Association of growth hormone and insulin-like growth factor I genotype with body weight, dominance of body weight, and mRNA expression in Korat slow-growing chickens

  • Objective: Growth hormone (GH) and insulin-like growth factor I (IGF-I) play a critical role in animal growth rates. We aimed to investigate the effect of GH and IGF-I genotypes on body weight (BW), dominance, and gene expression in slow-growing chickens at different ages. Methods: A total of 613 Korat chickens (KRs) were bred and divided into three groups by genotype - A1A1, A1A3, and A3A3 for GH and AA, AC, and CC for IGF-I. Chickens were weighed every two weeks, and liver and breast muscle tissues were collected at 10 weeks of age. Genetic parameters of KRs were estimated using ASReml software. The GH and IGF-I mRNA levels were measured by quantitative polymerase chain reaction. Significant differences between traits were analyzed using the generalized linear model. Results: A significant effect of GH genotypes on BW was found at most ages, and the A1A1 genotype had the highest value of BW. Compared with the A3A3 genotype, the A1A1 and A1A3 genotypes showed a higher dominance effect at 0 and 2 weeks, and genotype A1A1 had the highest value of dominance at 8 weeks of age. A difference in GH mRNA levels between genotypes was detected in breast muscle at 6 weeks and in the liver tissue at 2 weeks. In the case of IGF-I gene, the AA genotype had the highest BW at the beginning of life. Significant differences in BW dominance were found at 2 weeks. However, IGF-I mRNA levels were not different among genotypes in both breast muscles and liver tissues. Conclusion: Our results revealed that GH and IGF-I influence growth, but may not be involved in heterosis. GH can be used as a marker gene in selection programs for growth because the homozygous genotype (A1A1) had the highest BW at all ages. The IGF-I is not a useful marker gene for selection programs.

  • Duck industry had a rapid growth in recent years. Nevertheless, researches to improve duck house environment are still not sufficient enough. Moisture generation of duck house litter is an important factor because it may cause severe illness and low productivity. However, the measuring process is difficult because it could be disturbed with animal excrements and other factors. Therefore, it has to be calculated according to the environmental data around the duck house litter. To cut through all these procedures, we built several machine learning regression model forecasting moisture generation of litter by measured environment data (air temperature, relative humidity, wind velocity and water contents). 5 models (Multi Linear Regression, k-Nearest Neighbors, Support Vector Regression, Random Forest and Deep Neural Network). have been selected for regression. By using R-Square, RMSE and MAE as evaluation metrics, the best accurate model was estimated according to the variables for each machine learning model. In addition, to address the small amount of data acquired through lab experiments, bootstrapping method, a technique utilized in statistics, was used. As a result, the most accurate model selected was Random Forest, with parameters of n-estimator 200 by bootstrapping the original data nine times.

Physiochemical characteristics and fermentation ability of milk from Czech Fleckvieh cows are related to genetic polymorphisms of β-casein, κ-casein, and β-lactoglobulin

  • Objective: The aim of the study was to find a possible association between the ${\beta}-$ and ${\kappa}-casein$ and ${\beta}-lactoglobulin$ genotypes and important milk physiochemical and technological characteristics such as acidity, alcohol stability, the contents of some minerals and the parameters of acid fermentation ability (FEA) in Czech Fleckvieh Cattle. Methods: Milk and blood samples were collected from 338 primiparous Czech Fleckvieh cows at the same stage of lactation. The genotypes of individual cows for ${\kappa}-casein$ (alleles A, B, and E) and ${\beta}-lactoglobulin$ (alleles A and B) were ascertained by polymerase chain reaction-restriction fragment length polymorphism, while their ${\beta}-casein$ (alleles $A^1$, $A^2$, $A^3$, and B) genotype was determined using melting curve genotyping analysis. The data collected were i) milk traits including active acidity (pH), titratable acidity (TA), alcohol stability (AS); calcium (Ca), phosphorus (P), sodium (Na), magnesium (Mg), and potassium (K) contents; and ii) yoghurt traits including active acidity (Y-pH), titratable acidity (Y-TA), and the counts of both Lactobacilli and Streptococci in 1 mL of yoghurt. A linear model was assumed with fixed effects of herd, year, and season of calving, an effect of the age of the cow at first calving and effects of the casein and lactoglobulin genotypes of ${\beta}-CN$ (${\beta}-casein$, CSN2), ${\kappa}-CN$ (${\kappa}-casein$, CSN3), and ${\beta}-LG$ (${\beta}-lactoglobulin$, LGB), or the three-way interaction between those genes. Results: The genetic polymorphisms were related to the milk TA, AS, content of P and Ca, Y-pH and Lactobacilli number in the fresh yoghurt. The CSN3 genotype was significantly associated with milk AS (p<0.05). The effect of the composite CSN2-CSN3-LGB genotype on the investigated traits mostly reflected the effects of the individual genes. It significantly influenced TA (p<0.01), Y-pH (p<0.05) and the log of the Lactobacilli count (p<0.05). Conclusion: Our findings indicate that the yoghurt fermentation test together with milk proteins genotyping could contribute to milk quality control and highlight new perspectives in dairy cattle selection.

