The methanolic (MeOH) extract of A. fruticosa bark, which showed immune-regulatory activities, was separated to purify an active compared by means of a multi-stage column chromatography. This resulted in the isolation and characterization of an isoflavone glycoside named 4', $6-Dimethoxyisoflavone-7-O-{\beta}-D-glucopyranoside$. Immuno-regulatory activities of the crude extract of Amorpha fruticosa LINNE bark were compared with that of an isoflavone glycoside (4', $6-dimethoxyisoflavone-7-O-{\beta}-D-glucopyranoside$). The crude methanolic extract of A. fruticosa and purified single compound showed 16% of relatively low cytotoxicity at a maximum concentration of 1.0 g/L in cultivated normal human lung cell line (HEL299). Cell growth of human T cells was increased up to 15%, 0.5 g/L of the crude extract added group. This was higher than a single compound added one. On the other hand, specific production rates of IL-6 and $TNF-{\alpha}$ from T cell were higher in the purified compound treat group ($0.82{\times}10^{-4}\;pg/cell$ and $1.08{\times}10^{-4}\;pg/cell$, respectively), compared to 0.5 g/L of the crude extract added group ($0.65{\times}10^{-4}\;pg/cell$ and $0.84{\times}10^{-4}\;pg/cell$, respectively). In addition, the growth of NK-92MI cells incubated with the crude extract was higher up to 56% over the cells grown with a single compound (0.5 g/L). In overall, the crude extract showed relatively higher immuno-regulatory activities compared with a single compound, probably due to the synergic effect given by other substances existed in the crude extract. Even though the siolated compound stimulated higher secretion of cytokines from human T cells.
It has been proposed that oxidative modification of LDL (oxLDL) plays a significant role in the pathogenicity of atherogenesis. We tested the hypothesis that chitin and chitosan may function as antioxidants with respect to 0.1 mg cholesterol/ml LDL incubated with 5 $\mu$ M Cu$^2$$^{+}$alone or in the P338Dl mouse macrophage system using L-ascorbic acid as a standard classical antioxidant. The degree of oxLDL formation was ascertained by the relative electrophoretic mobility (rEM) in the combination of thiobarbituric acid reactive substances (TBARS) levels, and the cytotoxicity of oxLDL was detected by macrophage viability. The oxLDL uptake and foam cell formation of macrophages were measured by Oil Red O staining. Incubation with Cu$^2$$^{+}$and macrophages increased rEM of LDL and stimulated TBARS formation. Culture of macrophages with LDL in the presence 5 $\mu$ M Cu$^2$$^{+}$induced macrophage death. In cell-free system 200 $\mu$g/ml water-soluble chitosan and chitosan-oligosaccharide blocked oxLDL formation. Water-soluble chitosan and chitosan-oligosaccharide blocked oxLDL formation near-completely relative to L-ascorbic acid, whereas water-soluble chitin and chitin-oligosaccharide had no measurable antioxidant effect. In macrophage system water-soluble chitosan and chitosan-oligosaccharide blocked oxidation of LDL with a significant increase in cell viability, and decreased TBARS in medium. As for the inhibitory effect on macrophage foam cell formation, chitosan and its oligosaccharide, but not watersoluble chitin, revealed the effectiveness. The endothelial expression of lectin-like oxLDL receptor-1 (LOX-1) was tested by Western blot analysis, and chitosan, chitosan-oligosaccharide and chitin-oligosaccharide blocked LOX-1 expression. These results indicate that water-soluble chitosan and its oligosaccharide showed the inhibitory effect on Cu$^2$$^{+}$-induced LDL oxidation of macrophages, and chitosan, chitosan-oligosaccharide and chitin-oligosaccharide had blocking effect on oxLDL receptor expression in the human umbilical vein endothelial system. Thus, water-soluble chitosan and its oligosaccharides possess anti-atherogenic potentials possibly through the inhibition of macrophage LDL oxidation or endothelial oxLDL receptor expression depending on chemical types.l types.
마치현 추출물에 대한 생리활성 소재로서 응용 가능성을 확인하고자 하였다. 마치현 추출물의 측정 결과, 플라보노이드, 폴리페놀 함량과 DPPH radical 소거능을 확인하였으며, 세포실험 결과 HaCaT, RAW 264.7, RBL-2H3 세포에서 유의한 세포독성은 나타나지 않았으며. $H_2O_2$에 의한 유발되는 산화적 스트레스에 대한 HaCaT 세포는 마치현 에탄올 $100{\mu}g/mL$ 농도에서 83% 보호효과가 확인되었다. RAW 264.7 세포에 대한 마치현 추출물의 항염 효과를 확인 결과 저농도에서도 nitric oxide 생성이 억제되었으며, S. aureus, S. epidermidis, P. acnes 균에서도 마치현 추출물의 농도 의존적으로 항균 활성이 확인되었다. 본 연구는 마치현 추출물의 항산화, 항염증 및 항균 효과를 가지는 생리 활성 물질로서 활용 가능성을 확인하였다.
