This study was devised to observe the inhibitory effects of ethanol treatment precipitate (crude acidic polysaccharide) from Korean red ginseng on a lipolytic action of Toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been inoculated with sarcoma-180. The yields of crude acidic polysaccharide from Korean red ginseng was 63.5%. In vitro, at the concentration of 500rg /ml, the inhibition rate of lipolysis by the crude acidic polysaccharide of Korean red ginseng was 38.8% and the total inhibitory activity per gram of ginseng material was 4,928 nit. In vivo, the red ginseng polysaccharide(40mg/ml in saline soon.) 16ul/g of body weight was injected to the sarcoma-180 bearing mice once In 3 days until death. The effects against the extension of life span was little but body weight gain of sarcoma-180 bearing mice decreased significantly by administration of Korean red ginseng polysaccharide compared to those of the control group.
Toxohormone-L is a lipolytlc factor, found in ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma. A substance that inhibited the lipolytic action of Toxohormone-L was isolated from Korean red ginseng powder. This substance had a pectin-like a-1,4-polygalacturonan backbone with some acetoxyl groups, and so was an acidic polysaccharide. Acidic polysaccharide was found to inhibit significantly toxohormone-L-induced lipolysis at its concentration of 10$\mu\textrm{g}$/ml.
Effect of an acidic polysaccharide fraction In Korean white and red ginseng on lipolytic action of Toxohormone-L was studied. Crude acidic polysaccharide fraction was extracted from main and lateral root of Korean white and red ginseng separately and purified several times. Inhibitory effect of crude polysaccharide fraction was determined by unit (1 unit is loft inhibition rate per Is sample). Yields of purified crude polysaccharide fraction of main and lateral root of red ginseng were 2.9 and 2.2 times higher than those of white ginseng, respectively. Inhibitory effects of main root of white and red ginseng, 11.hen final reaction concentrations of sample were 50, 100, 200, 500 $\mu$g/ml, were 37.2가 and 23.7% higher than those of lateral root of white and red ginseng. Inhibitory effect of main root of red ginseng was 2.3 times higher than that of white ginseng.

In this study, the tripeptide GKH(Glycine-Lysine-Histidine) derived from Parathyroid hormone (PTH), which was known as lipolytic peptide, is attached to 9-poly Lysine(TAT) to be used as a cosmetic ingredient for slimming products. When Glycerol release, expressed as extracellular glycerol concentration, is lipolysis index, TAT-GKH at $10^{-5}$mo1/L induces approximately 41.5% maximal lipolytic effects in epididymal adipocytes isolated from rats, compared with basal lipolysis. Epididymal adipose tissues of male rats is assessed ex vivo by microdialysis. Probes are perfused with Ringer solution in which increasing concentrations of TAT-GKH. The perfusion of TAT-GKH induces lipolytic effect. Penetration study showed that TAT-GKH efficiently elevates 36 times higher penetration into the excised hairless mice skin than GKH. in vivo study showed that TAT-GKH had a better effect upon the relative volume of eye bag after 28 days of application on twenty(+2) healthy female volunteers. It was identified that TAT-GKH increases penetration enhancement and lipolytic effects in both in vitro, ex vivo and in vivo.

Lipolytic enzyme-producing bacteria were isolated from edible oil mill effluents on tributyrin agar medium. The shake-flask-scale studies yielded a promising isolate and it was identified as Pseudomonas sp. An OME using various microbiological observations such as cultural, microscopic, and biochemical tests was undertaken and confirmed using PIBWIN bacterial identification software. Lipolytic enzyme production was screened with oils such as sunflower, caster, coconut, tributyrin, and olive. Amongst these, olive oil showed an increased lipase production 6.1 U/ml. In view of the highest lipolytic enzyme production with olive oil, further optimizations were carried out using olive oil as a carbon source. Lipolytic enzyme production was optimized by a conventional 'one variable at a time' approach and the significant factors were further analyzed statistically using response surface methodology (RSM). The effect of physical factors such as incubation time, temperature, initial medium pH, and nutritional factors such as concentration of olive oil and yeast extract were examined for lipase production. Lipolytic enzyme secretion was strongly affected by three variables (incubation time, concentration of yeast extract and olive oil). Therefore, the interaction of these three factors was further optimized using response surface methodology. The optimized conditions of lipase production using response surface methodology yielded a maximum of 9.62 U/ml with optimum conditions for incubation, yeast extract and olive oil concentrations were found to be 48 hr, 0.3 g. and 0.9 ml. respectively.

