Effect of Divided Rhizome Size and Medium Type on Growth of Wasabia japonica Matsum. (분주 근경 크기와 배지 종류가 고추냉이 생육에 미치는 영향)

  • This experiment was investigated to effect of divided rhizome size and medium type on survival rate and growth of wasabi for 60 days in controlled growth room. In divided rhizome size of 5mm above, survival rate was 100% and their growth (plant height of 12cm and leaf number of 3${\sim}$4 per plant) was good at 30 days after wrapped-sphagnum treatment. Plant height was 20cm above and number of leaves increased in 1-2 per plant at 60 days after treatment. Survival rate and growth didn't show any effect on plant regulator of root-tone. In inorganic media (saprolite and aerated light stone) treatment, survival rate of wasabi in divided size of 5${\sim}$10mm showed 83% or above at 30 days in deep flow culture. Growth did not show significant difference of inorganic support media treatments. Therefore, it is possible for divided rhizome size of 5mm above to do production of seedlings by acclimatize for 30 days in hydroponics under controlled growth chamber.

Production of Baicalin, Baicalein, and Wogonin on Plant Tissue Culture of Scutellaria baicalensis (황금(Scutellaria baicalensis) 조직배양에 의한 Baicalin, Baicalein 및 Wogonin 생산)

  • Scutellaria baicalensis Georgi (SJ) is a perennial plant and its root has been used in oriental traditional medicine for treatment of fever, inflammation, diarrhea and anticancer effect, etc. In this study, plant tissue culture system for SJ was developed. Stem piece of younger plant was optimum explant for callus induction and growth on MS medium supplemented with NAA 1.0 ㎎/L plus BA 0.5 ㎎/L. Plantlet regeneration through callus culture was well on MS medium containing NAA 1.0 mg/L. SJ has been known biologically active substances such as baicalin, baiclein, and wogonin. This study was carried out to examine the effect of plant growth regulators for production of baicalin, baicalein, and wogonin through callus culture. The HPLC pattern of callus extract was identical to that of standard solution, it shows that the callus produced by tissue culture has the same flavonoids composition of SJ. Baicalin, baicalein, and wogonin production was 471.5~52.8 ㎍/g, 137.6~4.0 ㎍/g, and 16.6~1.3 ㎍/g, respectively, on MS media with nine different plant growth regulator combinations. This may indicate that plant tissue culture of SJ possible to produce the biologically active substances effectively

  • Deep excavations for development of subway systems in metropolitan regions surrounded by adjacent buildings is an important geotechnical problem, especialy in Tabriz city, where is mostly composed of young alluvial soils and weak marly layers. This study analyzes the wall displacement and ground surface settlement due to deep excavation in the Tabriz marls using two dimensional finite element method. The excavation of the station L2-S17 was selected as a case study for the modelling. The excavation is supported by the concrete diaphragm wall and one row of steel struts. The analyses investigate the effects of wall stiffness and excavation width on the excavation-induced deformations. The geotechnical parameters were selected based on the results of field and laboratory tests. The results indicate that the wall deflection and ground surface settlement increase with increasing excavation depth and width. The change in maximum wall deflection and ground settlement with considerable increase in wall stiffness is marginal, however the lower wall stiffness produces the larger wall and ground displacements. The maximum wall deflections induced by the excavation with a width of 8.2 m are 102.3, 69.4 and 44.3 mm, respectively for flexible, medium and stiff walls. The ratio of maximum ground settlement to maximum lateral wall deflection approaches to 1 with increasing wall stiffness. It was found that the wall stiffness affects the settlement influence zone. An increase in the wall stiffness results in a decrease in the settlements, an extension in the settlement influence zones and occurrence of the maximum settlements at a larger distance from the wall. The maximum of settlement for the excavation with a width of 14.7 m occurred at 6.1, 9.1 and 24.2 m away from the wall, respectively, for flexible, medium and stiff walls.

