Improved modeling of non-hepatic cellular uptake and degradation of low density lipoprotein

  • 인간 fibroblasts의 receptor를 통한 LDL의 섭취와 분해에 대하여 보다 개선되어진 수학적/동역학적 모델을 제시하였다. 관련된 동역학적 모델의 hierarchy를 통하여 세포 멤브레인 표면으로 recycle되는 receptor의 선택적 insertion 정도를 나타내는 파라미터, ${\alpha}$를 가지는 모델을 제안하였다. 여러 가지의 LDL 농도의 미디움과 여러 가지의 실험조건에서 모델예측을 수행하였는데, Brown과 Goldstein의 많은 실험데이터에 잘 일치하였다.

The Src/PLC/PKC/MEK/ERK Signaling Pathway Is Involved in Aortic Smooth Muscle Cell Proliferation induced by Glycated LDL

  • Low density lipoproteins (LDL) play important roles in the pathogenesis of atherosclerosis. Diabetes is associated with accelerated atherosclerosis leading to cardiovascular disease in diabetic patients. Although LDL stimulates the proliferation of arterial smooth muscle cells (SMC), the mechanisms are not fully understood. We examined the effects of native LDL and glycated LDL on the extracellular signal-regulated kinase (ERK) pathway. Addition of native and glycated LDL to rat aorta SMCs (RASMCs) stimulated ERK phosphorylation. ERK phosphorylation was not affected by exposure to the $Ca^{2+}$ chelator BAPTA-AM but inhibition of protein kinase C (PKC) with GF109203X, inhibition of Src kinase with PP1 ($5{\mu}M$) and inhibition of phospholipase C (PLC) with U73122/U73343 ($5{\mu}M$) all reduced ERK phosphorylation in response to glycated LDL. In addition, pretreatment of the RASMCs with a cell-permeable mitogen-activated protein kinase kinase (MEK) inhibitor (PD98059, $5{\mu}M$) markedly decreased ERK phosphorylation in response to native and glycated LDL. These findings indicate that ERK phosphorylation in response to glycated LDL involves the activation of PKC, PLC, and MEK, but is independent of intracellular $Ca^{2+}$.

갈근황련황금탕 부탄올 추출물의 혈중에서의 지질 개선효과 (Blood lipid lowering effect of butanol extract from Galkun-Whanglyeon-Whanggum-Tang)

  • The Galkun-Whanglyeon-Whanggum-Tang, an officially standardized mixture of traditional herbal medicines used in Korea and China, consists of Pueraria lobata, Scullellaria baicalensis, Coptis chinensis and Glycyrrhiza uralensis at a ratio of 6:9:3:2.4. In this study, we evaluated the effect of lowering lipid accumulation in blood by treatment of Galkun-Whanglyeon-Whanggum-Tang in Apo E(-/-) atherosclerotic animal model. ApoE/mice fed with 1.25% cholesterol, 7.5% cocoa butter and 0.5% sodium cholate diet were orally given vehicle or Galkun-Whanglyeon-Whanggum-Tang(10, 100 and 300 mg/kg/day) for 12 weeks. Serum levels of triglyceride(TG), total cholestrerol(TC), low-density lipoprotein(LDL) and high-density lipoprotein(HDL) were analyzed, and PPAR-${\alpha}$ and PPAR-${\gamma}$ were examined by Western blotting analysis. Galkun-Whanglyeon- Whanggum-Tang decreased serum levels of TG, TC and LDL, but not HDL in ApoE/mice. In parallel, Galkun-Whanglyeon-Whanggum-Tang treatment showed the increased activity of PPAR-${\alpha}$ and PPAR-${\gamma}$ in hepatocytes. In summary, Galkun-Whanglyeon-Whanggum-Tang can reduce lipid accumulation in blood, and this effect might be accompanied by the upregulation of PPAR-${\alpha}$ and PPAR-${\gamma}$ in Apo E(-/-) atherosclerotic mouse model.

