In Vitro α-Amylase, α-Glucosidase, Pancreatic Lipase, Xanthine Oxidase Inhibiting Activity of Agaricus bisporus Extracts

  • In this study, the α-amylase inhibitory activity, α-glucosidase inhibitory activity, pancreatic lipase inhibitory activity, and Xanthine Oxidase inhibitory activity of the fruiting body extracts of 5 varieties of Agaricus bisporus (AB) were confirmed. First, the α-amylase inhibitory activity of AB12, AB13, AB18, AB34, and AB40 methanol extracts was lower than that of acarbose, a positive control, in all concentration ranges. The α-glucosidase inhibitory activity of the AB40, AB13, and AB12 methanol extracts at the extract concentration of 1.0 mg/mL was 80.5%, 81.3%, and 78.5%, respectively, similar to that of acarbose, a positive control. The pancreatic lipase inhibitory activity of the methanol extract of Agaricus bisporus fruiting body was significantly lower than that of the positive control orlistat in the concentration range of 50~1.000 (mg/mL). The Xanthine Oxidase inhibitory activity was 0.5~8.0 mg/mL of each extract, which was significantly lower than that of the positive control allopurinol in the same concentration range. However, the Xanthine Oxidase inhibitory activity of AB13 and AB40 at 8.0 mg/mL was about 70%, which was higher than that of other mushrooms. In conclusion, five kinds of Agaricus bisporus fruiting bodies seem to have inhibitory effects on enzymes such as α-amylase, α-glucosidase, pancreatic lipase, and Xanthine Oxidase that degrade starch and protein. In particular, it has an inhibitory effect and a reduction effect on xanthine oxidase that causes gout, so it is expected that it can be developed and used as a food or health supplement with health functional properties through future research.

Separation and Purification of Lipase Inhibitory Peptide from Fermented Milk by Lactobacillus plantarum Q180

  • In this study, we separated and purified lipase inhibitory peptide from fermented milk by Lactobacillus plantarum Q180 with the aim of developing a new functional anti-lipase activity yogurt product. L. plantarum 180 was inoculated into 10% reconstituted skimmed milk and incubated at 37℃ until the pH of the culture reached pH 4.4. The lipase activity was measured using porcine pancreatic lipase. The lipase inhibitory peptides were gradually isolated by ultrafiltration, reversed phase column chromatography (RPC), reversed phase high-performance liquid chromatography (RP-HPLC), and gel permeation high-performance liquid chromatography (GP-HPLC) from the fermented milk by L. plantarum Q180. An ODS-AQ column was used for the RPC, a Vydac C18 column for the RP-HPLC, and a Superdex Peptide HR column for the GP-HPLC. The peptide was composed of Asp, Thr, Ile, Ser, Ala, and Gln, and the anti-lipase activity (IC50) was 2,817 ㎍/mL.

Extraction and Characteristics of Anti-obesity Lipase Inhibitor from Phellinus linteus

  • To develop a potent anti-obesity lipase inhibitor from mushroom, the lipase inhibitory activities of various mushroom extracts were determined. Methanol extracts from Phellinus linteus fruiting body exhibited the highest lipase inhibitory activity (72.8%). The inhibitor was maximally extracted by treatment of a P. linteus fruiting body with 80% methanol at $40^{\circ}C$ for 24 hr. After partial purification by systematic solvent extraction, the inhibitor was stable in the range of $40\sim80^{\circ}C$ and pH 2.0~9.0. In addition to lipase inhibitory activity, the inhibitor showed 59.4% of superoxide dismutase-like activity and 56.3% of acetylcholinesterase inhibitory activity.

제주 동백나무 겨우살이를 첨가한 막걸리의 항산화 및 항비만 활성 (In vitro antioxidant and lipase inhibitory activities of Makgeolli supplemented with Jeju Camellia Mistletoe (Korthalsella japonica Engl.) during fermentation)

  • 본 연구의 목적은 0, 1, 2, 및 4% (w/v)의 제주 동백나무 겨우살이를 첨가하여 발효시킨 막걸리의 함량 및 항산화 활성 그리고 pancreatic lipase 저해 활성을 평가하는 것이다. 총 페놀 및 플라보노이드 함량은 제주 동백나무 겨우살이의 함량에 따라 증가하는 경향을 보였다. 제주 동백나무 겨우살이의 첨가 결과 DPPH, hydrogen peroxide, nitric oxide 및 superoxide onion 라디칼 소거 활성과 환원력은 상당한 증가를 보였다. 또한, pancreatic lipase 저해 활성은 제주 동백나무 겨우살이 첨가에 의해 유의하게 증가하였다. 이러한 결과로 보아 제주 동백나무 겨우살이가 막걸리의 항산화 및 pancreatic lipase 저해 활성을 향상시키는 물질로 사료된다.

