Recently three proteins, playing central roles in the bidirectional transport of urate in renal proximal tubules, were identified: two members of the organic anion transporter (OAT) family, OAT1 and OAT3, and a protein that designated renal urate-anion exchanger (URAT1). Antibodies against these transporters are very important for investigating their expressions and functions. With the cytokine gene as a molecular adjuvant, genetic immunization-based antibody production offers several advantages including high specificity and high recognition to the native protein compared with current methods. We fused high antigenicity fragments of the three transporters to the plasmids pBQAP-TT containing T-cell epitopes and flanking regions from tetanus toxin, respectively. Gene gun immunization with these recombinant plasmids and two other adjuvant plasmids, which express granulocyte/macrophage colony-stimulating factor and FMS-like tyrosine kinase 3 ligand, induced high level immunoglobulin G antibodies, respectively. The native corresponding proteins of URAT1, OAT1 and OAT3, in human kidney can be recognized by their specific antibodies, respectively, with Western blot analysis and immunohistochemistry. Besides, URAT1 expression in Xenopus oocytes can also be recognized by its corresponding antibody with immuno-fluorescence. The successful production of the antibodies has provided an important tool for the study of UA transporters.
Many Lactobacillus strains have been promoted as good probiotics for the prevention and treatment of diseases. We engineered recombinant Lactobacillus casei, producing biologically active canine granulocyte macrophage colony stimulating factor (cGM-CSF), and investigated its possibility as a good probiotic agent for dogs. Expression of the cGM-CSF protein in the recombinant Lactobacillus was confirmed by SDS-PAGE and Western blotting methods. For the in vivo study, 18 Beagle puppies of 7 weeks of age were divided into three groups; the control group was fed only on a regular diet and the two treatment groups were fed on a diet supplemented with either $1\times10^9$ colony forming units (CFU)/day of L. casei or L. casei expressing cGM-CSF protein for 7 weeks. Body weight was measured, and fecal and blood samples were collected from the dogs during the experiment for the measurement of hematology, fecal immunoglobulin (Ig)A and IgG, circulating IgA and IgG, and canine corona virus (CCV)-specific IgG. There were no differences in body weights among the groups, but monocyte counts in hematology and serum IgA were higher in the group receiving L. casei expressing cGM-CSF than in the other two groups. After the administration of CCV vaccine, CCV-specific IgG in serum increased more in the group supplemented with L. casei expressing cGM-CSF than the other two groups. This study shows that a dietary L. casei expressing cGM-CSF enhances specific immune functions at both the mucosal and systemic levels in puppies.

Objective: The inhibitory leukocyte immunoglobulin-like receptors (LILRBs) play an important role in innate immunity. The present study represents the first description of the cloning and structural and functional analysis of LILRB1 and LILRB3 isolated from two genetically disparate chicken lines. Methods: Chicken LILRB1-3 genes were identified by bioinformatics approach. Expression studies were performed by transfection, quantitative polymerase chain reaction. Signal transduction was analyzed by western blots, immunoprecipitation and flow cytometric. Cytokine levels were determined by enzyme-linked immunosorbent assay. Results: Amino acid homology and phylogenetic analyses showed that the homologies of LILRB1 and LILRB3 in the chicken line 6.3 to those proteins in the chicken line 7.2 ranged between 97%-99%, while homologies between chicken and mammal proteins ranged between 13%-19%, and 13%-69%, respectively. Our findings indicate that LILRB1 and LILRB3 subdivided into two groups based on the immunoreceptor tyrosine-based inhibitory motifs (ITIM) present in the transmembrane domain. Chicken line 6.3 has two ITIM motifs of the sequence LxYxxL and SxYxxV while line 7.2 has two ITIM motifs of the sequences LxYxxL and LxYxxV. These motifs bind to SHP-2 (protein tyrosine phosphatase, non-receptor type 11) that plays a regulatory role in immune functions. Moreover, our data indicate that LILRB1 and LILRB3 associated with and activated major histocompatibility complex (MHC) class I and ${\beta}2-microglobulin$ and induced the expression of transporters associated with antigen processing, which are essential for MHC class I antigen presentation. This suggests that LILRB1 and LILRB3 are transcriptional regulators, modulating the expression of components in the MHC class I pathway and thereby regulating immune responses. Furthermore, LILRB1 and LILRB3 activated Janus kinase2/tyrosine kinase 2 (JAK2/TYK2); signal transducer and activator of transcription1/3 (STAT1/3), and suppressor of cytokine signaling 1 genes expressed in Macrophage (HD11) cells, which induced Th1, Th2, and Th17 cytokines. Conclusion: These data indicate that LILRB1 and LILRB3 are innate immune receptors associated with SHP-2, MHC class I, ${\beta}2-microglobulin$, and they activate the Janus kinase/signal transducer and activator of transcription signaling pathway. Thus, our study provides novel insights into the regulation of immunity and immunopathology.
