쥐에서 임신기, 수유기 및 이유후에 식이로 섭취한 어유가 뇌조직의 DHA 분포량에 미치는 영향 (Influence of Dietary Supplementation of Fish Oil at Different Life Cycle on the Incorporation of DHA into Brain in Rats)

  • The incorporation of docosahexaenoic acid(DHA) and arachidonic acid(AA) into brain and liver lipid has been compared in male pups from binth to 10 weeks old by feeding DHA-rich experimental diets or chow diets to dams from pregnancy in rats. The experimental DHA-rich diets contained 7g fish oil and 3g corn oil per 100g diet. There were three experimental groups, FO-I : Dams were fed DHA-rich diet during pregnancy and lactation, and their it pups fed the same diet until 10 weeks old. FO-II Dams fed chow diet during pregnancy and DHA-diet during lactation, and their pups fed the same DHA-diet until 10 weeks. FO-III : Dams fed chow diet during gestation and lactation, and then the pups fed DHA-diet after weaning. The relative % of DHA in hepatic lipid was about 12% with chow diets, but increased rapidly to 20-25% level when DHA-rich diets were supplied after weaning. The AA(%) of FO-III group was relatively high when a chow diet containing higher amount of linoleic acid was given, but there was no significant difference between the groups after feeding on a DHA-rich diet. When the DHA-rich diet was supplied from pregnancy(FO-I), the relative % of DHA in brain lipid was 13.7% at birth and continuously increased to a maximum level(17.2%) at 3-weeks and then was sustained until 5 weeks old. Similar levels of DHA incorporation were observed when DHA-rich diet was supplied from lactation(FO-II). However, the pups of FO-III group showed significantly lower levels of DHA incorporation(72%) at birth. These livels slowly increased and reached an 87% level of FO-I at 10 weeks when the pups ate DHA-rich diets after weaning. The relative % of AA in brain lipid was 10.4% in the FO-I group at birth, which was significantly lower than those of other groups, but there was no significant difference between groups after feeding DHA-rich diets in all groups. The Ah(%) level increased to maximum(11-12%) at 3-weeks and then was slightly reduced and was sustained at about 10% after S-weeks. Total amounts of DNA in the whole brain rapidly reached maximum level at 3-weeks and then was sustained at a constant level after S-weeks. DNA content was not significantly different between groups at birth, but it was significantly higher in FO-I and FO-II groups than in FO-III group at 3-weeks. However, DNA content in FO-III group was continuously increased to 80% level of FO-I at 10-weeks after feeding DHA-rich diet since weaning. In conclusion, the DHA(%) in whole brain was most effectively deposited when DHA-rich diet had been supplied during pregnancy and lactation in rats. However, DHA supplementation after weaning also improved the incorporaton of DHA into brain and content of DNA even though brain development was almost completed, which suggests that DHA supplementation might be necessary to improve brain development in humans during infancy as well as pregnancy and lactation. The Influence of Saturated Fats, ${\alpha}-linolenic$ Acid, EPA and DHA on the Lipid Hydroperoxide Level and Fatty Acid Composition in Liver Microsomes and in Plasma Lipid of Rabbits

  • To investigate the influence of saturated fats, ${\alpha}-linolenic$ acid, EPA and DHA on the lipid hydroperoxide concentration and fatty acid composition in liver microsomes and in plasma lipid of rabbits, the animals were fed on the perilla oil rich ${\alpha}-linolenic$ acid or sardine oil rich EPA and DHA diet for four weeks Were examined. The fatty acid composition of plasma lipid and liver microsomes of rabbits fed on the perilla oil diet was an accumulation of arachidonic acid(AA) 20:4 n-6, eicosapentaenoic acid(EPA) 20:5 n-3, and docosahexaenoic acid(DHA) 22:6 n-3, The fatty acid composition of plasma lipid and liver microsomes of rabbits fed on the sardine oil was an accumulation of ${\alpha}-linolenic$ acid(LNA) 18:3 n-3, and arachidonic acid(AA) 20:4. The p/s ratio of rabbits fed on the perilla oil diet changed from 7.4 to 2.27 for plasma lipid and 2.47 for liver microsomes. The concentration of lipid hydroperoxide was 3.48 nmol MDA/ml and 4.35 nmol MDA/ml for plasma lipid and liver microsomes, respectively, in perilla oil diet. The lipid hydroperoxide liver was 4.22 nmol MDA/ml and 67 nmol MDA/ml for plasma lipid and liver microsornes in sardine oil diet.

