Recent Update of Advanced Imaging for Diagnosis of Cardiac Sarcoidosis: Based on the Findings of Cardiac Magnetic Resonance Imaging and Positron Emission Tomography

  • Sarcoidosis is a multisystem disease characterized by noncaseating granulomas. Cardiac involvement is known to have poor prognosis because it can manifest as a serious condition such as the conduction abnormality, heart failure, ventricular arrhythmia, or sudden cardiac death. Although early diagnosis and early treatment is critical to improve patient prognosis, the diagnosis of CS is challenging in most cases. Diagnosis usually relies on endomyocardial biopsy (EMB), but its diagnostic yield is low due to the incidence of patchy myocardial involvement. Guidelines for the diagnosis of CS recommend a combination of clinical, electrocardiographic, and imaging findings from various modalities, if EMB cannot confirm the diagnosis. Especially, the role of advanced imaging such as cardiac magnetic resonance (CMR) imaging and positron emission tomography (PET), has shown to be important not only for the diagnosis, but also for monitoring treatment response and prognostication. CMR can evaluate cardiac function and fibrotic scar with good specificity. Late gadolinium enhancement (LGE) in CMR shows a distinctive enhancement pattern for each disease, which may be useful for differential diagnosis of CS from other similar diseases. Effectively, T1 or T2 mapping techniques can be also used for early recognition of CS. In the meantime, PET can detect and quantify metabolic activity and can be used to monitor treatment response. Recently, the use of a hybrid CMR-PET has introduced to allow identify patients with active CS with excellent co-localization and better diagnostic accuracy than CMR or PET alone. However, CS may show various findings with a wide spectrum, therefore, radiologists should consider the possible differential diagnosis of CS including myocarditis, dilated cardiomyopathy (DCM), hypertrophic cardiomyopathy, amyloidosis, and arrhythmogenic right ventricular cardiomyopathy. Radiologists should recognize the differences in various diseases that show the characteristics of mimicking CS, and try to get an accurate diagnosis of CS.

Seasonal Characteristics of the Longitudinal Wavenumber-4 Structure in the Equatorial Ionospheric Anomaly

  • Using the global total electron contents (TEC) measured by the TOPEX satellite from Aug. 1992 to Oct. 2005, we investigate the variations of the longitudinal wavenumber-4 (LW-4) structure in the equatorial anomaly (EA) crests with season, local time, and solar activity. Our study shows that the LW-4 structure in the EA crests ($5{\sim}20^{\circ}$ MLAT in both hemispheres) has clear four peaks at fixed longitude sectors during the daytime for both equinoxes and June solstice. In spite of being called a wavelike structure, however, the magnitudes and spatial intervals of the four peaks are far from being the same or regular. After sunset, the four-peak structure begins to move eastward with gradual weakening in its amplitude during equinoxes and this weakening proceeds much faster during June solstice. Interestingly, the longitudinal variations during December solstice do not show clear four-peak structure. All these features of the LW-4 structure are almost the same for both low and high solar activity conditions although the ion densities are greatly enhanced from low to high solar activities. With the irrelevancy of the magnetic activity in the LW-4, this implies that the large changes of the upper atmospheric ion densities, one of the important factors for ion-neutral interactions, have little effect on the formation of the LW-4 structure. On the other hand, we found that the monthly variation of the LW-4 is remarkably similar to that of the zonal component of wavenumber-3 diurnal tides at low latitudes, which implies that the lower atmospheric tidal forcing, transferred to the upper atmosphere, seems to have a dominant role in producing the LW-4 structure in the EA crests via the E-region dynamo.

Effects of Electromagnetic Radiation Exposure on Stress-Related Behaviors and Stress Hormones in Male Wistar Rats

  • Studies have demonstrated that electromagnetic waves, as the one of the most important physical factors, may alter cognitive and non-cognitive behaviors, depending on the frequency and energy. Moreover, non-ionizing radiation of low energy waves e.g. very low frequency waves could alter this phenomenon via alterations in neurotransmitters and neurohormones. In this study, short, medium, and long-term exposure to the extremely low frequency electromagnetic field (ELF-EMF) (1 and 5 Hz radiation) on behavioral, hormonal, and metabolic changes in male Wistar rats (250 g) were studied. In addition, changes in plasma concentrations for two main stress hormones, noradrenaline and adrenocorticotropic hormone (ACTH) were evaluated. ELF-EMF exposure did not alter body weight, and food and water intake. Plasma glucose level was increased and decreased in the groups which exposed to the 5 and 1Hz wave, respectively. Plasma ACTH concentration increased in both using frequencies, whereas noradrenaline concentration showed overall reduction. At last, numbers of rearing, sniffing, locomotor activity was increased in group receiving 5 Hz wave over the time. In conclusions, these data showed that the effects of 1 and 5 Hz on the hormonal, metabolic and stress-like behaviors may be different. Moreover, the influence of waves on stress system is depending on time of exposure.

