Preferential Peroxidase Activity of Prostaglandin Endoperoxide H Synthase for Lipid Peroxides

  • Prostaglandin endoperoxide H synthase (PGHS) catalyzes the committed step in prostaglandins and thromboxane A$_2$-- oxygenation of arachidonic acid to the hydroperoxy endoperoxide PGG$_2$, followed by reduction PGG$_2$to the alcohol PGH$_2$. The two reactions by PGHS -- cyclooxygenase and peroxidase -- occur at distinct but structurally and functionally interconnected sites. The peroxidase reaction occurs at a heme-containing active site located near the protein surface. The cyclooxygenase reaction occurs in a hydrophobic channel in the core of the enzyme. Initially a peroxide reacts with the heme group, yielding Compound I and an alcohol derived from the oxidizing peroxide. Compound I next undergoes an intramolecular reduction by a single electron traveling from Tyr385 along the peptide chain to the proximal heme ligand, His388, and finally to the heme group. Following the binding of arachidonic acid, Tyr385 tyrosyl radical initiates the cyclooxygenase reaction by abstracting the 13-pro(5) hydrogen atom to give an arachidonyl radical, which sequentially reacts with two molecules of oxygen to yield PGG$_2$. In order to characterize PGHS peroxidase active site, we examined various lipid peroxides with purified recombinant ovine PGHS proteins and determined the rate constants. The results have shown that twenty-carbon unsaturated fatty acid hydroperoxides have similar efficiency in peroxidation by PGHS, irrespective of either the location of hydroperoxy group or the number of double bonds. It was also confirmed by the subsequent study with PGHS peroxidase active site mutants.

솔잎 추출물의 항산화 효과 (Antioxidative Effects of Pine (Pinus denstifora) Needle Extracts.)

  • 식물성 성분 중에서 생리활성 인자를 탐색할 목적으로 솔잎의 수용성 추출물에 의한 항산화 활성을 DPPH법, thiocyanate법, TBA법 및 microsome 생 체 막 지 질 과산화물 생성정도를 TBARS법으로 측정 하고, 총 폴리페놀 함량을 측정하였다. 솔잎 수용성 추출물의 총 폴리페놀 화합물 함량은 1.61%이었으며, DPPH 측정법에서는 짙은 자색의 탈색되는 정도로 나타내는 전자 공여능이 솔잎 수용성 추출물의 0.1% 농도에서 BHT와 비슷한 수준으로 항산화 활성이 높게 나타났다. 간 microsome을 이용한 생체막 지질 과산화 억제정도는 솔잎 수용성 추출물의 0.1% (67.7%)>0.05% (63.1%)>0.01%(28.2%) 순으로 농도에 비례하여 증가하였다. Linoleic acid 산화 실험계를 이용한 thiocyanate법에서는 0.01%, 0.05%, 0.1%의 농도에서 모두 반응 7일째까지 대조구에 비해 매우 강한 항산화 활성을 보였으며, TBA법에서는 반응 6일째까지 는 0.1%>0.05%>0.01%의 순으로 농도 증가에 따라 항산화 활성도 높게 나타났다. 이상의 결과에서 솔잎 수용성 추출물 중에는 in vitro 항산화 실험에서 항산화 활성을 나타내는 생리활성 성분이 존재하는 것으로 볼 수 있으므로 천연 항산화제로서의 가능성을 시사하였다.

Isolation of a Natural Antioxidant, Dehydrozingerone from Zingiber officinale and Synthesis of lts Analogues for Recognition of Effective Antioxidant and Antityrosinase Agents

  • In the present study, the antioxidative and inhibitory activity of Zingiber officinale Rosc. Rhizomes-derived materials (on mushroom tyrosinase) were evaluated. The bioactive co mponents of Z. officinale rhizomes were characterized by spectroscopic analysis as zingerone and dehydrozingerone, which exhibited potent antioxidant and tyrosinase inhibition activities. A series of substituted dehydrozingerones [(E)-4-phenyl-3-buten-2-ones] were prepared in admirable yields by the reaction of appropriate benzaldehydes with acetone and the products were evaluated in terms of variation in the dehydrozingerone structure. The synthetic analogues were examined for their antioxidant and antityrosinase activities to probe the most potent analogue. Compound 26 inhibited Fe$^{2+}$-induced lipid peroxidation in rat brain homogenate with an IC$_{50}$ = 6.3${\pm}$0.4 ${\mu}$M. In the 1,1-diphenyl- 2-picrylhydrazyl (DPPH) radical quencher assay, compounds 2, 7, 17, 26, 28, and 29 showed radical scavenging activity equal to or higher than those of the standard antioxidants, like ${\alpha}$-tocopherol and ascorbic acid. Compound 27 displayed superior inhibition of tyrosinase activity relative to other examined analogues. Compounds 2, 17, and 26 exhibited non-competitive inhibition against oxidation of 3,4- dihydroxyphenylalanine (L-DOPA). From the present study, it was observed that both number and position of hydroxyl groups on aromatic ring and a double bond between C-3 and C-4 played a critical role in exerting the antioxidant and antityrosinase activity.

