The freezing susceptibilities of two larval stages (D-shaped and umbo) of the pearl oyster (Pinctada fucata martensii) were evaluated by the electron microscopy (light, transmission electron and scanning electron). The morphological shapes were examined from each pre-frozen or frozen-thawed stage of the cryopreserved larvae in liquid nitrogen by using the cryoprotectant, dimethyl sulfoxide ($Me_2SO$) mixed with sucrose. Although a portion of the shell was damaged, the hinge and prodissoconch were intact and clearly visible after preservation in liquid nitrogen. In addition, the cytoplasm of the frozen-thawed larvae maintained the normal organelle integrities, e.g., endoplasmic reticula, lipid droplets, mitochondria, nucleus and microvilli. However, some of the frozen-thawed larvae showed irregularly arranged cilia, rough shell surfaces and round-lumped cilium heads. These results indicate that P. fucata martensii larvae are susceptible to freezing, at least at those two critical developmental stages (D-shaped and umbo), and suggest a new industrial investigation including reduction method of cell injury for preserving microbial starter cultures need to be developed.

Individual starter culture were inoculated into liquid medium and incubated at $40^{\circ}C$ for 16 hours. Whole cell were obtained and evaluated for ${\beta}-galactosidase$ activity using orthonitrophenyl-${\beta}-D-galactopyranoside$ (ONPG) as substrate. S. thermophilus had more ${\beta}-galactosidase$ activity than other Lactobacilli did. To study the effect of storage temprature on enzyme activity of yoghurt, some samples of cultured yoghurt were stored under refrigeration $(4^{\circ}C)$, and the others under room temperature $(23^{\circ}C)$. At $4^{\circ}C$, yoghurt had ${\beta}-galactosidase$ activity and many viable bacteria in 1 month. After 20 days, yoghurt had maximum ${\beta}-galactosidase$ activity. At $23^{\circ}C$, yoghurt had ${\beta}-galactosidase$ activity by 5 days. As this experiment shown ${\beta}-galactosidase$ activity was ascribed to viable bacteria, especially S. thermophillus. Commercial yoghurt had lower ${\beta}-galactosidase$ activity. There were considerable variations with regard to the lactose hydrolyzing capabilities of commercial yoghurt samples.

Non-aflatoxigenic Aspergillus oryzae and aflatoxigenic A. flavus cannot be clearly identified by partial sequencing of the internal transcribed spacer (ITS) and 18S ribosomal ribonucleic acid (18S rRNA) regions. This study aimed to compare the accuracy among three aflatoxin detection methods using ultra-performance liquid chromatography (UPLC), high-performance liquid chromatography (HPLC), and an enzyme-linked immunosorbent assay (ELISA) kit and to select the non-aflatoxigenic Aspergillus sp. isolated from soybean paste. All analytical methods were suitable according to the international standards of Codex Alimentarius FAO-WHO (CODEX) or the Ministry of Food and Drug Safety (MFDS). UPLC exhibited the best of limit of detection (LOD) and limit of quantification (LOQ). Based on UPLC, HPLC, and the ELISA kit assay, the P5 and P7 strains isolated from soybean paste had 1,663.49, 1,468.12, and >20 ㎍/kg and 1,470.08, 1,056.73, and >20 ㎍/kg, respectively, detected and re-identified as A. flavus. In contrast, the P3 and P4 strains (A. oryzae), which were detected below the MFDS standards in all assays, were confirmed as non-aflatoxigenic fungi. Among the methods evaluated for quantitative analysis of aflatoxin, UPLC and HPLC are superior in terms of accuracy, and the ELISA kit rapidly detects low concentrations of aflatoxin. Furthermore, this study demonstrates that any Aspergillus sp. isolated for use as a fermentation starter should be analyzed for potential aflatoxin production using UPLC and HPLC for accurate quantitative analysis or ELISA for the rapid detection of low-level concentrations of aflatoxin.

Seeds of neem were collected from different parts of India and analyzed for their azadirachtin content by High Performance Liquid Chromatography (HPLC). In order to assess the effects of genotypic and geographical variation on azadirachtin content in cell cultures, callus development was attempted in the seeds containing high and low concentration of azadirachtin. The concentration of azadirachtin in callus cultures was significantly affected by the explant source. Seed kernels with higher azadirachtin content produced higher azadirachtin content in callus cultures and lower azadirachtin content was seen in callus cultures produced from seed kernels with low azadirachtin content. The protocol for development of elite stock culture of Azadirachta indica was established with the objective of selecting a high azadirachtin-producing cell line. The highest azadirachtin-producing cell line was selected and the effects of different media and illumination conditions on growth and azadirachtin production were studied in shake flask suspension culture. Detailed batch growth kinetics was also established. These studies provided elite starter culture and associated protocols for cultivation of A. indica plant cell culture in the bioreactor.

Pyrostarters play a role as a turbopump starter in liquid propellant propulsion systems by supplying pressurized gas to power turbines for engine start. A rupture disk in pyrostarters, which is usually installed behind a nozzle throat, not only isolates the charged solid propellants from the external environment but also improves the ignitability of the solid propellants by increasing a chamber pressure at the beginning of combustion. Experimental tests have been performed to study the effects of rupture disk thickness, depth and shape of scores, and pressure build-up rates on burst pressures and burst diameters. The experimental results show that the developed rupture disk fulfills the performance requirements expected in a real operational condition.

