Comparison and Evaluation on the Chemical Constituents of Progeny in T-DNA Inserted Rice

  • With the development of diverse agricultures worldwide, biofortified rice noted for its preferable marketability and palatability plays an important role in the world's agricultural economics and rice breeding programs. In this report, several $M_5$ of T-DNA inserted lines derived from the donor cultivars, 'Hwayong' and 'Dongjin', were selected for high or low protein, high lipid and low amylose content, respectively. The coefficients and ranges of variation for the chemical constituents between $M_4$ and $M_5$ T-DNA inserted lines were evaluated in comparison with those of the donor varieties. Results indicated that T-DNA insertion might be an effective way to generate useful variations for chemical composition of rice grains which could be used for the development of biofortified rice cultivars.

  • Twelve compounds with lipid peroxidation inhibitory activity were isolated from the stem bark of E. globulus. Their structures were assigned as a new aromatic monoterpene (1) and eleven known compounds, pinoresinol (2), vomifoliol (3), 3,4,5-trimethoxyphenol 1-O-$\beta$-D-(6'-O-galloyl)glucopyranoside (4), methyl gallate (5), rhamnazin (6), rhamnetin (7), eriodictyol (8), quercetin (9), taxifolin (10), engelitin (11), and catechin (12) on the basis of UV, mass, and NMR spectroscopic analyses. These compounds except vomifoliol significantly inhibited lipid peroxidation in rat liver microsome.

Effect of Diazinon, an Organophosphate Insecticide, on Plasma Lipid Constituents in Experimental Animals

  • There has been increasing interest in studying the various effects of organophosphate insecticides in humans and experimental animals. Only a few data are available on the effect of the organophosphate insecticide, diazinon, on lipid metabolism. The aim of this study was to evaluate the effect of diazinon on plasma lipid constituents in mammalian animals. The plasma levels of total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), triglycerides (TG), and phospholipids (PL) were measured in albino rats that were orally treated with a single dose of diazinon at a level of $LD_{50}$ or with repeated daily doses at the levels of $\frac{1}{2}$, $\frac{1}{8}$, and $\frac{1}{32}$ $LD_{50}$ for 2, 8, and 32 days, respectively. After a 24 h post-treatment with a single $LD_{50}$ dose of diazinon, TC was not significantly changed, the HDL-C and PL levels were significantly decreased, but the LDL-C and TG levels were significantly increased. Separate daily oral administrations of diazinon at $\frac{1}{2}$ $LD_{50}$, $\frac{1}{8}$ $LD_{50}$, and $\frac{1}{32}$ $LD_{50}$ doses resulted in a significant decrease in HDL-C and PL, with no significant change in TG. The LDL-C levels were significantly increased and TC showed no significant change with $\frac{1}{2}$ $LD_{50}$ and $\frac{1}{32}$$LD_{50}$ doses of diazinon, whereas a significant decrease in the levels of TC, HDL-C, as well as LDL-C, was observed with the $\frac{1}{8}$ $LD_{50}$ dose. These data suggest that diazinon may interfere with lipid metabolism in mammals.

The Effects of Dietary Patterns and Apolipoprotein E Phenotype on the Blood Lipid Profiles of Individuals from Cheju Area (제주지역 성인의 Apolipoprotein E Phenotype 분포와 식생활 및 혈청지질 농도의 관련인자 연구)

