Comparison of Biological Activities of Acer mono and A. okamotoanum Extracts by Water Extracton and Low Temperature High Pressure Extraction (고로쇠와 우산고로쇠 저온고압 수용성 추출물의 유용생리활성 비교)

  • Hepatoprotective and antioxidant activities of Acer mono and A. okamotoanum were compared after being extracted by low temperature and high pressure (LTHP) at 20 MPa and $60^{\circ}C$ for 15 minutes. Extraction yield of both A. mono and A. okamotoanum was increased about 40~43% by this extraction process. On scavenging activities, the bark of A. okamotoanum from this extraction process showed the highest activity as about 97%. This value was higher than that from conventional water extraction and A. mono extracts. Both of A. mono and A. okamotoanum showed high ability on nitrite scavenging, but decreasing tendency according to increasing of pH. On SOD-like test, A. okamotoanum had the highest activity as 46.28% at $1.0\;mg/m{\ell}$ concentration. A. okamotoanum extracted by LTHP also showed the highest activity as 197.38% in adding $1.0\;mg/m{\ell}$ concentration. Generally, the extracts from low temperature and high pressure extraction process are higher hepatoprotective and antioxidant activities than that from conventional water extraction. It can conclude that the bark of A. okamotoanum has better biological activities than other parts of A. mono.

Enhancement of Skin Immune Activities of Spirulina maxima by High Pressure Extraction Process (Spirulina maxima 초고압 추출물의 피부 면역 활성 증진)

  • A marine alga, Spirulina maxima, was extracted under high pressure and low temperature conditions at 500 MPa and $60^{\circ}C$ for 5 and 10 min. A high pressure of 500 MPa was applied to improve process yields because of low temperature extraction. This method resulted in highest higher extraction yield of 26.1% (w/w) in comparison to those results obtained from conventional extraction methods which produced a yield of 17.6% (w/w) from water. The extracts from this process also showed 19% of low cytotoxicity against human normal fibroblast cells in adding 1.0 mg/ml of the highest concentration. The crude extract significantly reduced the production of Prostaglandin $E_2$ ($PGE_2$) from CCD-986sk cells and increased nitric oxide production by macrophages. These higher activities of enhancing skin immune functions were found to have high antioxidant extract properties, like a 98% increase in DPPH radical scavenging activity. The extracts from the high pressure process showed a higher elution of active components than other processes and generated new compounds based on HPLC analysis. This clearly indicates that the extracts from high pressure and low temperature conditions have higher skin immune activation properties that have not been previously reported.

  • This study was performed to investigate a methodology of improving immune activities of Berberis koreana Palibin by ultra high pressure on low temperature extraction process. Extraction yield was enhanced up to more than 35% by above process, compare to the control and extraction average. The cytotoxicity on human kidney cell (HEK293) was showed below 20.4%, 21.6% in adding 1.0 mg/ml of the highest concentration. Generally, the extracts by ultra high pressure extraction process showed less toxicity about 5% than the other processes. It could tell that toxic materials that existing in the plant could be reduced or broken by ultra high pressure process due to can be broken bond such the hydrogen bond, the electrostatic bond, the van der Waals bond, and the hydrophobic bond, can be broken by high pressure. The result could be employed to develop a new type of functional food from B. koreana Palibin by low temperature high pressure process.

Enhancement of the Antioxidant and Anticancer Activities of Berberis koreana Bark by Using a Low Temperature and High-Pressure Extraction Process (저온고압 추출공정을 이용한 매자나무 수피의 항산화 및 항암활성 증진)

  • This study was conducted to characterize the enhanced antioxidant and anticancer activities of Berberis koreana bark following a low temperature and high pressure extraction process. The results indicate that the B. koreana bark extracted as described showed a 93% increase in DPPH radical scavenging activity. Inhibition activity of xanthine oxidase was highest by this extraction process. In addition the growth of human lung cancer cells (A549), human stomach cancer cells (AGS), human breast cancer cells (MCF-7) and human liver cancer cells (Hep3B) were inhibited by 70.8%, 86.2%, 84.3% and 62.5% respectively. These data indicate that this low temperature and high pressure extraction technique results in the efficient extraction of bioactive compounds from rigid plant materials. This process could also be combined with other techniques to improve extraction yields and identify new biologically active substances from relatively hard plants.