Epigallocatechin-3-gallate (EGCG) is the most abundant polyphenol molecule from green tea and is known to exhibit antioxidative as well as tumor suppressing activity. In order to examine EGCG tumor invasion and suppressing activity against adult T-cell leukemia (ATL), two HTLV-1 positive leukemia cells (HuT-102 and C91-PL) were treated with non-cytotoxic concentrations of EGCG for 2 and 4 days. Proliferation was significantly inhibited by 100 ${\mu}M$ at 4 days, with low cell lysis or cytotoxicity. HTLV-1 oncoprotein (Tax) expression in HuT-102 and C91-PL cells was inhibited by 25 ${\mu}M$ and 125 ${\mu}M$ respectively. The same concentrations of EGCG inhibited NF-kB nuclearization and stimulation of matrix metalloproteinase-9 (MMP-9) expression in both cell lines. These results indicate that EGCG can inhibit proliferation and reduce the invasive potential of HTLV-1-positive leukemia cells. It apparently exerted its effects by suppressing Tax expression, manifested by inhibiting the activation of NF-kB pathway and induction of MMP-9 transcription in HTLV-1 positive cells.
본 연구에서는 속팽윤성과 고흡수성을 갖는 초다공성 수화젤의 제조과정에서 생분해성 가교제를 이용하여 생분해성 초다공성 수화젤을 제조하고 특성분석을 수행하였다. 친수성 고분자인 poly(ethylene glycol)의 양말단에 D,L-lactide를 개환 중합시켜 PLA-PEG-PLA 삼중공중합체를 합성한 뒤, 양말단에 비닐기를 도입하여 생분해성 가교제를 합성하였다. 조성이 다양한 초다공성 수화젤을 제조하여 각각의 팽윤도, 팽윤속도 및 생분해성을 비교하였다. 중합된 고분자의 화학적 조성을 $^1H$-NMR, GPC, FT-IR 측정을 통해 확인하였고, 수화젤 표면 및 내부의 SEM 분석을 통해 수 백 ${\mu}m$ 크기의 공극들로 생성된 열린 채널구조의 초다공성 구조를 관찰하였다. 수은 다공도계로 수화절의 기공크기와 다공도를 측정하였고, 조성에 따라 물리회학적 성질이 조절될 수 있음을 알 수 있었으며, MTT분석에 의해 낮은 세포독성을 나타냄을 확인하였다.
신령버섯(Agaricus blazei Murill)의 액체 배양으로 다당체의 균사체외 분비를 유도하였으며, 버섯 균사체의 새포내 다당체와 세포외 분비 다당체의 면역증진활성을 in vitro 시험으로 비교하였다. 부분 정제된 세포내 다당체와 세포외 다당체의 총당 함량은 각각 85.6%와 95.3%였으며, ${\beta}$-glucan 함량은 각각 67.9%와 88.1%로 측정되었다. 면역활성 실험에는 시료의 닥 함량을 동일하게 맞추어 사용하였다. In vitro에서 균사체내외 다당체는 대식세포 주인 RAW 264.7을 활성화시켜 nitric oxide (NO) 생성을 농도 의존적으로 증가시켰으며, 각각 최대 53.9%, 53.1%의 비슷한 증가활성을 나타내었다. 또 균사채내외 다당체는 모두 RAW 264.7을 활성화시켜 염중성 cytokine류인 interleukin (IL)-$1{\beta}$, IL-6, tumor necrosis factor (TNF)-${\alpha}$의 생성을 증가시켰으며, 이때 3종의 cytokine 모두에서, 세포내 다당체에 비해 세포외 다당체를 처리했을때 저농도에서 높은 증가률을 나타내었다. 두 다당체는 in vitro 상에서 비장세포를 증식시키는 효과를 나타내었으며, 세포내 다당체가 농도 의존적으로 증식효과를 보인데 반해 세포외 다당체는 저농도에서 증식이 높았고, $250\;{\mu}g/ml$ 농도 이상에서는 더 높아지지 않았다. 두 다당체 모두 암세포인 B16F0 melanoma에 대한 직접적인 세포독성 효과는 나타내지 않았다. 신령버섯 균사체 배양으로 생성된 세포내외 다당체는 in vitro에서 모두 면역활성을 증가시키는 것으로 나타났으며 전반적으로 그 활성은 세포내 다당체보다 세포외 다당체가 우수한 것으로 판단되었다.