BACKGROUND/OBJECTIVES: Previous studies have reported that protein supplementation contributes to the attenuation of inflammation. Serious trauma such as burn injury usually results in the excessive release of inflammatory factors and organs dysfunction. However, a few reports continued to focus on the function of protein ingestion in regulating burn-induced inflammation and organ dysfunction. MATERIALS/METHODS: This study established the rat model of 30% total body surface area burn injury, and evaluated the function of blended protein (mixture of whey and soybean proteins). Blood routine examination, inflammatory factors, blood biochemistry, and immunohistochemical assays were employed to analyze the samples from different treatment groups. RESULTS: Our results indicated a decrease in the numbers of white blood cells, monocytes, and neutrophils in the burn injury group administered with the blended protein nutritional support (Burn+BP), as compared to the burn injury group administered normal saline supplementation (Burn+S). Expressions of the pro-inflammatory factors (tumor necrosis factor-α and interleukin-6 [IL-6]) and chemokines (macrophage chemoattractant protein-1, regulated upon activation normal T cell expressed and secreted factor, and C-C motif chemokine 11) were dramatically decreased, whereas anti-inflammatory factors (IL-4, IL-10, and IL-13) were significantly increased in the Burn+BP group. Kidney function related markers blood urea nitrogen and serum creatinine, and the liver function related markers alanine transaminase, aspartate aminotransferase, alkaline phosphatase, and lactate dehydrogenase were remarkably reduced, whereas albumin levels were elevated in the Burn+BP group as compared to levels obtained in the Burn+S group. Furthermore, inflammatory cells infiltration of the kidney and liver was also attenuated after burn injury administered with blended protein supplementation. CONCLUSIONS: In summary, nutritional support with blended proteins dramatically attenuates the burn-induced inflammatory reaction and protects organ functions. We believe this is a new insight into a potential therapeutic strategy for nutritional support of burn patients.
