Dietary Antioxidant Vitamins Intakes and Plasma Antioxidant Levels in Korean Elderly with Diabetes Living in Ulsan (울산 지역 당뇨 질환 노인의 항산화 영양소 섭취실태 및 혈액 항산화 영양상태)

  • Diabetic mellitus in an older population is associated with increased basal oxidative stress and free radical accentuated by hyperglycemic challenge. Enhanced free radical in diabetic elderly can cause the oxidative damage and such damage can be protected by antioxidant defense system. It is believed that vitamin C, A and E are the most abundant and effective antioxidants in human plasma. The purpose of this study was to determine the antioxidant status in Korean diabetic elderly using the case-control study. The antioxidant status was examined by determining plasma levels of antioxidant vitamins (vitamin C, A, E, ${\beta}$-carotene), total antioxidant status (TAS) and thiobarbituric acid reactive substance (TBARS) and intakes of vitamin C, A, ${\beta}$-carotene and retiol. Fasting glucose and HbA1c levels and serum lipid profiles (triglyceride (TG), total cholesterol, HDL-cholesterol and LDL-cholesterol) were also determined. Diabetic subjects were 122 elderly persons over 60 years old, visiting public health center, and control subjects were 96 healthy elderly persons living in Ulsan, Korea and they were matched by age, gender, smoking and drinking status. The diabetic and control subjects were divided into sub-groups according to the status of using diet therapy and vitamin supplement. The subjects were interviewed to collect data on their general characteristics, disease history, vitamin supplement, diet therapy and health-related habits by questionnaires. Their dietary intakes were obtained by means of semi-quantitative food frequency questionnaires (SQFFQ). Fasting plasma glucose and HbA1c levels were significantly higher in diabetes than in control subjects, and plasma total cholesterol level of diabetes was not significantly different from that of control subjects. However serum HDL cholesterol level of diabetes was significantly lower and serum TG level of diabetes was significantly higher than those of control group. The average vitamin A and ${\beta}$-carotene intakes of diabetes were significantly higher than those of control subjects. There was no significant difference in plasma vitamin C, ${\beta}$-carotene, and TBARS levels between two groups, but plasma vitamin A, E and TAS levels were significantly higher in diabetes than those in control group. Plasma vitamin A and TAS levels of diabetic subjects using diet therapy were higher than those of control using diet therapy, and plasma vitamin E, ${\beta}$-carotene and TAS levels of diabetic subjects using vitamin supplements were significantly higher than those of controls using vitamin supplements. These results suggested that diabetic mellitus could enhance antioxidant defences against reactive oxygen species and interest in healthy eating such as consumption of more antioxidant nutrients.

Effects of Various Mulberry Products on the Blood Glucose and Lipid Status of Streptozotocin-induced Diabetic Rats (오디의 가공형태에 따른 Streptozotocin 유발 당뇨쥐의 혈당 및 지질의 상태에 미치는 영향)

  • The study aim was to investigate the effects of three types of mulberry products on the blood glucose and lipid statuses and peroxidative state under diabetic condition. The three types were mulberry liquor prepared by adding 30% ethanol to the crushed fresh fruit, mulberry wine and vinegar by fermentation. For diet experiment the mulberry liquor (M-Liquor), wine (M-Wine), and vinegar (M-Vinegar) were prepared as powders by freeze-drying of the respective product and were added to the diet at the level of 1% and mulberry fruit powder (M-Powder) at the level of 5%. Sprague- Dawley female rats weighing $150{\pm}10\;g$ were randomly assigned to one normal group, and five diabetic groups induced by one intraperitoneal injection of streptozotocin (STZ) at the level of 50 mg/kg. The normal and diabetic control (DM-Control) groups were fed diet without the mulberry products. During twenty-one days of experimental diet, blood glucose was maintained at a low level in the M-Liquor group compared with the DM-Control group. However, serum insulin level was higher in both M-Liquor and M-Vinegar groups after the experimental diet period. Serum levels of total cholesterol and triglyceride (TG) were lower in M-Liquor but HDL-/total cholesterol ratios were higher in the four M groups. The TG liver level was lower in M-Powder and M-Vinegar groups but the cholesterol level was lower in M-Powder than in the DM-Control group. Serum levels of thiobarbituric acid reactive substances were not different among the groups but the liver levels of these substances were lower in the four M groups than in DM-Control. Serum GOT and GPT levels were not changed by the mulberry products. These results indicated that mulberry liquor is the most effective among the four mulberry products for normalizing blood glucose and lipid status and that all the mulberry products were effective in enhancing antioxidant defense in the diabetic state.

