Chitosan Stimulates Calcium Uptake and Enhances the Capability of Chinese Cabbage Plant to Resist Soft Rot Disease Caused by Pectobacterium carotovorum ssp. carotovorum

  • Chinese cabbage plant was grown hydroponically for 4 weeks in order to examine the temporal relationship of calcium concentration of the nutrient solution with calcium content in the leaf tissue and susceptibility of the tissue to soft rot disease by $Pectobacterium$ $carotovorum$ ssp. $carotovorum$ (Pcc). Calcium concentration from 0.5 to 32.0 mM was maintained for 1 week using Hoagland & Arnon solution. The calcium content of the leaf was proportionally increased to the concentration of the nutrient in the solution (r = 0.912). In contrast, the severity of soft rot symptom in the young leaves was inversely related with the amount of calcium supplied to the nutrient solution (r = 0.899). Water-soluble chitosan, prepared by hollow fiber filtration (> 100 kDa) was applied into the nutrient solution from 0.0 to 5,000 ppm. The chitosan of 10 ppm was the most effective to promote calcium uptake of the leaf, showing 155% of the control. The same chitosan solution prohibited most soft rot development of the leaf by Pcc, exhibiting only 53% of the control. Among different molecular weight fractions, chitosan fraction obtained from 30-100 kDa molecular weight cut-off promoted calcium uptake the most up to 163% of the control, and reduced the development of soft rot disease recording merely 36% of the control of the leaf tissue. The results obtained in the present study suggest that large scale production of water-soluble chitosan with an optimum molecular weight and its commercial application to Chinese cabbage production will be important to improve yield and quality of the crop.

  • This study was conducted to identify the possibility of paddy weeds served as the host plant of bacterial leaf blight, using various bacterial groups and inoculation methods. The results obtained can be summarized as follows. 1. Alopecurns spp., Setaria viridis P. Beauv., and Leersia juponica Makino were identified the most susceptible to bacterial leaf blight, similar to Milyang 23 which was used as a susceptible check variety. The others such as Digitaria adscendens Hem., Eleusine indic aGaertin., Cyperns serotinus Rottb, Cyperns difformis L. showed slight infection but most of broadleaf weeds were resistant to bacterial leaf blight. 2. Weed species showing early susceptibility maintained their susceptibility throughout the growth stages. Group I of bacterial leaf blight was the most effective to develop infection symptom to weeds. 3. Pin and scissor inoculation methods were more effective mean for infection than spray method which was used without wound.

Biological Characterization and Sequence Analysis of Cucumber mosaic virus isolated from Capsicum annuum

  • Whereas most of isolates of Cucumber mosaic virus(CMV) can induce green mosaic systemic symptoms on zucchini squash, foliar symptoms of a pepper isolate of CMV (Pf-CMV)-infected zucchini squash revealed systemic chlorotic spots. To assess this biological property, infectious full-length cDNA clones of Pf-CMV were constructed using long-template RT-PCR. The complete nucleotide sequences of RNA2 and RNA3 of Pf-CMV were determined from the infectious fulllength cDNA clones, respectively. RNA 2 and RNA3 of Pf-CMV contain 3,070 nucleotides and 2,213 nucleotides, respectively. Overall sequence homology of two RNAs revealed high similarity (90%) between CMV strains, and 60% similarity to those of Tomato aspermy virus and Peanut stunt virus strains. By sequence analysis with known representative strains of CMV, Pf- CMV belongs to a typical member of CMV subgroup IA. The virus has high evolutionary relationship with Fny-CMV, but the pathology of Pf-CMV in zucchini squash was quite different from that of Fny-CMV. The pesudorecombinant virus, F1P2P3, induced chlorotic spot leaf symptom and timing of systemic symptom in squash plants, similar to the plants infected by Pf-CMV. No systemic symptoms were observed when Pf-CMVinoculated cotyledons were removed at 5 days postinoculation (dpi) while Fny-CMV showed systemic symptom at 2 dpi. These results suggest that the pepper isolate of CMV possesses unique pathological properties distinguishable to other isolates of CMVs in zucchini squash.