The high pressure turbopump carries out supplying the oxidizer in the liquid propulsion rocket in the combustion chamber. Because an LRE requires a very short starting time , the turbine at the turbopump experiences high torque that was produced by the high pressure and the high temperature. The purpose of this study is to evaluate a turbine blade surface temperature profiles at initial starting 0 ${\sim}$ 0.5 sec. Using $Fine^{Tm}$/turbo, three dimensional Baldwin-Lomax turbulence models are used for numerically analysis. The turbine is composed of 108 blades total, but only 7 rotors were considered because of periodic symmetry effect. Because of interaction with a bow shock on the suction surface, the boundary layer separates from suction surface at inner area of turbine blade. The averaged temperature of the turbine blade tip at 1000 rpm is higher than that of 9000 rpm. Especially at 1000 ${\sim}$ 9000 rpm, temperatures increases on the hub side of the turbine blade tip. Moreover at 9000 rpm, the temperatures from the hub to the shroud of the blade tip increase as well.

A total of 38 energy balance trials were done for calves fed a liquid milk replacer, calf starter and second cut mixed hay. Milk replacer supplied 81% of the total dietary energy. Live weight of calves averaged 54.1 (S.D 6.2) kg and daily gain 0.37 (${\pm}0.23$)kg. The metabolizability of gross energy averaged 0.822. A regression was calculated relating energy retention (ER, $kJ/kg^{0.75}$) to the intake of metabolizable energy (IME, $kJ/kg^{0.75}$): ER = 0.72 (${\pm}0.12$) IME - 330, r = 0.702, P < 0.01, $S.E.{\pm}18.0$. Metabolizable energy for maintenance (MEm) was calculated to be $458kJ/kg^{0.75}$ when ER = 0. The amount of IME over MEm for an individual animal (Meg, $kJ/kg^{0.75}$) was regressed on averaged daily gain (ADG, kg): Meg = 413 (${\pm}91$) ADG + 0.2, r = 0.650, P < 0.01, $S.E.{\pm}21$. The amount of ME requirement for suckling calves was estimated using values obtained above.

Gumiganghwal-tang (GMT) is a traditional herbal prescription used for treatment of the common cold, pain, and inflammatory diseases. Variations in the amounts of bioactive components of GMT and GMT fermented with 10 Lactobacillus strains were investigated by high-performance liquid chromatography coupled with diode array detection (HPLC-DAD). Simultaneous qualitative and quantitative analyses of eleven bioactive compounds (prim-O-glucosylcimifugin, liquiritin, cimifugin, baicalin, liquiritigenin, wogonoside, baicalein, wogonin, butylphthalide, imperatorin, and isoimperatorin) were performed, with comparison of their retention times (tR) and UV spectra with those of standard compounds. The amounts of baicalin (8.71 mg/g), liquiritigenin (5.28 mg/g) and butylphthalide (5.10 mg/g) were the major compounds in GMT. We found that L. fermentum KFRI 145 fermented wogonoside and baicalin to their aglycones, wogonin and baicalein, respectively. These results indicated that L. fermentum KFRI 145 has potential as a functional starter culture for manufacturing fermented GMT.

Makgeolli is a traditional Korean alcoholic beverage. The flavor of makgeolli is primarily determined by metabolic products such as free sugars, amino acids, organic acids, and aromatic compounds, which are produced during the fermentation of raw materials by molds and yeasts present in nuruk, a Korean fermentation starter. In this study, makgeolli was brewed using the wild yeast strain Saccharomyces cerevisiae Y98-5, and temporal changes in the metabolites during fermentation were analyzed by ultra-high-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry. The resultant data were analyzed by partial least squares-discriminant analysis (PLS-DA). Various metabolites, including amino acids, organic acids, sugar alcohols, small peptides, and nucleosides, were obviously altered by increasing the fermentation period. Changes in these metabolites allowed us to distinguish among makgeolli samples with different fermentation periods (1, 2, 3, 6, 7, and 8 days) on a PLS-DA score plot. In the makgeolli brewed in this study, the amounts of tyrosine ($463.13{\mu}g/mL$) and leucine ($362.77{\mu}g/mL$) were high. Therefore, our results indicate that monitoring the changes in metabolites during makgeolli fermentation might be important for brewing makgeolli with good nutritional quality.
Self-contained portable remote-indicating magnetic compass was designed and tested. Its sensing part consists of main scale disk of rotary encoder with photo-etched 180, oopaque slits and three pairs of light sources and detectors seperated by a transparent index scale, so that any angular deflection of main scale disk against the bowl may be detected optically. The outputs from the sensing part are transformed into digital ship's course indication by electronics unit which consists of operational amplifier, phase discriminator, up-down counter and counting starter circuit. The results from testign the above compass with turntable at variable rate of turn are as following : 1) The main scale disk deflection against the bowl can be detected without any disturbing influence to the freedom of its north-seeking function. 2) The digital indication resolution is found to be $\circ \pm$1 degree. 3) Misrepresenting indication can be avoided by ensuring dimension uniformity and arrangement accuracy of slits on main and index scale disks and by centering pivot in the encoder disk. 4) Indication resolution should be improved by modifying the signal processing and by doubling the number of slits on compass card. 5) Further study should be made on developing non-conductive compass liquid, analogue repeater driving system and more compact processing element.