낙상예방 운동프로그램이 노인여성의 균형, 낙상효능감과 혈압 및 혈중지질에 미치는 영향 (The Effect of Exercise Program for Falls Prevention on Balance, Falls Efficacy, Blood Pressure and Blood Lipids in the Elderly Women)

  • Purpose: The purpose of this study was to investigate the effects of falls prevention exercise program to balance, falls efficacy, blood pressure and blood lipids in the elderly females. Methods: A total of twenty-six elderly women participated in this study. All subjects participated in exercise program based on pelvic stabilization and balance training on two times a week for twelve weeks. They were measured about Berg balance scale (BBS), Time up and go (TUG), Functional reach test (FRT), Sit to stand (STS) for balance, falls efficacy scale-international (FES-I) for falls efficacy, systolic blood pressure (SBp), diastolic pressure (DSp) for blood pressure, and total cholesterol (TC), triglyseride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C) for blood lipids. Results: There were significant improvements of BBS (p<.05), TUG (p<.001) FRT (p<.01), and STS (p<.001) in the balance after exercise program. There was significant improvements in FES-I (p<.001) in the falls efficacy, There were no statistical differences of SBp and DSp in the blood pressure. There were no statistical differences of TC, TG, HDL-C, and LDL-C in the blood lipids. Conclusions: Falls prevention exercise program was meaningful increasing balance ability and falls efficacy.

유도비만 흰쥐에서 발효 서목태가 미치는 생리활성 (Bioactivities of Fermented Rhynchosia nulubilis in Dietary Obese Rats)

  • This study was to examine the lipid metabolism of fermented Rhynchosia nulubilis (FRN) in obese rats. The abnormal content of blood lipids often results in metabolic diseases, such as obesity and hyperlipidemia. Seven weeks female Sprague-Dawley rats were divided into four groups and fed high fat diets for 44 days. Also FRN was administered orally for 44 days at 7.5 ml/kg of body weight of rats. The effects of the lipid metabolism were evaluated by total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL), low density lipoprotein (LDL), glutamic pyruvate transaminase (GPT) and glutamic oxalacetic transaminase (GOT) levels in sera. The levels of TC, TG, LDL and GPT in FRN-treated groups were lower than those in obese groups. While HDL levels were significantly increased. These results demonstrated that FRN had improving effects of lipid metabolism in the obese rats, suggesting that FRN would be used as an ingredient of the useful functional products.

Antihyperlipidemic Activity of the Ethyl-acetate Fraction of Stereospermum Suaveolens in Streptozotocin-induced Diabetic Rats

  • Objectives: Dyslipidemia in diabetes mellitus is a significant risk factor for the development of cardiovascular complications. The aim of this study was to evaluate the effect of the ethyl-acetate fraction of an ethanolic extract from Streospermum suaveolens on lipid metabolism in streptozotocin (STZ)-induced diabetic rats. Methods: Diabetes was induced by intraperitonial injection of STZ (50 mg/kg). Diabetic rats were treated with an ethyl-acetate fraction orally at doses of 200 and 400 mg/kg daily for 14 days. On the $15^{th}$ day, serum lipid profiles, such as total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), and high-density lipoprotein (HDL), were estimated in experimental rats. The atherogenic (AI) and the coronary risk (CRI) indices were also evaluated. Results: The ethyl-acetate fraction at doses of 200 and 400 mg/kg significantly (P < 0.001) and dose-dependently reduced serum cholesterol, triglycerides and LDL, but increased HDL towards near normal levels as compared to diabetic control rats. The fraction also significantly (P < 0.001) lowered the atherogenic index (AI) and coronary risk index (CAI) in a dose-dependent manner. Conclusion: The present study demonstrated that the ethyl-acetate fraction of Stereospermum suaveolens exhibits a potent antihyperlipidemic activity in hyperglycemic rats and suggests that the plant may have therapeutic value in treating the diabetic complication of hyperlipidemia.

노인의 규칙적 신체활동이 심혈관 기능에 미치는 영향 고찰 (Relationship between Physical activity and Cardiovascular Outcomes in the Korean Elderly: Review of Experimental Studies)

  • Purpose: The purpose of this study was to review the relationship between physical activity and cardiovascular outcomes in the Korean elderly. Methods: Experimental studies were located using PubMed, CINAHL, PsycINFO, Cochrane, RISS, KISS, and KoreaMed. The selected studies for analysis were 20 articles of cardiovascular outcomes (total cholesterol, triglyceride, high density lipoprotein (HDL), low density lipoprotein (LDL), blood pressure, and pulse rate) from 515 articles. Results: One-group pretest-posttest design was the most common. The main physical activity was an aerobic exercise. Five of 14 studies reported a significant improvement of total cholesterol. Four of 14 studies found relationship between exercise and triglyceride. Eight of 12 studies reported a significant improvement of HDL, whereas 3 of 9 studies reported a significant improvement of LDL. In over 60% of selected studies, reported significant improvement of blood pressure. Conclusion: Based on the review, it suggests that regular physical activity of the elderly may improve cardiovascular outcomes.

