Use of Phenolic Foam as a Medium for Production of Plug Seedlings of Paprika (Phenolic Foam 배지를 이용한 파프리카의 플러그묘 생산)

  • The study was conducted in a glasshouse to examine the possibility of producing paprika plug seedlings in a newly developed inert phenolic foam growing medium. Plug seedlings of 'Fascinato' paprika were grown in five media, Grodan rockwool (Grodan Co. Ltd., Denmark), UR rockwool (UR Co. Ltd., Korea), phenolic foam LC (Smithers Oasis Korea Co. Ltd., Korea), phenolic foam RC, and phenolic foam 3813-4 all in a pellet type. Seeds were germinated in a growth chamber ($25^{\circ}C$, 90% RH, dark) for 4-5 days and then seedlings were grown in a glasshouse with nutrient solution supplied by an overhead irrigation system. Seedling growths were measured 20 days after sowing. The medium pH was the highest in the Grodan rockwool, and medium EC was the highest in phenolic foam 3813-4, although no nutritional excess disorders were observed. Germination rates of paprika were higher than 90% in all the media. Plant height, stem diameter, T/R ratio, leaf area, and chlorophyll showed a similar to those in the rockwool medium. Number of leaves, length of the longest root and dry weights of shoot were not significantly different among treatments. Overall, phenolic foam LC and RC produced seedlings with a similar growth as the rockwool medium. The results obtained suggest that rockwool can be replaced with a new material such as phenolic foam in the commercial scale production of plug seedlings of 'Fascinato' paprika.

  • "Printing is an Art of reproduction & a Technique of However printing has extended its limitation from a genre of reproduction itself and now it stands on the turning point as an art such as painting or sculpture which expresses artist's originality. Early Printing has had relations in depth with press printing in terms of information recording, preservation, and transmission. It was acknowledged value in a way of information satisfaction different from how it is valued as a pure art today. But, later printing has transferred its function from a mean of reproduction to pure art due to the development of printing skills and photography invention. It can be said that the concept of modern printing is taking over its genealogy as a creative work not as just printing. Also its expression capability is widen to dimensional printing and high-tech multimedia from original tradition techniques. As we discussed above, modern painting is very open to various changes. This modern painting aspect can be seen as an extended interpretation of 'board' concept. This dissertation raises a question why monotype and monoprint couldn't secure its position in printing history in spite of numerous artists' tryouts in its way. Monotype and monoprint fundamentally based on intaglio technique in its history. Yet, its systematic study hasn't been worked out. This is because of the lack of recognition of monotype and monoprint's originality as printing. Especially in monoprint, it has known as an early stage in copperplate printing process which is an attempt to solve the technique limitation or trial work for edition. Likewise the reason why monotype and monoprint remains at the edge of printing border ambiguously is because of conceptual, technical characteristics which are against traditional printing. In traditional printing, the concept of board is important as a method of reproduction. Different from the fact, monotype and monoprint accept the form of medium 'board' conceptually out of limited condition as mentioned. Thus monotype and monoprint hasn't stand out for several reasons until late 20th century when it started come out to public as people starts to have interests that works from famous artists are actually based on monotype and monoprint. This dissertation likes to step into the monotype and monoprint theoretically which is not well known in domestic and try to study the meaning of monotype and monoprint as a printing medium which is also hasn't been considered sincerely. For this study the process follow as below. First, look into how monotype and monoprint has a concept and history. Next, check differences through comparison with traditional printing and how printing can be understood in what aspects at the same time. After, verify how monotype and monoprint have influence on the acceptance of extended concept of 'board'. This study will show the expressional possibility of monotype and monoprint which has already known as 'tableau printing' in today's situation where adventurous experiments of printing medium are going on with the development of technology.

  • A micropropagation system for black Locust(Robinia pseudoacacia) was established by using shoots and pin-punctured leaves of in vitro germinated seedlings. The greatest number of shoots (an average of 10.5 shoots) was obtained when shoot tips were cultured on MS medium supplemented with 1.0 mg/l BAP and 0.01 mg/l NAA. When pin-punctured leaf explants were cultured on the same medium, mean number of 13.5 shoots were produced. Shoot growth was accelerated by adding 50 mg/l of silver nitrate ($AgNO_3$), an anti-ethylene compound to the culture medium. Each shoot was excised from the mass and transferred onto half strength MS medium for rooting. Zygotic embryos at different developmental stages were cultured on LS medium supplemented with various growth regulators to induce somatic embryos. When cultured on LS medium with 1.0 mg/l 2,4-D. 14.3% of the zygotic embryos induced somatic embryos. Upon transfer onto the basal medium, somatic embryos sporadically converted into plantlets.

