생식기의 Candida albicans, Neisseria gonorrhoeae 및 Lactobacillus acidofillus에 대한 천연물질과 Polyphenol의 효과 (Effects of Natural Substances and Polyphenols on Candida albicans, Neisseria gonorrhoeae and L. acidofillus in the Reproductive Organ)

  • Antimicrobial studies on reproductive organs have been scarcely reported. Candida albicans and Neisseria gonorrhoea test to find out the natural substances of various concentrations in the antimicrobial experiments have been made. Antimicrobial effect of biopole as the natural compound biosynthesis matter that contain anti-inflammatory material, catechin polyphenols and lactic acid as a single natural substance on C. albicans were of great significance. Also the results of antimicrobial effects of biopole, catechin and lactic acid as a single natural substance on N. gonorrhoea, respectively, showed lower concentration than those of the antimicrobial effects on C. albicans. Through the survival of Lactobacillus acidofillus that acts for the protection of the genital tissue the importance of lactic acid was confirmed. Lactobacillus acidofillus protection and C. albicans firmly into disinfected to low concentrations of the natural mixture from biopole and catechin with lactic acid were found and the antimicrobial effects of this natural mixture on N. gonorrhoea were perfect. C. albicans and N. gonorrhoea to disinfect the optimal natural mixture from 2% concentration biopole, 0.2% concentration of catechin and 2% lactic acid were obtained. Through the survival of Lactobacillus acidofillus in the reproductive organs protectable effects were acquired to prevent the infections of reproductive tissue and the recurrence.

Loperamide로 유도된 변비 증상에 유산균 제제가 미치는 영향 (Effect of Lactic Acid Bacteria Powder on Loperamide-induced Constipation in Rat)

  • Loperamide를 통해 변비가 유발된 실험동물에 유산균 투여가 미치는 영향을 알아보기 위하여 14일 동안 저농도($10^7CFU/mL$ per kg of body weight)의 유산균과 고농도($10^9CFU/mL$ per kg of body weight)의 유산균을 투여한 후, 변비개선 효과를 측정하였다. 그 결과, 변비 유발 후 유산균을 투여한 실험군들의 경우, 대조군(CON)에 비하여 변의 개수, 변 중량 및 수분 함량이 유의적으로 증가하였다. 또한 소화관 이동률을 조사한 결과, 대조군(CON)에 비하여 변비 유발 후 유산균을 투여한 군에서 정상대조군(NOR)인 군과 유사한 이동성을 보이는 것을 확인하였다. 변비가 유발된 실험동물의 혈청 중성지방, 총 콜레스테롤 함량, HDL-콜레스테롤 함량에는 유산균 투여 유무 및 유산균의 농도에 대한 영향을 크게 받지 않은 것으로 보여진다. 고농도의 유산균을 섭취시킨 실험동물군(HIG)의 경우, 분변으로 유도된 아세트산과 프로피온산의 함량이 유의적으로 높게 나타났다. 또한 헤마톡실린 및 에오신 염색을 통한 장내 상피세포 관찰을 통해 유산균 분말 투여시 장 점막의 길이와 넓이가 유의적으로 증가하는 것을 확인하였다. 메타지노믹스 유전자 분석을 통한 미생물 분포의 상동성을 비교해 본 결과, 고농도의 유산균 투여군(HIG)이 정상대조군(NOR)과 가장 흡사한 분포 특성을 가지고 있음을 보여주었다. 이상의 결과에 따라 유산균 제제는 변비 개선 효과가 있다고 볼 수 있다.

인천연안 Vibrio parahaemolyticus의 분포 및 유산과 과산화수소 처리에 의한 항균효과 (Antimicrobial Effect of Lactic acid and Hydrogen Peroxide and Distribution of Vibrio parahaemolyticus from the Incheon Adjacent Sea)

  • This study was carried out to investigate the distribution of Vibrio parahaemolyticus in the Incheon adjacent sea, and antimicrobial effect on growth of Vibrio parahaemolyticus in lactic acid and hydrogen peroxide and combination of lactic acid and hydrogen peroxide. The detected strains were compared geographical, months and sample types. The distribution of Vibrio parahaemolyticus was high at Ganghwa county with 66.1%(336 samples), on 7-9 months with 72.4%(386 samples) and from tireland with 75.0%(90 samples), respectively. The minimun inhibitory concentration (MIC) of lactic acid in Vibrio parahaemolyticus were 1250 ppm at pH 6.5 and 7.0, 625 ppm at pH 6.0. respectively. The minimun inhibitory concentration (MIC) of hydrogen peroxide in Vibrio parahaemolyticus were 25 ppm at pH 6.5 and 7.0, 12.5 ppm at pH 6.0, respectively. MICs of combined treatment of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus were 625 ppm of lactic acid with 12.5 ppm of hydrogen peroxide. The correlations between MICs of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus were obtained through the coefficient of determination($R^2$). $R^2$ value were 1.0000. The antimicrobial effect of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus could be confirmed from the result of this experiment.