Approximate Analysis of Shear Wall-Frame Structure For Seismic Design (전단벽-골조 시스템의 내진설계를 위한 근사해석법)

  • A wall-frame structure resists horizontal load by the interaction between the flexural mode of the shear wall and the shear mode of the frame, which implies that the frame deflects only by reverse bending of the columns and girders, and that the columns are axially rigid. However, as the height of frame increases the shear mode of frame changes to flexural mode, which is due to the extension and shortening of the columns. An approximate hand method for estimating horizontal deflection and member forces in high-rise shear wall-frame structures subjected to horizontal loading is presented. The method is developed from the continuous medium theory for coupled walls and expressed in non-dimensional structural parameters. It accounts for bending deformations in all individual members as well as axial deformations in the columns. The deformations calculated from the presented approximate method and matrix analysis by computer program are compared. The presented approximate method is more accurate for the taller structures.

First Report of Leptosphaerulina saccharicola Isolated from Persimmon (Diospyros kaki) Tree Bark in Korea

  • A fungal strain, designated PTT-2, was isolated from the bark of the trunk of a persimmon (Diospyros kaki) tree in Cheongdo, Korea. The isolate showed morphological similarities with Leptosphaerulina saccharicola. Strain PTT-2 had more rapid growth on potato dextrose agar medium than on oatmeal agar, malt extract agar, and synthetic nutrient poor agar media, with colony sizes of 53.8 mm, 49.8 mm, 48.4 mm, and 28.1 mm after 7 days at $25^{\circ}C$ temperature, respectively. Strain PTT-2 produced ascospores, which had irregular wavy edges, oblong to ellipsoidal shape, hyaline appearance and $23.6{\times}10{\mu}m$ size. The black ascomata were developed on PDA medium, and asci were recorded. A BLAST search of the internal transcribed spacer (ITS) region, TEF1-${\alpha}$ and RPB2 gene sequences revealed that strain PTT-2 showed more than 99% nucleotide similarity with a strain of Leptosphaerulina saccharicola previously reported from Thailand. A neighbor-joining phylogenetic tree was constructed by concatenating the above-mentioned sequences, and showed that strain PTT-2 clustered in the same clade with L. saccharicola. Based on these findings, this is the first record of Leptosphaerulina saccharicola occurring in Korea.

Characteristics of a new Grifola frondosa Cultivar "Daebak" with stable pinheading and high yield (발이 안정 및 다수성 잎새버섯 신품종 '대박'의 특성)

  • 'Daebak', a new cultivar of Grifola frondosa, was bred by mating two monokaryotic strains isolated from 'F14309' and 'GMGF44062' at the Mushroom Research Institute, Gyonggi-Do ARES in 2017. The optimum temperature for mycelial growth of 'Daebak' was $25^{\circ}C$ on PDA medium. In bottle cultivation, the culture period of 'Daebak' was 57 days, which was 2 days shorter than that of 'Cham' (control). The pinheading rate of 'Daebak' was 98.4%, which was 24.8% higher than that of 'Cham' and its CV (Coefficient of variation) was 0.6%, 5.3% lower than that of 'Cham'. Regarding the growth characteristics of 'Daebak', the diameter and thickness of the pileus were 27.7 mm and 1.73 mm, respectively, and the diameter and height of the fruiting bodies cluster were 132 mm and 87.2 mm, respectively. The pileus was thinner but the fruiting bodies cluster was larger than that of 'Cham'. Fruiting bodies weighed 139 g per 1,100 ml bottle of 'Daebak', which was 28% higher than that for 'Cham', with a CV of 2.5%, which was 6.2% lower than that of 'Cham'. The yield per 10,000 bottles (used for cultivation) of 'Daebak' was 1,376 kg, 70% higher than that of 'Cham', with a CV of 3.0% that was 11.5% lower than that of 'Cham'. With respect to physical characteristics, the strength and brittleness of the fruiting body of 'Daebak' was less than that of 'Cham'. When considering the period available for sale, the shelf life of 'Daebak' was 42 days, which was 6 days longer than that of 'Cham'.