호박 물 추출물이 부종 및 고지혈증에 미치는 영향 (Effects of the Water Extract from Cucurbita maxima Duchesne on Inflammation and Hyperlipidemia in Rats)

  • 민간약으로 사용되는 당호박의 물 추출물에 대하여 실험한 결과 500mg/kg 투여군에서 carrageenan에 의한 부종에 대하여는 42.1%의 억제효과를 보였고, 고지방 식이에 의해 상승된 혈중 cholesterol과 triglyceride의 함량을 각각 20.1%와 35.3% 감소시킨 반면 HDL-cholesterol의 함량을 43.1% 증가시킴으로 고지혈증에 대하여 유의성 있는 억제효과를 나타내어 당호박의 부종 및 고지혈증의 치료에 대한 약용가치는 충분하다고 사료된다.

Effects of dietary lipid level and source in fishmeal-based diet on growth and body composition of grower sunshine bass, Morone chrysops $\times$ M. saxatilis raised in seawater

  • Effects of dietary lipid level and source (squid liver oil being rich in n-3 HUFA, soybean oil being rich in 18:2n-6, and linseed oil being rich in 18:3n-3) in fishmeal-based diet on growth and body composition of grower sunshine bass raised in seawater were investigated. Fifteen grower (an initial weight of 146.8$\pm$0.23 g) sunshine bass were randomly distributed into 27 of 250 L fiber reinforced plastic flow-through tanks. Fish were hand-fed to satiety twice daily for 6 days a week throughout the feeding trial. Survival was over 97% and not significantly affected by either dietary lipid level or lipid source (n-3 highly unusaturated fatty acid, HUFA). Weight gain of fish tended to improve with dietary n-3 HUFA level up to 2.9%, but sharply decreased at 3.5%. The best weight gain was obtained in fish fed the diet supplemented with 6% squid liver oil and 3% soybean oil. FER and PER were not significantly affected by either dietary lipid level or dietary lipid source. The lowest moisture content of the whole body was observed in fish fed the diet supplemented with 12% squid liver oil and highest for the diet supplemented with 9% linseed oil, respectively. Protein content of fish was not significantly affected by either dietary lipid level or dietary lipid source. However, lipid content of the whole fish tended to increase with an increase of either dietary lipid level or dietary n-3 HUFA level, except for fish fed the diet supplemented with 9% linseed oil. Ash content of fish fed the diet with no supplementation of oil was highest and lowest for the diet supplemented with 9% soybean oil, respectively. Significant differences in saturated fatty acids (16:0, 18:0 and 24:0), monoene (18:1n-9), 18:2n-6, 20:5n-3 and sum of n-3 HFUA of fish were observed. In considering these results, it could be concluded that supplementation of 9% oil combined with 6% squid liver oil and 3% soybean oil into fishmeal-based diet was the most recommendable for growth of grower sunshine bass raised in seawater.

  • The effects of the water extract from the stem bark of Acanthopanax senticosus were evaluated on hyperlipidemic rats induced by lipid rich diet or poloxamer-407. The water extracts, when administered orally for 3 consecutive days in hyperlipidemic rats induced by poloxamer-407 (1 ml of 30%), was found to cause a significant decrease in plasma cholesterol and triglyceride concentrations. The water extracts, when treated orally for 5 consecutive days also showed a significant inhibition of serum total cholesterol and triglyceride level in rats treated with lipid rich diet (15% cholesterol, 1% sodium cholate and 84% com oil). HDL-cholesterol, however, was increased significantly.