B. amyloliquefaciens 세포 성장에 미치는 포도당과 맥아당 농도의 영향에 관한 연구 (Effects of Concentrations of Glucose and Maltose on the Growth of Bacillus amyloliquefaciens)

  • 탄소원으로 초기 glucose 농도와 초기 maltose 농도를 5g/$\ell$, 109/$\ell$, 20g/$\ell$ 로 한 후 Bacillus a amyloliquefaciens(ATCC 23350 ) 균주를 이용하여 플라스크 배양시의 세포 성장과 생성물 생성에 대한 연구를 수행하였다. 균주의 최대 건조세포농도는 초기 maltose 농도가 증가할수록 증가하였으나, glu­c cose에서는 초기 농도가 109/$\ell$일 때 가장 높았다 최대 비성장속도는 초기 glucose농-토가 109/$\ell$ 일 때 $0.060(hr^{-1})$로 가장 높으며, maltose에서는 초기 농도가 5g/P 얼 때 $0.042(hr^{-1})$이였다. 균주에 의해 생성되는 최대 a-amylase 생산은 초기 maltose 농도가 109/P 얼 때 l49.0unit/ml로 가장 높았으며, 초기 maltose 농도가 증가할수록 증가하였다. 최대 비생성속도는 초기 maltose 농도가 5g/$\ell$ 일 때 3.26unit/ mg- hr로 가장 높았으며 , glucose보다는 maltose에서 더 높았다. 균주에 의해 생성되는 최대 ${\alpha}$-acetic acid는 탄소원의 초기 농도가 증가함에 따라 감소하며, 초기 glucose 농도 109/$\ell$ 일 때 0.94g/$\ell$ 로 가장 많이 생성되였고, lactic acid는 탄소원의 초 기 농도가 증가할수록 증가하였으며, 초기 maltose 농도가 20g/$\ell$일얼 때 8.20g/$\ell$ 로 가장 많이 생성되였 다.

Inhibition of Lactic Acid Bacteria in Kimchi Fermentation by Nisin

  • Sixty isolates of lactic acid bacteria found in kimchi, a traditional Korean dish of fermented vegetables, were tested for nisin sensitivity. Of the sixty isolates, all belonging to the genera Leuconostoc, Lactobacillus, and Pediococcus, fifty isolates were sensitive to nisin at a concentration of 100 IU/$m\ell$, and four isolates appeared to be resistant to nisin. This demonstrated that the nisin sensitivity of lactic acid bacteria found in kimchi varied considerably among isolates. In MRS broth containing nisin at concentrations of 100 to 300 IV/$m\ell$, the growth of sensitive isolates of Leuconostoc mesenteroides and Lactobacillus plantarum was inhibited for two to three days at 2$0^{\circ}C$. When nisin was added to kimchi preparations at a concentration of 100 IU/$m\ell$, the growth of lactic acid bacteria was delayed and reached a maximum two days later than that in kimchi without nisin. These results suggest the possible use of nisin in kimchi preparation, at recommended levels, to control the lactic acid fermentation. Scanning electron micrographs of a sensitive isolate L. plantarum revealed the formation of pores on cell surfaces followed by rapid cell wall destruction 1 h after the addition of nisin.

탄소원으로서 입국을 이용한 유산균 발효 (Lactic Acid Fermentation with Rice Koji as a Carbon Source)

  • Recently, several health benefits of rice wine, makgeolli, were known due to the interest on the traditional Korean liquor and the researches on the rice wine are increasing. Organic acids produced during the process of rice wine fermentation play important roles in the taste and flavor. In this study, we have examined the optimal conditions for lactic acid production in rice koji as a carbon source. Skim milk was also used as a supplementary ingredient for the optimization of lactic acid fermentation. Bacterial growth of Lactobacillus sakei was monitored under this condition. The pH, acidity of the culture and the ethanol tolerance of this bacterium were also tested. Through these experiments, we were able to optimize the growth condition of lactic acid bacteria by the addition of skim milk. This was also able to affect the change of pH, acidity, sugar concentration and alcohol tolerance, which might contribute to the improvement of the quality of rice wine. The optimal condition for the growth was 2 days with 10% (w/v) of skim milk concentration. With these results, it was confirmed that rice koji was an effective carbon source for the growth of lactic acid bacteria.