DL-2 하이드록시 3-메틸 부틸산물 배위자로 한 새로운 항암성 백금(II) 착체의 위암세포와 정상신장세포에 대한 선택적 세포독성 (Selective Cytotoxicity of Novel Platinum(II) Coordination Complexes Containing DL-2-Hydroxy 3-Methylbutyric Acid)

  • A new series of highly water soluble platinum(II) complexes[Pt(II)(DL-2-hydroxy-3-methylbutyrate)(trans-l-1,2-dimninocyc1ohexane)] (PC-1) and [Pt(II)DL-2-hydroxy-3-methylbutyrate](cis-1,2-diaminocyclohexane)](PC-2) were synthesized and characterized by their elemental analysis and by various spectroscopic techniques [infrared(IR), $^{13}C$-nuclear magnetic resonance (NMR)]. In vitro antitumor activity of new Pt(II)complexes was tested against MKN-45, MKN/ADM and MKN/CDDP human gastric adenocarcinoma cell lines using colorimetric MTT[3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyltetrazoliumbromide] assay for cell survival and proliferation. PC-1 and PC-2 showed active against MKN-45/P, MKN/ADM and MKN/CDDP human gastric cancer cell lines, and the antitumor activity of these compounds were comparable or superior to that of cisplatin. The nephrotoxicities of PC-1 and PC-2 were found quite less then that of cisplatin using MTT and [$^3H$] thymidine uptake tests in rabbit proximal tubule cells, human kidney cortical cells human renal cortical tissues. Based on these results, these novel platinum(II) complex compounds(PC-1 & PC-2) represent a valuable lead in the development of the new anticancer chemotherapeutic agents capable of improving antitumor activity and low nephrotoxicity.

PC-1 showed active against L-1210, P-388 leukemia, human lung, stomach, prostate, bladder and breast cancer cell lines, and the antitumor activity of these compounds were comparable or superior to those of PC-2 and displatin. The nephrotoxicities of PC-1 and PC-2 were found quite less than that of cisplatin using MTT and [3H] thymidine uptake in rabbit proximal tubule cells and human kidney cortical cells. On the Variability of the Ionospheric F2-Layer During the Quietest Days in December 2009

  • December 2009 was one of the quietest (monthly Ap=2) months over the last eight decades. It provided an excellent opportunity to study the day-to-day variability of the F2 layer with the smallest contribution due to geomagnetic activity. With this aim, we analyze hourly values of the F2-layer critical frequency (foF2) recorded at 18 ionosonde stations during the magnetically quietest (Ap=0) days of the month. The foF2 variability is quantified as the relative standard deviation of foF2 about the mean of all the "zero-Ap" days of December 2009. This case study may contribute to a more clear vision of the F2-layer variability caused by sources not linked to geomagnetic activity. In accord with previous studies, we find that there is considerable "zero-Ap" variability of foF2 all over the world. At most locations, foF2 variability is presumably affected by the passage of the solar terminator. The patterns of foF2 variability are different at different stations. Possible causes of the observed diurnal foF2 variability may be related to "meteorological" disturbances transmitted from the lower atmosphere or/and effects of the intrinsic turbulence of the ionosphere-atmosphere system.

코스모스(Cosmos bipinnatus) 꽃으로부터 phenolic 화합물의 분리 동정과 항아토피 효과 (Phenolic compounds from the flowers of Cosmos bipinnatus and their anti-atopic activity)