Effect of Different Vegetable Oils on Growth and Fatty Acid Profile of Rohu (Labeo rohita, Hamilton); Evaluation of a Return Fish Oil Diet to Restore Human Cardio-protective Fatty Acids

  • Two experiments in the sequential order were conducted to determine the effects of different dietary lipid sources on the growth and fatty acid composition of rohu (Labeo rohita) and to examine the viability of a return fish oil finisher diet in restoring the human cardio-protective fatty acid profile. In the first experiment, fish were fed either with coconut oil (D1), olive oil (D2), sunflower oil (D3), linseed oil (D4) and fish oil (D5) as the main lipid source in the isonitrogenous diet for 90 days. No significant differences in growth were observed. Among the experimental diets moisture content of fish varied significantly (p<0.05) between the groups. Dietary lipid sources had a profound influence on the fatty acid profile of the muscle and liver as tissue fatty acid profile reflected the dietary fatty acid composition. Increased amounts of eicosapentaenoic acid and docosahexaenoic acid were observed in tissue of fish fed D4 and arachidonic acid was observed in the tissue of fish fed D3. We have also detected the metabolites of n-3 and n-6 pathway in D4 and D3 groups respectively, which prompted us to conclude that rohu, can desaturate and elongate $C_{18}$ essential fatty acids to $C_{20}$ and $C_{22}$ HUFA. A second feeding trial was conducted using the animals from the five different treatment groups for the duration of 30 days with fish oil rich diet (D5). Feeding with fish-oil rich washout diet resulted in the near equalization of all the other treatment groups tissue fatty acid profiles to that of fish oil (D5) fed group. These results indicate that a finishing fish oil diet can be effectively used to restore the human cardioprotective fatty acid profile in rohu fed with vegetable oils as lipid source.

Effect of Dietary Lipid Sources on Body Fatty Acid Composition of Chinese Longsnout Catfish Leiocassis longirostris

  • We investigated the effects of dietary lipid sources on growth and fatty acid composition of juvenile Chinese longsnout catfish. Triplicate groups of fish (initial average weight, 3.8 g) were fed four diets containing either fish oil (FO), soybean oil (SO), linseed oil (LO) and lauric acid (LA) for 10 weeks. There were no differences among the groups in body weight, feed intake, feed efficiency, protein efficiency ratio, and body proximate composition of fish fed the diets containing different lipid sources (P > 0.05) during the study. However, fatty acids compositions of the whole body were influenced by dietary lipid source. Fish fed the SO diet had high concentration of linoleic acid, whereas those of fish fed the LO diet were rich in linolenic acid and arachidonic acid. Fish fed the FO diet had significantly (P < 0.05) higher levels of monounsaturated fatty acids such as 18:1n-9 and 20:1n-9 than those of fish fed the SO and LO diets. Eicosapentaenoic acid (EPA; 20:5n-3) and docosahexaenoic acid (DHA; 22:6n-3) composition of body were not influenced by dietary lipid source. The results suggest that each of FO, SO, LO or LA can be used as a lipid source in the diets of Chinese longsnout catfish without any negative effects on growth and feed utilization and these data demonstrate the potential impact which dietary fat composition can change the body fatty acid profile.

식이지방의 종류와 수준에 따라 쥐의 혈장과 조직의 Tocopherol 및 지질과산화상태에 미치는 영향 (Effect of Quality and Quantity of Dietary Fats on the Status of Tocopherol and Lipid Peroxidation of Plasma and Tissue in Rats)

