Secoiridoids, Iridoids and Flavonol Glycosides from Hydrangea paniculata Flowers and their C2C12 Myotube Hypertrophic Activity (나무수국 꽃의 Secoiridoid, Iridoid 및 Flavonol 배당체의 골격근세포 비대 유도 효능)

  • Five secoiridoids (1-3, 5, 10), a iridoid (4) three flavonol glycosides (7-9) and a coumarin (6), were isolated from the flowers of Hydrangea paniculata. Their chemical structures were elucidated as kingiside (1), morroniside (2), sweroside (3), loganin (4), vogeloside (5), umbelliferone (6), quercetin-3-O-sambubioside (7), quercetin-3-O-neohesperidoside (8), kaempferol 3-O-sambubioside (9) and secologanin dimethyl acetal (10), respectively, by spectroscopic analysis. All isolated compounds 1-10 were assessed for their ability to induce C2C12 myotube hypertrophy. Among them, loganin (4) and kaempferol 3-O-sambubioside (9) increase the diameter of C2C12 myotubes. All isolated compounds 1-10 were firstly reported from the flowers of Hydrangea paniculata, and the skeletal muscle hypertrophic activity of 4 and 9 was also reported for the first time.

  • Six iridoids were isolated from the 70% ethanol extract of Lonicera Caulis (Caprifoliaceae) and their structures were identified as epialyxialactone (1), secologanin dimethyl acetal (2), sweroside (3), loganin (4), loganic acid (5) and demethylsecologanol (6). The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. This is the first report of the iridolactone, epialyxialactone (1), from the Caprifoliaceae plants and loganic acid (5) and demethylsecologanol (6) from Lonicera Caulis.

Antioxidative Activity and Active Compound Analysis of the Extract and Fractions of Corni Fructus (산수유 추출물 및 분획물의 항산화 활성과 활성성분 분석)

  • In this study, analysis of active compounds that are believed to be highly relevant to antioxidant activity was carried out on the methanol extract and its solvent fractions of Corni fructus. The DPPH radical scavenging activity for the comparison of antioxidant activity was higher in order of aqueous fraction > methanol extract > ethyl acetate fraction > n-hexane fraction. It is similar to the order of total polyphenol contents in the samples. As a result of LC-MS analysis, phenolic acid compounds such as caffeic acid, gallic acid and chlorogenic acid and lognin, which is known as a representative active ingredient of Corni fructus, were identified as active compounds. And the antioxidative activity and the total polyphenol content of the extracts and solvent fractions were found to be related to the contents of the compounds. Particularly, it was confirmed that phenolic acid such as caffeic acid contributes to the antioxidative activity of the aqueous fraction of Corni fructus methanol extract.

  • Phlomis umbrosa and Dipsacus asperoides are distinct species, even though they have a similar appearance. Dipsacus asperoides is used as a Chinese medicinal plant for and has bone strengthening and fracture healing but Phlomis umbrosa has no effect on bone growth. Recently, these plants were used in children's food to improve their bone growth, without distinction in food. Intakes of Dipsacus asperoides in food may be dangerous, because it has never been used in food and its safety has never been tested in humans. We developed liquid chromatography with tandem mass spectrometry method to distinguish these plants in food. The method was validated for linearity, limits of detection, limits of quantification, accuracy and precision. In 5 of 17 samples, we identified Dipsacus asperoides, containing loganin $0.19-14.45{\mu}g/mL$, sweroside $0.13-4.61{\mu}g/mL$ and akebia saponin D $0.59-19.29{\mu}g/mL$. The developed method might be useful to identify Dipsacus asperoides in adulterated food.

Acute Oral Toxicity Study of Standardized Gami-Honghwa-Tang (KH-19) in Rats and Beagle Dogs (HPLC로 표준화한 가미홍화탕(KH-19)의 랫드와 Beagle견에 대한 단회경구투여독성시험)

  • Gami-Honghwa-Tang (KH-19), a herbal prescription for reducing the side effect of radiotherapy, is composed of eight crude herbs such as Rehmanniae Radix Preparata, Angelicae Gigantis Radix, Cnidii Rhizoma, Paeoniae Radix, Corni Fructus, Moutan Cortex Radicis, Lycii Fructus, Carthami Flos, and Glycyrrhizae Radix. In this study, marker substances in KH-19 were analyzed by high performance liquid chromatography-diode array detector (HPLC-DAD) and safety evaluation of standardized KH-19 was evaluated by good laboratory practices (GLP) guideline of Korea Food and Drug Administration. HPLC-DAD was employed to determine the quantities and the qualities of several marker substances such as 5-hydroxymethyl-2-furaldehyde (5-HMF), paeonol, loganin, paeoniflorin, glycyrrhizin, and decursin in the KH-19. In acute oral toxicity study on rat, transient inhibition of body weight was shown, but change in general symptom was not detected. No dead animal was observed up to 5,000 mg/kg in both male and female animals. In acute oral toxicity study on Beagle dog, transient vomiting, diarrhea, anorexia, and body weight reduction were observed, However, no dead animal was observed up to 2,000 mg/kg in both male and female animals.

