무화과(Ficus carica L.) 잎의 메탄올 추출물과 그 분획물의 살충활성 (Insecticidal Potential of Methanol Extract and Its Fractions from Fig (Ficus carica L.) Leaves)

  • 양한 환경조건 온실, 하우스 및 노지에서 재배된 무화과 잎의 메탄을 추출물 및 그 분획물의 살충활성을 검토한 결과 노지에서 재배된 무화과 잎의 메탄을 추출물($80{\sim}89%$ 억제)이 온실이나 하우스에서 재배된 것에 비해 점박이응애(T. urticae)에 대해 더 높은 활성을 보였다. 특히 노지 재배 무화과 잎의 메탄올 추출물에서 분획한 에틸아세테이트층과 헥산층의 점박이응애에 대한 살충활성은 각각 92%와 90%로 가장 높은 방제가를 보였다. 따라서 본 연구 결과는 노지재배 무화과 잎의 메탄올 추출물 및 그 분획물이 식물유래 해충방제 수단으로서 잠재적인 살충효과를 기대할 수 있을 것으로 본다.

하엽(荷葉) 추출물이 항산화 효능 및 피부노화에 미치는 영향 (Antioxidant and Anti-wrinkling Effects of Extracts from Nelumbo nucifera leaves)

  • Objective : The purpose of this study was to investigate anti-aging and antioxidant effects of extracts of Nelumbo nucifera leaves (NN-L) using ethanol on skin .Methods : Each part of leaves(NN-L), flowers(NN-F) and stem(NN-S) was extracted with 70% ethanol. We performed radical scavenging assay(DPPH, ABTS+, Superoxide anion radical), elastase inhibition assay, collagenase inhibition assay. NN-L extracts were tested for cell viability(MTT assay), MMP-1 inhibition and MMP-1 protein expression on CCD-986sk cells (human fibroblast line).Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. We measured its free radical scavenging activity, elastase inhibitory activity and expression of MMP-1 (matrix metalloprotease-1) in human fibroblast cells. Among the parts of Nelumbo nucifera, NN-L showed the highest antioxidant activities and in radical scavenging. DPPH, ABTS+ and Superoxide anion radical scavenging activity of NN-L at concentration of 1,000 μg/mL were 91.43%, 99.31% and 73.7% respectively. In vitro elastase and collagenase inhibition effects of NN-L at concentration of 1,000 μg/mL was 42.8% and 55.3% respectively. The ethanol extract of NN-L showed cell viability of 95.4% in 50 μg/mL concentration. In addition, The results from Western blot assay showed that NN-L decreased the expression of MMP-1 protein in a dose-dependent manner (by up to 35.0% at 50 μM).Conclusion : The findings suggest that the NN-L great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

Mineral, Nutritional, and Phytochemical Profile, Total Phenolic Content, and Radical Scavenging Activity of Philippine Bamboo "Bolo" Gigantochloa levis (Blanco) Merr. Leaves

  • The study is a pioneering effort to determine the mineral, nutritional, and phytochemical composition and phenolic content and to determine the free radical scavenging activity of Gigantochloa levis (Blanco) Merr, a native bamboo species (locally known as "bolo") in the Philippines. Proximate analysis showed that air-dried G. levis leaves contain 15.8% ash, 22.6% crude protein, 1.2% crude fat, 29.3% crude fiber, and 19.7% total sugar. Phytochemical tests indicated the presence of diterpenes, triterpenes, saponins, phenols, tannins, and flavonoids in both the ethanolic and aqueous leaf extracts, while phytosterols were only detected in the ethanolic extract. Folin-Ciocalteu assay determined the total phenolic content in gallic acid equivalents (GAE) to be $85.86{\pm}3.71$ and $32.32{\pm}1.01mg\;GAE/100g$ dried sample for the ethanolic and aqueous extracts, respectively. The total phenolic content in quercetin equivalents (QE) was $74.44{\pm}3.11$ and $29.43{\pm}0.85mg\;QE/100g$ dried sample for the ethanolic and aqueous extracts, respectively. The radical scavenging activity of the different solvent fractions containing varying concentrations of the extract was determined using the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay. The ethyl acetate and 1-butanol fractions were found to have the highest radical scavenging activity. Mineral analysis via Energy Dispersive X-Ray Spectrometry (EDS) of the ash of G. levis leaves showed that Si is the major component, followed by K and Mg. These results point to the potential of G. levis leaves as a source of minerals and bioactive compounds with medicinal value.

