클론된 웅성 특이 DNA절편에 의한 돼지의 성결정 (Sex Determination in Somatic and Embryonic Cells of the Pig by Cloned Male-Specific DNA Fragments)

  • 3.3kb 웅성특이 DNA(pEM39 plasmid DNA)가 성 특이 DNA 검색자로 활용되어질 수 있는가를 확인하기 위하여 구조적인 분석을 Southern blotting, DNA sequencing과 computer program 분석을 통하여 실시하였다. 전체 3.3kb에서 유래된 약 1kb 단위의 단편을 이용하여 표지된 짧은 DNA probe들은 Southern blot 분석에서 웅성특이성을 나타내었다. McGraw와 Jeon의 sequence에 대한 유사성 비교 자료로부터 여러 부분의 conserved region을 찾아내고 이것을 기초로 하여 5개의 primer set들을 선발하였다. Conserved region에 존재하면서 computer program에 의해서 선발되어진 PMS1과 2의 primer set가 최종적으로 PCR 분석을 위하여 선정되었다. 이 primer set를 사용한 PCR 분석에서, 1ng부터 10pg까지의 웅성 genomic DNA에서 PCR 산물을 얻을 수 있었으며, 자성의 경우는 어떠한 산물도 찾을 수 없었다. PCR에 이용할 수정란의 시료는 2 세포기의 수정란에서 얻었으며 순수 분리된 genomic DNA에서 확립된 조건에서 PCR을 수행하였다. 8개의 수정란을 분석한 결과 4개의 웅성과 4개의 자성 수정란을 확인하였다. 이러한 결과는 선정된 primer set가 돼지 수정란의 성을 조기 감별하는데 효율적인 DNA probe로 사용될 수 있다는 것을 암시한다.

우리나라 시.도별 자살 사망 분석 (A Study on Regional Differentials in Death Caused by Suicide in South Korea)

  • Purpose: The purpose of this study was to compare suicide mortality by region in South Korea. Method: Suicide mortality differentials were calculated for several mortality indicators by geographical regions from raw data of the cause of death from KNSO. Results: The results are as follows; the Crude suicide death rate was 22.63 per 100,000. The highest was in Kangwon showing 37.84% whereas, Chungnam, and Jeonbuk followed after. Suicide was 4.4% of all causes of death, but Inchon and Ulsan showed a higher proportion. The male suicide death rate was 31.12 per 100,000 and females 14.09. The ratio of gender suicide mortality was 2.21, per 100,000 and was the highest in Jeju. For age-specific suicide death rates, the rate increased as age advanced, showing 2.33 per 100,000 in 0-19years, 18.68 in 20-39, 30.48 in 40-59, 63.33 in 60 years and over. In Ulsan, Kangwon, and Inchon, age-specific suicide death rates of the 60 and over age group were higher than other regions, Daegu, Busan, and Kangwon showed a higher age-specific suicide mortality of the 40-59 age group, and Kangwon, Jeonnam, and Chungnam had a higher age-specific suicide mortality of the 20-39 age group. Conclusions: Suicide mortality differed by region. These results can be used for a regional health care plan and planning for suicide prevention by regions.

Prevalence of Dokha Use among Secondary School Students in Ajman, United Arab Emirates

  • Background: Dokha is a novel form of smoking in United Arab Emirates (UAE) on which there is very little published literature, especially among adolescents, and this form of smoking has been not been addressed adequately in the smoking cessation strategies in the UAE. Objectives: To assess the prevalence of dokha smoking among male secondary school students in Ajman UAE. Materials and Methods: A cross sectional survey was conducted among male secondary school students in Ajman, UAE. A total of 560 participants filled in self-administered questionnaires. Results: The prevalence of ever smokers was 39%; ever dokha smokers was 36% and current dokha smokers was 24%. The prevalence is very high when compared to other forms of smoking in the region and globally. Prevalence increased with increasing age and grade of students, prevalence of ever smokers, ever dokha smokers and current dokha smokers was lower in students in the science stream. 40% of the smokers used dokha, cigarettes and shisha, 30% used dokha and cigarettes, and 21% used dokha alone. 30% of the students smoked dokha on all days of the month. Conclusions: The prevalence of ever smokers, ever dokha smokers and current dokha smokers is very high. There is an urgent need for specific health promotion programs tailored to this age group on prevention of dokha smoking and policies restricting the availability of dokha to this age group.