Effects of Methanol Extract from Cornis fructus on Melanogenesis (산수유 메탄올 추출물이 B16/F10 Melanoma 세포주의 멜라닌 생성에 미치는 영향)

  • Cornis fructus has been traditionally used for treating allergy, asthma, hepatitis, and chronic nephritis. It major chemical constituents have been saponins, phenolic acids and loganin. The main aim of the present study was to examine the effect of methanol extract from Cornis fructus on melanogenesis. Cell were cultured in the presence of methanol extracts from Cornis fructus for 48 h, and there were estimated total melanin content as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. Methanol extract from Cornis fructus increased the melanin content and tyrosinase activity in a dose-dependent manner. Particularly, it was observed that only methanol extract $200\;{\mu}g/ml$ stimulated the melanin secretion in B16/F10 melanoma cells by 152% at 48 h treatment and the activity of tyrosinase was increased by 261% in the presence of same concentration.

Extract changes of Caulis Lonicerae Japonicae according to with or without Iron (인동(忍冬)의 기철(忌鐵) 및 반철(伴鐵) 추출시(抽出時) 추출물(抽出物)의 변화(變化))

  • Objectives : In case of herbs decoction, the ceramic or earthware pots was recommended, but not the metals, particularly iron or aluminum, which could cause unknown chemical reactions. In Korean Medical classics, it has been known that some herbs including Caulis Lonicerae Japonicae (CL) were not recommended to boil with iron pot. This study investigates the physical changes of extracts of CL with or without iron. Methods : CL was decocted with reflux cooling extraction system to prevent evaporation and volatilization. Content of polyphenol was detected by Folin-Denis method and the levels of loganin and chlorogenic acid were evaluated by UPLC. Results : The color of extract with glass beads (GB) is yellowish brown, and the iron beads (IB) is blackish brown. Polyphenol and chlorogenic acid levels were reduced in IB extracts. Conclusions : The color of extract was change to blackish brown, and polyphenol and chlorogenic acid levels were reduced in CL extract with iron. Therefore, iron pots is not suitable for CL extraction.

Quantitative Analysis of Twelve Marker Compounds in Palmijihwang-hwan using Ultra-Performance Liquid Chromatography Coupled with Electrospray Ionization Tandem Mass Spectrometry

  • An ultra-performance liquid chromatography (UPLC) coupled with electrospray ionization (ESI) tandem mass spectrometry (MS) method was established for quantitative analysis of twelve components, allantoin (1), morroniside (2), 5-hydroxymethyl-2-furfural (5-HMF) (3), loganin (4), coumarin (5), cinnamic acid (6), mesaconitine (7), cinnamaldehyde (8), hypaconitine (9), aconitine (10), alisol B (11), and alisol B acetate (12) in a Palmijihwang-hwan decoction. The twelve constituents were separated on a UPLC BEH C18 column ($2.1{\times}100mm$, $1.7{\mu}m$) at a column temperature of $40^{\circ}C$ by gradient elution with 0.1% (v/v) formic acid in water and acetonitrile as the mobile phase. The flow rate was 0.3 mL/min and the injection volume was $2.0{\mu}L$. Calibration curves of all compounds were acquired with values of the correlation coefficient ${\geq}0.99$ within the test ranges. The limits of detection and quantification for all analytes were 0.01 - 4.53 ng/mL and 0.03 - 13.60 ng/mL, respectively. The concentrations of the compounds 1 - 9 and 12 were 72.83, 4389.00, 4859.00, 3155.17, 223.67, 33.50, 1.97, 518.00, 2.25, and $25.00{\mu}g/g$, respectively. However, compounds 10 and 11 were not detected.

Neuroprotective Compounds Isolated from the Methanolic Extract of Lonicera japonica

  • A $CH_3Cl$ extract from the flower of Lonicera japonica (Lauraceae) significantly protected primary cultures of rat cortical cells injured by the excitotoxic amino acid, L-glutamate. Loganin (1), secoxyloganin (2), caffeic acid (3) rutin (4), hyperoside (5), quercetin-3-O-glucoside (6), lonicerin (7), kaempferol-3-O-rutinoside (8), luteolin-7-O-b-D-glucopyranoside (9), quercetin (10) and luteolin (11) were isolated by bioactivity-guided fractionation from the $CH_3Cl$ fraction and further separated using chromatographic techniques. Caffeic acid, lonicerin, kaempferol-3-O-rutinoside, quercetin and luteolin had significant neuroprotective activities against glutamate-induced neurotoxicity in primary cultures of rat cortical cells at concentrations ranging from $0.1{\mu}M$ to $10.0\;{\mu}M$.