The Discriminant Analysis of Blood Pressure - Including the Risk Factors - (혈압 판별 분석 -위험요인을 중심으로-)

  • The purpose of this study was to evaluate the usefulness of variables which were known to be related to blood pressure for discriminating between hypertensive and normotensive groups. Variables were obesity, serum lipids, life style-related variables such as smoking, alcohol, exercise, and stress, and demographic variables such as age, economical status, and education. The data were collected from 400 male clients who visited one university hospital located in Incheon, Republic of Korea, from May 1996 to December 1996 for a regular physical examination. Variables which showed significance for discriminating systolic blood pressure in this study were age, serum lipids, education, HDL, exercise, total cholesterol, body fat percent, alcohol, stress, and smoking(in order of significance). By using the combination of these variables, the possibility of proper prediction for a high-systolic pressure group was 2%, predicting a normal-systolic pressure group was 70.3%, and total Hit Ratio was 70%. Variables which showed significance for discriminating diastolic blood pressure were exercise, triglyceride, alcohol, smoking, economical status, age, and BMI (in order of significance). By using the combination of these variables, the possibility of proper prediction for a high-diastolic pressure group was 71.2%, predicting a normal-diastolic pressure group was 71.3%, and total Hit Ratio was 71.3%. Multiple regression analysis was performed to examine the association of systolic blood pressure with life style-related variables after adjustment for obesity, serum lipids, and demographic variables. First, the effect of demographic variable alone on the systolic blood pressure was statistically significant (p=.000) and adjusted $R^2$was 0.09. Adding the variable obesity on demographic variables resulted in raising adjusted $R^2$to 0.11 (p=.000) : therefore, the contribution rate of obesity on the systolic blood pressure was 2.0%. On the next step, adding the variable serum lipids on the obesity and demographic variables resulted in raising adjusted R2 to 0.12(P=.000) : therefore, the contribution rate of serum lipid on the systolic pressure was 1.0%. Finally, adding life style-related variables on all other variables resulted in raising the adjusted $R^2$to 0.18(p=.000) ; therefore, the contribution rate of life style-related variables on the systolic blood pressure after adjustment for obesity, serum lipids, and demographic variables was 6.0%. Multiple regression analysis was also performed to examine the association of diastolic blood pressure with life style-related variables after adjustment for obesity, serum lipids, and demographic variables. First, the effect of demographic variable alone on the diastolic blood pressure was statistically significant (p=.01) and adjusted $R^2$was 0.03. Adding the variable obesity on demographic variables resulted in raising adjusted $R^2$to 0.06 (p=.000) ; therefore, the contribution rate of obesity on the diastolic blood pressure was 3.0%. Non-enzymatic Antioxidant Status and Biochemical Parameters in the Consumers of Pan Masala Containing Tobacco

  • Background: Tobacco consumption is one of the leading causes of oral submucous fibrosis, oral cancer and even premature death. The present study was designed to compare the biochemical parameters and non-enzymatic antioxidant status and the lipid peroxidation products in pan masala tobacco users as compared with age-matched non-user controls. Methods: Pan masala and tobacco users of age $33.2{\pm}9.94$ years and age-matched controls ($31.2{\pm}4.73$ years) were enrolled for the study. Plasma levels of vitamin E, vitamin C, albumin, bilirubin, uric acid, glucose, urea, creatinine, aspartate amino transferase (AST), alanine amino transferase (ALT) were measured by standard methods. Serum malondialdehyde (MDA) levels were estimated as a measure of lipid peroxidation. Results: In the pan masala tobacco users, as compared to the controls, the level of vitamin C ($68.5{\pm}5.9$ vs $97.9{\pm}9.03{\mu}mol/L$, $p{\leq}0.05$) vitamin E ($18.4{\pm}5.3$ vs $97.9{\pm}9.03{\mu}mol/L$, $p{\leq}0.001$), albumin ($37.5{\pm}7.01$ vs $44.3{\pm}9.99g/L$, $p{\leq}0.001$), and malondialdehyde ($10.8{\pm}1.29$ vs $1.72{\pm}1.15nmol/ml$, $p{\leq}0.001$) were found to be significantly altered. Malondialdehyde was significantly correlated with vitamin E (r=1.00, p<0.001) and vitamin C (r=1.00, p<0.001) in pan masala tobacco users. Serum levels of AST ($31.0{\pm}16.77$ IU) and ALT ($36.7{\pm}31.3$ IU) in the pan masala tobacco users were significantly raised as compared to the controls (AST, $25.2{\pm}9.51$ IU, p=0.038; ALT, $26.2{\pm}17.9$ IU, p=0.038). Conclusion: These findings suggest that pan masala tobacco users are in a state of oxidative stress promoting cellular damage. Non-enzymatic antioxidants are depleted in pan masala tobacco users with subsequent alteration in the biochemical parameters. Supplementation of antioxidants may prevent oxidative damage in pan masala tobacco users.