Fumonisin B$_1$(FB$_1$)은 모든 동물종에서 간독성을 나타내며 흰쥐에서는 간암과 신장독성을 일으키는 발암물질이다. 본 연구에서는 FB$_1$의 반복투여 효과를 관찰하고자 수컷 Sprague-Dawley 흰쥐에 1 mg FB$_1$/kg을 1일(T$_1$) 혹은 2일(T$_2$), 3일(T$_3$)간 반복주사하였다. T$_1$군에서는 간장과 신장 다같이 독성작용이 발견되지 않았으나 간장에서는 대조군에 비해 5.5배의 증가된 세포분열상이 관찰되었다. 한편 T$_3$군에서는 병리조직학적 관찰과 혈청검사에서 간소엽 중심부에 병변이 있는 간독성이 관찰되었으며, 신장은 수질부 outer stripe에서 세포변성이 인정되었다. T$_3$군에서는 간효소를 포함하는 혈청검사치가 증가되었는데 특히 Cholesterol이 증가되었다. 따라서 FB$_1$의 반복 노출시에 신장보다는 간장에서 먼저 세포독성이 관찰되었으며 세포분열 역시 증가되었다.
본 연구에서는 인간 면역 세포인 B 세포와 T 세포 그리고 그들의 cytokinedml 분비에 대해서 조사하였다. 또한 일일 경구 투여량을 30 mg/mL 그리고 100 mg/mL의 농도로 조절하여 암컷 ICR 마우스에 투여하여 혈청에서의 항체 생성량을 조사하였다. 인간 정상 폐세포에 대한 세포독성 결과 모든 추출물이 24.15% 이하의 독성을 나타내었다. 인간 면역 세포인 B 세포와 T 세포 모두 초고압 추출공정에 의해 40% 이상 생육이 증진하였다. 인간 면역 세포인 B 세포와 T 세포에서 분비되는 cytokine(IL-6, TNF-${\alpha}$)의 양은 일반 열수 추출 공정과 비교하여 약 20-50% 증가하였다. 또한 고로쇠 추출물의 투여로 인해 총 항체 생성량도 증가하였다. 이와 같은 결과에서 고로쇠의 효과적인 최적의 추출 조건은 일반적인 공정 보다 $100^{\circ}C$이하의 온도에서 높은 압력과 함께 용매 추출공정을 이용하는 것이다.

Hepatocytes isolated from rats which have been pretreated with phenobarbital (80 mg/kg for 3 days), were able to take up N-acetylcysteine from surrounding medium and were able to synthesize the reduced glutathione ($GSH^{\ast}-3$) intracellularly. The N-acetylcysteine is quickly deacetylated after the uptake and increases the pool size of cysteine, which was very low initially (5 nmol/$10^6$ cells). From this increased intracellular cysteine pool, GSH was synthesized. Freshly isolated rat hepatocytes contained a high level of GSH (30 nmol/$10^6$ cells), but upon incubation with the diethylmaleate, it was markedly decreased (10 nmol/$10^6$ cells). The hepatocytes with depleted GSH have lost viability upon incubations with acetaminophen (5mM) and paraquat (2 mM). However, when the N-acetylcysteine (1 mM) was added to this incubation condition, these chemical induced hepatocellular necrosis were prevented for longer durations. This N-acetylcysteine dependent protective effect against the hepatotoxic chemicals was lost by adding methionine sulfoximine (10 mM), an inhibitor of GSH biosynthesis. Both the carbontetrachloride (5 mM) and chioroform (5 mM) added to the incubation medium caused rapid losses of GSH and cell viability, even without the prior depletion of cellular GSH. However, again, if the 1mM N-acetylcysteine was supplemented, the rates of losses of GSH and cell viability were retarded in both cases. Even though large amounts of the added N-acetylcysteine was present in the cell, N-acetylcysteine conjugate of acetaminophen was not formed. Instead, only large amounts of GSH conjugate of the drug was produced. Thus, it is concluded that the added N-acetylcysteine is taken up and utilized for resynthesis of GSH. In turn, this resynthesized GSH contributes to the protection against cytotoxicity inducible with hepatotoxic drugs.