In the tortoise, Amyda japonica, a cold-blooded animal readily available in this country, the role of catecholamines in the regulation of free fatty acids(FFA) metabolism was investigated in both in vivo and in vitro studies. 2. Norepinephrine elevated both FFA and glucose levels in plasma. 3. When $50{\mu}g/kg$ of Epinephrine, Norepinephrine and Isopropylarterenol were administered intravenously, the relative effectiveness of mobilizing FFA was in the descending order of potency-Epinephrine, Norepinephrine and Isopropylarterenol. 4. In order to exclude the 'tonic influence of the endogenous catecholamines', reserpine was given to some animals. Two days after the reserpine-treatment, glucose showed a significant increase over the solely vehicle treated controls, FFA but an insignificant one. Excised auricles from those animals showed a diminished response to tyramine. Seven days after the treatment, however, when the depletion of catecholamines from the tissue stores seemed to be complete, judged from the absence of the response of isolated auricles to tyramine, both FFA and glucose levels were definitely lowered. 5. In in vitro experiments Epinephrine enhanced the FFA-release from the adipose tissue. The effect increased proportionately with the concentration until a maximal effect was attained at a concentration of 1x $10^5$ g/ml. 6. The order of potency in releasing FFA from adipose tissue in vitro was the same as in vitro, i.e., Epinephrine, Norepinephrine and Isopropylarterenol, but the differences were much less marked. 7. Ergotamine exerted no lipolytic action, but inhibited the lipolytic effect ef Epinephrine significantly. 8. Nethalide showed a slight lipolytic effect per se but inhibited the Epinephrine-induced lipolysis significantly. 9. Catecholamines play an important role in regulating FFA metabolism in the cold-blooded animal, just as in the warm-blooded animals, and the tortoise, Amyda japonica, may be used in the studies of fat metabolism as well as the rat.
This study was devised to obserL'e the inhibitory effects of 7 kinds of ginsenosides on a lipolytic action of Toxohormone-L. The ginsenosides used in this experiment were -Rbl, -Rbl, - Rc, - Re, - Rgl, and - Rg2 prepared from Korean red ginseng. Toxohormene-L was partially purified by centrifigation from the ascites fluids of Sarcoma-180 bearing mice. In vitro test showed that the inhibitory effect of - Rb2 on the lipolysis by Toxohormone-L was highest percent among other treatments at concentration of 100 $\mu\textrm{g}$/ml and 500 $\mu\textrm{g}$/ml of reaction mixture. And total inhibitory activity (units) of - Rb2 was also highest among other treatments at the same concentration. However, in vivo test, body weight gain of Sarcoma-180 bearing mice decreased significantly by administration of - Rg2 compared to those of the control or other ginsenosides treated groups.
The effects of chromium picolinate supplementation in pig diet were evaluated by measuring the in vitro lipogenic and lipolytic activities in adipose tissue and the protein synthetic activity in liver acinar cell in culture. Thirty-two male and thirty-two female pigs were randomly assigned to one of four dietary groups: Control, 100 ppb, 200 ppb, and 400 ppb of Cr in the form of picolinate. The chromium picolinate supplementation (p < 0.01) increased the in vitro lipolytic activity in adipose tissue of pig, but had no effects on lipogenesis. The chromium picolinate effect was greater in female pigs than in male pigs on lipolytic activity. The results from the studies with the liver acinar cells in culture indicated that chromium picolinate supplementation increased protein synthetic activity (p < 0.05). It was observed through this experiment that chromium picolinate functions not only on fat degradation but also on retained protein synthesis.

Objectives: In this study, the lipolytic effects of Eriobotrya folium extract (EFE) on local fat was investigated in high fat diet (HFD)-induced obesity mouse and 3T3-L1 adipocytes. Methods: C57BL/6J mice (5 weeks) were fed HFD for 6 weeks to induce obesity. EFE (20 mg/ml, $100{\mu}l$) or saline ($100{\mu}l$) as a normal control was injected into left inguinal fat pad region, 3 times per a week for last 2 weeks. After sacrifice, body weight, and histological changes of the inguinal fat pad were evaluated. The expressions of hormone-sensitive lipase (HSL) and adipose triglyceride lipase (ATGL) in inguinal fat pad were analyzed by Western blotting. Also, lipid accumulation and lipases release were determined in 3T3-L1 adipocytes by oil red o staining. Results: EFE significantly reduced the weight of inguinal fat pad and the size of adipocytes in HFD-induced obesity mice compared to control. The treatment of EFE up-regulated the expressions of HSL and ATGL in inguinal fat pads of obesity mice, as well as 3T3-L1 adipocytes. In addition, EFE inhibited the lipid accumulation in 3T3-L1 adipocytes in a dose dependent manner. Conclusions: EFE showed lipolytic effect on local fat of HFD-induced obesity mice by up-regulation of the lipases secretion. This suggests that EFE could be considered as anti-obese substance with lipolytic property on local fat.