A Study of Nutrient Intake, Serum Lipid and Leptin Levels of Elementary School Students with Different Obesity Index in Kyunggi Area (경기지역 일부 초등학생의 성별과 비만도에 따른 영양소 섭취상태와 혈청 Leptin 및 혈청 지질에 관한 연구)

  • The purpose of this study was to investigate the relationship among nutritional status, serum lipids and leptin of 134 elementary school students with different obesity index residing in Kyunggi-do, Korea. Subjects were assigned to under weight, normal weight or over weight group according to the their obesity index. Their nutritional status, serum lipid and leptin levels were evaluated based on 24-hr dietary recalls, anthropometric measurements and blood analysis. The mean weight, height and obesity index of total subjects was 150.8cm, 45.6kg and 5.4%. The mean % of body fat was higher while LBM and TBW (total body water) were lower in over weight subjects than those of other two groups. Total food, plant protein, carbohydrate, and vitamin C intakes of underweight male students were lower than those of overweight male students. The consumption of total fat was the highest in the female normal weight subjects and the lowest in female overweight subjects. Male students consumed more plant protein and vitamin C than female students. The mean daily intake of vitamin B$_2$, calcium and zinc of all subjects did not meet the Korean RDA. Also, iron intake of female students was lower than the Korean RDA. The mean level of serum leptin was the highest in male and female overweight subjects (p<0.001). The HDL-cholesterol level was the highest in the underweight group of male subjects (p<0.05) and in the normal weight group of female subjects (p<0.001) The serum level of leptin and atherogenic index (AI) of females were higher than those of males. Serum leptin of total subjects had significantly positive correlation with obesity index, weight, SBP, body fat, cholesterol intake, serum triglyceride, and AI. However, serum leptin was negatively correlated with LBM, TBW, intake of plant protein, vitamin B$_1$, vitamin C and serum HDL-cholesterol. Obesity index was also positively correlated with weight, SBP, body fat, serum triglyceride and AI. To summarize the results, the vitamin B$_2$, calcium, zinc, iron and fiber intakes should be supplied to elementary school students, especially those who were overweight. The average serum leptin level of students showed remarkably positive correlation with obesity index, body fat, serum lipids and AI. Also, it was suggested that the serum leptin might be related to the dietary factors like cholesterol, plant protein, vitamin B$_1$ and vitamin C of elementary school students.

Effect of Dietary Supplementation of Vitamin A and Chronic Consumption of Ethanol on Oxidative Damage and Antioxidant System in Rats (비타민 A 보충 식이 및 에탄올의 만성적 급여가 흰쥐의 체내 산화적 손상과 항산화체계에 미치는 영향)

