Production of monoclonal antibody(MAb) for lymphocytes of Korean native cattle 1. MAb to T lymphocytes (한우 림프구에 대한 단일항체 생산 1. T 림프구에 대한 단일항체)

  • This study was undertaken to develop the monoclonal antibody(MAb) for lymphocytes of Korean native cattle by the cell hybridization of myeloma P3/NS-1/ 1-Ag-4-1 and spleen cells from BALB/c mice hyperimmunized with nylon wool column eluted peripheral T lymphocytes of Korean native cattle. The isotype of MAb KCT-14 against T lymphocyte was mouse $IgG_1$. KCT-14 positivity of mononuclear cells(MNC) from peripheral blood lymphocytes, nylon wool nonadherent and adherent-lymphocytes was 41.7%, 58.4% and 22.6%, respectively. And that of mesenteric lymph node-, spleen and thymus-MNC was 43.3%, 40.2% and 33.6%, respectively. Immunoperoxidase staining of frozen tissue sections showed that the MAb positive cells were located in the medulla of the thymus and in the paracortical area and the mantle zone of the germinal center in the lymph nodes. These results indicated that KCT-14 was one of the MAb for investigate of T lymphocyte subpopulations in the Korean native cattle.

  • In the study of investigate the expression and effects of adrenomedullin (AM) in the tissue injured by ethanol in our laboratory, there were lymphosarcomas involving mesenteric lymph node and intestine discovered in experimental mice. Lymphosarcoma is probably the second most common spontaneous neoplastic disease, the incidence of which is dependent on the strain of mouse. The diagnosis of lymphosarcoma was based on gross and light microscopic studies. Crossly, the excised intestine and mesentery showed soft-to-firm, cream-colored, displayed protuberance into the lumen. The neoplastic cells had mainly infiltrated into the mucosal and mesenteric lymph node, and have round hyperchromatic nuclei and a small amount of cytoplasm. The origin of these tumor cells was determined by immunohistochemistry positive of CD20, CD3 and Vimentin.

Evaluation of Mucosal Immunity in BTBR T+tf/J Mice Resembling Autism Spectrum Disorder (자폐증 유사증상 발현 마우스의 점액 면역에 대한 연구)

  • Objectives: This study was undertaken in order to evaluate a potential mechanism involved in gastro-intestinal problems observed in autistic subjects and uses an animal model of autism investigation. Methods: BTBR T+tf/J, a mouse strain with typical socio-behavioral characteristics of autistic subjects and FVB mice with highly social behaviors as the control strain were used. Both genders of mice aged three weeks and six months were used from four separate litters for each strain. Serum was prepared following cardiac puncture, and mesenteric lymph nodes were collected for in vitro stimulation and enumeration of major immune cell proportion. Results: The level of serum IgA was significantly enhanced in six-month-old BTBR mice compared with three-week-old BTBR, which was not observed with the FVB control mice. The serum IgE level was also higher among BTBR mice than among age-sex matched FVB mice, respectively. Considering the ratio of interleukin-4 vs interferon-gamma production from mesenteric lymph node T cells, skewedness toward type-2 reactivities was observed. In addition, the proportion of B cells in mesenteric lymph nodes was significantly higher in BTBR mice than in FVB mice. Conclusion: Upregulation of mucosal immunity related with enhanced type-2 immune reactivity observed in BTBR mice could be involved with the etiology of gastro-intestinal abnormalities in autism.

An Acute Outbreak of Calf Salmonellosis Caused by Salmonella dublin (Salmonella dublin에 의한 소의 살모넬라증의 발생)

  • An epizootic of calf diarrhea occurred in an extensive dairy farm located in Gangwondo province in December of 1985. The patients showing fever, chills, complete anorexia and watery diarrhea sometimes dysentery were concentrated among the calves ranged from 8 days to 3 months old and many of them died in acute course. The five carcases were examined by pathological and bacteriological means. The predominant gross lesions were edematous swelling of the mesenteric lymph nodes, profuse catarrhal or diphtheritic enteritis and extensive purulent bronchopnenomonia or lobar fibrinous pneumonia. Microscopically the lesions of the liver and spleen were characterized by coagulative necrotic foci and granulomatous nodules. The spleen and lymph node showed also proliferative changes of reticular cells and involution of intestinal lymphatic nodules were noticed. The strains of Salmonella(S) species isolated from the liver, spleen, bile juice, peritoneal fluid and thoracic fluid were identified serologically as S. dublin. These clinical and pathological findings of the disease were those of acute enteritis form of salmonellosis and characterized by high morbidity and mortality among the calf herd. The report also signifies the first description of an epizootic of bovine salmonellosis caused by S. dublin in Korea.