The Comparison of Extraction Process for Enhancement of Immunomodulating Activities of Ulva pertusa kjellman (구멍갈파래의 면역활성 증진을 위한 추출방법 비교)

  • The purpose of this study was to investigate the immunomodulatory effect of Ulva pertusa kjellman extract after undergoing a low temperature and high-pressure extraction process. First, the extracts obtained under the extraction conditions of 150 MPa and $80^{\circ}C$showed a relatively high antioxidant activity, with 90% super oxide radical activity compared to the extracts from conventional extraction process with water at $100^{\circ}C$. This extract also improved the growth of both human immune B and T cells up to $14.5{\times}10^4$ cells/mL and $14.2{\times}10^4$ cells/mL compared to $9.1{\times}10^4$ cells/mL in adding the extracts from conventional processes. It was found that the extracts obtained at 100 MPa and $60^{\circ}C$ showed better activities in NK cell growth and NO production from macrophage as $11.8{\times}10^2$ cells/mL and 30.0 ${\mu}M$. Overall, the extracts from high pressure and low temperature extraction process had relatively higher immune activation activity, possibly because the low temperature and high pressure extraction process may have higher yields of active compounds and have less damage to useful ingredients from relatively weak marine natural resources, such as Ulva pertusa kjellmann than that from the conventional extraction system.

Development of Microwave Extraction Method for the Active Ingredients and Functional Constituents of Paeonia Root (마이크로웨이브 추출방법을 이용한 작약의 유효성분 추출 및 생리활성 측정)

  • The heat-mediated reflux apparatus extraction of Paeonia lactiflora Pall. has been widely used as a traditional extraction method. In this paper, the microwave apparatus extraction method of Paeonia Radix was performed and the active ingredients and functional constituents were compared with the reference extraction method. The most effective extraction condition of albiflorin using the microwave was 120℃ in 50% methanol, and paeoniflorin was maximally extracted at 60℃. The reduced level of paeoniflorin molecule at high-pressure and high-temperature extraction condition was caused by the molecular instability. Additionally, the microwave extraction of 50% methanol extracts at 150℃ showed the highest functional constituents determined by in vitro DPPH radical scavenging activity, polyphenol concentration, and tyrosinase inhibition assay. The microwave apparatus was adapted as a rapid, low-cost, and environmentally friendly method to extract active ingredients and the practical extraction conditions of Paeonia Radix can be used in industrial applications.

A Study on the Relief of Shell Wall Thinning of Low Pressure Type Feedwater Heater Around the Extraction Nozzle Identified (저압형 급수가열기 추기노즐에서 동체 감육 완화에 관한 연구)

  • The current machinery and tools of secondary channel of the nuclear power plants were produced in the carbon-steel and low-alloy steel. What produced with the carbon-steel occurs wall thinning effect from flow accelerated corrosion by the fluid flow at high temperature, high pressure. Several nuclear power plants in Korea have experienced wall thinning damage in the area around the impingement baffle-installed. Wall thinning by flow accelerated corrosion occurs piping system, the heat exchanger, steam condenser and feedwater heaters etc,. Feedwater heaters of many nuclear power plants have recently experienced sever wall thinning damage, which will increase as operating time progress. This study describes the comparisons between the numerical results using the FLUENT code and experimental data of down scale model.