  • Alcohol is well known agent which can damage the human tissues such as liver via stimulating lipid peroxidation. On the other hand, carotenoids in addition to vitamins A, C and I play important roles in protecting these oxidative damages as well as preventing the production of free radicals. This study was carried out to investigate the effect of dietary vitamin A on lipid peroxidation and antioxidants status in ethanol-treated rats. In the experiment, male Sprague-Dawley rats weighing 160~180 g were given a liquid diet containing 36% of total calories as ethanol for 7 weeks. The pair-fed control rats received an isocaloric amount of diet containing sucrose instead of ethanol on the following day Additionally, the liquid diet contained adequate amount of $\beta$-carotene, retinyl acetate or 13-sis-reinoic acid except vitamin A-deficient diet. The results obtained are as follows. The levels of plasma and hepatic lipid peroxide were increased after chronic ethanol feeding in rats. Retinyl acetate supplementation significantly reduced lipid peroxidation induced by ethanol feeding Glucose 6-phosphatase activity was significantly reduced in rats fed vitamin A-deficient diet with ethanol and alkaline phosphatase activity was significantly induced in rats fed 13-cis-reinoic acid diet with ethanol. Catalase and alcohol dehydrogenase activities did not show a consistent tendency in experiment groups. The hepatic antioxidant enzyme activities did not significantly changed by chronic ethanol feeding groups. The striking decrease in conversion of $\beta$-carotene to retinol was observed in rats fed a $\beta$-carotene diet with ethanol feeding The level of retinol and retinoic acid in plasma and liver was decreased after chronic ethanol administration Based on this result, these data suggest that ethanol feeding enhances oxidative stress especially in those fed a vitamin A-deficient diet, and vitamin A supplementation, especially, retinyl acetate intake can prevent enhanced lipid peroxidation and related damage to some extent.

Effects of Mulberry Leaf Extract Feeding on Lipid Status of Rats Fed High Cholesterol Diets (뽕잎추출물이 고콜레스테롤 식이 흰쥐의 지질대사에 미치는 영향)

  • This study was to investigate the effect of mulberry leaf water extract on lipid metabolism of rats fed high cholesterol diets. Sprague-Dawley male rats were randomly assigned to two normal groups; mulberry extract-free (N), 0.16% mulberry leaf extract (NM) groups and high cholesterol groups with four different levels of mulberry leaf extract; 0% (HC), 0.08% (HCL), 0.16% (HCM), and 0.32% mulberry leaf extract (HCH) groups. Serum levels of triglyceride, cholesterol, LDL-cholesterol and AI index in mulberry leaf extract supplemented groups were significantly lower than the HC group (p<0.05). The level of HDL-cholesterol in the HC group was significantly (p<0.05) reduced, compared with N group, but it was increased by mulberry leaf extract supplementation. Mulberry leaf extract had no effect on the UDP-glucuronyl transferase activity. Also, there was no significant difference in the level of liver cholesterol between the HC and mulberry leaf extract supplemented groups, while there was significant difference in the levels of liver total lipid and triglyceride. The HCM and HCH groups had more significant reduction in the activity of lipoprotein lipase in epididymal fat tissue than the HC group. The levels of total lipid, triglyceride and total cholesterol in epididymal fat tissue of HCL, HCM and HCH groups were decreased compared to HC group. The levels of total lipid, triglyceride and total cholesterol in feces from HCL, HCM and HCH groups were higher than those of HC group.

Concentrations of PBDE Congeners in Breast Milk and Predictors of Exposure in Seoul Residents (서울 거주 산모 모유 중 PBDEs 이성질체 농도 및 노출 요인에 관한 연구)

  • Objectives: This study was designed to determine the levels of polybrominated diphenyl ethers (PBDEs) in breast milk and to evaluate the relations with factors affecting these levels. Methods: The congener levels of PBDE in 22 samples of breast milk were analyzed using a high resolution gas chromatograph with a high resolution mass detector. In accordance with our standard operating procedures, the recoveries of internal standards had to range between 68% and 118%. Since the distribution of PBDE concentrations is close to log-normal, the data were logarithmically transformed before analysis. Test subjects were healthy primipara and multipara mothers with a mean age of 32 (SD = 2.7) in 2006. Results: Seven PBDE congeners (BDE-28, 47, 99, 100, 153, 154, and 183) were detected and identified in all of the pooled breast milk samples, indicating widespread contamination from PBDEs in the environment in Korea. Residue levels of total PBDEs (sum PBDEs from tri- to hepta-BDE) ranged from 0.84-13.1 ng/g lipid with median and geometric mean levels of 2.6 ng/g lipid and 2.74 ng/g lipid, respectively. PBDE congeners 47, 99 and 153 markedly predominated and accounted for about 75% of the amount of the PBDE congeners analyzed. BDE-47 was the dominant congener in most samples, whereas BDE-153 was predominant in a few (n = 7/22). BDE-47 was highly correlated with total PBDEs (r = 0.987, p < 0.01). In analyses of the differences of the means of log transformed breast milk PBDE levels for groups of potential covariates, only breast milk BDE-47 and BDE-99 levels were significantly associated with fish (p < 0.05) and meat consumption (p < 0.01). However, we did not find significant correlations between PBDE levels and maternal age, body mass index (BMI), parity, job presence and smoking status. Conclusions: Our findings are mainly limited due to the small sampling size and low doses of PBDEs exposure. Background and human exposure data of PBDEs is lacking, and longitudinal investigations into the environment and biota are encouraged to determine the health impact on future populations in Korea.

