  • The full genome sequence of a Korean white spot syndrome virus (WSSV, isolate: WSSV-GoC18) is presented here. We obtained a total of 12,320,554 reads with 291,172 bases, 170 gene, and 170 coding DNA sequence, which were assembled in 1 contig. Phylogenetic analysis revealed that the WSSV-GoC18 was closely related to Chinese isolate (WSSV-PC) and distinctly different with previously reported a Korean isolate (WSSV K-LV1). The complete genome sequence of WSSV isolates will be of great help in molecular epidemiological studies, contributing to molecular diagnosis and disease prevention in shrimp aquaculture.

  • White spot syndrome virus (WSSV) is a strong causative agent for high mortality in cultured and wild shrimps. From this study, the WSSV prevalence in marine organisms around shrimp farm as well as live feed-mediated transmission of WSSV to farmed shrimps were investigated. Based on nested-PCR method, WSSV was detected in wide array of marine organisms including Perinereis aibuhitensis (81.3% of prevalence rate, 13/16), Enedrias fangi (100%, 16/16), Ruditapes philippinarum (20%, 2/10), crab larvae (100%, 10/10), copepoda (30%, 3/10), Periophthalmus modestus (50%, 5/10), Pachygrapsus crassipes (10%, 1/10), Helice tridens (20%, 2/10) and Neomysis sp. (70%, 7/10). On the other hand, WSSV was not detected in Bullacta exarata, Uca arcuata, and Reishia clavigera. The percent prevalence of WSSV in wild shrimps, Fenneropenaeus chinensis was only 6%, but markedly increased up to 56% after a feeding trial using polychaete worms for one month, indicating that the live feed is one of significant carriers of WSSV to shrimps under practical farming conditions.

  • Dinoflagellates are considered one of the most abundant and diverse groups of marine microplankton and viruses are recognized as one of the significant factors affecting the plankton dynamics. Here, we report basic characteristics of a new dinoflagellate-infecting virus, Heterocapsa pygmaea DNA virus (HpygDNAV) which infects a toxic dinoflagellate, H. pygmaea. HpygDNAV is a polyhedral large virus (ca. 160-170 nm in diameter) propagating in its host's cytoplasm. Because of the virion size, appearance in thin sections, and propagation characteristics, HpygDNAV is assumed to harbor a large double-stranded DNA genome; i.e., HpygDNAV is most likely a nucleocytoplasmic large DNA virus (NCLDV) belonging to the family Phycodnaviridae. Its infectivity is strain-specific, rather than species-specific, as is the case for other algal viruses. The burst size and latent period are estimated to be roughly 100-250 infectious units $cell^{-1}$ and < 96 h, respectively.

  • The susceptibility of five different marine fish to iridovirus IVS-1 infection was analyzed and found a higher the cumulative mortality in the order of rock bream (Oplegnathus fasciatus), red sea bream (Pagrus major), sea perch (Lateolabrax sp.), rockfish (Sebastes schlegeli) and black porgy (Acanthopagrus schlegeli). However, the concentrations of virus in the infected spleens of these species did not differ significantly. To determine the release of iridovirus from infected fish into culturing seawater, rock bream were challenged with iridovirus IVS-1 and the concentration of virus in the water was analyzed using PCR. Over the 10 days of the analysis, the linear relationship between the number of dead fish and viral DNA concentration found in culturing seawater should be considered direct evidence of horizontal iridovirus transmission.

  • White spot syndrome virus (WSSV) causes the most serious epizootic in cultured penaeid shrimp. The epizootic started in 1992, and spread through east and south east Asia and into other shrrimp growing countries of the region. In order to circumvent the epizootic, in many Asian countries, freshwater prawn, Macrobrachium rosenbergii is being widely considered as an alternative species to marine shrimp. (omitted)