A Cold-Tolerant and Medium-Maturing Italian Ryegrass (Lolium multiflorum Lam.) New Variety, 'Kowinmaster' (내한 중생 이탈리안 라이그라스 신품종 '코윈마스터')

  • This experiment was carried out to breed a cold-tolerant variety of Italian ryegrass (Lolium multiflorum Lam.) in Grassland and Forage Crops Division, National Institute of Animal Science, RDA, Suwon from 1999 to 2006. ew variety, Kowinmaster as a diploid variety was light-green in leaf color and had medium and semi-erect growth habit in late autumn and early spring, respectively. Kowinmaster was 13th May in heading date as a medium-maturing variety. Especially, Kowinmaster was similar to Florida 80 in flag leaf width, longer 3cm in flag leaf length and longer 6cm in plant length than those of control variety, Florida 80, respectively. Stem width of Kowinmaster was similar to that of Florida 80. Cold tolerance of Kowinmaster was better than that of Florida 80. Dry matter yield (9,521 kg/ha) of Kowinmaster was 2% more than that of Florida 80. Feed value of Kowinmaster was 12.1% in crude protein, 70.9% in In vitro dry matter digestibility and 32.9% in acid detergent fiber that were similar to those of Florida 80, respectively, but in neutral detergent fiber, Kowimaster was 56.0% that was 1% lower than that of Florida 80.

Plant Regeneration by in vitro Tissue Culture in Korean Soybean (Glycine max L.) (기내 배양을 통한 국내 콩(Glycine max L.) 품종의 식물체 재분화)

  • Plant regeneration via organogenesis and somatic embryogenesis was investigated in Korean soybean cultivars including Cheongja 3, Jinpumkong 2, Taekwangkong and Uram. Cotyledon, cotyledon+hypocotyl and hypocotyl segments of 7-day-old seedlings were cultured on MS medium containing various concentration (0, 1, 2 and 4 ㎎/L) of BA and TDZ. The results showed that MS medium supplemented with BA 2.0 ㎎/L yielded the highest shoot formation ratio of 83.3%. In 4 cultivars, Taekwangkong showed the highest ratio of shoot formation. When various sizes of immature cotyledons (S: 1∼ 2 ㎜, M: 3∼5 ㎜, L: 6∼8 ㎜) were tested on MS medium containing 2,4-D 40 ㎎/L for somatic embryogenesis, the optimum size for embryogenic callus induction was 3∼5 ㎜ in length of immature cotyledons. In 4 cultivars, Taekwangkong showed the highest percentage of embryogenic callus induction. The results indicate that Taekwangkong is the best soybean cultivar for plant regeneration via organogenesis and embryogenic callus induction among the 4 cultivars.

Perception on Impact of Climate Change on Forest Ecosystem in Protected Area of West Bengal, India

  • In the present exploration we identified perception of forest dependent communities in relation to impact of climate change on forest ecosystem in and around Chilapatta reserve forest in northern part of West Bengal, India. Purposive sampling method was used for selection of area and random sampling method was used for selection of respondent. The data collection in this study was through questionnaire based personal in-depth interviews. Almost all the respondents (94%) were farmers and rest had occupation other than farming. Almost all the respondents perceived negative impact of climate change on forest though the level of perception varies from very low to medium (0.23-0.52) based on average perception score after assigning score to individual statements. The level of perception on impact of climate change on forest ecology and forest flora of the community is low and very low as the average perception score is 0.39 and 0.23, respectively while, it is medium (0.52) for forest fauna. Alternately their perception on decreased stream/river flow and quick drying of seasonal streams or water bodies is based on their livelihood experience as they depend on these for their domestic and irrigation water use and fish catch for family diet.

Elastic local buckling of thin-walled elliptical tubes containing elastic infill material

  • Elliptical tubes may buckle in an elastic local buckling failure mode under uniform compression. Previous analyses of the local buckling of these members have assumed that the cross-section is hollow, but it is well-known that the local buckling capacity of thin-walled closed sections may be increased by filling them with a rigid medium such as concrete. In many applications, the medium many not necessarily be rigid, and the infill can be considered to be an elastic material which interacts with the buckling of the elliptical tube that surrounds it. This paper uses an energy-based technique to model the buckling of a thin-walled elliptical tube containing an elastic infill, which elucidates the physics of the buckling phenomenon from an engineering mechanics basis, in deference to a less generic finite element approach to the buckling problem. It makes use of the observation that the local buckling in an elliptical tube is localised with respect to the contour of the ellipse in its cross-section, with the localisation being at the region of lowest curvature. The formulation in the paper is algebraic and it leads to solutions that can be determined by implementing simple numerical solution techniques. A further extension of this formulation to a stiffness approach with multiple degrees of buckling freedom is described, and it is shown that using the simple one degree of freedom representation is sufficiently accurate for determining the elastic local buckling coefficient.