뽕잎추출액코팅농도에 따른 뽕잎쌀밥의 품질에 관한 연구 (The Study on the Characteristic of Cooked Rice According to the Different Coating Ratio of Mulberry Leaves Extracts)

  • The purpose of this study was to assess the optimum coating ratio for rice, using various ratios of mulberry leaves extract, 1.0, 1.5, and 2.0%, and to determine the optimum ratio of added water, in proportion to the total weight of mulberry rice. The moisture content of the soaked rice, and the optimum water uptake rate, moisture content of the cooked rice, as well as its blue and color values, mechanical characteristics, internal structure and sensory evaluation, were analyzed. The statistical data analyses were completed using the SAS program. The results are summarized as follows: The moisture content of mulberry rice was less than that of raw rice. The average optimum water uptake of the soaked mulberry rice at the different water temperatures, 10, 20 and 30, was 20% of the total weight of the raw mulberry rice. As for the results of the sensory evaluation,; 140% water, in proportion to the total weight of raw mulberry rice, was judged to be the optimum. The average moisture content of the cooked mulberry rice was 45∼50%, but there was no significant difference in the various coating ratios. The blue value of the cooked mulberry rice awas highest on the first day of cooking. The L- and a-values decreased with increasing coating ratio, but the b-value increased under the same conditions. As for the mechanical characteristics,; the adhesiveness, hardness and springiness decreased during 2 days of storage. The internal structure of the mulberry rice, observed by SEM, showed a close structure on increasing the coating ratios of mulberry leaves extracts. It was concluded that the optimum coating ratio of mulberry rice and ratio of added water for cooking wereas 1.5 and 140%, respectively, in proportion to the total weight of raw mulberry rice.

경쟁 수종들의 잎 추출 수용액이 가문비나무 종자발아 및 유묘생장에 미치는 타감효과 (Allelopathic Effect against Seed Germination and Seedling Growth of Picea jezoensis of Leaf Aqueous Extracts from Competition Species)

  • 본 연구는 가문비나무 집단 내 치수형성이 잘 이루어지지 않는 이유를 찾고자 실시하였다. 가문비나무 집단 내 치수형성을 방해하는 타감물질의 존재여부를 확인하고, 타감작용을 일으키는 수종을 확인하기 위해서 타감물질을 추출하여 종자발아 및 유묘생장에 미치는 영향을 조사하였다. 수용성 추출액은 GC/MS를 사용하여 수용성 페놀화합물의 유무를 확인하였다. 가문비나무 집단에서 채취한 주변 경쟁수종 잎의 수용성 추출물은 가문비나무 종자의 발아를 감소시켰으며, 유묘의 생장을 저해하였다. 추출액의 수용성 페놀화합물을 분석한 결과, borneol, Camphor, Longifolen, Longifolenaldehyde, Norbornene, Ketobornane, 1,8-Cineole, ${\alpha}$-Cadinol이 검출되었는데, 이들은 monoterpenoids 화합물로서, 타감물질로 잘 알려진 화합물들이다. 결론적으로, 가문비나무 집단 내 치수형성에 영향을 미치는 인자중의 하나가 주변 경쟁식생들의 타감효과에 의한 것으로 판단된다.

Anti-inflammatory Activity of Extracts from Ultra-Fine Ground Saururus chinensis Leaves in Lipopolysaccharide-Stimulated Raw 264.7 Cells