Evaluation of L-Lactic Acid Production in Batch, Fed-batch, and Continuous Cultures of Rhizopus sp. MK-96-1196 Using an Airlift Bioreactor

  • Various processes which produce L-lactic acid using ammonia-tolerant mutant strain, Rhizopus sp. MK-96-1196, in a 3L airlift bioreactor were evaluated. When the fed-batch culture was carried out by keeping the glucose concentration at 30g/L, more than 140 g/L of L-lactic acid was produced with a product yield of 83%. In the case of the batch culture with 200g/L of initial glucose concentration, 121g/L of L-lactic acid was obtained but the low product yield based on the amount of glucose consumed. In the case of a continuous culture, 1.5g/L/h of the volumetric productivity with a product yield of 71% was achieved at dilution rate of $0.024\;h^{-1}$. Basis on these results three processes were evaluated by simple variable cost estimation including carbon source, steam, and waste treatment costs. The total variable costs of the fed-batch and continuous cultures were 88% and 140%, respectively, compared to that of batch culture. The fed-batch culture with high L-lactic acid concentration and high product yield decreased variable costs, and was the best-suited for the industrial production of L-lactic acid.

재조합 대장균의 세포성장에 대한 인산암모늄 농도의 영향 (Effect of Ammonium Phosphate Concentration on the Growth of Recombinant E. coli)

  • 인산암모늄의 첨가에 따른 재조합 대장균의 성장과 그 부산물 생성 관계를 알기 위하여 인산암모늄의 청가 상태에 따른 영향을 검토하였다. 인산암모 늄은 세포 성장에 중요한 작용을 하고 있다. 배양 실시후 8시간에서 12시간 전후에서 인산 암모늄이 거의 흡수되었고 탄소원이 세포에 흡수되는 속도와 인산암모늄이 세포에 흉수되는 속도가 거의 비슷한 현 상으로 진행되었다. 인산 암모늄의 초기농도를 O.5g/ L로 사용할 때 세포 최대 농도는 4.2g/L로 최고를 보였다. 그러으로 세포 성장에 펼요한 인산 암모늄 의 첨가를 초기 인산암모늄 농도 O.5g/L 전후로 첨가함이 적당하다. 그리고 인산암모늄에 trypton을 첨가하지 않는 LB 배지와 trypton을 첨가할 LB 배지에서 세포 성장은 비슷한 결과로 성장되었으며 부산물들의 생성은 acetic acid의 경우는 Trypton을 첨가하지 않으면 많이 생성되고 lactic acid의 경우 는 trypton을 첨가할 때 매우 높게 축적되어졌다. 그러므로 trypton은 세포성장에 그렇게 중요하지 않 는 결과를 보였으며 저해제인 부산물을 적게 나오게 하려면 trypton을 첨가함이 좋고 lactic acid 생성 관계는 더욱 연구되어져야 한다.

고생산성 젖산생성균 분리 및 배양 최적화 (Isolation of Microorganism with HIgh Productivity and Cultivation Optimization for Lactic Acid Production)

  • In order to screen microorganism producing lactic acid with high productivity from nature, we used a medium containing 100 g/l glucose and selected several microorganisms producing more than 80 g/l L-lactic acid. We investigated their physiological characteristics and compared them. The best microorganism was identified as Lactobacillus casei subsp. rhamnosus. The optimum pH for growth and production of lactic acid was 6.0 and this strain showed the highest growth rate at around 30$\circ$C , but the optimum temperature for lactic acid production was 45$\circ$C . The growth was inhibited proportionally from 50 g/l to 300 g/l of glucose and the maximal cell mass increased according to increasing the concentration of corn steep liquor (CSL) protein up to 30 g/l. In batch fermentation for lactic acid production, we produced 128 g/l L-lactic acid with 20 g/l CSL protein and 150 g/l glucose in 35 hours. In pH-stat fed-batch fermentation, we were able to produce 183 g/l L-lactic acid.

젖산과 옥살산을 이용한 폐 이차 리튬이온 전지 양극 활물질로부터 희유금속들의 회수 (Recovery of Rare Metals from the Waste Secondary Lithium Ion Battery Cathode Active Materials Using Lactic Acid and Oxalic acid)

  • 젖산을 이용하여 안전하게 양극활물질에 있는 Co, Mn, Ni을 침출할 수 있는 방법을 개발하였다. 양극활물질을 젖산으로 침출 시켰을 때, 젖산의 농도는 1N과 4 N 이상에서 보다는 2 N에서 가장 높은 효율을 보였다. 양극활물질을 단계적으로 젖산 용액에 첨가 하였을 때, 최대 용해도는 2 N의 젖산 용액에서 1 L 당 30 g이었다. 젖산 용액에 옥살산을 첨가하였고 희유금속들은 1 L 당 4 g에서 가장 경제적인 회수율을 보였다. 본 연구를 바탕으로 해서, 양극활물질로부터 희유금속들을 회수하기 위한 최적의 조건은 무게비로 옥살산과 양극활물질이 7 : 1이라는 것을 확인할 수 있었다. 첨가하여, 옥살산에 의해서 생성된 침전물은 Co, Ni, Mn 3성분이 결합되어 있는 다핵 결정성 물질이었다.