Selection of Superior Strains from Collected Ear Mushrooms for Artificial Cultivation and Their Optimal Condition of Mycelial Growth (목이류 인공재배를 위한 우량균주 선발 및 균사생장 최적 조건 구명)

  • This study was carried out to obtain morphological and physiological characteristics of ear mushrooms for an artificial cultivation. Eighteen strains were cultivated with bag culture and classified into mainly five groups such as brown, black, white, purple and others group. The highest yield was shown in 43007 strain as 98.3 g/bag and 43009, 43016, 43025 and 44035 strains were more than 60 g/bag. Among collected strains, 43007, 43009 and 43035 were selected in this study as superior strains. Three selected strains were investigated for optimal mycelial growth conditions. MCM and GPYM media were selected for the favorable culture medium. The carbon sources of 43007, 43009 and 43035 on mycelial growth were maltose, fructose and glucose, respectively and peptone was selected as a nitrogen source. Highest mycelial growth was observed when the C/N ratio was 10 for 43007 and 20 for 43009 and 43035.

Introduction of a speedy growing trait into Pleurotus eryngii by backcrossing (큰느타리(새송이)버섯에서 역교배를 통한 속성형질의 도입)

  • Two strains which have good traits in quality and speedy growing were selected based on previous report(Ryu, 2007) to breed a new strain carrying the traits in it. KNR2322 carrying high quality and KNR2503 carrying a speedy growing trait were cultivated to harvest monokaryons. The monokaryotic mycelia were characterized in growth rate, color, and stability. Selfing of monokaryons from KNR2322 and crossing between KNR2322- and KNR2503-derived monokaryons were performed. Two parental lines, $KNR2322-4{\times}37-6{\times}12(B)$ showing thremmatological values in weight and quality, 68.5g and 6.5 and $KNR2322-30{\times}KNR2503-60(A)$ was harvested in 13.5 days after scraping old medium were selected. Crossing between A and B resulted in two high quality and speedy growing strain, A8B8 and A8B10. Their days for harvest after scraping, weight, and quality were 14.0 days, 88.8g and 92.7g, 7.0 and 7.1. Originality tests were conducted with genomic DNAs of A8B8 and Keuneutari 3ho(commercial strain) by DNA fingerprinting a confrontation culture. The results showed polymorphism and a inhibition line between them. It suggested that the new strain have originality from the commercial strain. A8B8 have been registered in Korea Seed and Variety Service with as "Saesongi 1ho".

Ex situ acclimatization of asymbiotically germinated seedlings of endangered lady's slipper orchid(Cypripedium macranthos Sw.) (멸종위기종 복주머니란 발아특성 및 실생묘 현지외 적응성 검토)

  • We use two different methods for laboratory propagation from seed of lady's slipper orchid(Cypripedium macranthos Sw.); immature seed which also called green capsule or fully mature seed about 120~130 days from pollination. In green capsule culture, the seed pods should be collected within precisely right time. The right time of seed collection could be diverse under the wether conditions or nutritional factors of the plants. In fully matured seed culture, the more complicated procedures are needed to break the dormancy of the seed; thermal or chemical treatment. The seedlings in this study were easily germinated from immature seeds in Harvais medium; 53 days after pollination(DAP) in Cypripedium pubescens, DAP 65 in C. parviflorum and C. macranthos. The germinated seedlings were transplanted to hormone free media immediately to avoid abnormal growth of seedlings. When the seedlings have roots with a minimum length of around 2-3cm and have visible dormant buds, the seedlings were removed from the flask and stored in refrigerator for vernalization. To examine the correlation of seedlings and maternal plants, the 125 seedlings of C. macranthos were transplanted in the soil bed at a distance of 20-100 cm from mother plants on April 20. The survival rate of seedlings were 92% in 20 cm distance from the ripe plants, and 56 % in 100 cm distance. The seedlings which were transplanted near mother plants showed vigorous growth in plant height, leaf width, and especially dormant buds. Considering the existence of mycorrhiza which is a symbiotic association between a fungus and the roots of a orchid vascular, the various fungus from mother plants could affect the growth of the seedlings. These results indicate the possibility of high and stable production and practical industrialization of endangered lady's slipper orchids.