  • The purpose of this study was to determine the relation between serum lipid profiles, apolipoprotein E phenotype, and dietary patterns in a cross-section of healthy individuals from Cheju-Do. Age, gender, anthropometric measurements, blood pressure, dietary consumption, drinking / smoking habits and menopausal status were surveyed. Total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, fasting blood glucose, and insulin levels were measured from overnight fasting blood. The study involved a total of 286 individuals(147 men and 139 women) between the ages of 20 and 60 years old. All of the subjects were recruited from a population of healthy individuals living in Cheju-Do. The results of the study are as follows : 1) Among the males, those in their 20's had the maximum food intake, while those in their 40's had the minimum food intake. For the females, food intake was the highest for those in their 30's. Energy and nutrient intakes were directly proportional to the amount of food intake. Men in their 30's were heavier than other men and women in their 40's were heavier than other women. The activity index for men in their 20's and 30's appeared to be lower than that of men above 40. The activity index of women in their 20's appeared to be lowest among all aged groups, and the index appeared to increase from the age of 30 onwards. 2) In terms of changes In serum constituents with age, men in their 40's appeared to have the highest levels of serum constituents such as lipids, glucose, and insulin. Men in their 50's showed the highest levels of serum LDL-cholesterol and glucose. Men in their 30's showed peak levels of serum triglycerides. On the other hand, women in their 50's appeared to have peak levels of serum total cholesterol, LDL-cholesterol, and triglycerides. There was no ch:ange with age in HDL-cholesterol and insulin levels for men and women. The percentage of the subjects had the following apo E phenotypes : E3/3, 91.3% ; E3/2, 5.4% ; E4/3, 2.5% ; E4/2, 0.7%. Lee's reserch with Korean female college students showed that the percentage of ApoE3/3, E3/2, E 4/2, E4/3, and E4/4 were 84.8%, 6.7%, 6.7%, 0.9%, 0.9%, respectively. The number of samples with ApoE mutation was so small that there was no statistical significance in the relation between apolipoprotein E phenotype and se겨m lipids. 3) To investigate the relati onship between weight and serum constituents, the subjects of this study were divided into three groups by BMI underweight, normal weight, and overweight. The serum constituents of men and women below the age 40 in the overweight groups belonged to the normal domain. On the other hand, serum cholesterol levels of both men and women above the age 40 in the overweight groups remained in the borderline-high region(above 200mg/dl), and the mean value of LDL-cholesterol(above 130mg/dl) and triglycerides of men were above normal. Fasting blood glucose levels also remained in the borderline-high region. Total cholesterol levels of women above the age 40 in the overweight group was in the borderline-high region.   • Herbal medicines have been used since the dawn of civilization to maintain health and to treat diseases. Diabetes mellitus is one of the leading cause of death in many developed countries. The incidence of diabetes is increasing at an alarming rate in India. It was estimated that India which had 19.4 million diabetes in 1995 is expected to register a near threefold increase by. Many plants reported to be useful for the treatment of diabetes mellitus in ayurvedic medicine, are being tested for their hypoglycemic activity in experimental animals Lagerstroemia flos- reginae is one such plant commonly found as shade trees in Kerala. In Ayurveda both root and leaves are used in the treatment of diabetes. The main objective of this study was to assess the antidiabetic effect of the alcohol extracted leaves of Lagerstroemia flos- reginae in alloxan induced diabetic rats in terms of controlling blood glucose level, lipid profile, bilirubin, uric acid in serum and lipid peroxides and glutathione in the liver of the experimental animals. The present study has been undertaken to observe the protective effect of the active constituents of Lagerstroemia flos- reginae leaf extracts against alloxan induced diabetes in experimental animal model. The activity of the active constituents was compared with Daonil -a standard drug.

Protective Effects of Jisilhaebaekgyeji-tang and Constituents Extract on Cultured Rat Myocardial Cell treated by XO/HX (XO/HX에 의해 손상된 배양 심근세포에 대한 지실해백계지탕과 구성약물 추출물의 방어효과)

  • To certify the protective effect of herbal medicine against oxygen free radical-induced myocardiotoxicity, cytotoxicity was measured using LDH activity and TBARS assay in the presence of Jisilhaebaekgyejitang(JHGT) extracts or single constituents of this prescription, In the present study, xanthine oxidase/hypoxanthine (XO/HX) resulted in a cell damage such as increases in LDH activity in culture medium and lipid peroxidation in cultured myocardial cells. In the effect of JHGT extract and its single constituents, which are Fructus Ponciri Seu Aurantii Immaturus (FPSAI), Cortex Magnoliae Officinalis (CMO), Bulbus Allii Macrostemi (BAM), Ramulus Cinnamomi (RC) and Fructus Trichosanthis (FT), they showed the prevention from the XO/HX-induced cardiotoxicity by the decrease of LDH activity and lipid peroxidation. From these results, they show that XO/HX is cardiotoxic in cultured myocardial cells derived from neonatal rat, and it suggests that JHGT, FPSAI, PT, CMO, BAM, RC and FT extracts are positively effective in the blocking in XO/HX-induced cardiotoxicity.