  • Bioactive components of ultra-fine ground Saururus, the extraction yield increases when the leaves are ultra-fine ground. Comparison of normal-ground and ultra-fine ground Saururus chinensis leaves showed that the solid content and antiinflammatory activity of ultra-fine ground extracts was higher than that of normal-ground extracts. Lipopolysaccharide (LPS)-stimulated Raw 264.7 cells were treated with different concentrations of Saururus chinensis extract and the amount of nitric oxide (NO) was determined; LPS-treated cells produced 2 times more NO than cells that were not treated with LPS. Moreover, the NO production in cells treated with Saururus chinensis extract was inhibited in a concentration-dependent manner. Because the stimulant-induced NO production is regulated by the inducible nitric oxide synthase (iNOS), we measured the iNOS protein level to elucidate the mechanism by which the NO production was inhibited. We found that the amount of iNOS decreased dose-dependently. It was reduced by 53% at a Saururus chinensis extract concentration of $100{\mu}g/mL$. The protein expression of cyclooxygenase-2 (COX-2) in LPS-treated Raw 264.7 cells was inhibited by 31% at $100{\mu}g/mL$ of Saururus chinensis extract. Gel shift of the nuclear factor kappa B-DNA complex occurred in LPS-treated cells and the intensity of the band decreased gradually in a concentration-dependent manner. Ultra-fine ground Saururus chinensis extract had a concentration-dependent inhibitory effect on the production of prostaglandin $E_2$, tumor necrosis factor ${\alpha}$, interleukin $1{\beta}$ (IL-$1{\beta}$), IL-6, and IL-8 in LPS-treated Raw 264.7 cells, i.e., at $50{\mu}g/mL$ of Saururus chinensis extract, their levels were decreased by 53, 67, 52, 37, and 21% respectively.

마치현 부위별 에탄올 추출물의 항산화 활성 (Antioxidant Effects of Ethanol Extracts from Different Parts of Portulacae Herba)

  • Objectives : This study reviewed the antioxidant activity of each part of Portulacae Herba (PH) in order to better analyze the possibility of PH being used as a natural material. Methods : The root, stem, and leaves of the PH were separated, dried at $50^{\circ}C$, put in a 70% ethanol and then extracted three times every six hours. Antioxidant activities was performed by measuring the total polyphenol and total flavonoid contents, DPPH, ABTS, nitrite, hydroxyl radical scavenging activity, and $Fe^{2+}$ chelating. Results : The total polyphenol and total flavonoid contents was highest in the root, then the leaves, and lastly the stem. DPPH and ABTS radical scavenging activities for the root extract indicated the highest activity followed by the leaf and stem extracts. It also showed similar activity to that of the ascorbic acid. Hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity resulted to be highest in the root extract and then the leaf and stem extracts. Nitrite scavenging activity decreased with higher pH levels, and activity was highest in the order of root extract, leaf extract, and stem extract. Also, in reaction to a solution with a pH 1.2 level, the root extract showed similar scavenging activity to that of BHT. Conclusions : Therefore, based on the above results, it was verified that the roots of PH have a high antioxidant level, and they can be considered to have potential to be applied to functional food, cosmetics, and medicine with antioxidant efficacy.

Suppression of β-Secretase (BACE1) Activity and β-Amyloid Protein-Induced Neurotoxicity by Solvent Fractions from Petasites japonicus Leaves

  • Alzheimer's disease (AD) is a neurodegenerative disorder characterized by neuronal loss and extracellular senile plaques containing $\beta$-amyloid peptide (A$\beta$). The deposition of the A$\beta$ peptide following proteolytic processing of amyloid precursor protein (APP) by $\beta$-secretase (BACE1) and $\gamma$-secretase is a critical feature in the progression of AD. Among the plant extracts tested, the ethanol extract of Petasites japonicus leaves showed novel protective effect on B103 neuroblastoma cells against neurotoxicity induced by A$\beta$, as well as a strong suppressive effect on BACE1 activity. Ethanol extracts of P. japonicus leaves were sequentially extracted with methylene chloride, ethyl acetate and butanol and evaluated for potential to inhibit BACE1, as well as to suppress A$\beta$-induced neurotoxicity. Exposure to A$\beta$ significantly reduced cell viability and increased apoptotic cell death. However, pretreatment with ethyl acetate fraction of P. japonicus leaves prior to A$\beta$ (50 ${\mu}M$) significantly increased cell viability (p<0.01). In parallel, cell apoptosis triggered by A$\beta$ was also dramatically inhibited by ethyl acetate fraction of P. japonicus leaves. Moreover, the ethyl acetate fraction suppressed caspase-3 activity to the basal level at 30 ppm. Taken together, these results demonstrated that P. japonicus leaves appear to be a useful source for the inhibition and/or prevention of AD by suppression of BACE1 activity and attenuation of A$\beta$ induced neurocytotoxicity.