Studies on the Potato Virus X and Potato Leaf Roll Virus for Disease-free Seed Potato Production (무병종서 생산을 위한 감자X바이러스 및 엽권바이러스에 관한 연구)

  • A series of experiment was carried out to study on the production of disease-free seed potatoes at the Alpine Experiment Station from 1960 to 1968, which initiated a study of comparison on degeneration of plain warm region and high altitude products and the effect of latent potato virus X (PVX) and potato leaf roll virus(PLRV) on degeneration. Particular observations were made on some aspect of the nature of potato virus disease and its control such as concentrations of PVX, range of host plants, physical properties such as concentrations of PYX, range of host plants, physical properties and carrying effect of insects, by investigating 9 different areas of the main potato producing regions (Kimhae, Taegu, Choongju, Taejoen, Suwon, Kwangju, Chonju, Cheju and Chinju). Highly purified anti-serum was separated and tested for control of the virus disease and also various method of prevention and control of PLRV were observed, using cultivation of sprouted seed tubers, early harvesting method, and systemic chemicals. The results obtained are summarized as follows; 1. Potato yield in the plain region decreased by 32.8~66.3% in the first year cultivation of seed potatoes from colder region, and the rate of virus infection was 92.9 to 95.4%. 2. Plants of three families including, 20 species were susceptible to the PVX, and among the plants Salvia officinalis of a habits only was the carrier while the symptom of Digitalis purpurea of Screphulariaceae was masked. Necrosis and ring spot was occurred in most pJants of the Solanaceae and ring spot symptom also was observed in Nicotiana tabacum L. var. White Burley and in N. glutinosa. 3. The 8$C_2$ strain of virus had the following physical properties; thermal inactivation point, 68-$72^{\circ}C$ : dilution inactivation point, above 1, 000, 000 dilution: ageing in vitro, 240-360 days: and ageing in dry plant tissue, 30 days. 4. Myzus persicae and Oxya spp. did not transmit the 8$C_2$ strain of potato virus. 5. Virus was purified through the ammonium sulphate isolating method, and higher titer value, 1/2048 was obtained through anti-serum test. 6. Inhibition Chenopodiacae on the virus infection of potato was remarkable, and inhibition of local lesion host also was observed. 7, By earlier planting of sprouted seed tubers, growth period could be prolonged by 10 to 12 days. 8. Earlier harvest decreased much the rate of virus infection of seed potatoes. 9. According to the results of aphid control trial using systemic soil insecticides at Kangnung and Taekwanlyung, PSP 204, Disyston and Thimet was effective to aphid control. In particular, control effect of twice treatments of PSP 204 was great. 10. Treatmental effect of those chemicals lasted about 60-70 days. However, single foliar application of emulsified chemicals was not effective to potato virus control. 11. The effect of PSP 204, Disyston, and Thimet on the control of potato leaf roll virus was great, particularly in the case of two treatments of PSP 204, at Kangnung as well as at Taekwanlyung. Higher negative correlationship between the control effect of potato leaf roll virus and potato yield was observed showing the value r=-0.85 at Kangnung, and r=-0.87 at Taekwanlyung. 12. Differences in the control effects among PSP 204, Disyston, and Thimet was not noticed.

  • This study was carried out to investigate the effect of ozone gas on paddy rice at the different growth stage. Seokwang variety of rice plant was exposed to 0.5 ppm ozone gas for 4 hours at rooting, maximum tillering, ear formation and heading stages. after ozone gas fumigation, damage symptom, percentage of destroyed leaf, chlorophyll content and peroxidase activity of rice plant were observed. The results obtained are as follows. 1) Typical symptom of ozone gas damage appeared greyish or reddish brown subtle spots within rice leaf vein. 2) Yield loss by ozone gas exposure at different growth stage was in the order of maximum tillering stage>rooting stage>ear formation stage>heading stage. 3) Chlorophyll damage and leaf destruction was the highest at maximum tillering stage, while damage of leaf and chlorophyll were not found at heading stage. 4) The damage by ozone gas fumigation was higher at the growth stage with higher N content in plant, and N content was decreased after ozone gas exposure.

Growth, Deficiency Symptom and Tissue Nutrient Contents of Leaf Perilla (Perilla frutesens Britt) Influenced by Phosphorus Concentrations in Fertigation Solution (인산 시비농도가 잎들깨의 생육, 결핍증상 및 무기원소 함량에 미치는 영향)

  • This study was carried out to investigate the effect of phosphorus concentrations in fertilizer solution on growth and development of nutrient deficiency in leaf perilla (Perilla frutesens). The nutrient concentrations in above ground plant tissue, petiole sap and soil solution of root media were also determined. Phosphorus deficiency resulted in a slow growth, lustreless leaves, suffused purple tining in older leaves and falling prematurely. Elevation of P concentrations in fertilizer solution increased the crop growth at 75 days after transplanting. The fresh weight in 0, 0.5 and 4.0 mM treatments were 0.48 g, 9.28 g, and 25.5 g, respectively, and dry weights were 0.06 g, 1.46 g and 4.13 g, respectively. The P concentrations in above ground plant tissue and petiole sap in 4.0 mM treatment were 1.78% and $2.040mg{\cdot}kg^{-1}$, respectively, at 75 days after transplanting. The soil P concentration in 4.0 mM treatment was $1.26mg{\cdot}kg^{-1}$ when it was determined by the 1:2 (sample:water) method. These results indicated that P concentrations higher than 0.3% in above ground plant tissue, $900mg{\cdot}kg^{-1}$ in petiole sap, and $0.57mg{\cdot}kg^{-1}$ in soil solution should be maintained to ensure proper growth of leaf perilla (Perilla frutesens).

