인체 흑색종 세포(SK-MEL-28 Cell Line)에서 Cisplatin, Heptaplatin, 그리고 Sulpla에 의한 Apoptosis의 유도 (Induction of Apoptosis by Cisplatin, Heptaplatin and Sunpla in Human Melanoma (SK-MEL-28) Cell Line)

  • A wide variety of cancer chemotherapeutic agents have been shown to induce programmed cell death (PCD, APOPTOSIS) in various tumor cell lines in vitro. cis-Malonato [(4R,5R)-4,5-bis(aminomethyl)-2-isoprpopyl-1,3-dioxolane] platinum(II) (heptaplatin), which is a new drug approved by KFDA in 1999, in a novel platinum-based antitumor agent with clinical potential against stomach cancer and the 3rd generation of the cisplatin. This study was performed to know how heptaplatin and cisplatin and sunpla (mixture of heptaplatin and mannitol) affect on SK-MEL-28 cell line, and how they induce the apoptosis. At EM analysis, the morphology of the cell was changed by treatment of the cisplatin, heptaplatin and sunpla. Apoptotic body formed around plasma membrane, and chromatin condensation represented in nucleus. This phenomenon is one of the characteristic of the apoptosis. The DNA of SK-MEL-28 cell line truncated by cisplatin and sunpla treatment was identified on 2% agarose gel electrophoresis. TUNEL assay was performed to know whether SK-MEL-28 cell die as apoptosis or necrosis by cisplatin, heptaplatin and sunpla. At this result, fluorescence intensity increased according to increase of time and concentration. Therefore, it was identified that cislatin, heptaplatin and sunpla induced apoptosis. Fas expressed on SK-MEL-28 cell membrane by cisplatin, heptaplatin and sunpla was identified by using flow cytometer and the expression of bcl-2(anti-apoptotic gene) decreased according to increase of concentration of the cisplatin, heptaplatin and sunpla. Cisplatin, heptaplatin and sunpla induced apoptosis against SK-MEL-28 cell line, and the apoptotic mechanism was identified as Fas-mediated apoptosis and decreased bcl-2 expression.

식도암의 외과적 요법에대한 임상적 고찰 (Surgical Results of Esophageal Cancer)

  • From January 1984 to December 1991, One hundred sixty five patients with carcinomoa of the esophagus were treated surgically at the department of Thoracic and Cardiovascular Surgery, Seoul National University Hospital. Among them, hospital records were available in 121 patients and were included in this study. There were 115 men and 6 women, with ages ranging from 40 years to 79 years[mean age of 59.2 years]. The most frequent preoperative symptoms included dysphagia[72.7%], weight loss[60.3%], chest pain or discomfort[14.9%], general malaise[13.2%]. All were treated surgically: 100 patients were managed by curative or palliative resection with reconstruction, and 6 by palliative bypass surgery. In 15 patients, explorative thoracotomy or laparotomy was only done due to unresectability. [operability: 87.6%, resectability: 82.6%] All specimens[those from resectable 100 cases] were sent to pathology, and histopathologic examinations were done; squamous cell carcinomas were found in 95 cases, adenocarcinoma in l. Adenosquamous carcinomas were found in 3, and malignant melanoma in l. Postoperative complications occurred in 34 cases; anastomotic site leakage[10], which was followed by empyema in 9 of them, wound problem[7], hepatic failure[6], pneumonia [3], post-operative bleeding[3], chylothorax[2], post-operative stricture[2], sepsis[1], and tracheobronchial fistula[1]. Hospital deaths were in 6 cases[Hospital mortality: 5.0%]. During the follow up period, 26 patients were proven to be recurrence of cancer locally or distantly. The one, two, and five-year actuarial survival raf.es were 71.3$\pm$4.5%, 57.4$\pm$5.6%, 34.7$\pm$8.9%, respectively. The data from this study suggested that esophagectomy with reconstruction of gastrointestinal tract could be performed with a low operative mortality and a few serious postoperative complications and achieved reasonable long term palliation for carcinoma of the esophagus.