Carcass characteristics and meat quality of purebred Pakchong 5 and crossbred pigs sired by Pakchong 5 or Duroc boar

  • Objective: This study investigated carcass characteristics and meat quality of purebred Pakchong 5, crossbred pigs sired by Pakchong 5, and crossbred pigs sired by Duroc. Methods: Forty-eight pigs (average body weight of 22.25 kg) were composed of three groups as purebred Pakchong 5 (PP), Large $White{\times}Landrace$ pigs sired by Pakchong 5 (LWLRP), and Large $White{\times}Landrace$ pigs sired by Duroc (LWLRD). Each group consisted of eight gilts and eight barrows. At 109-day-raising period, pigs were slaughtered, and carcass characteristics were evaluated. Longissimus thoracis (LT) muscles from left side of carcasses were evaluated for meat quality and chemical composition. Data were analyzed using general linear model procedure, where group, sex, and their interaction were included in the model. Results: The PP had greater carcass, total lean, and ham percentages than crossbred pigs (p<0.05). LWLRP had thicker backfat and more carcass fat percentage than LWLRD (p<0.05). There were no differences (p>0.05) on cutting percentages from tender loin, loin, boston butt, and picnic shoulder among groups. The PP and LWLRP had larger loin eye area (LEA) than LWLRD (p<0.05). Gilts had more loin percentage and lower $L^*$ value than barrows (p<0.05). No meat color parameters ($L^*$, $a^*$, and $b^*$) were affected by groups (p>0.05). PP and LWLRP had larger muscle fiber diameters than LWLRD (p<0.05). However, water holding capacity, Warner-Bratzler shear force values, and chemical composition of LT were not affected by group or sex (p>0.05). Conclusion: Pakchong 5 purebred has good carcass and lean percentages. Compared to Duroc crossbred pigs, Pakchong 5 crossbreds have similar carcass and lean percentages, larger LEA, and slightly more carcass fat, with comparable meat quality and chemical composition. Pakchong 5 boars are more affordable for very small- to medium-scale pig producers.

발광미생물을 이용한 중금속 6종의 생물독성 평가 및 모델링 (Bio Toxicity Assessment and Kinetic Model of 6 Heavy Metals Using Luminous Bacteria)

  • 북미, 유럽 뿐 아니라 우리나라에서도 유해물질 관리제도가 도입되는 등 매년 크게 증가하는 화학물질의 생산, 유통으로 인한 독성 피해에 대응하고 있다. 유해물질의 생물독성을 평가하는 방법은 널리 소개되어 왔으나, 모델링을 위한 정량적이고 신속한 정보가 요구된다. 본 연구에서는 발광 미생물인 Vibrio fischeri를 이용하여 아연, 구리, 크롬, 카드뮴, 수은, 납 등 중금속 6종에 대한 독성 평가를 실시하고, 독성 모델링을 통해 생존율을 재현하였다. 발광 미생물의 활성 정도 및 중금속에 의한 독성은 photomultiplier가 내장된 감광 장치를 이용하여 상대발광도를 측정, 독성의 영향을 판단하였다. 30분 내 저해율이 20% 미만에 이르는 농도 및 $EC_{50}$ 등을 통해 독성을 평가하였다. 그 결과, 생물 독성은 수은, 납, 구리, 크롬, 아연, 카드뮴 순으로 나타났다. 시간별 $EC_{50}$의 추세선을 통해 얻은 $EC_{{50},{\infty}}$는 독성 모델링의 주요 파라미터와 선형 관계가 높았다. 실험결과로부터 DAM, TDM 모델링에 필요한 주요 파라미터를 얻었으며, 재현된 독성 영향은 아연, 구리, 크롬, 카드뮴 등에서 오차율 평균 10.2%이었다. Vibrio fischeri를 및 감광 장치를 이용한 미생물 독성 측정 방법은 짧은 시간에서 $EC_{{50},{\infty}}$ 및 독성 모델링 구현에 필요한 주요한 인자들을 신속하게 얻을 있어 유용하게 활용될 수 있을 것으로 기대된다.

