the experimental diets for ten days, all birds' body weight and feed intake were measured, and representative eight birds were selected from each treatment group. Small intestinal lamina propria cells were isolated using flow cytometry to examine the frequency of immune cells. Cecal feces were harvested for 16s rRNA gut microbiota analysis and fecal IgA levels. Here, we found that 0.5% TO supplementation increased CD3+CD4+ T cells in the small intestine, but decreased CD3+CD8+ T cells in the small intestine. Gut microbial analysis showed that TO supplementation significantly increased the alpha diversity of the gut microbiome. Taxonomic analysis showed that TO treatment increased the abundance of Firmicutes and decreased that of Bacteroidetes at the phylum level. The distribution of Enterobacteriaceae containing many harmful bacteria at the family level, was lower in the TO group. In the LEfSe analysis, the TO group had a significantly enriched abundance of Agathobaculum at the genus level. Overall, results show that Tetradesmus sp. supplementation influences intestinal T-cell immunity and induces the expansion of beneficial gut microbes in pre-starter broiler chickens.
연소배가스에 존재하는 질소산화물의 제거를 위해서는 촉매 환원, 흡수, 흡착 등 화학적 기술이 적용되고 있는데, 장기적으로는 환경친화적이고 에너지 소모가 적은 생물학적 공정의 개발 및 이용이 필요하다. 본 논문에서는 연소 배가스에 존재하는 질소산화물을 제거하기 위한 생물학적 공정의 기술동향을 살펴보고 각각의 장단점을 고찰하였다. 질산화와 탈질 기작을 이용하는 박테리아 시스템과 광합성 미세조류를 이용하는 시스템으로 구분하여 기술의 원리와 현재의 기술 수준을 논하였다. 두 경우 모두 처리속도를 높이기 위해서는 불용성의 일산화질소를 일단 적절한 흡수제에 고농도로 포집시킨 후 미생물에 의하여 분해 또는 고정하는 방향이 바람직하며, 배가스 중 $CO_2$와 NOx를 동시에 고정이 가능하고 별도의 탄소원이 요구되지 않는 미세조류의 활용이 기대된다.
This study investigated the effect of high concentration of free ammonia on microalgal growth and substrate removal by applying real wastewater nitrogen ratio. To test of this, the conditions of free ammonia 1, 3, 6, 9, 12, 15 mg-N/L are compared. After 3 days of incubation, algal growth of Chlorella vulgaris and carbon removal rate are respectively lower in the reactors of FA 12, 15 mg-N/L compared to the others. This indicates that the high concentration of free ammonia, in this case, above 12 mg-N/L, has negative effect on algal growth and metabolic activity. Also, high concentration of free ammonia causes the proton imbalance, ammonium accumulation in algae and has toxicity for these reasons. So, we have to consider free ammonia in applying the microalgae to wastewater treatment system by the way of diluting wastewater or controlling pH and temperature.
Chlorella is a eukaryotic organism that can be used as an industrial host to produce recombinant proteins. In this study, a salt-inducible promoter (SIP) was isolated from the freshwater species Chlorella vulgaris PKVL7422 from the screening of genes that were upregulated after salt treatment. Several cis-acting elements, including stress response elements, were identified in the isolated SIP. Moreover, the Gaussia luciferase gene was cloned after the SIP and transformed into C. vulgaris to test the inducibility of this promoter. Reexamination of transcriptome of C. vulgaris revealed that genes involved in the synthesis of methyl jasmonic acid (MeJA), gibberellin (GA), and abscisic acid (ABA) were upregulated when C. vulgaris was treated with salt. Furthermore, the expression level of recombinant luciferase increased when the transformed C. vulgaris was treated with salt and MeJA, GA, and ABA. This study represents the first report of the C. vulgaris SIP and highlights how transformed microalgae could be used for robust expression of recombinant proteins.

Herein, different extracts of Scenedesmus deserticola JD052, a green microalga, were evaluated in vitro as a potential anti-aging bioagent. Although post-treatment of microalgal culture with either UV irradiation or high light illumination did not lead to a substantial difference in the effectiveness of microalgal extracts as a potential anti-UV agent, the results indicated the presence of a highly potent compound in ethyl acetate extract with more than 20% increase in the cellular viability of normal human dermal fibroblasts (nHDFs) compared with the negative control amended with DMSO. The subsequent fractionation of the ethyl acetate extract led to two bioactive fractions with high anti-UV property; one of the fractions was further separated down to a single compound. While electrospray ionization mass spectrometry (ESI-MS) and nuclear magnetic resonance (NMR) spectroscopy analysis identified this single compound as loliolide, its identification has been rarely reported in microalgae previously, prompting thorough systematic investigations into this novel compound for the nascent microalgal industry.