Identification of Species and Sex of Korean Roe Deer (Capreolus pygargus tianschanicus) Using SRY and CYTB Genes

  • The nucleotide sequences of a male-specific marker sex determining region Y (SRY) gene and a mitochondrial cytochrome B (CYTB) gene were characterized and analyzed to establish a molecular method for identification of species and sex of Korean roe deer (Capreolus pygargus tianschanicus). Similarity search result of SRY sequences showed very similar result to those reported in Moose (Alces alces) and Reindeer (Rangifer tarandus), both of which had 95.9% similarity in identity. CYTB genes were very similar to those reported in Siberian roe deer (C. pygargus pygargus) which had 98.6% similarity and not to European roe deer (C. capreolus), suggesting that the DNA samples tested were of Siberian roe deer lineage. Polymerase chain reaction (PCR)-based sex typing successfully discriminated between carcasses of male and female roe deer. Males had SRY band on agarose gels and females did not. The result of this molecular sex typing provided similar information with that obtained by genital organ observation. Therefore, this molecular method using male specific marker SRY and mitochondrial CYTB genes would be very useful for identification of the species and sex of the carcass remains of roe deer.

MRI와 MRA를 이용한 허혈성 뇌혈관 질환의 뇌혈관별 분포에 대한 연구 (Intracerebral Regional and Vasculature-Specific Distributions of Ischemic Cerebrovascular Diseases: Using MRI and MRA)

  • 본 연구에서는 MRI와 MRA를 이용하여 우리나라 사람의 뇌혈관 질환 중 뇌혈관의 파열에 의한 뇌혈관 질환을 제외하고 뇌혈관이 좁아지거나 막힌 허혈성 뇌혈관 질환의 분포(호발부위)를 분석하여 임상에서 활용할 수 있도록 하였다. 뇌혈관이 좁아지거나 막힌 허혈성 뇌혈관 질환만의 분포를 분석한 이유는 뇌혈관의 파열에 의한 출혈성 뇌혈관 질환과 뇌경색(brain infarction) 등 허혈성 뇌혈관 질환의 발생학적 기전(mechanism)이 다르기 때문이며 그 결과는 아래와 같다. 1. 연구대상자 총 626명 중 성별로는 남자가 55.0%로 여자 45.0%보다 높은 분포를 보였다. 2. 위치별 허혈성 뇌혈관 질환의 분포에서 우측 뇌혈관이 37.5%로 가장 많았으며, 좌측 뇌혈관 35.1%, 양쪽 뇌혈관 27.3% 순으로 나타났다. 3. 허혈성 뇌혈관 질환의 뇌혈관 별 분포는 ICA가 38.9%로 가장 많았으며, 다음으로 MCA 35.7%, PCA 13.4%, ACA 6.0%, VA 3.3% 순으로 나타났다. 4. A-com에 질환이 있는 경우는 한 명도 없었으며 P-com 에도 남자 1명만이 발생했다. 5. MCA에는 여자가 54.6%로 남자 42.2%보다 많아, 성별에 따라 통계적으로 유의미한 차이를 보였다($x^2$=9.64, p < .01). 6. ICA에서는 남자가 56.4%로 여자 46.8%보다 많아 통계적으로 유의미한 차이를 보였다($x^2$=5.71, p < .05). 7. BA에서도 남자가 2.3%로 여자 0.4%보다 많아 성별에 따라 유의미한 차이를 보였다($x^2$=4.25, p < .05). 8. 연령에 따른 혈관별 질환의 분포에 대한 차이는 없었으며 협착과 폐색의 분포에서는 폐색보다 협착이 많았다.