Physical, chemical composition and umami compound of dried immature and mature roes of skipjack tuna (Katsuwonus pelamis)

  • In this study we investigate physical and chemical characteristics of immature and mature skipjack tuna (Katsuwonus pelamis) roes in fresh and dried forms. Fresh roes were studied for histological structure and also dried by three methods: hot air drying (HD), vacuum drying (VD) and freeze drying (FD). The obtained roe powders were analysed for proximate composition, color value, fatty acid composition, amino acid profile, equivalent umami concentration (EUC) and protein pattern. Unyolked oocytes were more common in immature roes, while fully yolked oocytes were more common in mature roes. All dried tuna roes contained high content of protein and lipid (69.31%-70.55% and 11.14%-16.02%, respectively). The powders obtained by FD provided the highest lightness value (L*). The main fatty acid found in all roe powders was docosahexaenoic acid (DHA) (23.49%-27.02%). Glutamic acid, leucine, and aspartic acid were the three most abundant amino acids found in the powders (13.58-14.61, 8.06-8.42, and 7.81-8.39 g/100 g of protein, respectively). The mature roe powder obtained from HD provided the highest EUC value (73.09 g monosodium glutamate/100 g of samples). The protein band at molecular weight of 97 kDa (vitelline) represented the major protein. Therefore, dried tuna roe could be a functional ingredient source of protein and lipid rich in DHA and it also has potential to be used as taste enhancer with umami compound.

유색미와 향미 품종의 지방질함량 및 지방질의 조성 (Contents of Total Lipids and Their Composition in Colored and Aromatic Rices Cultivars)

  • 유색미인 흑진주벼 등 5종, 일반미인 동진벼, 향미인 향남벼을 대상으로 이들 품종의 현미 중에 함유된 지방질의 함량과 지방질의 조성을 비교 연구하였다. 1. 총지방질 함량은 건물기준으로 2.04 - 4.68%로 Tohoku 149호, 한산흑미, 상해향혈라 순으로 많았고 흑남벼, 향남벼와 동진벼에서는 비교적 적었으며 그들간에는 차이가 없었다. 2. 총지방질의 조성은 sterol ester & hydrocarbon, triglyceride, free fatty acid, diglyceride, free sterol 등 5종이 동정되었다. 그중 triglyceride의 비율이 가장 높았고 그 다음으로 diglyceride 와 free sterol의 비율이 높았다. 일반미와 유색미 및 향미간에 비교했을때 triglyceride의 비율은 향미와 유색미가 일반미보다 낮은 반면 diglyceride의 비율은 향미와 유색미에서 일반미인 동진벼에 비해 월등히 높았다. 3. 총지방질 중 지방산 조성은 모든 품종 다같이 linoleic acid, oleic acid 및 palmitic acid가 주된 지방산이었고 linolenic, stearic acid은 소량이 함유되어 있었으며, erucic acid는 일반미인 동진벼에는 함유되어 있지 않았다. 양적으로 많은 지방산을 품종별로 비교해 보면 oleic acid는 한산흑미가 41%로 가장 많았고, 그외 품종은 32-37% 범위이었다. 또한 linoleic adid 향남벼와 상해향혈라는 각각 37%그외 품종은 32 - 36%의 범위이었다.

진피 복합제 복용과 운동이 비만 여성의 체격, 체지방 및 혈중지질에 미치는 영향 (The Effects of Dried Orange Peel Compound intaking and Exercise Training on the Physique, Body Fat, and Concentration of Blood Lipid in Obese Women)

  • The purpose of this research was to investigate the effects of dried orange peel compound intaking and exercise training on the physique, body fat, and concentration bloodlipid in obese women. Ninety-seven volunteers(mean age 21.47 ${\pm}$ 1.47 yrs), all over 30% of body fat, agreed to Participate and were placed in 4 groups: dried orange peel compound intaking and exercise group(n=25), only exercise group(n=25), only dried orange peel compound intaking group(n=24), and control group(n=20). The subjects who joined this project were asked to take dried orange peel compound(2 times/day, 30 ml/time) or participate in exercise program(3 times/week, 70 min/day), depending on particular group for 12 weeks. They were tested on their height, weight, circumference of chest, abdomen, hip, and thigh, body fat%, total-cholesterol(TC), HDL-C, LDL-c, and triglyceride(TG) at the beginning and the end of the 12-weeks program. For data analysis, paired t-tests and ANCOVAs were used. The intaking of dried orange peel compound was influenced on weight, circumference of chest, abdomen, hip, and thigh, body fat%, HDL-C, LDL-C, and TG. The exercise participation had almost same effects on those dependent variables. Furthermore, the dried orange peel compound intaking along with exercise training was the most effective on those dependent variables among them. Therefore, these results indicate that dried orange peel compound intaking, like exercise, may be useful for controling weight, body fat, HDL-C, LDL-C, and TG.