Eisenia bicyclis 에탄올 추출물로부터 분리한 Dieckol의 Lipase 저해 Mode (Lipase Inhibitory Mode of Dieckol Isolated from Eisenia bicyclis Ethanol Extract)

  • 대황 ethyl acetate 분획은 $IC_{50}$ 값이 1.31 mg/ml으로 에탄올 추출물보다 높은 lipase 저해활성을 나타내었다. Silicagel column chromatography를 이용하여 ethyl acetate 분획으로부터 얻은 sub-fraction 중 EA1 fraction이 $IC_{50}$ 값 0.54 mg/ml로 가장 높은 저해활성을 보였으며 더 나아가 HPLC를 이용하여 8개의 sub-fraction으로 분리하였다. HPLC fraction 중 fraction 5가 $IC_{50}$ 값 0.37 mg/ml로 높은 lipase 저해활성을 보였다. 이 fraction 5는 분자량 741.1로 $C_{36}H_{22}O_{18}$ 화합물인 phlorotannin derivative dieckol로 확인되었으며 lipase에 대해 비경쟁적 저해타입을 나타내었다. 이상의 결과로, 대황 에탄올 추출물로부터 분리한 dieckol의 lipase 저해 활성 및 저해 타입 확인을 통해 식품 산업에 항비만 소재로서 응용할 수 있을 것으로 사료되어진다.

Rhei Rhizoma and Chunghyuldan Inhibit Pancreatic Lipase

  • Pancreatic lipase-inhibitory activity of the rhizome of Rhei Rhizoma and its antihyperlipidemic activity were measured. Rhei Rhizoma inhibited pancreatic lipase with $IC_{50}$ value of 6.5 mg/ml (triolein as a substrate). Rhei Rhizoma significantly inhibited serum TG level in corn oil feeding-induced mice, and serum TG and cholesterol in Triton WR-1339-induced hyperlipidemic mice. However, Rhei Rhizoma did not show the hypolipidemic activity in high cholesterol diet-induced hyperlipidemic mice. When in vitro pancreatic lipase-inhibitory and in vivo antihyperlipidemic activities of Whangryunhaedoktang (WT) and Chunghyuldan (CD), which is consisted of ingredients of WT and Rhei Rhizoma, were measured, CD exhibited more potent inhibitory activities than WT. Therefore these results suggest that antihyperlipidemic activity of Rhei Rhizoma and CD may be more or less originated from the inhibition of pancreatic lipase.

In Vitro Inhibitory Effect of Triterpenoidal Saponins from Platycodi Radix on Pancreatic Lipase

  • In the process of investigating anti-obesity effect of Platycodi Radix, we found that aqueous extract of Platycodi Radix might inhibit intestinal absorption of dietary fat by inhibiting pancreatic lipase (PL) activity. In order to clarify the anti-obesity mechanism of Platycodi Radix, activity-guided isolation was performed to find active components. The total saponin fraction of Platycodi Radix appeared to have a potent inhibitory activity against the hydrolysis of triolein emulsified with phosphatidycholine by pancreatic lipase in vitro. Based on these results, further purification of active components yielded 10 known triterpenoidal saponins, among these compounds, platycodin A, C, D, and deapioplatycodin D exhibited significant inhibitory effects on PL at the concentration of $500\;{\mu}g/mL$ with 3.3, 5.2, 34.8, and $11.67\%$ pancreatic lipase activity vs control, respectively. Platycodin D was found to inhibit the PL activity in a dose-dependent manner. Therefore, the anti-obesity effect of Platycodi Radix might be due to the inhibition of pancreatic lipase by its saponins.

Evaluation of Cytotoxicity, Carbohydrate, and Lipid Inhibitory Activity of Codonopsis lanceolata using Different Solvent Fractions

  • This study was conducted to evaluate the cytotoxicity and ${\alpha}-Amylase$, ${\alpha}-Glucosidase$, pancreatic lipase inhibition in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The values of $IC_{50}$ against Calu-6 cell showed a high effect in n-hexane fraction ($10.13{\mu}g/mL$) whereas DW fraction exhibited the weakest inhibition on cell viability, having an $IC_{50}$ value of over $1,000{\mu}g/mL$. The values of $IC_{50}$ against HCT-116 cell showed the highest activity in n-BuOH fraction ($102.01{\mu}g/mL$), followed by n-hexane fraction ($145.85{\mu}g/mL$), methylene chloride fraction ($332.02{\mu}g/mL$), ethyl acetate fraction ($462.93{\mu}g/mL$) and DW fracion ($>1,000{\mu}g/mL$). ${\alpha}-Amylase$ inhibitory activity in methylene chloride fraction and ethyl acetate fraction was found to have a higher inhibitory effect with 24.5% and 25.6% than the other fractions. The highest ${\alpha}-Glucosidase$ inhibitory activity was observed from the ethyl acetate fraction extract, while the extract of DW fraction showed the lowest level of inhibitory activity at given experiment concentration. The pancreatic lipase inhibitory activity of C. lanceolata was found to have a higher the effect in ethyl acetate fraction. Inhibition of lipase activity of the ethyl acetate fraction and n-hexane fraction showed a relatively high, while the extract of DW fraction showed the lowest level at given experiment concentration. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on carbohydrate, lipid Inhibitory activity and anticancer activity.

지질분해효소 저해물질 함유 약용식물의 선발 및 저해물질 추출조건 (Screening of Medicinal Plants containing Lipase Inhibitor and Optimal Extraction Conditions)

  • This study was performed to develop new anti-obesity agents for functional foods from medicinal plants and herbs. Methanol extracts from 560 kinds of medicinal plants and herbs, and their lipase inhibitory activities were investigated. Methanol extract from Desmodium oxyphyllum Dc showed the highest lipase inhibitory activity at 74.2%. This lipase inhibitory activity was maximally extracted when powder of Desmodium oxyphyllum Dc was treated with 80% methanol (1 : 20) for 48 h.