Development of Late Bolting and New Deep Red Leaf with Wrinkled Lettuce 'Chunpungjeokchukmyeon' (추대가 늦고 색깔이 진한 새로운 잎상추 '춘풍적축면' 육성)

  • A new cultivar of lettuce (Lactuca sativa L.) with wrinkled traverse elliptic and deep red leaf, 'Chunpungjeokchukmyeon' which has late bolting and deep red expression leaf color was developed from a cross between 'Pojabijeokchukmyeon' (red leaf color and late bolting) and 'Meokchima' (Deep red and low yield). The cross and selection for advanced lines had been done by the pedigree method during 2000-2007. The advanced lines were evaluated for yield and adaptability at several locations in Korea (Gangwon-do, Gyeonggi-do, Chungcheongbuk-do, Jeollabuk-do, Gyeongnam-do, and Jeju-do) from 2008 to 2009. The 'Chunpungjeokchukmyeon' has gray seed color and traverse elliptic leaves. The type of matured stage is medium shape between 'chukmyeon' and 'chima' leaf lettuce. Compared to 'Dukseomjeokchukmyeon', marketable yield of 'Chunpungjeokchukmyeon' was higher by 6% (at 372 g per plant) and 'Chunpungjeokchukmyeon' has particularly improved expression of deep red leaf color in high temperature cultivation in the field. The shelf-life of 'Chunpungjeokchukmyeon' was three weeks longer than 'Dukseomjeokchukmyeon' at 4$^{\circ}C$. The anthocyanin content of 'Chunpungjeokchukmyeon' was higher than that of 'Dukseomjeokchukmyeon' with 17.5 mg/100g. The BSL (latucin+8-deoxylactucin+lactucopicrin) content of 'Chunpungjeokchukmyeon' is lower than that of 'Dukseomjeokchukmyeon'. Furthermore, its taste is better, more crispy, and sweeter than those of 'Dukseomjeokchukmyeon'. So we recommend that new cultivar 'Chunpungjeokchukmyeon' can be suitable for cultivation in spring season than summer season.

Characteristics of the gene resources and selected strains of Agrocybe cylindracea (버들송이버섯(Agrocybe cylindracea)유전자원 및 선발계통의 특성)

  • Agrocybe spp. belongs to the Agaricales order, Bolbitiaceae family, and Agrocybe genus. In Korea, so far, it has been cultivated through bottle cultivation; therefore, this study was conducted for the development of a new cultivar using the bag cultivation method for quantitative improvement. Thirty-three gene resources of Agrocybe spp. were collected and their quantity and characteristics of bag cultivation were examined. Next, 5 kinds of crossing parents were selected based on the cultivation period and shape of the fruit body. Seven strains were selected through 3 cross combinations. The 7 selected strains and the comparative cultivar 'Cham' were cultivated in a bag. As a result, the cultivation period was 49 days for 'JBAC15-1' and 50 days for 'JBAC15-6' which are 4 days and 3 days less than the cultivation period of the comparative cultivar 'Cham'(53 days), respectively. Cultivation periods of other strains except for 'JBAC15-1' and 'JBAC15-1' were longer than that of the comparative cultivar'Cham'. The best ratio of primordia formation among the selected strains was found to be that of 'JBAC15-1' with 96.1% followed by 'JBAC15-6' with 94.5%. These rates were 3.1% and 1.5% higher than the ratio of primordia formation of the comparative cultivar 'Cham', which is found to be 93.0%. The quantity was maximum in the 'JBAC15-1' cultivar with 176.8 g per bag followed by 'JBAC15-6' with 168.7 g per bag. The quantities were 10% and 5% more than the comparative cultivar 'Cham' with 160.7 g per bag. Based on these results, 'JBAC15-1' and 'JBAC15-6', which had shorter cultivation periods and more quantities than the comparative cultivar 'Cham' were finally selected. For the selected strains of 'JBAC15-1' and 'JBAC15-6', mycelial growth was observed to be optimal on PDA medium and the optimum temperature was $27.5^{\circ}C$. The optimum pH was pH 5 for 'JBAC15-1' and pH 6 for 'JBAC15-6'. The color of the pileus of the fruit body was dark brown in 'JBAC15-1' and 'Cham' and light brown in 'JBAC15-6'. The pileus was hemispherical in shape in both 'JBAC15-1' and 'Cham'. However, the colors of the stem were different - light brown in 'JBAC15-1', white in 'JBAC15-6', and ivory in the comparative cultivar 'Cham'.