The Effects of Dietary Patterns and Apolipoprotein E Phenotype on the Blood Lipid Profiles of Individuals from Cheju Area (제주지역 성인의 Apolipoprotein E Phenotype 분포와 식생활 및 혈청지질 농도의 관련인자 연구)

  • The purpose of this study was to determine the relation between serum lipid profiles, apolipoprotein E phenotype, and dietary patterns in a cross-section of healthy individuals from Cheju-Do. Age, gender, anthropometric measurements, blood pressure, dietary consumption, drinking / smoking habits and menopausal status were surveyed. Total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, fasting blood glucose, and insulin levels were measured from overnight fasting blood. The study involved a total of 286 individuals(147 men and 139 women) between the ages of 20 and 60 years old. All of the subjects were recruited from a population of healthy individuals living in Cheju-Do. The results of the study are as follows : 1) Among the males, those in their 20's had the maximum food intake, while those in their 40's had the minimum food intake. For the females, food intake was the highest for those in their 30's. Energy and nutrient intakes were directly proportional to the amount of food intake. Men in their 30's were heavier than other men and women in their 40's were heavier than other women. The activity index for men in their 20's and 30's appeared to be lower than that of men above 40. The activity index of women in their 20's appeared to be lowest among all aged groups, and the index appeared to increase from the age of 30 onwards. 2) In terms of changes In serum constituents with age, men in their 40's appeared to have the highest levels of serum constituents such as lipids, glucose, and insulin. Men in their 50's showed the highest levels of serum LDL-cholesterol and glucose. Men in their 30's showed peak levels of serum triglycerides. On the other hand, women in their 50's appeared to have peak levels of serum total cholesterol, LDL-cholesterol, and triglycerides. There was no ch:ange with age in HDL-cholesterol and insulin levels for men and women. The percentage of the subjects had the following apo E phenotypes : E3/3, 91.3% ; E3/2, 5.4% ; E4/3, 2.5% ; E4/2, 0.7%. Lee's reserch with Korean female college students showed that the percentage of ApoE3/3, E3/2, E 4/2, E4/3, and E4/4 were 84.8%, 6.7%, 6.7%, 0.9%, 0.9%, respectively. The number of samples with ApoE mutation was so small that there was no statistical significance in the relation between apolipoprotein E phenotype and se겨m lipids. 3) To investigate the relati onship between weight and serum constituents, the subjects of this study were divided into three groups by BMI underweight, normal weight, and overweight. The serum constituents of men and women below the age 40 in the overweight groups belonged to the normal domain. On the other hand, serum cholesterol levels of both men and women above the age 40 in the overweight groups remained in the borderline-high region(above 200mg/dl), and the mean value of LDL-cholesterol(above 130mg/dl) and triglycerides of men were above normal. Fasting blood glucose levels also remained in the borderline-high region. Total cholesterol levels of women above the age 40 in the overweight group was in the borderline-high region.   • A study was conducted to investigate the status of organic phosphorus in soils originated from different parent materials and under different uses(Virgin, rice soil, upland). The results are summarized as a following. 1. Regardless of parent materials and land uses, the contents of different forms of organic phosphorus are in the order of unidentified organic-P>Inositol-P>Sugar-P>Lipid-P. 2. In case of Inositol-p, the contents were in the order of rice soil>upland>virgin soil. 3. The contents of Sugar-P and Lipid-P in cultivated soils are higher than in uncultivated soil. 4. RNA and DNA are not identified in any soil when detected by ultra-violet absorption method. 5. There is highly significant correlation between the content of soil organic matter and that of organic phosphate. However, there is remarkable difference in the pattern of correlation between cultivated soils and uncultivated soils. 6. Comments were made on the factors on plant availability of various organic phosphorus.