  • Despite a variety of development in fish farming during the last decades, fish diseases by bacteria, virus, and parasites are still major problems in aquaculture. Aquaculture of Hounder fish is widely performed around Korea as well as Jeju island, due to relatively stable seed production, short farming period, and a higher value in market.   • Stimulator of interferon gene (STING) is induced by various inflammatory agents, such as lipopolysaccharide and microbial pathogens, including virus and bacteria. In this study, we obtained a full-length cDNA of a STING homolog from olive flounder using rapid amplification of cDNA ends PCR technique. The full-length cDNA of Paralichthys olivaceus STING (PoSTING) was 1442 bp in length and contained a 1209-bp open reading frame that translated into 402 amino acids. The theoretical molecular mass of the predicted protein sequence was 45.09 kDa. In the PoSTING protein, three transmembrane domains and the STING superfamily domain were identified as characteristic features. Quantitative real-time PCR revealed that PoSTING expressed in all the tissues analyzed, but showed the highest level in the spleen. Temporal expression analysis examined the significantly upregulated expression of PoSTING mRNA after viral hemorrhagic septicemia virus (VHSV) stimulation. In contrast, no significant changes in the PoSTING expression were detected in Edwardsiella tarda-challenged group compared to the un-injected control. The expression of P. olivaceus type I interferon (PoIFN-I) was also highly upregulated upon VHSV challenge. These results suggest that STING might be involved in the essential immune defense against viral infection together with the activation of IFN-I in olive flounder.

  • 복부팽만, 안구돌출 및 체색혹화의 외부증상을 대표적으로 나타내며, 세균 및 기생충과 같은 병원체 감염의 원인이 아닌 버나바이러스성으로 판단되어지는 남해안 양식산 넙치, 조피볼락, 농어를 대상으로 RT-PCR 법에 의한 체내 감염 바이러스 검출을 행하였다. IPNV 및 MBV만을 선택적으로 확인 검출 할 수 있으며 감염어체를 사용한 검출에서 배양 세포법(12/50)에 비하여 높은 검출 감도(46/50)를 나타내는 RT-PCR 진단법을 도입하였다. 아울러 우리나라 남해안 양식장에 버나바이러스의 감염이 폭넓게 진행되고 있음을 확인할 수 있었다.

Characterization of histone gene expression in sevenband grouper, Hyporthodus septemfasciatus against nervous necrosis virus infection

  • Recent studies revealed that histone proteins are involved in innate immune responses during pathogen invasion as well as DNA packing. This study characterized the histone genes (H2A.V) of sevenband groupers and analyzed gene expression in NNV-infected sevenband groupers. The open reading frame (ORF) of H2A.V is 387 bp which encoded 128 amino acid residues. The deduced amino acid sequence of H2A.V harbor a highly conserved domain for H2A/H2B/H3 and H2A_C binding domain. Quantitative real-time PCR analysis showed that H2A.V had a high gene expression level in the brain and blood after being NNV-infected. An increase in extracellular histone protein in the blood has been identified as a biomarker for vascular function in humans. More research is required to understand histone's immune response at the protein level or in aquatic animals.

Designing a novel mRNA vaccine against Vibrio harveyi infection in fish: an immunoinformatics approach

  • Vibrio harveyi belongs to the Vibrio genus that causes vibriosis in marine and aquatic fish species through double-stranded DNA virus replication. In humans, around 12 Vibrio species can cause gastroenteritis (gastrointestinal illness). A large amount of virus particles can be found in the cytoplasm of infected cells, which may cause death. Despite these devastating complications, there is still no cure or vaccine for the virus. As a result, we used an immunoinformatics approach to develop a multi-epitope vaccine against most pathogenic hemolysin gene of V. harveyi. The immunodominant T- and B-cell epitopes were identified using the hemolysin protein. We developed a vaccine employing three possible epitopes: cytotoxic T-lymphocytes, helper T-lymphocytes, and linear B-lymphocyte epitopes, after thorough testing. The vaccine was developed to be antigenic, immunogenic, and non-allergenic, as well as having a better solubility. Molecular dynamics simulation revealed significant structural stiffness and binding stability. In addition, the immunological simulation generated by computer revealed that the vaccination might elicit immune reactions in the actual life after injection. Finally, using Escherichia coli K12 as a model, codon optimization yielded ideal GC content and a higher codon adaptation index value, which was then included in the cloning vector pET2+ (a). Altogether, our experiment implies that the proposed peptide vaccine might be a good option for vibriosis prophylaxis.