Immunogenic Cell Death Induced by Ginsenoside Rg3: Significance in Dendritic Cell-based Anti-tumor Immunotherapy

  • Cancer is one of the leading causes of morbidity and mortality worldwide; therefore there is a need to discover new therapeutic modules with improved efficacy and safety. Immune-(cell) therapy is a promising therapeutic strategy for the treatment of intractable cancers. The effectiveness of certain chemotherapeutics in inducing immunogenic tumor cell death thus promoting cancer eradication has been reported. Ginsenoside Rg3 is a ginseng saponin that has antitumor and immunomodulatory activity. In this study, we treated tumor cells with Rg3 to verify the significance of inducing immunogenic tumor cell death in antitumor therapy, especially in DC-based immunotherapy. Rg3 killed the both immunogenic (B16F10 melanoma cells) and non-immunogenic (LLC: Lewis Lung Carcinoma cells) tumor cells by inducing apoptosis. Surface expression of immunogenic death markers including calreticulin and heat shock proteins and the transcription of relevant genes were increased in the Rg3-dying tumor. Increased calreticulin expression was directly related to the uptake of dying tumor cells by dendritic cells (DCs): the proportion of CRT+CD11c+cells was increased in the Rg3-treated group. Interestingly, tumor cells dying by immunogenic cell death secreted IFN-γ, an effector molecule for antitumor activity in T cells. Along with the Rg3-induced suppression of pro-angiogenic (TNF-α) and immunosuppressive cytokine (TGF-β) secretion, IFN-γ production from the Rg3-treated tumor cells may also indicate Rg3 as an effective anticancer immunotherapeutic strategy. The data clearly suggests that Rg3-induced immunogenic tumor cell death due its cytotoxic effect and its ability to induce DC function. This indicates that Rg3 may be an effective immunotherapeutic strategy.

  • Due to the fact that it reliably results in positive outcomes, lymph node flap transfer is becoming an increasingly popular surgical procedure for the prevention and treatment of lymphedema. This technique has been shown to stimulate lymphoangiogenesis and restore lymphatic function, as well as decreasing infection rates, minimizing pain, and preventing the recurrence of lymphedema. In this article, we investigate possible additional benefits of lymph node flap transfer, primarily the possibility that sentinel lymph nodes may be used to detect micrometastasis or in-transit metastasis and may function as an additional lymphatic station after the excision of advanced skin cancer.

팔성 양조용수의 세포독성 및 항암활성 (The Cytotoxicity and Antitumor Activity of Palsun Brewing Water)

  • In the present study, we have evaluated cytotoxic effects, antitumor activities and metastasis inhibitory effects of Palsun brewing water in NIH 3T3 cells, human epitheloid carcinoma cells, and human skin melanoma cells. The light microscopic study showed morphological changes, AG-NOR (argyrophylic nucleolar organizer region) by silver chloride stain, and glycoprotein by PAS (periodic acid stain) reaction of the treated cells. Disruptions in cell organelles were determined by colorimetric methods; MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide) and SRB (sulforhodamine B protein) assays. These results suggest that Palsun Brewing Water retains no cytotoxic effects in NIH 3T3 cells and a growth-inhibitory activity in cancer cell lines.

Antiviral, Antimicrobial, and Cytotoxic Properties of Peptavirins A and B Produced by Apiocrea sp.14T

  • Two peptaibol antibiotics, peptavirins A and B, which exhibited strong inhibitory effect against Tobacco mosaic vials (TMV) infection, were isolated from steam-cooked rice culture of Apiocrea sp.14T. The peptavirins were identified as new derivatives of chrysospermins, which are 19-mer and have been reported to be produced in a fungal isolate. The physicochemical properties of the peptavirins were mostly identical with chrysospermins A through D except for the UV absorption spectrum. The peptavirins inhibited the growths of the Grampositive bacteria tested, including the plant pathogenic bacterium, Corynebacterium lilium, and the fungus, Aspergillus niger. Peptavirin A was somewhat cytotoxic to cancer cell lines, especially K562 (leukemia) and UACC 62 (melanoma), whereas peptavirin B only exhibited slight cytotoxicity.