  • This study was carried out to investigate the effect of potassium concentrations in fertigation solution on growth and development of nutrient deficiency symptoms of leaf perilla (Perilla frutesens). The nutrient concentrations in above ground plant tissue, petiole sap and soil solution of root media were also determined. Potassium deficiency symptoms developed in older leaves with marginal necrosis. The brown areas on the lower leaves enlarged rapidly and the margins became scorched. Elevation of K concentrations in the fertigation solution up to 8 mM increased the crop growth in leaf length, stem thickness, and fresh and dry matter production of above ground plant tissue. However, that decreased the chlorophyll contents. The 8.0 mM K treatment which showed the greatest growth had 5.01 g in dry weight and 2.76% in K content of above ground plant tissue, suggesting that maintaining K content higher than 1.7% is necessary for good growth of Perilla frutesens. The K concentrations in petiole sap and soil solution of 8.0 mM treatment were $12,289mg{\cdot}kg^{-1}\;and\;11.65mg{\cdot}L^{-1}$, respectively. These indicated that K fertilization to maintain higher than $8,700mg{\cdot}kg^{-1}$ in petiole sap and $4.5mg{\cdot}L^{-1}$ in soil solution are necessary to ensure good crop growth.

Leaf Spot of Kalanchoe (Kalanchoe blossfeldiana) Caused by Stemphylium lycopersici (Stemphylium lycopersici에 의한 칼란코에 점무늬병)

  • Leaf spot disease of Kalanchoe (Kalanchoe blossfeldiana) occurred at the farmer's vinly house in Gangseo-gu, Busan Metropolitan City, Korea, 2006. The diseased plants with typical symptom were collected and the casual agent were isolated. Its mycological characteristics and pathogenicity were examined. The results were as follows. The typical symptoms of the disease appeared as small brownish or dark brown spot on both sides of the leaves. The spots tended to develop from lower leaves. The spots gradually enlarged into conspicuous necrotic lesions 1-5 mm in diameter. Colonies of the causal fungus formed on potato dextrose agar were velvety, gray or grayish brown in color, Conidia were cylindrical or obclavate to oblong in shape, brown in color, $24{\sim}65\;{\times}\;12{\sim}23\;{\mu}m$ in size and had 1-4 transverse septa, The optimum temperature for growth of the fungus was about $25-30^{\circ}C$. Conidiophores were brown in color, $32{\sim}135\;{\times}\;4{\sim}8\;{\mu}m$ in size and had 1-7 transverse septa. The fungus was identified as Stemphylium lycopersici (Enjoji) Yamamoto based on its symptom and mycological characteristics. This is the first report of leaf spot of Kalanchoe caused by S. lycopersici in Korea.

  • A pot experiment was carried out to investigate the effect of boron on the growth, symptom, yield, and content of boron in Perilla(Perilla frutescens var. japonica HARA), Melon(Cucumis melo L.) and Cucumber(Cucumis sativus L.). The results obtained are as follows. Perilla. melon and cucumber uptake a lot of boron from the soil, and toxic effects of boron result in leaf tip yellowing by progressive necrosis. Yield of perilla, melon and cucumber are decreased at the $3{\sim}5mg\;kg^{-1}$, $5{\sim}10mg\;kg^{-1}$ of boron applicated in soil, respectively. Boron content in the leaf is the highest among various parts plant, and the tip and margin of the leaf contained more boron than central part of the leaf. Boron applicated was moved and accumulated to the plant and soil.

Undescribed Fungal Leaf Spot Disease of Pepper Caused by Cercospora capsici in Korea (고추의 미기록병(未記錄病)인 Cercospora capsici에 의(依)한 고추 반점병(斑點病))

  • An undescribed Cercospora leaf spot of pepper grown in a greenhouse was observed at Jinju area in 1983. The symptom of this disease showed frog eyes spot on lower leaves of pepper. Leaves and petiole of pepper plant inoculated with spore suspension of this pathogen app­eared leaf spot after 2 weeks and exhibited cicular, brown frog eyes spot. This fungus was grown slowly on PDA and produce condia on infected leaves. The occurrence of this disease was responsi­ble for factors with high temperature and humidity conditions in the ill-ventilated greenhouse. This causal fungus was identified as Cercospora capsici Heald et. Wolf from the shape and length of the conidiophores, the length of the condia, and pathogenic behaviors.