  • The study was to compare the effect of dietary fatty acids on fatty acid profile in tissue and the status of tocopherol and lipid peroxidation, and superoxide dismutase and glutathione peroxidase activities at two fat levels. Male Sprague Dawley rats weighing average 350g(17 weeks) were fed either low fat(LF, 4.3% w/w, 10% kcal) or high fat(HF, 20.8%, w/w, 40% kcal)diet for 6 weeks. The fats used were beef tallow as a source of saturated fatty acid, corn oil for n-6 linoleic acid, perilla oil for n-3 $\alpha$-linolenic acid and fish oil for n-3 eiocosapentatenoic acid(EPA) and n-3 docosahexaenoic acid(DHA). Palsma tocopherol was significantly reduced by fish oil compared to beef tallow at body fat level. However, there was no significant effect on the levels of plasma MDA, RBC MDA and tocopherol, and RBC hempolysis by the type and amount of dietary fat. The peroxidizibility index of fatty acid profile in plasma and liver was increased and liver MDA level was significantly increased by fish oil when dietary fat level was increased. The activities of SOD and GSHPx tended to be increased by perilla oil and fish oil at both fat oil significantly reduced the incorpration of c20:4 and increased the incorporation of c20:5 into liver compared to corn oil. The incorporation of n-3 fatty acids into tissue by perilla oil rich in $\alpha$-linolenic acid was significantly higher tan corn oil and its effect was improved with higher amount of perilla oil in diet by high fat diet. Overall, the lipid peroxidation of tissue could be prevented by tocopherol supplementation when dietary fat level was low in diet. However, at high fat diet, tocopherol supplementation might not be enough to prevent the lipid peroxidation in tissue since the potential for lipid peroxidation was tended to be increased with higher incorporation of higher unsaturated n-3 fatty acids into tissue. Therefore, it could not be recommended to consume large amount of fish oil even with excess amount of tocopherol supplemented to the high fat diet.

조피볼락의 사료원으로 갈색거저리의 영양적 가치 및 소화율 평가 (Nutritional Value and Digestibility of Tenebrio molitor As a Feed Ingredient for Rockfish (Sebastes schlegeli))

  • Tenebrio molitor larvae, also known as yellow mealworms (MW), are rich in protein and lipid and can serve as a potential alternative protein and energy source in commercial aquafeeds. Therefore, this study attempts to evaluate the effects of different drying methods on the nutritional value of MW meal. For this, live MW were cold-anaesthetized before being subjected to three different types of drying methods, including freeze-drying, oven-drying at $60^{\circ}C$ and air-drying at room temperature for three days, and compared for proximate composition and energy content. An in-vivo digestibility test was then conducted to evaluate the nutrient digestibility of MW meal in diets for rockfish, Sebastes schlegeli. A test diet was prepared by mixing the MW meal with a reference diet (Ref) in a 30:70 ratio with chromium oxide as an inert marker at the inclusion level of 0.5%. Rockfish with mean body weight of 150 g were stocked into a fecal collection system equipped with fiberglass tanks of 400 L capacity. Each group of fish was fed one of the experimental diets to apparent satiation for 4 weeks. The results of the proximate analysis showed that drying methods had no significant effect on crude protein, crude lipid, ash and energy contents of MW. Despite being a rich source of protein and lipid, MW meal was deficient in certain amino acids, particularly methionine, and highly unsaturated fatty acids, particularly 22:6n-3 (DHA) and 20:5n-3 (EPA). MW meal showed high digestibility values for protein (93%), lipid (97%) and energy (88%). These results may indicate that MW meal is a nutritious and acceptable feed ingredient, with comparable digestibility values to conventional animal and plant feedstuffs such as fish meal and soybean meal, in practical diet for rockfish at grower stage.

빨간배추가 고지방식이를 급여한 렛트의 장기무게 및 혈청지질 수치에 미치는 영향 (Effect of Red Chinese Cabbage on the Organ Weight and Serum Lipid Levels of Rats Fed High Fat Diet)

  • Red Chinese cabbage is rich in anthocyanins, with beneficial effects on anti-obesity and lipid profiles. The purpose of this study was to examine the anti-obesity effects of red Chinese cabbage. Four-weeks old Spague-Dawley male rats were divided into four groups for six weeks as follow: Normal Diet (ND), High Fat Diet (HFD), HFD + Red Chinese Cabbage Diet (RCD), HFD + Red Chinese Cabbage Juice (RCJD). After six weeks, the liver weight was significantly higher in the HFD group than in the other groups, and the RCD and RCJD groups showed similar values to the ND group. The epididymal fats were significantly higher in all high fat diets (HFD, RCD, RCJD) than in the ND group and there was statistical significance in the post-tests when comparing the RCD and RCJD groups with the HFD group. Triglycerides significantly decreased in the RCD group. In conclusion, red Chinese cabbage is rich in anthocyanins, with beneficial effects on organ weight and lipid profiles.