여대생의 체지방률분류 비만도에 따른 체성분, 혈압, 혈중지질 및 혈당 (Body Composition, Blood Pressure, Blood Lipids, and Glucose according to Obesity Degree by Body Fat Percentage in Female University Students)

  • Purpose: The purpose of this study was to investigate body composition, blood pressure, blood lipids, and glucose according to obesity degree by body fat percentage in female university students. Methods: A total of 231 subjects were participated between July and December 2010. The anthropometric measures and blood analyzer were used. Data were analyzed using descriptive statistics, ANOVA, and Pearson's correlation coefficient with SPSS 18.0. Results: Percentage of body fat (PBF)-defined obesity had higher prevalence than body mass index (BMI)-defined obesity. Weight, BMI, PBF, fat mass, Waist to hip ratio (WHR), Systolic blood pressure (SBP), Diastolic blood pressure (DBP) in obesity group were higher than normal group and Soft lean mass and Low density lipoprotein (LDL) in obesity group were lower than normal group. There were significant correlations between PFB, SBP, DBP, Triglyceride (TC), High density lipoprotein (HDL), and LDL. Conclusion: In conclusion, when screening for obesity in female university students, body fat should be considered. It is needed to develop obesity management program according to obesity degree and to identify the effectiveness.

청년층 성인남성에서 흡연, 음주, 비만지표가 심뇌혈관계 위험인자에 미치는 영향 (Influencing Factors on Cardio-cerebrovascular Disease Risk Factors in Young Men: Focusing on Obesity Indices)

  • Purpose: The purpose of this study was to investigate influences of drinking, smoking, and obesity indices on cardio-cerebrovascular disease risk factors in Korean young men. Methods: The subjects were 234 young men, aged 20 to 39 years. Body mass index (BMI), percent body fat (PBF), body fat mass (kg), and waist hip ratio (WHR) were measured as obesity indices. Systolic blood pressure (SBP), diastolic blood pressure (DBP), fasting blood sugar (FBS), total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) were measured as cardio-cerebrovascular disease risk factors. The data were analyzed by t-test, ANOVA, Pearson's correlation coefficient, and multiple regression analysis using SAS 9.4. Results: Statistically significant factors were BMI for SEP; age, degree of drinking, body fat mass for DBP; WHR for TC; WHR and age for TG; age, degree of smoking and PBF for HDL-C; WHR, age and degree of smoking for LDL-C. Conclusion: Factors identified in this study need to be considered in cardio-cerebrovascular disease prevention programs for young men.

집단체조프로그램이 여성노인의 신체조성, 혈청지질에 미치는 효과 (The Effects of Group Exercise on Fat Distribution, Lipid Profiles in Elderly Women)

  • Purpose: This study was aimed to identify the effects of group exercise on physiological functions in Elderly. Method: The design of research was one group pretest-posttest design. The subjects were sixty two elderly women persons over sixty years old to live in C-GUN, The exercise period was 12-weeks and frequency was three times per week. the exercise duration was sixty minites. In order to evaluate the effect of exercise, we measured %body fat, fat mass(FM), fat free mass(FFM), body mass index(BMI), body metabolic rate(BMR), Total Cholesterol(TC), Triglyceride(TG), high density lipoprotein-c(HDL-C), low density lipoprotein-c(LDL-C) before and after the 12 week's group exercise program. The data analyzed by the paired t-test using SPSS package. Result: There was significant difference in FFM(p=.000), BMR(p=.000), HDL-C(p=.010). There was no significant difference in weight(p=.122), %body fat(p=.062),, FM(p=.704), BMI(p=.136), TC(p=.125), TG(p=.064), LDL-C(p=.575) Conclusion: This group exercise for elderly women can be recommended as an effective nursing intervention for elderly women in a community.