옥수수수염, 들깻잎 및 포도줄기 복합추출물의 UVB 조사 피부 손상 및 Compound 48/80 유도 가려움증 개선 효과 (Improvement Effect of Corn Silk, Perilla Leaf and Grape Stem Extract Mixture against UVB-Induced Skin Damage and Compound 48/80-Induced Pruritus)

  • This study was conducted to evaluate the synergistic protective effects of mixtures of corn silk, perilla leaf and grape stem extract (CPG mixture) against UVB-induced skin damage and compound 48/80-induced pruritus in mice. The results showed that treatment with CPG mixture exhibited much stronger suppressive effect on erythema and melanin index as well as melanin formation than treatment with ascorbic acid (AA) in UVB-irradiated mice. Moreover, the treatment with CPG mixture showed ameliorative effect on immune cell infiltration and collagen fiber destruction in UV-irradiated mice. The treatment with CPG mixture inhibited glutathione (GSH) depletion, lipid peroxidation and production of pro-inflammatory cytokines in UVB-irradiated mice. Furthermore, the treatment with CPG mixture inhibited compound 48/80-induced scratching behavior and histological changes in mice. Taken together, these results indicated that CPG mixture has potentials as functional and therapeutic materials against skin damage and itch-related skin diseases.

L-Carnitine의 수준이 다른 Lysine 제한식이 섭취가 비만유도 성숙쥐의 체중과 지질대사에 미치는 영향 (Effect of Lysine-Limited Diets Containing Different Levels of L-Carnitine on Body Weight and Lipid Metabolism in Obesity-Induced Adult Rats)

  • This study was performed to investigate the effect of lysine-limited diets containing different levels of L-carnitine on body weight and lipid metabolism in obesity-induced adult rats. Eight-month-old male Sprague-Dawley rats (n = 90) were raised for one month with high fat diet (40% fat as calorie) to induce obesity. After induction of obesity, rats weighing 739.5 g were randomly blocked into three groups according to the body weight and raised for eight weeks with control diet (Co), 50% lysine-limited diet (-L), 50% lysine limitation with 0.3% pivalate diet (-L + P). Each of three groups was allotted to 0.0% L-carnitine (0.0% CT), 0.5% L-carnitine (0.5% CT) and 2.5% L-carnitine (2.5% CT) groups, respectively. The levels of AST, ALT, total protein and albumin in plasma were within the normal range. Daily food intake and calorie intake tended to be lower in 2.5% CT groups than those of other groups regardless lysine limitation or pivalate intake. And body weight gain and calorie efficiency ratio (weight gain (g) /calorie intake (100 kcal)) were significantly the lowest in 2.5% CT groups among all experimental groups regardless of lysine limitation or pivalate intake. The weights of perirenal, epididymal fat pads and brown adipose tissue in 2.5% CT groups were significantly lower than 0.0% CT groups. Plasma total lipid, triglyceride, total cholesterol concentrations in all groups were not significant by experimental compound. HDL-cholesterol concentrations in -L + P +2.5% CT group were highest in -L + P groups. Levels of hepatic total lipid, triglyceride and total cholesterol in 2.5% CT groups were tend to be lower those than in 0.0% CT groups regardless of dietary lysine limitation and pivalate intake. Fecal total lipid excretions of 2.5% CT groups were significantly lower than in 0.0% CT groups in all experimental groups. But fecal triglyceride excretions of 2.5% CT groups were significantly higher than 0.0% CT groups regardless of lysine limitation and pivalate. In conclusion, there was no difference on body weight and lipid metabolism by dietary lysine limitation and pivalate intake. And feeding of 2.5% L-carnitine was more effective than feeding of 0.5% L-carnitine and 0.0% L-carnitine in reduction of body weight, body fat and lipid metabolism.