배추과 작물의 자가불화합성 유전자의 발현 및 조절 (Expression and regulation of self-incompatible genes in Brassica)

  • In most self-incompatible plant species, recognition of self-pollen is controlled by a single locus, termed the S-locus. The self-incompatibility (SI) system in Brassica is controlled sporophytically by multiple alleles at a single locus, designated as S, and involves cell-cell communication between male and female. Two highly polymorphic S locus genes, SLG (S locus glycoprotein) and SRK (S receptor kinase), have been identified, both of which are expressed predominantly in the stigmatic papillar cell. Gain-of-function experiments have demonstrated that SRK solely determines S haplotype-specificity of the stigma, while SLG enhances the recognition reaction of SI. The sequence analysis of the S locus genomic region of B. campestris (syn. rapa) has led to the identification of an anther-specific gene, designated as SP11/SCR, which is the male S determinant. Molecular analysis has demonstrated that the dominance relationships between S alleles in the stigma were determined by SRK itself, but not by the relative expression level. In contrast, the expression of SP11/SCR from the recessive S allele was specifically suppressed in the S heterozygote, suggesting that the dominance relationships in pollen were determined by the expression level of SP11/SCR. Furthermore, recent studies on recessive allele-specific DNA methylation of Brassica self-incompatibility alleles demonstrate that DNA methylation patterns in plants can vary temporally and spatially in each generation. In this review, we firstly present overview of self incompatibility system in Brassica and then describe dominance relationships in Brassica self- incompatibility regulated by allele-specific DNA methylation.

펑크헤어스타일의 근원 고찰 -닭 볏 머리형을 중심으로- (Studies on the Origin of Punk Hair Style -With Specific Emphases on Cockcomb Hair Style-)

  • The objective of this research is to identify and examine the punk hair styles which, with their various, rather vague names, help to describe the twentieth century subordinate culture or subculture and also their place today as one of the mainstream male hair trends. The scope of this research will stretch to include also: North-east and around that areas native Americans'hair styles which, are thought to have strong links with the Punk hair tend. The focus of this research will remain with the so-called Mohican or Mohawk hair styles(collectively referred to as Punk styles), which have originated mainly from native American warrions residing in North-east and South-east region of the United States. This study clarified a few questions in doubt concerning the origin of cockcomb hairstyle. Firstly, the cockcomb hairstyle known to represent punk hairstyle was influenced the American Indian's Roach style. Secondly, since Roach style is referred as Mohawk tribe's hairstyle in some refernces, the view of seeing cockcomb hairstyle as a Mohawk style is considered more persuasive. Thirdly, although Roach style was a usual hairstyle through North-east and South-east region of the United States, the people in South-east region did Roach hair style because of the North-east's influences. Lastly alike the Punkers had several kinds of cockcomb hairstyle. American Indians'Roach style also differed in shape concerning of their regions and their kindred. In addition, the reason why the most common term to refer punk hairstyle became Mohican is considered as that the film 'Last Mohican' has influenced on it. That film can appeal their history and play a good role of mediator to let the public know better about themselves, 'Mohican' naturally became the name of the specific hair style.

최근의 사망패턴에 관한 고찰 (Recent Pattern of Mortality in Korea)