High rate deposition of poly-si thin films using new magnetron sputtering source

  • After LeComber et al. reported the first amorphous hydrogenated silicon (a-Si: H) TFT, many laboratories started the development of an active matrix LCDs using a-Si:H TFTs formed on glass substrate. With increasing the display area and pixel density of TFT-LCD, however, high mobility TFTs are required for pixel driver of TF-LCD in order to shorten the charging time of the pixel electrodes. The most important of these drawbacks is a-Si's electron mobiliy, which is the speed at which electrons can move through each transistor. The problem of low carier mobility for the a-Si:H TFTs can be overcome by introducing polycrystalline silicon (poly-Si) thin film instead of a-Si:H as a semiconductor layer of TFTs. Therefore, poly-Si has gained increasing interest and has been investigated by many researchers. Recnetly, fabrication of such poly-Si TFT-LCD panels with VGA pixel size and monolithic drivers has been reported, . Especially, fabricating poly-Si TFTs at a temperature mach lower than the strain point of glass is needed in order to have high mobility TFTs on large-size glass substrate, and the monolithic drivers will reduce the cost of TFT-LCDs. The conventional methods to fabricate poly-Si films are low pressure chemical vapor deposition (LPCVD0 as well as solid phase crystallization (SPC), pulsed rapid thermal annealing(PRTA), and eximer laser annealing (ELA). However, these methods have some disadvantages such as high deposition temperature over $600^{\circ}C$, small grain size (<50nm), poor crystallinity, and high grain boundary states. Therefore the low temperature and large area processes using a cheap glass substrate are impossible because of high temperature process. In this study, therefore, we have deposited poly-Si thin films on si(100) and glass substrates at growth temperature of below 40$0^{\circ}C$ using newly developed high rate magnetron sputtering method. To improve the sputtering yield and the growth rate, a high power (10~30 W/cm2) sputtering source with unbalanced magnetron and Si ion extraction grid was designed and constructed based on the results of computer simulation. The maximum deposition rate could be reached to be 0.35$\mu$m/min due to a high ion bombardment. This is 5 times higher than that of conventional sputtering method, and the sputtering yield was also increased up to 80%. The best film was obtained on Si(100) using Si ion extraction grid under 9.0$\times$10-3Torr of working pressure and 11 W/cm2 of the target power density. The electron mobility of the poly-si film grown on Si(100) at 40$0^{\circ}C$ with ion extraction grid shows 96 cm2/V sec. During sputtering, moreover, the characteristics of si source were also analyzed with in situ Langmuir probe method and optical emission spectroscopy.

  • The most components and piping of the secondary side of domestic nuclear power plants were manufactured carbon-steel and low-alloy steel. Flow accelerated corrosion leads to wall thinning (metal loss) of carbon steel components and piping exposed to the flowing water or wet steam of high temperature, pressure, and velocity. The feedwater heaters of many nuclear power plants have recently experienced sever wall thinning damage, which increases as operating time progress. Several nuclear power plants in Korea have also experienced wall thinning damage in the shell wall around the impingement baffle. This paper describes the comparisons between the numerical analysis results using the FLUENT code and the experimental results based on down-scaled experimental facility. The experiments were performed based on several types of impingement baffle plates which are installed in low pressure feedwater heater.

Antioxidative and antimicrobial activities of Oenothera biennis extracted by different methods (추출방법을 달리한 달맞이꽃 추출물의 항산화 및 항균 활성)

  • A effect of extraction methods, including stirrer extraction method (SE), ultrasonification extraction method (USE), reflux extraction method (RE), autoclave extraction (AE) and low temperature high pressure extraction (LE) method on the antioxidative and antimicrobial activities of ethanol extracts of Oenothera biennis was investigated. The extraction yield (46.33%), total polyphenol (463.05 mg GAE/g) and flavonoid (71.71 mg RHE/g) content of Oenothera biennis extract obtained by RE were higher than those from other extraction methods. The antimicrobial activity of Oenothera biennis extract was only observed against Bacillus cereus among other tested organisms (Bacillus cereus, Staphylococcus aureus, Escherichia coli and Salmonella Typhimurium). Oenothera biennis obtained by RE showed the best DPPH radical scavenging ability (74.40%), ABTS radical scavenging ability (65.29%), reducing power (1.370 ($OD_{700}$)) and ferrous ion chelating ability (90.14%) compared with other tested extraction methods tested. The RE method was the most efficient method for extracting crude antioxidant and antimicrobial substances from Oenothera biennis. These results suggested that Oenothera biennis obtained by RE could be used as a bioactive and functional material in the food industry.