A Case of Visceral T Cell Lymphoma with Prominent Histiocyte Infiltration in a Dog

  • A 13-year-old intact male English Springer Spaniel presented with anorexia. Physical examination revealed a palpable abdominal mass without peripheral lymphadenopathy. Ultrasonography revealed hepatosplenomegaly and a markedly enlarged hepatic lymph node. Fine-needle aspiration of the splenic and nodal lesions revealed atypical round cells admixed with numerous histiocytes. The dog was euthanized owing to deteriorating condition despite a month of chemotherapy with lomustine. Histopathology revealed obliteration of the normal architecture of the liver, spleen, kidney, and hepatic and mesenteric lymph nodes by CD3+ neoplastic lymphocytes, accompanied by extensive F4/80+ histiocytic infiltration. This report describes a rare presentation of T-cell lymphoma with prominent histiocytic infiltration that may initially be misdiagnosed as histiocytic neoplasia in a dog.

  • The purpose of this research was to investigate the immunoregulatory effect and the leukemia cell apoptosis of EtOH extract of OGAPI(OGP). The proliferation of cultured splenocytes, thymocytes and mesenteric lymph node cells were enhanced by the addition of OGP. Splenic and thymic T lymphocytes, especially TH and Tc cells were significantly increased in OGP-administered mice. OGP markedly increased the production of γ-interferon in mice serum and accelerated the phagocytic activity in peritoneal macrophages. OGP treatment enhanced the apoptosis of L1210 mouse leukemia and Jurkat, Molt4 human leukemia cells, and increased the expression of apoptosis-related ICE, c-myc, p53 gene in Jurkat cell. These results suggest that OGP have an immunoregulatory action and anti-cancer activity.

Kinetics of IFN-${\gamma}$ and IL-17 Production by CD4 and CD8 T Cells during Acute Graft-versus-Host Disease

  • Graft-versus-host disease (GVHD) is a fatal complication that occurs after allogeneic hematopoietic stem cell transplantation. To understand the dynamics of CD4 and CD8 T cell production of IFN-${\gamma}$ and IL-17 during GVHD progression, we established a GVHD model by transplanting T cell-depleted bone marrow (TCD-BM) and purified T cells from B6 mice into irradiated BALB.B, creating an MHC-matched but minor histocompatibility (H) antigen-mismatched transplantation (B6 ${\rightarrow}$ BALB.B GVHD). Transplantation-induced GVHD was confirmed by the presence of the appropriate compositional changes in the T cell compartments and innate immune cells in the blood and the systemic secretion of inflammatory cytokines. Using this B6 ${\rightarrow}$ BALB.B GVHD model, we showed that the production of IFN-${\gamma}$ and IL-17 by CD4 T cells preceded that by CD8 T cells in the spleen, mesenteric lymph node, liver, and lung in the BALB.B GVHD host, and Th1 differentiation predated Th17 differentiation in all organs during GVHD progression. Such changes in cytokine production were based on changes in cytokine gene expression by the T cells at different time points during GVHD development. These results demonstrate that both IFN-${\gamma}$ and IL-17 are produced by CD4 and CD8 T cells but with different kinetics during GVHD progression.