충청지역 주요 수종의 수고-흉고직경 생장모델에 관한 연구 (Height-DBH Growth Models of Major Tree Species in Chungcheong Province)

  • 본 연구는 5차 국가산림자원조사(National Forest Inventory, NFI) 표본점 자료를 이용하여 충청도 지역에 분포하는 주요 수종에 대한 수고-흉고직경 생장모델을 개발하고자 하였다. 충청도 지역 고정표본지 내에서 수고와 흉고직경이 측정된 주요 수종의 총 임목 본수는 2,681본이었으며, 무작위로 생장모델의 개발을 위해 90% 자료와 모델 타당성 검정을 위해 10% 자료로 나누어서 분석하였고, 본 연구에서 제시된 최종모형의 추정된 계수는 100% 자료를 이용하였다. 8개 주요 수종에 대한 생장모델들의 적합성 검정은 결정계수($R^2$), 추정치의 오차인 평균제곱근오차(RMSE), 평균편의(MD), 절대평균편의(AMD)와 직경급별로 평균편의(MD)를 비교 분석하였다. 본 연구 결과에 의하면, 6개 주요 생장식의 결정계수는 모두 94% 이상의 높은 설명력을 나타냈으며, 특히 C-R 생장모델과 Weibull 생장모델은 다른 모델에 비해 좋은 결과를 나타냈다. 직경급 30 cm 이하에서는 소나무, 리기다소나무, 굴참나무, 신갈나무가 상대적으로 가장 작은 평균편의를 나타낸 반면, 직경급 30 cm 이상에서는 신갈나무, 상수리나무, 졸참나무가 큰 평균편의를 나타냈다. 또한 본 연구의 결과로 제시된 6개 주요 생장식에서 추정한 수고를 임목자원평가 프로그램에 적용하여 간재적을 분석한 결과, 직경급 30 cm까지는 큰 차이를 보이지 않지만, 30 cm 이상인 대경목의 경우 추정된 간재적은 큰 차이를 보이므로, 생장모델 선정에 주의를 기울여야 한다.

Effects of a multi-strain probiotic on growth, health, and fecal bacterial flora of neonatal dairy calves

  • Objective: The aim of this study was to investigate the effects of dietary supplementation with a multi-strain probiotic (MSP) product containing of Bifidobacterium animalis, Lactobacillus casei, Streptococcus faecalis, and Bacillus cerevisiae on growth, health, and fecal bacterial composition of dairy calves during the first month of life. Methods: Forty Holstein calves (24 female and 16 male) at 2 d of age were grouped by sex and date of birth then randomly assigned to 1 of 4 treatments: milk replacer supplementation with 0 g (0MSP), 2 g (2MSP), 4 g (4MSP), and 6 g (6MSP) MSP per calf per day. Results: Supplementation of MSP did not result in any significant differences in parameters of body measurements of calves during the 30 d period. As the dosage of MSP increased, the average daily gain (p = 0.025) and total dry matter intake (p = 0.020) of calves showed a linear increase. The fecal consistency index of the 2MSP, 4MSP, and 6MSP group calves were lower than that of the 0MSP group calves (p = 0.003). As the dosage of MSP increased, the concentrations of lactate dehydrogenase (p = 0.068) and aspartate aminotransferase (p = 0.081) in serum tended to decrease, whereas the concentration of total cholesterol increased quadratically (p = 0.021). The relative abundance of Dorea in feces was lower (p = 0.011) in the 2MSP, 4MSP, and 6MSP group calves than that in the 0MSP group calves. The relative abundance of Dorea (p = 0.001), Faecalibacterium (p = 0.050), and Mitsuokella (p = 0.030) decreased linearly, whereas the relative abundance of Prevotella tended to increase linearly as the dosage of MSP increased (p = 0.058). Conclusion: The MSP product can be used to reduce the diarrhea, improve the performance, and alter the composition of the fecal bacteria in neonatal dairy calves under the commercial conditions.

근골격계 질환에 대한 양약 및 한약 병용의 간과 신장에 대한 안전성: 후향적 관찰 연구 (Hepatic and Renal Safety of Concurrent Use of Conventional and Herbal Medications for Musculoskeletal Disorders: A Retrospective Observational Study)

  • Objectives This study aimed to investigate whether the concurrent use of conventional and herbal medications affects liver and kidney function, by examining blood test data. Methods We retrospectively reviewed the electronic medical records of 590 inpatients with musculoskeletal diseases between 2013 and 2017. We investigated cases of drug-induced liver injury (DILI) according to the Roussel Uclaf Causality Assessment Method criteria and cases of drug-induced kidney injury (DIKI) based on the Kidney Disease Improving Global Outcomes definition. Results One case (0.17%) of DILI and one case (0.17%) of DIKI were identified. Significant improvements in serum laboratory data were observed after the concurrent use of both types of medications (p<0.05). The kappa coefficients ranged from 0.26 to 0.72, indicating that the values after the concurrent use of conventional and herbal medications showed a fair similarity to the baseline values of the patients. The linear regression test showed that female sex and high body mass index (BMI) were risk factors for an increase in the serum blood levels of liver function parameters. Conclusions The concurrent use of conventional and herbal medications for musculoskeletal disorders is relatively safe; however, clinicians should exercise caution when prescribing these medications to female patients and patients with a high BMI because of their potential effect on hepatic function.