Yellowish Friable Embryogenic Callus (YFEC) Production and Plant Regeneration from Immature Embryo Cultures of Domestic Maize Cultivars and Genotypes (Zea may L.) (국내 옥수수 품종 및 계통의 미숙배 배양으로부터 Yellowish Friable Embryogenic 캘러스 (YFEC) 생산과 식물체 재생)

  • Immature embryos of 3 cultivars (Du Me Chal, Mi Baek Chal, Heug Jeom Chal) and 5 genotypes (HW1, KL103, HW3, HW4, KW7) were cultured on medium containing MS salts, Eriksson's vitamins, 1 mg/L 2,4-dichlorophenoxyacetic acid, 25 mM proline, 100 mg/L casamino acid, 3 mM MES, 1.7 mg/L $AgNO_3$ and 20 g/L sucrose (SIM). Frequency of somatic embryo formation on explant of immature embryos showed in HW1 (45.20%), KL103 (5.75%), HW3 (37.20%), HW4 (30.10%), KW70 (55.20%), Mi Baek Chal (18.74%), Heug Jeom Chal (22.41%), Du Me Chal (36.72%) and Hi II type (<10%), respectively. Yellowish friable embryogenic callus (YFEC) such as type II callus of Hi II genotype only produced from the HW3 and Heug Jeom Chal, whereas other cultivars and genotypes were directly formed somatic embryos with late-embryonic stages or expanded yellowish compact somatic embryo with morphological abnormality. The yellowish friable embryogenic callus (YFEC) could be proliferated on the same medium, which were maintained embryogenic capacity for 6 months over. Upon transfer to first regeneration and second regeneration medium, somatic embryos converted to plantlets at a frequency of approximately 100%. However, the expanded somatic embryos with abnormal morphology were slowly proliferated when subcultured on the same medium, and some of them were degenerated or converted to plantlets at a frequency of approximately 25%. Accordingly, The Heug Jeom Chal and HW3 genotype will be further used for development of high frequency transformation system in domestic maize germplasm.

Factors influencing shoot regeneration from petal explant in spray mum 'Purple ND' (스프레이국화 '퍼플엔디'의 꽃잎 배양에 있어서 식물체 재분화요인)

  • This experiment compared the regeneration conditions of the radiation mutant spray chrysanthemum 'purple ND'. The four different flower blooming stages (S1: 10% opened flower, S2: 30% opened flower, S3: 50% opened flower, and S4: 70% opened flower) and different petal parts (TBOP: the basal of petal and TEOP: the end of petal) were used to compare regeneration conditions between plants grown in MS medium supplemented with IAA and BAP. The highest adventitious shooting rate was identified in plants grown on the IAA $1.0mg{\cdot}L^{-1}$ and BAP $2.0mg{\cdot}L^{-1}$ when using the end of petal at the S2 stage. It displayed 79.2% regeneration and produced 33.4 shoots. Rooted plantlets were successfully established in the greenhouse, showing the same morphological characteristics of vegetative and reproductive organs with those of the mother plant. Flow cytometry analysis revealed no ploidy variation between the regenerated plants and the mother plant grown under greenhouse conditions.