A GFP-labeled Human Colon Cancer Metastasis Model Featuring Surgical Orthotopic Implantation

  • Colorectal cancer has become a major disease threatening human health. To establish animal models that exhibit the characteristics of human colorectal cancer will not only help to study the mechanisms underlying the genesis and development effectively, but also provide ideal carriers for the screening of medicines and examining their therapeutic effects. In this study, we established a stable, colon cancer nude mouse model highly expressing green fluorescent protein (GFP) for spontaneous metastasis after surgical orthotopic implantation (SOI). GFP-labeled colon cancer models for metastasis after SOI were successfully established in all of 15 nude mice and there were no surgery-related complications or deaths. In week 3, primary tumors expressing GFP were observed in all model animals under fluoroscopy and two metastatic tumors were monitored by fluorescent imaging at the same time. The tumor volumes progressively increased with time. Seven out of 15 tumor transplanted mice died and the major causes of death were intestinal obstruction and cachexia resulting from malignant tumor growth. Eight model animals survived at the end of the experiment, 6 of which had metastases (6 cases to mesenteric lymph nodes, 4 hepatic, 2 pancreatic and 1 mediastinal lymph node). Our results indicate that our GFP-labeled colon cancer orthotopic transplantation model is useful with a high success rate; the transplanted tumors exhibit similar biological properties to human colorectal cancer, and can be used for real-time, in vivo, non-invasive and dynamic observation and analysis of the growth and metastasis of tumor cells.

Kinetics and Biological Function of Transforming Growth Factor-$\beta$ Isoforms in Bovine and Human Colostrum

  • Colostrum contains various kinds of cytokines including TGF-$\beta$ that has potent regulatory effects on cells of the immune system. We compared the levels of TGF-$\beta$1 and TGF-$\beta$2 in bovine and human colostrum. Based on the isoform-specific ELISA, bovine colostrum collected on day 1 post-delivery retained $53.71{\pm}29.55\;ng/ml$ of TGF-$\beta$1 and $40.41{\pm}21.78\;{\mu}g/ml$ of TGF-$\beta$2 (n=4), while in human, $381.45{\pm}158.24\;ng/ml$ of TGF-$\beta$1 and $41.47{\pm}9.63\;ng/ml$ of TGF-$\beta$2 (n=5). Thus, dominant TGF-$\beta$ isoforms were completely opposite between human and bovine colostrum samples. The concentrations of both isoforms declined as lactation proceeded. Biological activities of the colostrum samples were determined using an MV1LU cell line. Consistent with the result from the immunoassay, TGF-$\beta$1 in human and TGF-$\beta$2 in bovine colostrum were responsible for the anti proliferative activity against MV1LU cells. Furthermore, bovine colostrum increased IgA secretion by LPS-stimulated mesenteric lymph node (MLN) cells, and this effect was abrogated by either anti­TGF-$\beta$2 antibody or combined anti-TGF-$\beta$1/$\beta$2 antibody, but not by anti- TGF-$\beta$1 antibody alone. Similarly, TGF-$\beta$2 in bovine colostrum enhanced the Ig germ line (GL) promoter activity, which is the earliest event toward IgA isotype switching. Taken together, these results suggest that TGF-$\beta$ isoforms, differentially expressed in human and bovine colostrum, may promote IgA isotype production in the neonatal intestine.

Inhibitory Effect of Scutellaria baicalensis Root Extract on Chemical Mediator Release and Immune Response

  • Inhibitory effect of Scutellaria baicalensis ethanol extracts (SR) on chemical mediator release and immunoglobulin (Ig) production from Sprague-Dawley rats originated cells as type I allergic reaction was examined. SR showed concentration-dependent inhibition on basal and concanavalin A (ConA)-stimulated Ig production. In the mesenteric lymph node lymphocytes, the inhibitory effect of SR on the IgE production in the presence of Con A was stronger than these on IgA and IgG production. Moreover, tumor necrosis factor-alpha $(TNF-{\alpha})$ production-inhibiting effect of SR in the presence ConA was observed. However, SR did not affect the production of $interferon-{\gamma}$. SR also inhibited histamine release from the peritoneal exudate cells stimulated with a calcium ionophore A23187. In the case of leukotriene B4, SR markedly inhibited it at the concentration of 100 mg/ml. From these results, ethanol extracts obtained from Scutellaria baicalensis may have an anti-allergic effect on the intestinal system of rats.