연구배경: Interleukin-12 (IL-12) 는 자연살해세포와 T 림프구 활성을 증가시킬 뿐 아니라 증식을 촉진시키고 mterferon-$\gamma$와 granulocyte-macrophage colony stimulating factor 그리고 tumor necrosis factor-$\alpha$ (TNF-$\alpha$)의 생성을 유도한다고 알려져 있으며 여러 동물실험에서 악성종양의 치료에 상당한 효과가 있음이 알려져 있다. 이런 결과는 종양내 염증세포의 침윤이 증가함과 밀접한 관련이 있다고 밝혀져 있지만 이들 염증세포가 종양내로 이동하는 단계에 IL-12가 미치는 영향에 대해서는 잘 알려진 바가 없다. 본 연구에서는 IL-12가 종양의 성장 및 전이에 미치는 영향과 활성화된 내피세포에 발현되는 것으로 알려진 E-selectin의 발현에 미치는 영향을 관찰하고자 하였고 또한 E-selectin의 발현이 IL-12에 의해 생산이 증가되고 여러 유착분자의 발현을 유도하는 작용이 있는 것으로 알려진 TNF-$\alpha$와 연관이 있는지 알아보고자 하였다. 방법: C57BL/6 마우스를 대상으로 Lewis 폐암세포주를 피하주사하여 암세포를 이식한 후 각 군마다 각각 IL-12 또는 TNF-$\alpha$ 생리식염수를 복강내 투여하였다. 암세포이식 28일째 마우스를 도살하여 폐전이 결절의 수를 측정하였으며 피하종양에 대해 CD4, CD8, CD16, E-selectin에 대한 면역화학조직염색을 시행 하였다. 결과: IL-12를 투여한 군에서 대조군에 비해 암세포 이식부위의 종양 크기가 감소하였으며 폐전이 결절의 수도 감소하였다. IL-12를 투여한 군에서 대조군에 비해 CD4+ 및 CD8+ 그리고 CD16+ 세포의 종양내 침윤이 증가하였다. IL-12를 투여하지 않은 대조군에서 E-selectin은 발현되지 않았으나 IL-12 투여군에서 종양내 혈관내피세포에서의 E-selectin 발현이 증가되었다. TNF-$\alpha$ 투여시 종양내 혈관내피세포에서의 E-selectin 발현 증가가 관찰되었으며 또한 종양내 CD4+ 및 CD8+ 그리고 CD16+ 세포의 침윤이 증가되었다. 결론: 이상의 결과로 보아 IL-12는 Lewis 폐암모델에서 종양내 염증세포의 침윤을 일으켜 종양의 성장과 전이를 억제하는 작용을 나타내며 염증세포의 침윤에는 E-selectin이 관여할 것으로 생각된다. 또한 E-selectin의 발현증가에는 IL-12 에 의해 생성이 유도되는 TNF-$\alpha$가 작용할 가능성을 추정할 수 있겠다.
본 연구는 기존의 불가사리 콜라겐 유래 펩타이드, 저분자화된 펩타이드, 저분자화 된 펩타이드의 젤라틴 나노입자의 비교를 통해 면역 활성의 증진을 확인하였다. 불가사리 펩타이드는 Sephadex G-75 gel 크로마토그래피 분리를 통해 5-7 kDa 범위의 분자량 범위의 시료를 얻었고, MALDI-TOF MS 분석에 의해 펩타이드의 정확한 분자량을 측정하였다. 0.1% collagenase를 처리한 펩타이드의 분자량은 기존의 불가사리 콜라겐 유래 펩타이드에 비해 약 2,000 m/z까지 분자량이 감소되는 효과를 보였다. 이는 2개의 폴리펩티드결합으로 상호 결합되어 감겨있던 콜라겐형구조가 collagenase의 작용으로 풀어진 후 효소의 영향을 받아 가수분해되었기 때문인 것으로 사료된다. 이후 $20^{\circ}C$에서 젤라틴으로 시료를 나노입자화 하여 TEM 및 DLS로 분포와 크기를 각각 확인하였다. 이후 인간섬유아세포에 대한 세포독성 측정 결과, 나노입자화 한 ALPG가 최고 농도인 1.0 mg/mL의 농도에서 11.64%로 가장 낮게 나타났다. 각 시료의 면역물질의 분비 정도를 확인하기 위해 macrophage에서의 $NO^-$ 분비를 측정하였는데 역시 나노 입자화 한 ALPG가 $40\;{\mu}M$로 가장 높은 $NO^-$ 생성량을 보였다. 또한 불가사리 나노 시료가 저분자 추출물과 비교해 B cell 생육도를 10% 이상 향상시켰다. UV 자극에 의해 형성되는 염증물질로 잘 알려진 prostaglandin의 생성을 정량하기 위해 $PGE_2$ 방법을 이용하였는데, 나노입자를 가해준 모든 군에서 농도 의존적으로 $PGE_2$의 생성이 감소하였고 역시 ALPG가 860 pg/mL로 $PGE_2$를 가장 적은 농도로 생성하였다. 마지막으로 confocal 현미경을 이용해 인간 면역 세포에 나노입자가 얼마나 효과적으로 침투하는지를 관찰하였는데 나노 시료의 경우 입자의 크기가 50-200 nm로 분산됨에 따라 일반 불가사리 펩타이드에 비해 세포 침투의 용이성이 크게 증진되는 것을 확인하였다.