Ecological toxicity testing of the whole-effluent from the ozone ballast water treatment system was conducted as specified in the quality assurance project plans (QAPP). The growth inhibition test with microalgae, acute aquatic toxicity test with the Rotifer reproduction, toxicity test (or population growth) with the Rotifer, survival and growth toxicity test with larval fish and sediment toxicity test with amphipod were carried out to evaluate ecological toxicity on the movile test barge.
The present study determined the effect of dietary cultivated microalgae (Parachlorella sp.) on the growth and immune responses of pre-starter broilers. A total of 320 one-day-old birds (Ross 308) were allocated to 4 treatments with 8 blocks in a randomized complete block design. The four experimental diets consisted of a corn-soybean meal-based control diet, and three diets contained 0.5%, 1.0%, and 1.5% microalgae powder at the expense of cornstarch in the control diet. After feeding the experimental diets for 7 days, the body weight and feed intake of all birds were measured, and 8 birds were randomly selected from each treatment. Peripheral blood mononuclear cells (PBMCs) and serum were harvested for immune profile assessment, including cytokines and cell migration receptors. No differences in growth performance were observed among the treatments. The birds that were fed diets containing graded levels of microalga showed a linear increase in the mRNA expression of cytokine genes in PBMCs, including that of IL2, IL1β, and IL18 (P<0.05). With respect to the chemokine receptor genes in PBMCs, mRNA expression of CCR2, CCR9, and ITGA4 changed quadratically (P<0.05), but that of CCR7 increased linearly (P<0.01). Cytokine protein secretion in blood, including that of IL-1β and IL-6, increased linearly (P<0.01) with an increase in the microalgal content. Overall, the present results show that the indigenous microalgae powder used in this study could stimulate immunity with no detrimental effects on the growth performance of pre-starter broiler chickens.
바이오디젤을 농작물보다 미세조류에서 생산할 때 여러 가지 장점이 있다. 미세조류는 적은 공간에서도 배양이 가능하며 지방 함유량이 높아 생산량이 매우 높다. 그러나 아직까지는 생산 공정 비용이 높다는 문제점을 가지고 있다. 미세조류에서 바이오디젤을 생산하는 공정에 필요한 비용 중 미세조류 배양액에 첨가되는 당이 차지하는 비중이 높다. 본 연구에서는 바이오디젤 생산용 Chlorella protothecoides의 배양액에 첨가될 당을 농업부산물 배추로부터 생산하는 공정을 연구 개발하였다. 먼저 리그노셀룰로우스 바이오매스를 이용하기 위해서 분쇄된 배추에 H2SO4로 처리하였을 때 0.5%가 가장 당 생산량이 많았지만 다음 단계에서 효소의 활성을 저해하였기 때문에 0.25% H2SO4 용액을 50℃에서 24시간 처리하는 것이 가장 적절한 조건임을 알 수 있었다. 셀룰로스를 분해하기 위하여 Trichoderma harzianum균주를 배양하여 배양액에 분비된 셀룰로스 분해효소(cellulases)를 사용하였다. T. harzianum 배양 조건은 28℃/pH 7/130 rpm에서 5일간 배양으로 최적화하였다. H2SO4 용액으로 전처리 된 배추는 T. harzianum 배양액을 효소 활성도 0.24 FPU/ml로 희석하여 45℃/pH 5/130 rpm 조건에서 약 3일동안 처리하였을 때 가장 효율적인 당 생산량을 이룰 수 있었다. 다른 농업부산물을 같은 공정 과정으로 처리하였을 때 배추가 가장 높은 당 생산률을 나타내어 활용도면에서 우수하다고 여겨진다. 배추의 수분함유량이 최대(95%)인 갓 폐기된 배추 잎의 경우 가장 당의 생산량이 많아서 가능한 빨리 처리하는 것이 바람직하였다. 지속 가능한 바이오디젤 생산을 위하여 미세조류를 배양할 때 본 연구에서 최적화한 조건에 따라 농업부산물 배추를 처리함으로써 비용 절감을 이룰 수 있을 것으로 기대된다.
