Isolation and Characterization of Burkholderia cepacia EB215, an Endophytic Bacterium Showing a Potent Antifungal Activity Against Colletotrichum Species (탄저병균에 길항력이 우수한 식물내생세균 Burkholderia cepacia EB215의 분리 및 특성 규명)

  • In order to develop a new microbial fungicide using endophytic bacteria for the control of anthracnoses occurring on various crops, a total of 260 bacterial strains were isolated from fresh tissues of 5 plant species. After they were cultured in broth medium, their antifungal activities were tested for in vivo antifungal activity against cucumber anthracnose caused by Colletotrichum orbiculare. As the results, liquid cultures of 28 strains showed potent antifungal activities more than $90\%$ against cucumber anthracnose. At 3-fold dilutions of liquid cultures, 18 strains inhibited the development of cucumber anthracnose of more than $70\%$. They were further tested for in vivo antifungal activity against red pepper anthracnose caused by C. coccodes and in vitro antifungal activity against C. acutatum, a fungal agent causing red pepper anthracnose. Among 18 strains, a bacterial strain EB215 isolated from cucumber roots displayed the most potent antifungal activity against Colletotrichum species. It was identified as Burkholderia cepacia based on its physiological and biochemical characteristics, Biolog test and 16S rDNA gene sequence. It also controlled effectively the development of rice blast (Magnaporthe grisea), rice sheath blight (Corticium sasaki), tomato gray mold (Botrytis cinerea), and tomato late blight (Phytophthora infestans). Studies on the characterization of antifungal substances produced by B. cepacia EB215 are in progress.

Control effects of new triazoyl quinolines KSI-4315 and KSI-4317 against barley powdery mildew and wheat leaf rust (신규의 triazoyl quinoline 화합물 KSI-4315와 KSI-4317의 보리 흰가루병과 밀 붉은녹병에 대한 방제효과)

  • Disease control activities of 230 triazolyl quinoline derivatives were investigated against six plant diseases such as rice blast, rice sheath blight, tomato gray mold, tomato late blight, wheat leaf rust (WLR) and barley powdery mildew (BPM). New triazolyl quinolines, KSI-4315 and KSI-4317 exhibited a great in vivo control activities against WLR and BPM, and then were selected for further tests such as preventive, curative, systemic, and persistence against WLR and BPM. The KSI-4315 and KSI-4317 contained MeS moiety and MsO moiety in carbon 4-position, respectively. They possessed both preventive activity and curative activity against WLR and BPM. KSI-4317 showed the better control activity than KSI-4315 against BPM, while KSI-4315 represented the better antifungal activity against WLR. Good persistence of KSI-4315 and KSI-4317 were also observed against WLR and BPM. Persistence of KSI-4315 was similar to that of KSI-4317 on WLR, but KSI-4317 was superior to KSI-4315 on BPM in its persistence. Systemic disease control of KSI-4315 and KSI-4317 was investigated by examining translaminar activity from leaf-under-surface to leaf-upper-surface, systemic activities by leaf to leaf movement and the effect of drenching treatment. Systemicities of KSI-4315 and KSI-4317 were not observed in wheat, but KSI-4317 showed more predominant systemicity than KSI-4315 in barley. These results suggest that KSI-4317 would potentially control WLR and BPM in the fields.

Isolation of an Rx homolog from C. annuum and the evolution of Rx genes in the Solanaceae family

  • The well-conserved NBS domain of resistance (R) genes cloned from many plants allows the use of a PCR-based approach to isolate resistance gene analogs (RGAs). In this study, we isolated an RGA (CapRGC) from Capsicum annuum "CM334" using a PCR-based approach. This sequence encodes a protein with very high similarity to Rx genes, the Potato Virus X (PVX) R genes from potato. An evolutionary analysis of the CapRGC gene and its homologs retrieved by an extensive search of a Solanaceae database provided evidence that Rx-like genes (eight ESTs or genes that show very high similarity to Rx) appear to have diverged from R1 [an NBS-LRR R gene against late blight (Phytophthora infestans) from potato]-like genes. Structural comparison of the NBS domains of all the homologs in Solanaceae revealed that one novel motif, 14, is specific to the Rx-like genes, and also indicated that several other novel motifs are characteristic of the R1-like genes. Our results suggest that Rx-like genes are ancient but conserved. Furthermore, the novel conserved motifs can provide a basis for biochemical structural. function analysis and be used for degenerate primer design for the isolation of Rx-like sequences in other plant species. Comparative mapping study revealed that the position of CapRGC is syntenic to the locations of Rx and its homolog genes in the potato and tomato, but cosegregation analysis showed that CapRGC may not be the R gene against PVX in pepper. Our results confirm previous observations that the specificity of R genes is not conserved, while the structure and function of R genes are conserved. It appears that CapRGC may function as a resistance gene to another pathogen, such as the nematode to which the structure of CapRGC is most similar.