  • In this paper, an attempt has been made to examine the pattern of mortality in Korea during 1970~80. By applying the age-sex specific mortality rates quoted from 1978~79 life tables for Korea published by NBOS, EPB to those of the West pattern of regional model life tables and the far eastern pattern of model life tables for developing countries, life expectancy at birth were calculated. Also the author reviewed the trends of death rates, life expectancy and cause of death using vital registration data and other materials. Summarized results are as follows; 1. Crude death rates in Korea was reduced to one fifth in the 1983 compared to that in 1920's. Life expectancy also improved to almost double in 1985 compared to 1920's. But the difference in the life expectancy between male and female increased during that period and it was recorded as 6.4 years in 1985. This discrepancy was mainly due to the different tempo of decreasing in mortality level by sex, particularly, for the age 40 and above. 2. For the pattern of mortality in Korea, it showed that female mortality could accounted closer to the West pattern model life tables. There were high similarity between actual pattern prevalent in Korea and West pattern. And its coefficient of variance was also very low. However for the case of male, it was difficult to find the exact model life tables for explaining the actual situation on the male mortality pattern which means exist considerable dissimilarity in older ages. The Far eastern pattern of U.N. model life tables show better results than West pattern, however, the deviation of the pattern to actual was severe. Also in Far eastern pattern, high coefficient of variance was existed. Furthermore it was found in the paper that the mortality level of Korean male for the age 40 and above were much higher than that of Far eastern pattern which was reflected the high mortality of the male adult in Far east region. 3. The analysis of cause of death showed that circulatory disease such as cerebrovascular disease and hypertensive disease accounted for the leading cause of death in Korea for the age 40 and above. There should he paid special attention to chronic retrogressive diseases for the older age groups. For younger age groups, injury and poisoning were reported as important cause of death.

Multiplex PCR을 이용한 은행나무 수나무 식별용 SCAR 마커 개발 (Development of SCAR Marker for Identifying Male Trees of Ginkgo biloba using Multiplex PCR)

  • 은행나무는 이식성이 좋고 열악한 환경에서도 잘 자라기 때문에 도심 가로수나 조경수로 매우 적합한 수종이다. 은행나무는 암수딴그루 식물로 수나무와 암나무의 생식기관(수꽃과 암꽃)의 비교를 통하여 성을 식별할 수 있으나, 꽃이 달리기까지 약 20년 이상이 필요하다. 가로수용 은행나무는 주로 꽃이 생성되기 이전에 식재되기 때문에 암나무와 수나무 구분 없이 가로수로 식재되어왔다. 가로수로 식재된 암나무에서 열리는 은행나무 열매는 악취를 발산하고 거리 오염을 야기하고 있다. 따라서 본 연구에서는 유시에 수나무를 선별하기 위하여 기존에 보고된 수나무 특이 RAPD 변이체의 염기서열을 기반으로 수나무에서만 675 bp의 PCR 산물을 증폭하는 SCAR 마커(SCAR-GBM)를 개발하였다. SCAR-GBM 마커의 우성 마커 특성에 기인한 위음성(false-negative)문제를 해결하고 식별 효율을 향상시키기 위하여 SCAR-GBM 마커와 mtDNA의 atp1 영역을 증폭하는 범용 프라이머를 동시에 적용하는 multiplex PCR을 이용하였다. 그 결과 암나무와 수나무에서 공히 atp1 영역에 해당하는 1,039 bp의 PCR 산물이 증폭되었으며, 수나무에서만 특이적으로 SCAR-GBM 마커가 증폭되었다. 전국 8개 지역에서 채취된 암나무와 수나무 각 80개체에 대한 분석을 통하여 SCAR-GBM 마커와 multiplex PCR 방법의 재현성이 확인되었다.

뇌실주위 배아종 - 증례보고 - (Periventricular Germinoma - Case Report -)

  • Germinomas of the central nervous system are rare embryonal tumors(accounting for less than 1% of intracranial neoplasms) that may be located in the pineal region, in the floor of the third ventricle, or in the suprasellar area. We report a case of germinoma developed in periventricular deep white matter without pineal region tumors or suprasellar masses. The 19-year-old male patient presented with slowly progressing headache, dizziness, photophobia, and dysarthria. Initial brain MRI revealed a irregular and dense enhancement from lateral ventricles to 4th ventricle. The stereotactic biopsy of tumor and histologic examination revealed the germinoma. Craniospinal axis radiation therapy was performed. After radiation therapy patient was improved and no neurologic sequelae was seen at discharge. Periventricular germinomas without pineal or suprasellar lesion are very rare. The radiation therapy, as in our case, is beneficial as with other intracranial germinomas. Stereotactic biopsy of periventricular germinoma provides precise pathologic diagnosis and thus allows more specific management.