Palmitate처리된 인간 간세포주 HepG2 세포에서 piperine의 지질 축적과 인슐린 저항성 기전에 대한 연구 (Effects of Piperine on Insulin Resistance and Lipid Accumulation in Palmitate-treated HepG2 Cells)

  • 간의 지질 축적과 인슐린 저항성은 비알콜성 지방간 환자에게서 증가한다. Piperine은 후추(Piper nigrum)와 필발(인도산 후추, P. longum)의 주요 성분으로 항암, 항비만, 항 당뇨병, 항염증 및 항산화 등의 생리활성이 보고되었다. 그러나 piperine의 인간 간세포 HepG2 세포에서 지질 축적과 인슐린 저항성의 억제제로서의 연구는 보고된 바가 없다. 본 연구의 목적은 지질 축적 및 인슐린 저항성에 대한 piperine의 효과를 palmitate처리된 HepG2 세포에서 잠재적인 분자 기전을 밝히는 것이다. 그 결과 piperine처리군은 지질 함량을 감소시켰고, 지방 형성 표적 유전자인 SREBP-1c와 FAS의 발현을 억제함으로써 palmitate처리된 세포내 지질 축적을 감소시켰다. 게다가 piperine처리군은 지방산 산화에 관련된 CPT-1과 인산화된 ACC 및 인산화된 IRS-1 (Tyr632)와 Akt의 레벨을 증가시켰다. 또한, piperine처리군은 인산화된 IRS-1 (Ser307)의 레벨을 감소시켰다. 결론적으로 palmitate처리된 HepG2 세포에서 piperine은 SREBP-1와 FAS발현의 감소 및 CPT-1과 ACC 인산화의 증가 및 인산화된 IRS-1(Try632)와 Akt 신호전달 경로를 조절함으로써 지질 축적 및 인슐린 저항성을 개선함을 확인하였다. 따라서 piperine의 지질 축적 및 인슐린 저항성을 예방하는 약물로써 가능성이 제시되었다.

용매침지(溶媒浸漬)에 의하여 추출(抽出)된 쌀알의 유지획분(油脂劃分)의 산화(酸化)에 대(對)하여 (On the Oxidation of Rice Lipid Fractions Extracted from the Whole Grain by Immersion)

  • Hexane 및 ethanol을 이용(利用)하여 쌀알미립(米粒)에서 추출(抽出)한 유지(油脂)을 Folch's법(法)에 의해 정제(精製)하고, 이들을 자외선조사(紫外線照射) 또는 $40^{\circ}C$암소(暗所)에 저장하여 쌀유지(油脂)의 산화양상(酸化樣相)을 경시적(經時的)으로 살펴 본 바, 다음과 같은 결과(結果)를 얻었다. 1. 산소흡수(酸素吸收)에 의한 중량증가현상(重量增加現像)은 자외선조사(紫外線照射)에 의하여, 처리(處理) 3일(日) 이내(以內)에 현저한 중량증가(重量增加) 원래(元來) 시료중량(試料重量)의 0.6% 이상)를 보였으나 , $40^{\circ}C$암소(暗所)에서는 45일(日) 이후(以後)에 비로소 현저한 중량증가(重量增加)가 있었다. 각처리별(各處理別) 유도기간(誘導期間)은 hexane유출유지(抽出油脂)가 자외선조사(紫外線照射)에서는 1일(日), $40^{\circ}C$암소(暗所)에서는 30일(日)이었고, ethanol유출유지(抽出油脂)는 자외선조사紫外線照射에서는 2(日), $40^{\circ}C$암소(暗所)에서는 40일(日)이었으며, hexane유출유지(抽出油脂)가 ethanol유출유지(抽出油脂)보다 그 안정성(安定性)이 다소 낮았다. 한편 각 처리별 변패취는 유도기간(誘導期間)보다 약간 늦게 확인 되었다. 2. 각처리별(各處理別)로 총(總)carbonyl화합물(化合物) 및 malonaldehyde(M.A.)의 변화(變化)를 살펴 본 바, 자외선조사구(紫外線照射區)에서는 carbonyl가(價)가 $2{\sim}4$일(日)째, M.A. 는 7일경(日頃)에 그생성량(生成量)이 각각 정점(頂點)을 이루었다가 다시 감소(減少)되었고,$40^{\circ}C$암소(暗所)에서는 carbonyl가(價) 및 M.T. 생성량(生成量) 공(共)히 55일(日)까지 서서히(徐徐)히 증가(增加)하였다. 3. 전체적으로 보아 hexane유출유지(抽出油脂)가 ethanol유출유지(抽出油脂)보다 산화반응(酸化反應)이 더 빨랐으며 이는 지방산조성(脂肪酸組成) 및 미확인(未確認)된 항산화물질(抗酸化物質)의 존재(存在)에 의한 가능성(可能性)으로 고려된다. 그리고 자외선조사(紫外線照射)는 이미 알려진 바 있지만 유지(油脂)의 산화(酸化)를 급격히 촉진(促進)시키고 있었다. 4. Hexane에 의해 유출(抽出)된 쌀유지(油脂)를 유출직후(油出直後)와 $40^{\circ}C$ 55일간 저장처리(貯藏處理)한 후의 지방산조성(脂肪酸組成)을 살펴본 바 linoleic 및 linolenic acid가 현저히 감소(減少)하였고, 상대적(相對的)으로 oleic, stearic, palmitic acid 등(等)이 증가(增加)하였다.
