Parasites found in Pacific oysters, Crassostrea gigas from Korean waters

  • Pathological conditions of Pacific oysters were surveyed in some coastal areas of Korea. Samples of 30 to 45 oysters were collected in Kamakman, Masan and in Gosung Bay. Protozoan includes gregarine-like organism, Marteilioides chungmuensis, Haplosporidium; ciliates Ancitrocoma and Metazoa (termatode, nematode, copepod) were observed in samples from most of the sampling areas. Marteilioides chungmuensis was found in all sampling sites. Protozoan, Halosporidium sp. and Ancistrocoma sp. occurred in oysters from Gosung Bay (year 2000) with very low prevalence. Trematode Gymnophalloides was frequently found in oysters from all studied areas. Inflammation was frequently observed in oysters infected with M. chungmuensis and Pseudomyicola. The damaged structures were found in digestive tubes of oysters infected by Halosporidium sp. and Pseudomyicola sp. Some of the parasites and pathological status could associate with the conditions of oysters and the environment.

Further observations on the genetics and morphometrics of Coolia santacroce (Dinophyceae)

  • Coolia santacroce is a newly described epibenthic dinoflagellate species collected from the U.S. Virgin Islands. The original description indicates this species is unique from others in the Coolia monotis complex due to the relative size of the apical pore complex, broad range of pore sizes, and ribosomal DNA. The original description was done based on the isolation and cultivation of one isolate of the organism. In this study, we report three more isolates of Coolia santacroce collected from the Bahamas. Morphological observations were made using scanning electron microscopy that do not correspond to those from the original description, indicating the variability of the morphological features. However, analysis of the D1 / D2 regions of the large subunit rDNA places the three strains in a strongly supported monophyletic clade with the type specimen.

  • 공업적 응용 가능성을 검토하기 위하여 한국의 근해 연얀저토에서 분리한 청색색소를 생성하는 Streptomyces sp.의 성질을 조사하였다. 탄 소원은 가용성 전분으로서 1% 였고, pH 7.0 이 상의 약 얄칼리성에셔 최대성장을 하였으며, 다량의 청색색소를 생생하였다.

은편모류 Teleaulax amphioxeia의 윤충류 Brachionus plicatilus 및 이매패류 유생 2종에 대한 먹이 기능성 평가 (Growth Performance of the Rotifer Brachionus plicatilus and the Larvae of Two Bivalves Fed on the Cryptophyte Teleaulax amphioxeia)

  • The aim of this study was to verify the performance of Teleaulax amphioxeia as prey for Brachionus plicatilis and for the larvae of the bivalves (Pacific oyster Crassostrea gigas and Manila clam Ruditapes philippinarum) to select the best of five T. amphioxeia strains. The five strains of T. amphioxeia were collected from the coasts of Korea, purecultured and then fed to the rotifer and bivalve larvae. The density and fecundity rate of the rotifer fed on strain 01 were the highest. The instantaneous growth rate of the rotifer fed strain 08-2 was the highest. The maximum density and fecundity rate of the rotifer fed strain 08-2 were significantly higher than those fed Chlorella ellipsoidea, while instantaneous growth rate showed the opposite pattern. Survival rates of the Manila clam larvae fed each strain of Teleaulax showed no significant difference between strains. Survival rates of the Pacific oyster fed Teleaulax were significantly higher than those fed Chaetoceros sp., but lower than those fed I. galbana. This study indicates that strain 08-2 showed better growth for B. plicatilis and better survival for the bivalve larvae than did the other strains, and that T. amphioxeia can also be used as a prey organism for rotifers or bivalve larvae.

수온, 염분, 사육밀도 및 먹이공급이 왕우럭(Tresus keenae) 유생의 성장과 생존에 미치는 영향 (Effects of Water Temperature, Salinity, Rearing Density and Food Supply on the Growth and Survival of the Surf Clam, Tresus keenae Larvae)

  • 본 연구는 수온, 염분, 사육밀도 및 혼합 먹이생물 공급량과 같은 환경요인이 왕우럭 Tresus keenae의 유생사육 시 생존 및 발생에 미치는 영향을 조사하기 위하여 수행하였다. 연구 결과, 왕우럭 유생사육의 최적 조건은 수온 20℃에서 24℃, 염분 30 psu, 사육밀도 1 ml당 5개체 및 먹이생물 I. galbana, C. simplex 및 T. suecica 혼합 공급량 10×104 세포/ml이였다. 본 연구 결과를 통해 밝혀진 왕우럭 유생 최적의 사육 환경요인은 빠른 성장 및 높은 생존을 통해 생산성 향상에 기여하리라 생각된다.