위성기반 증발산량 및 토양수분량 산정 국내 연구동향 (Research Status of Satellite-based Evapotranspiration and Soil Moisture Estimations in South Korea)

  • 최근 수문 및 수자원 분야에서 위성영상의 활용성이 높아짐에 따라 관련 전용 위성 개발연구와 연계하여 위성을 활용한 증발산량과 토양수분량 산정 연구의 필요성이 강조되고 있다. 본 연구에서는 이러한 위성을 기반으로 증발산량 및 토양수분량의 국내 연구현황과 그 산정 방법론을 조사하여 현재까지의 연구동향을 파악하고자 하였다. 국내 연구현황을 세부 방법론 별로 살펴본 결과 일반적으로 증발산량의 경우는 Surface Energy Balance Algorithm for Land (SEBAL), Mapping Evapotranspiration with Internalized Calibration (METRIC)과 같은 에너지수지 기반 모형과 Penman-Monteith (PM) 및 Priestley-Taylor (PT) 산출식을 기반으로 산정되었으며, 토양수분량의 경우 능동형(AMSR-E, AMSR2, MIRAS, SMAP) 및 수동형(ASCAT, SAR)와 같은 마이크로파 센서를 통한 산정이 주를 이루었다. 통계적 측면에서는 증발산량 및 토양수분량 공통적으로 회귀식 및 인공지능을 이용한 산출사례를 찾을 수 있었다. 또한 위성기반 자료들을 이용한 Evaporative Stress Index (ESI), Temperature-Vegetation Dryness Index (TVDI), Soil Moisture Deficit Index (SMDI) 등의 다양한 지표를 산정하여 가뭄 특성파악에 적용한 연구 사례도 다수 있었으며, 지표모형(Land Surface Model, LSM)을 기반으로 하여 위성 다중센서에서 얻을 수 있는 주요 자료들을 활용해 증발산량과 토양수분량의 수문순환인자를 산출하기도 하였다. 본 논문에서는 이렇게 기존 연구사례 조사 및 내용파악 과정을 통해 위성을 활용한 주요 세부 방법론을 비교·검토 제시함으로써 관련 연구분야 기준 참고자료로의 활용 및 향후 위성기반 관련 수문순환 자료 산출 고도화 연구의 초석을 다지고자 한다.

탄도형 충격파 치료기의 음향 출력 시험을 위한 Dry Test Bench의 신뢰성 및 유용성 (Reliability and utility of a Dry Test Bench for testing the acoustic output from a ballistic shock wave therapeutic device)

  • 탄도형 체외 충격파 치료기의 출력 에너지를 측정하는 Dry Test Bench(DTB)의 신뢰성을 검증하기 위해, 상용 탄도형 충격파 치료기에 대해 Laser Doppler Vibrometer(LDV)로 측정된 충격파의 음향 에너지와 비교했다. 실험 결과, DTB로 측정된 역학적 에너지는, 동일한 출력 설정에서, 5 % 이내의 변동성을 보이며, 치료기의 전 출력 설정 범위에서 LDV로 측정된 충격파의 음향 에너지와 선형적인 상관성(adj. R2 = 0.991)을 확인했다. 두 측정 방법의 상관성과, LDV을 이용하여 공기 및 수중에서 측정된 충격파 음향 에너지의 상관성(adj. R2 = 0.995)을 통합하면, DTB 측정으로부터 수중에서 발생된 충격파의 energy flux density를 평균 7.85 % 오차로 추정된다. DTB는 치료기의 출력에너지에 대한 정보만 제공하기 때문에, IEC61846 및 IEC63045에서 요구하는 다양한 충격파의 음향 출력을 시험하는 도구로 적합하지 않다. 그러나 측정 원리가 단순하고 사용이 용이한 DTB는 제조사 및 사용자가 탄도형 Extracorporeal Shock Wave Therapy(ESWT) 치료기의 성능을 관리하는 목적으로 유용하게 사용될 수 있을 것으로 기대된다.