  • 코스모스 꽃을 MeOH : H2O = 4 : 1 용매로 추출하여, 얻어진 추출물을 EtOAc n-BuOH 및 water로 용매 분획 하였다. 이 중 n-BuOH 분획으로부터 silica gel (SiO2)과 octadecyl silica gel(ODS) column chromatography로 정제하여 2종의 화합물을 분리하였다. NMR, MS 및 IR 등의 스텍트럼 데이터를 통해 화합물의 화학구조를 benzyl O-β-ᴅ-glucopyranoside (1)와 and 2-phenylethyl O-β-ᴅ-glucopyranoside (2)로 동정하였다. 분리한 두 화합물의 자외선(UVB)으로 광노화가 유도된 HaCaT 세포주를 이용한 항아토피 효능을 평가하기 위해 실험을 진행하였으며, 1-1,000 ㎍/mL의 농도범위에서 2종의 화합물에서 모두 독성을 나타내지 않았다. TARC생성억제활성을 통한 항아토피 활성결과에서는 2종의 화합물에서 모두 농도의존적인 TARC 억제활성이 나타났지만, 특히 화합물 1에서 낮은 농도범위인 10 ㎍/mL에서도 유의적인 활성이 나타났으며, 다른 농도범위에서도 타 화합물에 비해 억제활성이 높은 것으로 나타나, 항아토피 활성이 가장 우수한 것으로 확인되었다. 이 결과를 통해 기능성 화장품 원료로 활용이 가능할 것으로 사료된다.

  • Electric activity of cardiac muscles generates magnetic fields. Magnetocardiography (or MCG) technology, measuring these magnetic signals, can provide useful information for the diagnosis of heart diseases. It is already about 40 years ago that the first measurement of MCG signals was done by D. Cohen using SQUID (superconducting quantum interference device) sensor inside a magnetically shielded room. In the early period of MCG history, bulky point-contact RF-SQUID was used as the magnetic sensor. Thanks to the development of Nb-based Josephson junction technology in mid 1980s and new design of tightly-coupled DC-SQUID, low-noise SQUID sensors could be developed in late 1980s. In around 1990, several groups developed multi-channel MCG systems and started clinical study. However, it is quite recent years that the true usefulness of MCG was verified in clinical practice, for example, in the diagnosis of coronary artery disease. For the practical MCG system, technical elements of MCG system should be optimized in terms of performance, fabrication cost and operation cost. In this review, development history, technical issue, and future development direction of MCG technology are described.

다양한 노출 매트릭스를 통한 송전선로 주변과 비 주변 거주 초등학교 학생의 극저주파 자기장 노출량 평가에 관한 연구 (Exposure Assessment of Extremely Low Frequency Magnetic Fields by variable exposure matrices for the Selected Primary Schoolchildren Living Nearby and Away from a Overhead Transmission Power Line)

  • The objectives of this study were to analyze and compare 24 hrs personal exposure levels of MF at microenvironments such as home, school, educational institute, internet pc game room, transportation, and other places according to time activity patterns using various metrics for children attending the primary schools located near and away from the power lines, and to characterize the major microenvironments and impact factors attributed personal exposure level. The study was carried out for 44 children attending a primary school away from the lines(school A) and 125 children attending a school away from 154 kV power lines(school B), all who aged 12 years and were 6 grade, from July 2003 to December 2003. All participants filled in a questionnaire about characteristics, residence, use of electrical appliances and others. Children wore a small satchel in which EMDEX II and Lite (Enertech, Co. Ltd) and a diary of activity list for period of registration in 20 minutes blocks. All statistical calculations were made with the SAS System, Releas 6.12. The summary of results was presented below. First, about the characteristics of subjects, there no differences between two groups. The subject almost spent about 56 % of their time at home and about 20~25 % of their time at school. Fifty percent of children spent 2 hours at private educational institutes. Second, the personal exposure measurements of children in school B was statistically higher than those of children in school A by various metrics such as arithmetic mean, geometric mean, percentile(5, 25, 50, 75, 95), maximum, rate of change metric, constant field metric. The arithmetic and geometric mean magnetic fields during the time the children were at school B were 0.98 and $0.86{\mu}T$ and were about 23 times higher than those of children were at school A. In conclusion, the significant major determinants of personal exposure level is the distance from the power line to microenvironments.

Validation of protein refolding via 1-dimensional 1H-15N heteronuclear single quantum correlation experiments

  • Many proteins are expressed as an insoluble form during the production using Escherichia coli (E. coli) system. Although various methods are applied to increase their amounts of soluble expression, refolding is the only feasible way to obtain a target protein in some cases. Moreover, protein NMR experiments require 13C/15N-labeled proteins that can only be obtained from E. coli systems in terms of cost and technical difficulty. The finding of appropriate refolding conditions for a target protein is a time-consuming process. In particular, it is very difficult to determine whether the refolded protein has a native structure, when a target protein has no enzymatic activity and its refolding yield is very low. Here, we showed that 1-dimensional 1H-15N heteronuclear single quantum correlation (1D 1H-15N HSQC) experiment can be efficiently used to screen an optimal condition for the refolding of a target protein by monitoring both the structure and concentration of the refolded protein.