신속한 항생제 감수성 테스트 법의 개발을 위한 Water-Soluble Tetrazolium Salt의 적용 (Application of water-soluble tetrazolium salt for development of rapid antimicrobial susceptibility testing methods)

  • In this study, we conducted to the development of a rapid antimicrobial susceptibility test method using WST-1 which is known to water-soluble tetrazolium salt, in order to rapidly response against bacterial diseases in fish. Eight of antibiotics which are permissioned for marine organism from government were used to rapid antimicrobial susceptibility testing using the WST-1. As a result, a similar tendency was verified compare to conventional antibiotic susceptibility test results. Generally, the antibiotic susceptibility test method required about 3 days (72 hours) for determine the effective antibiotics, however, we have confirmed that the our method using WST-1 was required at least 36 hours in this study. Consequentially, our method will contribute to development of rapid antimicrobial susceptibility testing for bacterial diseases in fish.

Cochlodinium polykrikoides의 성장에 미치는 물리$\cdot$화학적 요인과 영양염 이용 (Physico-Chemical Factors on the Growth of Cochlodinium polykrikoides and Nutrient Utilization)

  • 우리나라 남해안에서 매년 유해 편모조류인 Cochlodinium polykrikoides 적조가 발생하고 있으며 많은 수산피해를 일으키고 있다. 본 연구는 본 종에 대한 적조발생기구를 구명하는데 있어 생물학적인 기초를 확립하기 위해 실시되었다. 실험실 배양실험을 통해 물리 화학적 환경요인에 대한 C. polykrikoides의 성장을 조사하였고, 그 결과를 요약하면 다음과 같다. 해양에서 발생하는 C. polykrikoides 자연군집의 동역학중에서 물리$\cdot$화학적 환경요인의 상대적인 중요성을 평가하기 위해 수온, 염분, pH, 조도 조건별로 무균배양을 통한 본 종의 성장을 조사하였다. 그 결과는 $25^{\circ}C,\;40\%_{\circ}$, pH 7.5, 7,500 lux에서 가장 성장률이 높았다. 그리고, $19\~28^{\circ}C$에서도 성장이 잘 이루어져 광온성(eurythermal) 종으로 확인되었으며, 발생해역의 염분 범위인$30\~35\%$의 환경조건을 만족하는, 즉 내만의 저염분 보다는 외양의 고염분 환경에서 발생하여 성장하는 종으로 확인되었고, 광염성 (euryhaline) 종이다. 그리고, 높은 일사량을 요구하며 현장해역의 pH 범위에서 성장이 원활히 이루어졌다. 무기태 영양염인 질소원으로 질산질소와 암모니아질소를, 인원으로 인산인을 이용하여 C. polykrikoides의 영양염에 대한 성장실험을 하였다. 그 결과, C. polykrikoides의 영양요구량은 질산질소, 암모니아질소, 인산인에 대해 각각 $40, 50, 5{\mu}M$이었고, 성장에 의한 반포화상수 (Ks)값은 각각 $2.10,\;1.03,\;0.57{\mu}M$로 확인되었다. 이러한 결과로부터 C. polykrikoides는 타와편모조류에서 보고된Ks 값보다 대체적으로 낮았고, 빈영양성 (oligotrophic) 종은 아니며, 부영양성 (eutrophic) 종에 해당하지만 암모니아질소 Ks 값은 뚜영양성 종보다 낮은 값을 가지는 것으로 나타났다. 이것은 C. polykrikoides 적조는 내만의 부영양화된 해역보다는 부영양상태가 높아지고 있는 해역에서 발생할 가능성이 매우 높은 것을 나타낸다. 또한, C. polykrikoides는 질산질소보다 암모니아질소를 성장시 보다 유효한 영양염으로 선택하여 소비하였다 이것은 암모니아질소가 타 영양염보다 성장에 보다 중요한 요인으로 작용하며, C. polykrikoides 적조는 해양에서 증가한 암모니아질소 농도와 연관되는 것으로 나타났다.