Practical Usability of Smoke Generator Containing Rice Chaff as a Combustible carrier (왕겨를 가연성 담체로 하는 훈연제의 실용성 연구)

  • Smoke rods were prepared for 2 insecticides and 5 fungicides using powdered rice chaff as a combustible carrier, and their burning characteristics were investigated. The distribution of active ingredient was investigated after the application of the granular smoke generator containing fenarimol in the connected vinyl plastic house growing cucumber. The protective effects of fungicide smoke generators were evaluated against cucumber gray mold (Botrytis cinerea) and tomato late blight (Phytophthora infestans), and compared with commercial wettable powders. Smoke rods containing powdered rice chaff showed high smoking rates of AI that were higher than commercial smoke pellets. The deposition of fenarimol on the ground of vinyl plastic house was $26.2{\pm}7.7\;ng/cm^2$ when averaged from 9 sites, and application uniformity was found. Also, the protective effects of 5 fungicidal smoke rods against plant diseases were so similar to the commercial wettable powders with no phytotoxicity that smoke rod formulations containing rice chaff as a combustible carrier could be used as an effective formulation for pesticides.

Antifungal activities for derivatives of 4-isopropyl-3-methylphenol and 5-isopropyl-3-methylphenol against plant pathogenic fungi (4-Isopropyl, 5-isopropyl-3-methylphenol 유도체들의 합성과 식물 병원균에 대한 항균 활성)

  • Fifty compounds such as ester, sulfonyl ester, carbamate, ether and phosphoyl ester derivatives of 4-isopropyl-3-methylphenol(I) and 5-isopropyl-3-methylphenol(II) were synthesized. These derivatives were identified by IR, GC/MS and $^1H$-NMR spectra. Their in vitro antifungal activities were tested against 10 plant pathogenic fungi. Among them, several compounds showed potent in vitro antifungal activity. The selected compounds showing potent in vitro antifungal activity were tested for their in vivo antifungal activities against 5 plant diseases such as rice blast, rice sheath blast, cucumber anthracnose, cucumber gray mold and tomato late blight. As a result, 4-isopropyl-3-methylphenyl(2-amino-thiazole-4-yl)methoxyiminoacetate(I-7a) showed a potent in vivo antifungal activity against rice blast. Both methyl (4-isopropyl-3-methylphenoxy)acetate(I-4d) and methyl (5-isopropyl-3-methylphenoxy)acetate(II-4d) effectively inhibited the development of cucumber gray mold.

Biological activities of Fusarium isolates from soil and plants (토양 및 식물체로부터 분리한 Fusarium속 균주들의 생물활성)

  • In order to select potent bioactive isolates, 70 Fusarium isolates obtained from soil and 21 plant species were screened by antifungal, insecticidal, herbicidal, and duckweed bioassays after culturing in potato dextrose broth and rice solid media. Eight (11.4%) of the 70 liquid broth cultures showed disease-controlling activities more than 80% against at least one of the 6 plant diseases tested. Fusarium sp. FO-68 isolate exhibited the most potent antifungal activity; it controlled rice blast, wheat leaf rust, and barley powdery mildew with control values more than 95%. Out of 70 solid cultures, 21 (30.0%) controlled at least one plant disease more than 80% and F. equiseti FO-68 isolate showed disease-controlling activities more than 95% against 3 plant diseases such as rice blast, tomato late blight, and wheat leaf rust. As for tile insecticidal activities, 2 liquid and 1 solid cultures showed potent insecticidal activities against pest insects more than 80%, Liquid cultures of F. oxysporum FO-61 and Fusarium sp. FO-80 isolates exhibited insecticidal activities more than 80% against green peach aphid and diamondback moth, respectively. The solid culture of Fusarium sp. FO-510 isolate had 80% insecticidal activity against green peach aphid. However, none of liquid and solid cultures of the 70 Fusarium isolates showed potent herbicidal activities against 10 upland weeds. As the results of duckweed assay, 3 liquid cultures showed 70% growth inhibitory activity at concentrations less than 1.25% of culture supernatants and 9 solid cultures had a potent inhibitory activity against duckweed growth. On the other hand, there was a significant correlation between antifungal activities and herbicidal activities against duckweed of both liquid and solid cultures of tile 70 Fusarium isolates.