Antioxidant Constituents from the Stem of Sorghum bicolor

  • The EtOAc soluble fraction from the stem of Sorghum bicolor showed a strong free radical scavenging activity. Five major compounds were isolated from this fraction. They were identified by spectral data as methyl ferulate (1), methyl p-hydroxycinnamate (2), p-hydroxybenzaldehyde (3), tricin (4), and quercetin 3,4 -dimethyl ether (5). Among these compounds, 1 exhibited a strong, free radical scavenging activity on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) with an $IC_50$ value of 0.7 $\mu$M. We further studied the effects of these isolated compounds on the lipid peroxidation in rat liver microsomes induced by non-enzymatic method. All five compounds showed anti-lipid peroxidation activity ($IC_50$ values of 0.5, 0.4, 0.3 and 0.3 $\mu$ M, respectively).

  • The EtOAc and n-BuOH soluble fractions from the aerial part of Setaria viridis showed a strong free radical scavenging activity. Six major compounds were isolated from these fractions. They were identified by spectral data as tricin (1), p-hydroxycinnamic acid (2), vitexin 2"-Ο-xyloside (3), orientin 2"-Ο-xyloside (4), $tricin-7-Ο-{\beta}-D-glucoside$ (5) and vitexin 2"-Ο-glucoside (6). Among these compounds, 4 and 5 exhibited strong free radical scavenging activities on 1, 1-diphenyl-2-picrylhydrazyl (DPPH). We further studied the effects of these isolated compounds on the lipid peroxidation in rat liver microsomes induced by non-enzymatic method. As expected, 4 and 5 exhibited significant inhibition on $ascorbic/Fe^{2-}$ induced lipid peroxidation in rat liver microsomes.ver microsomes.

Changes of Lipids in Chestnut during Storage (밤의 저장중 지방질 조성의 변화)

  • Studies were carried out to investigate the changes of lipids in chestnut (Castanea crenata Sieb. et Zucc) during storage at $20^{\circ}C$ for 9 weeks and $1^{\circ}C$ for 15 weeks. Lipid composition of fresh chestnuts were 43.4% of nonpolar lipid (NPL), 26.5% of glycolipid (GL) and 30.1% of phospholipid (PL) in total lipid (TL). Nonpolar lipid was composed of triglyceride (TG), 1,2-diglyceride (1,2-DG), 1,3-diglyceride (1,3-DG), sterolester (SE), sterol (S) and free fatty acid (FFA). Main constituents of glycolipid were digalactosyldiglyceride (DGDG), monogalactosyldiglyceride (MGDG), sterylglycoside (SG), and acylsteryglycoside (ASG). Main constituents of phospholipid were phosphatidylcholine (PC), phosphatidylethanolamine (PE) and phosphatidylinositol (PI). The content of nonpolar lipid was decreased after 5 weeks of storage, and glycolipid content was increased during 7 weeks and then decreased. Phospholipid was the most increased lipid during storage at $20^{\circ}C$ and $1^{\circ}C$. The contents of TG, SE, S, DGDG and MGDG were increased during storage at $20^{\circ}C$ and $1^{\circ}C$. The major fatty acids were linoleic acid, oleic acid, palmitic acid and linolenic acid in TL, NPL, GL, PL, TG, DG, ASG, PC and PE. The content of palmitic acid was relatively higher in the PL. Linoleic acid among them was increased during storage at $20^{\circ}C$ and $1^{\circ}C$, while oleic acid was decreased.