통영 연안 가두리 양식용 그물에 가입되는 오손생물군집의 시·공간적 비교 (Spatial-temporal Variation of Fouling Organisms Recruited on a Fishing Net based on Community Structure, Tongyeong, Southern Coast of Korea)

  • To understand the composition and community changes of benthic species by water depth (5 m, 20 m, 30 m) in the cage facilities, net substrate made of fish cages were installed at intervals of 2 months from March to September 2019, which is the main period of sessile organism recruitment. Water temperature and salinity varied in the ranges of 1~5℃ and 1~4 mg/L at 5 m from June to August, respectively, and less difference was observed according to water depth in other investigation periods. A total of 37 benthic animals were recruited, and Kamptozoa unid., Caprella scaura and Jassa slatteryi were verified during the entire study period. Mytilus galloprovincialis, a Mediterranean mussel, showed seasonality dominant only from spring to early summer (March to July). The community structure classified by temporal and spatial factors, and Bugula neritina, Kamptozoa unid., and Sertularella sp. contributed to the formation of the community. The depth of 5 m was exposed to environmental changes in water temperature and salinity temporally, so there was a seasonal variation in the composition of the sessile organism, but at depths of 20 m and 30 m, there was less significant environmental change and it showed relatively stable than 5 m.

Changes in Caenorhabditis elegans Exposed to Vibrio parahaemolyticus

  • Vibrio parahaemolyticus, which owes its origin to the marine environment, is considered as one of the most common causes of infectious diarrhea worldwide. The present study investigated the pathogenicity of V. parahaemolyticus against the model organism, Caenorhabditis elegans. Infection in the host was localized with GFP-tagged V. parahaemolyticus using confocal laser scanning microscopy. The times required for causing infection, bacterial load in intestine, chemotactic response, and alteration in pharyngeal pumping were analyzed in the host system. In addition, the regulation of innate immune-related genes, lys-7, clec- 60, and clec-87, was analyzed using real-time PCR. The role of immune-responsible pmk-1 was studied using mutant strains. The pathogenicity of environmental strain CM2 isolated from the Gulf of Mannar, India was compared with that of a reference strain obtained from ATCC. The pathogen infected animals appeared to ward off infection by up-regulating candidate antimicrobial genes for a few hours after the exposure, before succumbing to the pathogen. For the first time, the pathogenicity of V. parahaemolyticus at both the physiological and molecular levels has been studied in detail using the model organism C. elegans.

Protoplast Production from Sphacelaria fusca (Sphacelariales, Phaeophyceae) Using Commercial Enzymes

  • Sphacelaria is a filamentous brown algal genus that can be epibiotic on macroalgae, marine plants, and sea turtles. Its important role in benthic ecosystems, exposure to different stressors (e.g., grazing), and use as a model organism make Sphacelaria ideal for assessing physiological responses of organisms to environmental inputs. Single-cell RNA sequencing is a powerful new probe for understanding environmental responses of organisms at the molecular (transcriptome) level, capable of delineating gene regulation in different cell types. In the case of plants, this technique requires protoplasts ("naked" plant cells). The existing protoplast isolation protocols for Sphacelaria use non-commercial enzymes and are low-yielding. This study is the first to report the production of protoplasts from Sphacelaria fusca (Hudson) S.F. Gray, using a combination of commercial enzymes, chelation, and osmolarity treatment. A simple combination of commercial enzymes (cellulase Onozuka RS, alginate lyase, and driselase) with chelation pretreatment and an increased osmolarity (2512 mOsm/L H2O) gave a protoplast yield of 15.08 ± 5.31 × 104 protoplasts/g fresh weight, with all the Sphacelaria cell types represented. Driselase had no crucial effect on the protoplast isolation. However, the increased osmolarity had a highly significant and positive effect on the protoplast isolation, and chelation pretreatment was essential for optimal protoplast yield. The protocol represents a significant step forward for studies on Sphacelaria by efficiently generating protoplasts suitable for cellular studies, including single-cell RNA sequencing and expression profiling.