왕느릅나무(Ulmus macrocarpa)의 껍질을 말린 유근피는 오랫동안 부종, 감염 및 염증 제어의 목적으로 사용되어져 왔음에도 불구하고 잠재적 면역조절 효과에 관해서는 연구가 이루어진 바 없다. 본 연구에서는 전통 약용자원에서 새로운 면역기능 증가 신소재 발굴의 일환으로 유근피 열수 추출물의 면역 조절 효능을 RAW 264.7 대식세포 모델을 이용하여 조사하였다. 이를 위한 대식세포의 활성화 관련 지표로서 NO, TNF-α, IL-1β 및 IL-10의 생성량 변화를 조사하였다. 비록 유근피 추출물이 처리된 RAW 264.7 대식세포에서 IL-1β의 유의적인 유리는 관찰할 수 없었으나, NO, TNF-α 및 IL-10의 생성은 세포독성을 나타내지 않는 범위에서 유근피 추출물 처리 농도 의존적으로 증가되었으며, 이는 또한 iNOS, TNF-α 및 IL-10의 단백질 발현 증가와 연관되어 있었다. 아울러 유근피 추출물은 LPS에 의한 과도한 NO의 생성 억제능도 함유하고 있었으며, 유근피 추출물에 의한 대식세포의 활성화에는 NF-κB와 PI3K/Akt 및 MAPKs 등과 같은 면역 활성을 유도하는 신호전달계의 활성화가 연관되어 있음을 알 수 있었다. 따라서 본 연구의 결과는 유근피 추출물이 대식세포 활성화를 통한 면역 증강제로서의 개발 가능성이 매우 높음을 시사한다.
상추는 가장 선호하는 녹색 채소 중 하나이다. 상추는 폴리페놀성 화합물을 비롯한 다양한 성분을 함유하고 있으며, 항균, 항산화, 항염증 등의 생리활성을 가지고 있는 것으로 알려져 있다. 본 연구에서는 건조상추의 에탄올 추출물(DLE)을 제조하고 이들의 항염증 활성을 연구하였다. DLE의 항염증 활성을 측정하기 위하여 LPS로 활성화된 마우스 대식세포 RAW 264.7 세포주에서 nitric oxide (NO) 생성을 측정하였다. DLE는 세포주의 생존에는 영향을 미치지 않으면서 NO 생산을 현저하게 저해하였다. DLE에 의해 염증 유전자인 iNOS와 COX-2의 유전자와 단백질의 발현이 모두 감소하였으며, 6개의 염증관련 cytokine 유전자($IL-1{\alpha}$, $IL-1{\beta}$, IL-1F6, $TNF-{\alpha}$, CSF2, 그리고 CXCL10)의 발현이 모두 감소하였다. 또한, DLE의 처리는 MAPKs 경로의 인산화를 모두 저해하였으며, $NF-{\kappa}B$ p65의 핵으로의 이동을 저해하였다. 이러한 결과는 DLE의 항염증 활성은 MAPKs 경로와 $NF-{\kappa}B$ 경로를 조절함으로써 이루어짐을 시사한다. 또한, DLE는 농도의존적으로 reactive oxygen species (ROS)의 생산을 저해하였으며, hemeoxygenase-1 (HO-1) 단백질의 발현을 증가시켰으며, HO-1의 전사조절인자인 Nrf2의 핵으로의 이동을 증가시켰다. 결론적으로, 이러한 연구결과는 DLE가 염증관련 유전자의 발현을 감소시키며, MAPKs, $NF-{\kappa}B$, 그리고 Nrf2/HO-1 등 다양한 경로를 조절함으로써 항염증 활성을 가지는 것을 제시한다.