  • Two diterpenes and one new sucrose isovaleryl ester having inhibitory effects on melanogenesis in B16 mouse melanoma were isolated from Euphorbiae Lathyridis Semen which has been used in traditional medicine for cancer, tumors and warts. New sucrose isovaleryl ester was identified as ${\alpha}-D-glucopyranoside$, $3,4,6-tris-O-(3-methyl-l-oxobutyl)-{\beta}-D-fructofuranosyl$, 2,6-bis(3-methylbutanoate) and two diterpenes were identified as ingenol-20-palmitate and 5,10-diacetyl-3-benzoyllathyrol$(Euphorbia\;factor\;L_3)$ from their spectral data.

산수유 헥산 추출물의 항균효과 및 세포독성 (Screening of Cytotoxicity and Antimicrobial Effects of Hexane Extracts from Cornis fructus)

  • Cornis fructus was extracted by successive extraction and then fractionated with hexane extract to get active fractions. This study was performed to determine the cytotoxic effect of hexane extract from Cornis fructus on NIH 3T3 fibroblasts and cancer cell lines using MTT assay. Hexane extract showed cytotoxic effect against A549, B16 melanoma and MDA-MB-231. Futher fractionation with hexane extract was performed to obtain effective fraction, fraction 3 showed the cytotoxic effect against A549 and MDA-MB-231. In antimicrobial test of each fraction of hexane extract, fraction 5 showed antimicrobial activities against P. putida and P. aeruginosa.

  • The purpose of this study was to investigate the anti-melanogenic effects of the extracts from Decaschistia intermedia craib (EDI). In this study, we examined the effects of EDI on mushroom tyrosinase activity in in vitro, melanin contents, and expression levels of mRNA and proteins of melanogenesis-related genes in B16F10 melanoma cells. The treatment of EDI significantly decreased both tyrosinase activity and melanin contents in B16F10 cells with dose-dependent manner. In addition, we found that the expression of mRNA or proteins of melanogenic proteins, such as, a-melanocyte-stimulating hormone (a-MSH)-induced microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein-1 (TRP-1), and TRP-2 was significantly downregulated with dose-dependent manner in the EDI-treated B16F10 cells compared to controls. Our results suggest that the EDI inhibits cellular melanogenesis through downregulation of a-MSH-stimulated melanin synthesis. Thus EDI may potentially be an effective whitening agent.

Inhibitory Effects of Pine Cone (Pinus densiflora) on Melanogenesis in B16F10 Melanoma Cells

  • The pathological condition of excessive melanogenesis causing freckles, melasma, senile lentigo, pigmented acne scars, and cancer has a critical impact on the wellness of individuals. The mechanism of melanogenesis is related to the expression of melanogenic enzymes. Here, we evaluated the inhibitory effect of pine cone (Pinus densiflora) extracts on melanogenesis. P. densiflora, the Korean Red Pine, is the predominant tree species in the cool, temperate forests of northeast Asia, occurring in pure stands across Korea, Japan, and parts of northern China and Russia. P. densiflora leaves, pollen, and bark have been widely used for traditional medicine, or edible purposes. However, pine cones are rarely used as natural raw materials, although they contain many bioactive phytochemicals. The pine cone ethyl acetate fraction (PEF) showed no toxicity to B16F10 cells at a concentration of less than $100{\mu}g/mL$. PEF inhibited the expression of microphthalmiaassociated transcription factor (MITF), tyrosinase and tyrosinase-related factors in B16F10 cells treated with 3-Isobutyl1-methylxanthine (IBMX). These results suggest that pine cones can be used as an effective natural melanogenesis inhibitory agent.
